Beyond the cortex: Cerebellar contributions to apraxia of speech in stroke survivors.

Apraxia of Speech (AOS) is a motor speech disorder affecting speech planning and coordination, often co-occurring with aphasia after left hemisphere stroke. While commonly linked to cortical damage, the cerebellum's role remains underexplored. This study examined associations between cerebellar gray matter volume and AOS severity in individuals with chronic left hemisphere cortical stroke. Participants from the Aphasia Recovery Cohort (N = 142) completed structural MRI and AOS assessment using the Apraxia of Speech Rating Scale. Regional cerebellar gray matter volumes were extracted from T1-we

Folding of the cerebellar cortex is clade-specific in form but universal in degree.

Like the cerebralcortex, the surface of the cerebellum is repeatedly folded. Unlike the cerebralcortex, however, cerebellar folds are much thinner and more numerous; repeatthemselves largely along a single direction, forming accordion-like folds transverseto the mid-sagittal plane; and occur in all but the smallest cerebella. We haveshown previously that while the location of folds in mammalian cerebral cortex isclade-specific, the overall degree of folding strictly follows a universalpower law relating cortical thickness and the exposed and total surface areas predictedfrom the minimization o

Presynaptic ionotropic receptors in the cerebellar cortex: Just the tip of the iceberg?

The presence of ionotropic receptors to neurotransmitters in presynaptic structures is well documented in many synapses of the mammalian brain. However, due to technical limitations, the actual prevalence of presynaptic ionotropic receptors, as well as their potential functional roles, have remained largely uncertain. The relatively simple and regular organization of neurites in the cerebellar cortex has offered a unique opportunity to bridge this gap of knowledge, by systematically probing the presence and role of presynaptic ionotropic receptors at various synapses. In the present review, we

Accumulation of amyloid-β in the cerebellar cortex of essential tremor patients.

The accumulation of insoluble amyloid-beta (Aβ) peptides is associated with neurodegenerative disorders, such as Alzheimer's disease (AD). As essential tremor (ET) could involve neurodegenerative processes in the cerebellum, we quantified soluble and insoluble Aβ in cerebellar cortices from patients diagnosed with ET (n=9), compared to Controls (n=16) or individuals with Parkinson's disease (n=10). Although ante-mortem cognitive performance was not documented, all individuals included had the diagnosis of AD ruled out by a neuropathologist. ELISA-determined concentrations of insoluble Aβ42 in

Is essential tremor a Purkinjopathy? The role of the cerebellar cortex in its pathogenesis.

Essential tremor (ET) encompasses a group of progressive neurological diseases in which the primary clinical feature is kinetic tremor of the arms. There is accumulating evidence to suggest that the cerebellum is involved in the pathogenesis of ET; the clinical presentation, neurophysiological data, and functional and metabolic abnormalities revealed by neuroimaging studies all point toward the dysregulation of cerebellar circuits. Recent neuropathological findings at postmortem demonstrate that Purkinje neurons, and some brainstem neurons, play an integral role in the pathogenesis of this com

The primary vestibular projection to the cerebellar cortex in the pigeon (Columba livia).

The cerebellar cortex of the pigeon receiving direct vestibular afferents was delineated by anterograde transport of [3H]-amino acids injected into the vestibular nerve. Labelled mossy fiber rosettes in the granular layer were concentrated in lobule X (nodulus) and to a lesser extent, in the ventral portion of lobule IXd (uvula and paraflocculus). A few solitary labelled rosettes were also found in more dorsal portions of lobule IX, as well as in the anterior lobe between lobule II and IV. The lingula remained unlabelled. Discrete injections of [3H]-leucine into the cristae of each of the thre

Responses evoked in the cerebellar cortex by stimulation of the caudate nucleus in the cat.

1. Responses evoked in the cerebellar cortex following stimulation of caudate nucleus are described.2. The evoked responses recorded from the surface of the cerebellar cortex were found to be of two types, one with a short (4-6 msec) latency and one with a longer (12-17 msec) latency.3. The short latency response was maximal in the lobulus simplex, the longer latency response was maximal in paramedian lobule.4. Following lesions in the inferior olive the longer latency response was absent.5. Recordings from within the cerebellar cortex showed that the short latency response was uniformly distr

Focal Ischaemic Infarcts Expand Faster in Cerebellar Cortex than Cerebral Cortex in a Mouse Photothrombotic Stroke Model.

It is generally accepted that the cerebellum is particularly vulnerable to ischaemic injury, and this may contribute to the high mortality arising from posterior circulation strokes. However, this has not been systematically examined in an animal model. This study compared the development and resolution of matched photothrombotic microvascular infarcts in the cerebellar and cerebral cortices in adult 129/SvEv mice of both sexes. The photothrombotic lesions were made using tail vein injection of Rose Bengal with a 532 nm laser projected onto a 2 mm diameter aperture over the target region of th

Diffuse Cerebral Cortex, Cerebellar Cortex and Basal Ganglia Injury: a Rare MR Imaging Manifestation of Heat Stroke.

We present a case of heat stroke with rare magnetic resonance (MR) imaging manifestations. In addition to abnormal high signals in bilateral cerebellum and basal ganglia seen on T2-weighted MR images (T2WI), the case showed diffuse cerebral cortical high signals on T2WI and diffusion-weighted images (DWI). Heat stroke with diffuse hypoperfusion to the brain due to diversion of blood circulation was hypothesized as the cause of brain injury, including the cerebral cortex.

Cytoarchitectural changes in the developing cerebellar cortex of the laggard mutant mouse.

