Cerebrospinal Fluid Proteome Reveals Dysregulation of Lysosomal and Axonal Proteins in Neurosyphilis.

Neurosyphilis (NS), caused by Treponema pallidum, results in irreversible neurological damage. This study elucidated NS pathogenesis via cerebrospinal fluid (CSF) proteomic analysis comparing NS, suspected neurosyphilis (spNS), and non-neurosyphilis (nNS) controls by using liquid chromatography-tandem mass spectrometry (LC-MS/MS), public data set PXD033034, and ELISA validation. Of 1261 quantified proteins in the CSF, 234 were differentially expressed in NS, with 189 downregulated and enriched in lysosomal, axon guidance, and neurodegeneration pathways. Three previously identified CSF biomarke

Low synaptic and neurosecretory proteins in cerebrospinal fluid in early parkinsonian disease.

BACKGROUND: The early pathogenesis of Parkinson's disease (PD) and the atypical parkinsonian disorders multiple system atrophy (MSA) and progressive supranuclear palsy (PSP) is poorly understood, but presynaptic and axonal dysfunction are hypothesized to play a prominent role. OBJECTIVE: To identify synapse- and/or axonal dysfunction as indicated by cerebrospinal fluid (CSF) biomarker profiles and their clinical correlates in early-stage PD, MSA, and PSP. METHODS: Liquid chromatography mass spectrometry and enzyme-linked immunosorbent assay analyses of CSF samples from patients with early-st

Tau proteins in the cerebrospinal fluid of patients with subacute sclerosing panencephalitis.

Neurodegenerative diseases characterized by cytoskeletal deformation and neurofibrillary tangles are associated with altered levels of tau and related proteins in cerebrospinal fluid (CSF). Neuronal or glial fibrillary tangles have been shown in 20% of subacute sclerosing panencephalitis (SSPE) patients. We therefore investigated CSF samples from 60 newly diagnosed SSPE and 31 neurological control patients for total tau (t-tau), phosphorylated tau (p-tau), and S100-B levels by ELISA. There was no difference between patient and control groups in t-tau and S100-B levels. p-Tau was lower in the S

Brain-specific proteins in cerebrospinal fluid for the diagnosis of neurodegenerative diseases.

Neurodegenerative disorders have traditionally been classified according to clinical criteria, e.g. as dementia syndromes (the best known is Alzheimer's disease) or as movement disorders (e.g. Parkinson's disease). Another subdivision is based on recent insights into the respective pathogenetic mechanisms, leading to the recognition of so-called tauopathies and alpha-synucleinopathies. It is this increased knowledge of the underlying (neuro)pathological mechanisms that has sparked interest in studies aimed at the identification of disease-specific biomarkers in cerebrospinal fluid (CSF) for th

[Evaluation of cerebrospinal fluid proteins in patients with subacute sclerosing panencephalitis (SSPE)].

CSF proteins of 62 patients with SSPE aged 6 to 26 years were analysed by polyacrylamide gel electrophoresis. The total IgG concentration in the CSF was increased to 16.8 +/- 10 mg/dl and the IgG index was raised to from 2.7 +/- 0.5 to 3.6 +/- 3.0. All SSPE patients had pathological subfractions gamma 4 and gamma 5. The per cent proportion of the gamma 1 subfraction was raised only in the youngest patients. A tendency was observed for higher values of the prealbumin:albumin index and the prealbumin: transferrin index. The youngest SSPE patients had a tendency for greater total protein concentr

Cerebrospinal fluid cells and proteins in patients occupationally exposed to organic solvents.

Cerebrospinal fluid (CSF) cells and proteins were determined for 33 patients exposed to industrial organic solvents. A lymphoid reaction, i.e., a pathologically elevated number or percentage of enlarged lymphoid cells was observed in one-third of the patients, more often in patients with chronic intoxication (40%) than in those currently exposed to organic solvents (32%). An almost significant decrease of small lymphocytes in the CSF was observed among patients who had a past history of chronic solvent intoxication but no recent exposure. No cytological evidence of tissue destruction was found

Cerebrospinal fluid levels of tau and phospho-tau-181 proteins during pregnancy.

OBJECTIVES: During pregnancy various interactions occur between structural alterations of the maternal brain and placental metabolism. The aim of the present study was to investigate whether cerebrospinal fluid concentrations of tau and phospho-tau-181 protein vary during normal pregnancy and in women with preeclampsia and HELLP syndrome. STUDY DESIGN: We measured cerebrospinal fluid biomarkers, tau and phospho-tau-181 protein levels in 90 pregnant women electively assigned for regional anaesthesia during pregnancy or for cesarean section using enzyme-linked immunosorbent assays. RESULTS: Ce

Brain-specific proteins in the cerebrospinal fluid of severely asphyxiated newborn infants.

