Invasive pulmonary mycosis due to Chaetomium globosum with false-positive galactomannan test: a case report and literature review.

In this case, the authors report Chaetomium globosum as a cause of invasive pulmonary infection in a patient with Wegener's granulomatosis. Fungal hyphae (KOH and Calcofluor) were seen on direct microscopy of lung biopsy sample and bronchoalveolar lavage (BAL) sample. C. globosum isolated on culture clinched the diagnosis of invasive pulmonary infection by Chaetomium spp. A positive galactomannan of serum and BAL was repeatedly seen and was utilised for follow-up and as prognostic marker in patient management. The patient was successfully treated with liposomal amphotericin B followed by voric

Protein interfaces of the conserved Nup84 complex from Chaetomium thermophilum shown by crosslinking mass spectrometry and electron microscopy.

A key building block of the nuclear pore complex (NPC) is the Nup84 subcomplex that has been structurally analyzed predominantly in the yeast system. To expand this analysis and gain insight into the evolutionary conservation of its structure, we reconstituted an octameric Nup84 complex using the subunits from a thermophile, Chaetomium thermophilum (ct). This assembly carries Nup37 and Elys, which are characteristic subunits of the orthologous human Nup107-Nup160 complex but absent from the yeast Saccharomyces cerevisiae. We found that Elys binds cooperatively to the complex requiring both Nup

Green Foxtail-Derived Endophytic Fungus Chaetomium novozelandicum SV7 Produced Orsellinic Acid Ester Glycosides Active Against Rice Blast Pathogen.

Plants encounter various fungi during their life history, with some of them pathogenic, some beneficial, and some neither, forming complex plant-fungi interactions. Endophytic fungi are a type of fungi that live inside the plant, and are often beneficial to the host. In this study, the endophytic fungi of green foxtail (Setaria viridis) were explored for their defensive role against the pathogenic fungus Magnaporthe oryzae, which causes rice blast, one of the most devastating crop diseases. The endophytic fungi were isolated, taxonomically identified, fermented, and chemically extracted. The f

An epigenetic modifier enhances the generation of anti-phytopathogenic compounds from the endophytic fungus Chaetomium globosporum of Euphorbia humifusa.

Endophytic fungi are striking resources rich in bioactive structures with agrochemical significance. In order to maximize the opportunity of search for bioactive compounds, chemical epigenetic manipulation was introduced to enhance the structural diversity of the fungal products, and an UPLC-ESIMS and bioassay-guided separation was used to detect novel bioactive metabolites. Consequently, four previously undescribed compounds including two cyclopentenones (globosporins A and B) and two monoterpenoid indole alkaloids (globosporines C and D), as well as three known compounds, were isolated from

Chaetomium globosum: Spotting a scarce saprophyte in bacterial-fungal co-infection of the sinuses in a post-COVID-19 patient.

Atypical fungal co-infections in post-COVID-19 patients may have been underreported due to limited diagnostic methods. We present a case of Chaetomium globosum sinusitis in a 55-year-old post-COVID-19 patient with pain in the left side of the face, mimicking rhino-cerebral mucormycosis. CT-paranasal sinuses showed mucosal thickening of left paranasal sinuses, biopsy of which grew a velvety, white colony. It was confirmed as Chaetomium globosum. The patient responded to oral Posaconazole therapy for three months. Prompt identification of atypical fungal agents is critical for appropriate treatm

Cocultivation and pH shape the metabolite profile of Chaetomium globosum SCF-24: insights from gas chromatography-mass spectrometry analysis.

This study investigated the effects of cocultivation and pH on the metabolite profile of Chaetomium globosum SCF-24. Axenic and coculture fermentation systems were established at pH 5-8, and resulting extracts were analyzed by gas chromatography-mass spectrometry (GC-MS). A hierarchical clustered heatmap was used to compare metabolite profiles based on presence-absence data. GC-MS detected 51 putatively identified primary and secondary metabolites, some of which were detected across all treatments, whereas others were condition-specific. Metabolite profiles also differed between axenic culture

Nonsteroidal Anti-inflammatory Drugs (NSAIDS) Inhibit the Growth and Reproduction of Chaetomium globosum and Other Fungi Associated with Water-Damaged Buildings.

