Patients with inflammatory bowel disease (IBD) exhibit a dysregulated bile acid pool, characterized by increased primary and decreased secondary bile acids, largely due to gut microbiota dysfunction. However, the impact of colitis on hepatic bile acid synthesis remains poorly understood. In this study, analyses of public datasets, in-house patient samples, and an animal model revealed that colitis enhances flux through the classical bile acid synthesis pathway while suppressing the alternative pathway. Oral administration of chenodeoxycholic acid (CDCA) redirected bile acid synthesis toward th

The effect of administration of primary and secondary bile acids on absorption of cholesterol was investigated in 15 volunteers. Eight Caucasians with radiolucent gallstones were studied before and after administration of chenodeoxycholic acid (all eight) and cholic acid (eight before, six after treatment) for 3 months, and seven healthy subjects were studied before and five were studied after administration of deoxycholic acid for 6 weeks. The hourly absorption of [3H]cholesterol was measured for 24 hours in a 20-cm duodenal segment by use of an intestinal perfusion technique. Fasting serum c

Chenodeoxycholic acid (CDCA), an endogenous agonist of the farnesoid X receptor, is involved in lipid homeostasis and intestinal barrier regulation. This study examined how dietary CDCA alleviates high-energy diet (HED)-induced metabolic disorders in Wenchang chickens and determined the optimal supplemental dose. A total of 525 Wenchang hens were randomly assigned to five groups: Con (basal diet), HED, and HED supplemented with 50, 100, or 200 mg/kg CDCA (≥ 98% purity). The experiment spanned 42 days. Compared with the control group, HED significantly increased abdominal fat percentage, serum

UNLABELLED: We present a two-week old girl who was diagnosed with cerebrotendinous xanthomatosis (CTX), an inborn error of bile acid synthesis, after a diagnostic workup for convulsions which were shown to be caused by a parechovirus encephalitis. The diagnosis of CTX was confirmed with CYP27A1 mutation analysis. She was started on chenodeoxycholic acid (CDCA) supplementation, which inhibits cholestanol production through a feedback mechanism, at the advised dosage of 15 mg/kg/day. Within 6 weeks, she developed jaundice with hepatomegaly. CDCA supplementation was stopped after which liver size

BACKGROUND/AIMS: Unconjugated bilirubin (UCB) can be neurotoxic in jaundiced neonates and in patients with Crigler-Najjar syndrome. UCB toxicity may culminate in cell death, however, the occurrence of apoptosis has never been investigated. Ursodeoxycholic acid (UDCA) is a strong modulator of the apoptotic threshold in both hepatic and nonhepatic cells. The aims of this study were to determine whether apoptosis plays a role in neural cell death induced by UCB, and to investigate the ability of UDCA to prevent cell death.
METHODS: Cultured rat astrocytes were incubated with UCB (17 and 86 micro

BACKGROUND AND PURPOSE: Surgical ventricular reconstruction (SVR) partially reverses left ventricular remodelling in postinfarction heart failure, yet residual cardiac dysfunction persists through unknown mechanisms. We investigated ferroptosis involvement in post-SVR pathology and explored pharmacological interventions.
EXPERIMENTAL APPROACH: Myocardial infarction (MI) was induced in C57BL/6 mice followed by SVR at 4 weeks. Cardiac function was assessed by echocardiography and pressure-volume catheterization. Ferroptosis biomarkers were quantified. Connectivity Map analysis identified candid

Non-alcoholic fatty liver disease (NAFLD) is the most prevalent chronic liver disease globally. Chenodeoxycholic acid (CDCA), a key component of bile acids, improves hepatic lipid metabolism. The present study aimed to investigate whether CDCA regulates hepatic lipid metabolism by interacting with pyruvate kinase (PK), and to elucidate the underlying molecular mechanism. High-fat cell and murine models were generated. The levels of total cholesterol (TC), triglycerides (TG), superoxide dismutase (SOD) and malondialdehyde (MDA) in cells and animal livers were measured. BODIPY and Oil Red staini

Fibrates, hypolipidemic agents, are reported to be effective in treatment of primary biliary cirrhosis. However, the mechanism involved in therapeutic benefits of fibrates in primary biliary cirrhosis remains unknown. In contrast, hepatic regulated upon activation, normal T-cell expressed and secreted (RANTES) is increased in patients with primary biliary cirrhosis and bile acids up-regulate RANTES expression in hepatocytes. The role of fibrates in bile acid-induced RANTES expression was investigated in human hepatoma cells; 100 microM of bezafibrate and fenofibrate decreased expression of che

Thirteen patients (9 women, 4 men) with solitary (7) or multiple (6) radiolucent gallstones were treated with 1.0 g/day CDCA over a period of 7--17 months (mean 10.2). Routine liver function tests showed no changes compared to pre-treatment values, except a moderate elevation of SGPT activity in one patient. Serum cholesterol and serum triglyceride levels remained unchanged during therapy. 21 liver biopsies in 11 patients compared to pre-treatment controls studied by light and electron microscopy revealed no lesions in hepatic structure, especially no signs of cholestasis. Only some unspecific

Valproic acid (VA) is a commonly prescribed medication for epilepsy and other neurological conditions. Although effective, VA use can lead to neurotoxicity, especially with chronic use. This study aimed to investigate the potential neuroprotective properties of indole-3-acetic acid (IAA) and chenodeoxycholic acid (CDCA) in an animal model of VA-induced brain injury. Rats received intraperitoneal injections of VA at a dose of 500 mg/kg/day for 3 weeks. Concurrently, they were orally treated with IAA (40 mg/kg/day) and/or CDCA (90 mg/kg/day). The results showed significantly increased oxidative