UNLABELLED: Upcoming trials of neuroprotective strategies in severely asphyxiated newborn infants emphasize the need for early and objective markers of both good and bad long-term prognosis. Traditional markers such as neurological depression and seizures are not specific. AIM: To study whether measurement in the cerebrospinal fluid of some proteins known to be specific to the central nervous system was in covariance with the clinical course and long-term prognosis. METHODS: Twenty-two asphyxiated infants were included in the study and compared with a control group of 8 infants without signs

Abnormal proteins in the cerebrospinal fluid of patients with Creutzfeldt-Jakob disease.

We studied more than 300 cerebrospinal fluid proteins from 21 patients with Creutzfeldt-Jakob disease. We also examined cerebrospinal fluid from 100 normal controls and more than 400 patients with various neurologic disorders other than Creutzfeldt-Jakob disease. Four abnormal proteins that were identified in the patients with Creutzfeldt-Jakob disease were absent in the normal persons. Two of these proteins (Mr [relative molecular mass], 40,000; pl [isoelectric point], 5.7 and Mr 40,000; pl 5.9) were also present in some patients with multiple sclerosis, herpes simplex encephalitis, schizophr…

Differences in cerebrospinal fluid proteins between patients with schizophrenia and normal persons.

A survey of cerebrospinal fluid (CSF) proteins from normal persons and patients with schizophrenia reveals differences between the two populations. Computer-assisted densitometry of 68 proteins, resolved by two-dimensional electrophoresis and made visible by silver staining, shows six changes. Compared with their occurrence in the normal group, two proteins are increased in the schizophrenic patients by 22% and 27%, while four proteins are decreased by 29%, 46%, 20%, and 37% (p less than 0.005). Furthermore, two additional 40 000-Da proteins are found in CSF from 31.5% of the schizophrenic pat

[Cerebrospinal fluid proteins in the course of subacute sclerosing panencephalitis in children].

In cerebrospinal fluid samples obtained from lumbar tap from 31 children with subacute sclerosing panencephalitis the concentration of total protein was determined and electrophoretic separation of protein fractions on paper was carried out from 1 to 28 months after the onset of first clinical symptoms. Moreover, in 22 children the concentration of IgG was determined by electroimmunodiffusion. The reference group included 22 children without organic diseases of the central nervous system. In nearly half the samples of the cerebrospinal fluid obtained from these children a rise was observed in

Cerebrospinal fluid myelin basic protein and multiple sclerosis.

We have previously reported that myelin basic protein appears in CSF during acute attacks of multiple sclerosis. These studies have been extended to over 700 patients, 91 with multiple sclerosis. The data continues to indicate that myelin basic protein is released into cerebrospinal fluid during acute attacks of multiple sclerosis. We are currently characterizing the basic protein in the CSF.

Paediatric ventriculoperitoneal shunt obstruction and cerebrospinal fluid protein.

BACKGROUND: Ventriculoperitoneal shunt (VPS) insertion is a life-saving intervention for hydrocephalus. Device failure is extremely common, and carries immense morbidity for patients, their families, and the healthcare system. Predicting shunt obstruction remains a substantial challenge. Clinically, cerebrospinal fluid protein (CSFp) is suspected to contribute to VPS obstruction, this is widely debated, and definitive evidence is lacking. We assess the relationship of CSFp in predicting VPS failure in children. METHODS: A retrospective review of VPS procedures at the Queensland Children's Hos

Increased protein glycation in cerebrospinal fluid of Alzheimer's disease.

Accumulation of advanced glycation end products occurs in the brain with ageing and was proposed to be involved in pathogenesis of Alzheimer's disease. We studied changes in the level of an early glycation product, an Amadori product, in cerebrospinal fluid (CSF) in ageing and in late-onset Alzheimer's disease. The work was carried out on 99 consecutive patients. The concentration of Amadori product in CSF correlated with CSF glucose concentration but was not changed with age (n = 70). In contrast, level of CSF Amadori product was 1.7-fold higher in Alzheimer's disease patients (n = 29) as com

Human cerebrospinal fluid protein database: edition 1992.

Two-dimensional electrophoresis maps of human cerebrospinal fluid proteins are presented in the form of labeled images. 931 protein spots are identified in spinal fluid from a normal volunteer. Distinct spots that represent variants of the same protein, especially posttranslational modifications, are estimated to reduce the 931 different spots to < 200 different proteins. 248 spots of 29 protein groups have been identified and are indicated on enlargements of specific gel regions. The distribution of protein abundance, mass, charge and shape characteristics of these normal 931 spinal fluid spo…

Protein pattern of cerebrospinal fluid in various neurological diseases.

CSF/serum albumin and immunoglobulin G ratios were determined in 520 patients suffering from various neurological diseases. Blood-brain barrier impairment was detectable in most cases of spinal tumour, meningitis, Guillain-Barré syndrome and in two-thirds of the patients with cerebral infarctions. A local IgG formation in the CNS has to be assumed for some cases of meningitis considering the course of the protein dysequilibrium. Autochthonous IgG production together with a barrier dysfunction was found in patients with encephalitis, meningoradiculitis and neurosyphilis. In cases of multiple sc