Indoor mold due to water damage causes serious human respiratory disorders, and the remediation to homes, schools, and businesses is a major expense. Prevention of mold infestation of building materials would reduce health problems and building remediation costs. Nonsteroidal anti-inflammatory drugs (NSAIDs) inhibit yeasts and a limited number of filamentous fungi. The purpose of this research was to determine the possible inhibitory activity of nonsteroidal anti-inflammatory drugs (NSAIDs) on germination, fungal growth, and reproduction of Chaetomium globosum and other important filamentous f

Pathogen-activated Chaetomium globosum G3 enhances iron competition and other antagonistic mechanisms to suppress maize seedling blight causal agent Fusarium verticillioides.

Fusarium verticillioides (F. verticillioides) is a prevalent soilborne fungal pathogen that seriously threatens the production of maize (Zea mays). Species within the Chaetomium genus are recognized as potential biocontrol agents for managing plant pathogens. However, the biocontrol efficacy and underlying inhibitory mechanisms of Chaetomium species against maize seedling blight caused by F. verticillioides have yet to be comprehensively evaluated and fully elucidated. This study demonstrates that Chaetomium globosum (C. globosum) G3 reduced the disease index of F. verticillioides on maize see

Diversity and biological activities of endophytic fungi associated with Catharanthus roseus.

BACKGROUND: The present study involves diversity and bioactivity of the endophytic fungal community from Catharanthus roseus inhabiting the coastal region. This study has been conducted hypothesizing that the microbial communities in the coastal regions would tolerate a range of abiotic stress such as salinity, humidity, temperature and soil composition, and it may produce new metabolites, which may possess bioactive property. Therefore in the current study, the cytotoxicity and free radical scavenging potential of the fungal organic extracts have been investigated. Moreover, the apoptotic and

Sterilization of food packaging by UV-C irradiation: Is Aspergillus brasiliensis ATCC 16404 the best target microorganism for industrial bio-validations?

In food industries UV-C irradiation is used to achieve decontamination of some packaging devices, such as plastic caps or laminated foils, and of those smooth surfaces that can be directly irradiated. Since its effectiveness can be checked by microbial validation tests, some ascospore-forming molds (Aspergillus hiratsukae, Talaromyces bacillisporus, Aspergillus montevidensis, and Chaetomium globosum) were compared with one of the target microorganisms actually used in industrial bio-validations (Aspergillus brasiliensis ATCC 16404) to find the species most resistant to UV-C. Tests were carried

Insights into the catalytic mechanism of formate dehydrogenases from different microbial sources.

Four formate dehydrogenases (FDHs) from Pseudomonas sp. 101, Myceliophthora thermophila, Chaetomium thermophilum, and Ogataea parapolymorpha were recombinantly produced, purified, and characterized to investigate their catalytic properties and reaction mechanisms. The enzymes were studied for their ability to oxidize formate to carbon dioxide (CO2) coupled with NAD+ reduction. In contrast, their CO2 reduction activity was undetectable under the tested conditions. Oxidative reactions revealed significant differences in catalytic efficiency and substrate specificity, prompting further investigat

Butenolides from a cultured microfungus, Acremonium sp.

Two related butenolides have been isolated from a culture broth of a strain of the fungus, Acremonium sp. Although the lactone acid (3) is a known compound, the structure of the lactone diacid (4), deduced by spectroscopic analysis, is assigned for the first time. This compound appears to be identical to a partly characterised metabolite isolated, along with 3, from Chaetomium indicum in 1953.

Phylogenetic analysis of the complete mitochondrial genome of Madurella mycetomatis confirms its taxonomic position within the order Sordariales.

BACKGROUND: Madurella mycetomatis is the most common cause of human eumycetoma. The genus Madurella has been characterized by overall sterility on mycological media. Due to this sterility and the absence of other reliable morphological and ultrastructural characters, the taxonomic classification of Madurella has long been a challenge. Mitochondria are of monophyletic origin and mitochondrial genomes have been proven to be useful in phylogenetic analyses. RESULTS: The first complete mitochondrial DNA genome of a mycetoma-causative agent was sequenced using 454 sequencing. The mitochondrial gen