Two epimeric bile acid conjugates, 5-aminosalicylic acid-chenodeoxycholic acid (5-ASA-CDCA) and 5-aminosalicylic acid-ursodeoxycholic acid (5-ASA-UDCA), were synthesized to deliver 5-ASA to the large intestine by oral administration. The movement of the conjugates down the gastrointestinal tract and the anti-inflammatory effects on ulcerative colitis were investigated by administering the conjugates to guinea-pigs with an inflammatory bowel disease induced by 2% degraded carrageenan solution. The conjugates were protected from deconjugation in stomach and small intestine and reached the caecum

Conventional type 1 dendritic cells (cDC1s) play a pivotal role in initiating CD8+ T-cell-mediated antitumor immunity, and their intratumoral abundance strongly correlates with immunotherapy efficacy. Here, we demonstrate that orally administered chenodeoxycholic acid (CDCA), a clinically approved metabolic modulator, accumulates in the tumor interstitial fluid (TIF) of tumor-bearing mice as its metabolite taurochenodeoxycholic acid (TCDCA), thereby potentiating the antitumor functions of cDC1s and CD8+ T cells through a gut microbiota-independent mechanism. Mechanistic studies revealed that C

Chenodeoxycholic acid (CDCA) is an essential drug for patients with rare metabolic disease cerebrotendinous xanthomatosis (CTX). To ensure continuation of treatment, the Amsterdam UMC hospital pharmacy developed pharmacy compounded CDCA capsules when the authorized CDCA capsules were no longer available for Dutch patients. This study reports the safety of pharmacy compounded CDCA through pharmacovigilance monitoring and assesses its effectiveness by evaluating biochemical outcome measures, both in patients who were previously treated with authorized CDCA and subsequently switched to the compou

BACKGROUND & AIMS: Di-alpha hydroxy bile salt, sodium chenodeoxycholate (CDC), and bile acid binding have unclear effects on colonic transit in health and disease.
METHODS: We performed 2 randomized, double-blind, placebo-controlled studies. In healthy volunteers (20 per group), we evaluated the effects of oral placebo, 500 mg, or 1000 mg of CDC (delayed-release, each given for 4 days) on gastrointestinal and colonic transit. A second trial compared the effects of colesevelam (1.875 g, twice daily) versus placebo in 24 patients (12 per group) with diarrhea-predominant irritable bowel syndrome

Biliary cholesterol saturation indices (SI's) were measured in fasting duodenal bile from (i) obese and non-obese individuals with and without cholesterol gallstones, (ii) obese individuals undergoing weight reduction and (iii) obese gallstone patients receiving chenodeoxycholic acid (CDCA) therapy. Biliary lipid secretion rates were also measured in three obese subjects before and during 11 days starvation. The mean SI in fifteen non-obese controls (0.89 +/- SEM 0.06) was significantly lower than that in the twenty-four obese without (1.14 +/- 0.07; P less than 0.01), and in the twenty-nine n

Hamsters were fed chenodeoxycholic acid (CDC), ursodeoxycholic acid, (UDC), or no bile acid. [14C]Sucrose-labeled hamster low density lipoprotein (LDL) and methylated human LDL were infused intravenously to study LDL receptor-dependent and LDL receptor-independent organ uptake, respectively, of LDL. Biliary CDC increased during both CDC and UDC treatment. The UDC enrichment of bile after UDC feeding was relatively small. Bile acid synthesis was suppressed after both bile acid treatments. Under the condition of an acute bile fistula, the hamster LDL uptake increased in the liver, heart, and adr

Background: Cerebrotendinous xanthomatosis (CTX) is an inborn disorder of bile acid synthesis which causes progressive accumulation of toxic metabolites in various organs, particularly in brain and tendons. Most cases are diagnosed and treated in the second or third decade of life, when neurological involvement appears. We describe a case of CTX presenting as neonatal cholestasis. Results: The child presented cholestasis at 2 months of life. In the following months jaundice slowly disappeared, with a normalization of bilirubin and aminotransferases, respectively, at 6 and 8 months. A LC-Mass S

The aim of this study was to investigate the key bile acid metabolites and the target of action during calcium oxalate (CaOx) kidney stone formation using a multi-omics approach. CaOx kidney stone models in Sprague-Dawley (SD) rats were established by 1% ethylene glycol (EG). In CaOx kidney stone rat models, targeted metabolomics analysis revealed four differentially expressed bile acids in serum, and 11 differentially expressed bile acids in kidneys, and chenodeoxycholic acid (CDCA) was identified as the key metabolite in the formation of CaOx kidney stones. Transcriptomics of rat kidneys rev

Cholestatic liver diseases are characterized by excessive bile acid accumulation in the liver. Endothelial cells (ECs) shape the local microenvironment in both normal conditions and liver injury, yet their role in cholestasis is unclear. Through a comparative analysis of single-cell RNA sequencing data from various murine models of liver injury, we identify distinctive Myc activation within ECs during obstructive cholestasis resulting from bile duct ligation (BDL). Myc overexpression in ECs significantly upregulates P-selectin, increasing neutrophil infiltration and worsening cholestatic liver
