Molecular characterization, evolutionary dynamics, and pathogenic assessment of chicken infectious anemia virus isolates circulating in Egyptian poultry.

As a result of its severe immunosuppressive nature, CIAV persists as a critical pathogen causing substantial economic concerns in global poultry production. The emergence of genetic variants and reports of suspected vaccine sub-optimal protection in Egyptian broiler flocks necessitates an urgent investigation of currently circulating strains. In this study, 165 samples were collected from six Egyptian governorates (2024-2025). PCR screening revealed 38 positive cases, and six isolates were selected for full-length sequencing of the VP1 and VP2 genes. Molecular analysis identified the isolates

Development and immunological evaluation of a ferritin nanoparticle vaccine displaying chicken infectious anemia virus VP1-VP2 proteins.

BACKGROUND: Chicken infectious anemia virus (CIAV) causes immunosuppression, aplastic anemia, and lymphoid atrophy in young chicks, causing major economic losses to the poultry industry. Vaccination is the primary strategy for disease control. This study aimed to develop a ferritin-based nanoparticle vaccine displaying CIAV VP1-VP2 proteins and evaluate its protective efficacy in chicks. RESULTS: Using the SpyTag003/SpyCatcher003 system, we expressed ferritin-SpyCatcher003 (SC-FN) in insect cells and SpyTag003-VP2-VP1Nd129 in E. coli, achieving in vitro assembly of FN-VP2-VP1Nd129 nanoparticl

Evaluation of chicken infectious anemia virus and associated risk factors with disease and production losses in broilers.

A case-control study was performed to determine the significance of chicken infectious anemia virus (CIAV) as a risk factor associated with secondary disease in commercial broilers and to identify the significance of production losses associated with CIAV. The study also examined the relationship between bursal and thymic atrophy and the presence of CIAV. Cases were defined as submissions to the Alabama Veterinary Diagnostic Laboratories with a history of clinical disease and with a diagnosis of coccidiosis, gangrenous dermatitis, or respiratory disease. Controls were selected from submissions

Research Note: A novel peptide-based ELISA for efficient detection of antibody against chicken infectious anemia virus.

Chicken infectious anemia virus (CIAV) is the pathogen of chicken infectious anemia. Currently, due to the lack of effective diagnostics technology and prevention approach, CIAV has spread globally and caused huge economic losses to poultry industry. In this study, a novel peptide-based ELISA (pELISA) for efficient detection of antibody against CIAV was developed. The peptide (25CRLRRRYKFRHRRRQRYRRRAF45) used in pELISA was highly conserved in VP1 protein of different CIAV isolates. The specificity and reproducibility showed that the pELISA only reacted with sera against CIAV, not with sera aga

Molecular characterization and pathogenicity study of a highly pathogenic strain of chicken anemia virus that emerged in China.

Chicken infectious anemia (CIA) is caused by chicken anemia virus (CAV). Recently, severe anemia has emerged in layer chickens (8 to 10-week-old) on poultry farms in China. However, the etiological characteristics and pathogenic potential of CAV in chickens at 6 weeks or older are not well understood. In this study, we isolated a CAV strain, termed SD15, from two-month-old chicken with severe anemia and analyzed the genetic evolution relationship. We found that strain SD15 had the highest homology (98.9%) with CAV18 strain. Comparison with 33 reference strains revealed 16 amino acid mutations

Development of a quadruplex RT-qPCR for the detection of avian leukosis virus, chicken infectious anemia virus, avian reovirus, and fowl adenovirus.

Avian leukosis virus (ALV), chicken infectious anemia virus (CIAV), avian reovirus (ARV), and fowl adenovirus (FAdV) are important viral pathogens that can transmitted horizontally and vertically, and induce immunosuppression to the poultry flocks. They exhibit diverse pathogenic characteristics in clinical settings, and pose continuous threat to the health of poultry flocks. Here, the specific primers and probes were designed for the env gene of ALV, the VP1 gene of CIAV, the M1 gene of ARV, and the ORF1 gene of FAdV. The RNA standards for ALV, and ARV, and the plasmid DNA standards for CIAV,

Development and preliminary application of a multiplex qPCR assay for the detection of avian leukosis virus subgroup J, reticuloendotheliosis virus, and chicken infectious anemia virus.

Avian leukosis virus subgroup J (ALV-J), Reticuloendotheliosis virus (REV), and Chicken infectious anemia virus (CIAV) are important breeder-derived pathogens (BDPs) that continue to threaten poultry health and productivity. These viruses are frequently associated with immunosuppression and coinfections, which complicate eradication efforts, particularly in indigenous chicken breeds. In this study, a TaqMan-based multiplex qPCR (M-qPCR) assay was developed for the simultaneous detection of ALV-J, REV, and CIAV. The assay exhibited excellent linearity (R2 > 0.99), high repeatability (intra-/int

A novel chicken infectious anemia virus vaccine candidate: complete attenuation, strong immunogenicity, and a built-in DIVA marker.