The laggard (lag) mutant mouse, which arises from a mutation in the Kif14 gene, begins to exhibit ataxia and impaired growth after the first postnatal week and subsequently dies prematurely around two weeks of age. In this mutant mouse, the layered architecture of the cerebellar cortex, cerebral cortex, dentate gyrus, and olfactory bulb is disrupted at the cellular level. The aim of this study was to identify the effect of Kif14 mutation on the development of the cerebellar cortex. Abnormalities in the cytoarchitectonics of the developing cerebellar cortex were assessed using hematoxylin-eosin

Role of cerebellar cortex in associative learning and memory in guinea pigs.

Time-related cognitive function refers to the capacity of the brain to store, extract, and process specific information. Previous studies demonstrated that the cerebellar cortex participates in advanced cognitive functions, but the role of the cerebellar cortex in cognitive functions is unclear. We established a behavioral model using classical eyeblink conditioning to study the role of the cerebellar cortex in associative learning and memory and the underlying mechanisms. We performed an investigation to determine whether eyeblink conditioning could be established by placing the stimulating e

Cerebellar and prefrontal cortex contributions to adaptation, strategies, and reinforcement learning.

Traditionally, motor learning has been studied as an implicit learning process, one in which movement errors are used to improve performance in a continuous, gradual manner. The cerebellum figures prominently in this literature given well-established ideas about the role of this system in error-based learning and the production of automatized skills. Recent developments have brought into focus the relevance of multiple learning mechanisms for sensorimotor learning. These include processes involving repetition, reinforcement learning, and strategy utilization. We examine these developments, con

The role of interneurons in shaping Purkinje cell responses in the cerebellar cortex.

The well established anatomy of the cerebellar cortex has led to suggestions that cerebellar molecular layer interneurons laterally inhibit Purkinje cells. In support of the anatomical predictions, on-beam excitation and off-beam inhibition of Purkinje cells have been shown to occur when the surface of the cerebellum is electrically excited. Patchy excitation of Purkinje cells with flanking inhibition of sagittally oriented Purkinje cells have also been demonstrated following peripheral stimulation in vivo. To extend these observations, we mapped the functional connectivity between granule cel

Glutamate receptors in the cingulate cortex, hippocampus, and cerebellar vermis of alcoholics.

This study tests the hypothesis that glutamate receptors are altered in the brains of alcoholics as a result of chronic alcohol neurotoxicity. Excessive release of the excitatory neurotransmitter glutamate may damage postsynaptic neurons by increasing calcium flux through N-methyl-D-aspartate (NMDA) receptor-gated ion channels. Alcohol has opposite effects on the NMDA receptor, depending on the duration of exposure. Acute exposure to alcohol inhibits ion flow through NMDA receptors, whereas chronic exposure upregulates the number of these receptors and thereby increases ion flow. Acute withdra

Lipofuscin in the cerebellar cortex of albino rats: an electron microscopic study.

The ultrastructure of autofluorescent, PAS-positive lipofuscin in Purkinje, granule, Golgi epithelial, basket and stellate, microglial and perivascular cells in the cerebellar cortex of senescent rats is described. The membrane-bounded pigment is composed of three elements: 1) electron-lucent homogeneous droplets, 2) a granular matrix and 3) intensely osmiophilic patches. The proportions of these three components vary between cell types and one can grossly differentiate a neuronal and a glial lipofuscin. The lipofuscin granules of stellate and perivascular cells are different from lipofuscin o

Cross-species single-cell spatial transcriptomic atlases of the cerebellar cortex.

The molecular and cellular organization of the primate cerebellum remains poorly characterized. We obtained single-cell spatial transcriptomic atlases of macaque, marmoset, and mouse cerebella and identified primate-specific cell subtypes, including Purkinje cells and molecular-layer interneurons, that show different expression of the glutamate ionotropic receptor Delta type subunit 2 (GRID2) gene. Distinct gene expression profiles were found in anterior, posterior, and vestibular regions in all species, whereas region-selective gene expression was predominantly observed in the granular layer

Low-intensity ultrasound directly modulates neural activity of the cerebellar cortex.

BACKGROUND: Low-intensity ultrasound is a noninvasive neuromodulation technique with the potential to focally manipulate deep brain activity at millimeter-scale resolution. However, there have been controversies over the direct influence of ultrasound on neurons, due to an indirect auditory activation. Besides, the capacity of ultrasound to stimulate the cerebellum remains underestimated. OBJECTIVE: To validate the direct neuromodulation effects of ultrasound on the cerebellar cortex from both cellular and behavioral levels. METHODS: Two-photon calcium imaging were used to measure the neuron

Multizonal Cerebellar Influence Over Sensorimotor Areas of the Rat Cerebral Cortex.

The cerebral cortex requires cerebellar input for optimizing sensorimotor processing. However, how the sensorimotor cortex uses cerebellar information is far from understood. One critical and unanswered question is how cerebellar functional entities (zones or modules) are connected to distinct parts of the sensorimotor cortices. Here, we utilized retrograde transneuronal infection of rabies virus (RABV) to study the organization of connections from the cerebellar cortex to M1, M2, and S1 of the rat cerebral cortex. RABV was co-injected with cholera toxin β-subunit (CTb) into each of these cort

Cerebellar stimulation fails to modulate motor cortex plasticity in writing dystonia.

BACKGROUND: Primary dystonia is characterized neurophysiologically by reduced inhibitory mechanisms and abnormal regulation of plasticity responses. The potential of anodal cerebellar transcranial direct current stimulation as a therapeutic tool in writing dystonia was examined, after the observation that cerebellar stimulation reduces responses to an associative plasticity protocol in healthy subjects. METHODS: Ten patients with writing dystonia completed a two-part study (sham and anodal) in which cerebellar stimulation was given simultaneously with paired associative stimulation. Electroph