Chicken infectious anemia virus (CIAV) is a globally important immunosuppressive pathogen of poultry, causing anemia, impaired immunity, and production losses. Current live-attenuated vaccines have practical limitations: incomplete attenuation restricts safe use to older birds (e.g. ≥ 6 weeks), while over-attenuation can reduce in-vivo replication and manufacturable vaccine titers. To address these gaps, we serially passaged the virulent Korean field strain 17AD008 in MDCC-MSB1 cells for 200 generations, generating two attenuated derivatives, P45 and P100. Pathogenicity and replication were ev

Development and characterization of a potential diagnostic monoclonal antibody against capsid protein VP1 of the chicken anemia virus.

Chicken anemia virus (CAV) is an important viral pathogen that causes anemia and severe immunodeficiency syndrome in chickens worldwide. In this study, a potential diagnostic monoclonal antibody against the CAV VP1 protein was developed which can precisely recognize the CAV antigen for diagnostic and virus recovery purposes. The VP1 gene of CAV encoding the N-terminus-deleted VP1 protein, VP1Nd129, was cloned into an Escherichia (E.) coli expression vector. After isopropyl-β-D-thiogalactopyronoside induction, VP1Nd129 protein was shown to be successfully expressed in the E. coli. By performing

Pathological and virological analysis of concurrent disease of chicken anemia virus infection and infectious bronchitis in Japanese native chicks.

A concurrent infection of chicken anemia virus (CAV) and infectious bronchitis virus (IBV) was detected in Japanese native chicks in 2017, in which a high mortality rate (97.7%) was recorded in a small flock of 130 chicks exhibiting poor growth. Histological examination revealed that the affected chicks exhibited two different pathological entities: one was severe hematopoietic and lymphocytic depletion with abnormally large cells containing intranuclear inclusion bodies of CAV, whereas the other was renal tubular necrosis due to IBV infection. Immunohistochemistry detected CAV antigens in the

High prevalence of norovirus GII.P16/GII.2 and chicken anemia virus in two acute gastroenteritis outbreaks in Huzhou, China.

Acute gastroenteritis (AGE) is one of the most frequently occuring illnesses in children and adults worldwide. In February 2017, two AGE outbreaks occurred in two adjacent schools in Huzhou city, Zhenjiang province of China. We detected high percentages of recombinant norovirus GII.P16/GII.2 in one school and chicken anemia virus (CAV) in another school using next generation sequencing (NGS) and specific PCR. The results highlight the importance of continuous surveillance of GII.P16/GII.2, and suggest the need of further studies on whether CAV causes AGE. Keywords: acute gastroenteritis; norov

Oral infection with chicken anemia virus in 4-wk broiler breeders: lack of effect of major histocompatibility B complex genotype.

The pathologic consequences of chicken anemia virus (CAV) oral inoculation in 4-wk-old broiler breeders of different major histocompatibility B complex (MHC) genotypes were evaluated. MHC B complex was determined by hemagglutination and sequence-based typing. Clinical signs, serology, gross lesions, histopathologic analysis, and CAV genome quantification were used to evaluate disease progression. Clinical disease was not apparent in the inoculated broilers throughout the experimental period. At 14 days postinoculation, antibodies against CAV were detected in 26.4% (29/110) of the inoculated bi

Chicken infectious anemia virus exploits host CK2α as an essential factor for its replication via VP2 Ser182/Asp183-mediated interaction.

UNLABELLED: Chicken infectious anemia virus (CIAV) induces severe immunosuppression in chickens. The nonstructural protein VP2 of CIAV is shown to play critical roles in viral assembly and viral replication. However, the molecular mechanisms by which CIAV VP2 hijacks host machinery to promote efficient viral replication remain elusive. Here, we showed that casein kinase 2 alpha (CK2α) subunit was identified to interact with CIAV VP2 to promote viral replication. We found that knockdown of cellular CK2α by RNAi in MDCC-MSB1 or inhibition of its kinase activity by an inhibitor significantly supp

A New Strategy for the Detection of Chicken Infectious Anemia Virus Contamination in Attenuated Live Vaccine by Droplet Digital PCR.

Chicken infectious anemia virus (CIAV) causes the atrophy of bone marrow hematopoietic and lymphoid tissues in chicks, leading to huge economic losses all over the world. The using of attenuated vaccine contaminated with CIAV increased the mortality and the pathogenicity of other diseases in many farms. However, it is difficult to detect the CIAV contamination by general detection technology due to the extremely low dose of CIAV in vaccines. In this study, we established a new method called droplet digital Polymerase Chain Reaction (ddPCR) to detect CIAV contamination of vaccines more sensitiv

A recombinant human adenovirus type 5 vectored vaccine co-expression of VP1 and VP2 of chicken anemia virus elicits robust protective immune responses.

Chicken anemia virus (CAV) causes significant economic losses in the poultry industry worldwide. As current vaccines do not confer complete protection, the development of a highly effective and safe vaccine is of paramount importance. Control strategies relying on live attenuated vaccines carry a risk of reversion to virulence, while conventional inactivated or subunit vaccines preferentially induce a Th2-biased humoral immune response. There often fail to elicit the critical Th1-mediated cellular immunity required to clear infected cells. This immunological imbalance underscores the need for
