Molecular mechanisms and energetic costs of recovery from freezing in the polyextremophile midge Belgica antarctica.

Extracellular freezing challenges insects in several ways, including mechanical damage from ice crystals, exposure to stressful levels of cold and cellular dehydration. While stress-response mechanisms activated during and immediately after freezing are well studied, less is known about how freeze-tolerant organisms recover from freezing in the longer term and whether this recovery carries energetic costs. Here, we tracked changes in gene expression and energy stores over the course of 15 days following extracellular freezing using the world's southernmost insect, Belgica antarctica (Diptera:

[Botulism in birds living in an aquatic environment in Nové Mlýny in the Breclav District].

Epizootic of wild water birds caused by the toxin Clostridium botulinum of C type occurred in extensive shallow artificial lakes of the Nové Mlýny dam system from May to October 1988 and later on from February to early April 1989. In total more than 3,000 dead or dying birds of 44 species were found, the following orders were prevailing: Anseriformes (56%), Lariformes (33%), Charadriiformes (6%) and Ralliformes (4%). Botulotoxin was detected at high concentrations in sarcophagic fly larvae of Calliphora vomitoria and Lucilia sericata which were collected in bird cadavers, but it was not found

Determination of the protein expression profiles of Propsilocerus akamusi (Tokunaga) Malpighian tubules response to cadmium stress by iTRAQ coupled LC-MS/MS.

UNLABELLED: Propsilocerus akamusi (Tokunaga) is an important prey species for fish and aquatic birds, which represent an important link in the aquatic food chain as other species of Chironomidae. Malpighian tubules (MT) play an important role in metabolism and detoxification of harmful compounds and metal accumulation. Our previous studies have reported that P. akamusi have an extraordinary resistance to Cd. In this paper, iTRAQ-based proteomics technology was first used to study the differential proteomics of MT of Propsilocerus akamusi under sublethal Cd stress for different time courses. 11

Quorum-sensing signaling by chironomid egg masses' microbiota, affects haemagglutinin/protease (HAP) production by Vibrio cholerae.

Vibrio cholerae, the causative agent of cholera, is commonly isolated, along with other bacterial species, from chironomid insects (Diptera: Chironomidae). Nevertheless, its prevalence in the chironomid egg masses' microbiota is less than 0.5%. V. cholerae secretes haemagglutinin/protease (HAP) that degrades the gelatinous matrix of chironomid egg masses and prevents hatching. Quorum sensing (QS) activates HAP production in response to accumulation of bacterial autoinducers (AIs). Our aim was to define the impact of chironomid microbiota on HAP production by V. cholerae. To study QS signaling,

Hypersensitivity to larvae of chironomids (non-biting midges). Cross-sensitization with crustaceans.

In 2,368 consecutive adult patients with asthma and/or rhinitis the incidence of positive skin prick test (SPT) with a chironomid extract (CHIR) (produced from "red feather mosquito larvae" used as fish food) was 14% (26% in atopics and 4% in non-atopics). RAST with chironomid was positive in 4% of 110 consecutive sera (8% in atopic sera). Significant correlations were found between RAST and SPT results with chironomid and between SPT results with CHIR and with various crustaceans. Correlations were also found reciprocally among SPT results with different crustaceans and between some crustacea

Cold mountain stream chironomids (Diptera) of the genus Diamesa indicate both historical and recent climate change.

Chironomids of the genus Diamesa (Meigen, 1835, Diptera: Chironomidae) inhabit cold, oxygen-rich running waters. We have investigated the presence of Diamesa and other freshwater macroinvertebrates at 22 stream sampling sites in 3 European high mountain regions (the Central Pyrenees, the Ötztal Alps, and the Tatra Mountains) to establish suitable temperature conditions for Diamesa dominance. It has been generally accepted that their high abundance was linked to the presence of glaciers; however, we have shown that in the Tatra Mountains, where there are no glaciers, the conditions for the domi

Mechanisms influencing the impact of microplastics on freshwater benthic invertebrates: Uptake dynamics and adverse effects on Chironomus riparius.

Chironomids inhabit freshwater benthic ecosystems which are prone to microplastic contamination. This work aimed at understanding the factors and mechanisms influencing microplastic uptake and related adverse effects on Chironomus riparius, by exploring an extensive project database, conducting a literature review, and performing an agent-based model to explore trends in data. Results reveal that high concentrations of small microplastics fill the gut of fourth instar C. riparius (99.7 %). Ingested microplastics had an average size of 38-61 μm, presenting slower elimination rates than undigest

Metallothionein modulation in relation to cadmium bioaccumulation and age-dependent sensitivity of Chironomus riparius larvae.

The goal of this study was to contribute to understanding of the mechanisms behind sensitivity differences between early and late instar larvae of Chironomus riparius and to address the influence of the differences in standard testing approaches on the toxicity evaluation. A 10-day contact sediment toxicity test was carried out to assess sensitivity to cadmium exposure in relation to different age and laboratory culture line origin of test organisms. Chironomid larvae of early (OECD 218 method) and late instar (US-EPA600/R-99/064 method) differed substantially in sensitivity of traditional end

Metal Exposure and Sex Shape the Fatty Acid Profile of Midges and Reduce the Aquatic Subsidy to Terrestrial Food Webs.

Aquatic micropollutants can be transported to terrestrial systems and their consumers by emergent aquatic insects. However, micropollutants, such as metals, may also affect the flux of physiologically important polyunsaturated fatty acids (PUFAs). As certain PUFAs have been linked to physiological fitness and breeding success of terrestrial consumers, reduced fluxes from aquatic systems could affect terrestrial populations and food webs. We chronically exposed larvae of the aquatic insect Chironomus riparius to a range of environmentally relevant sediment contents of cadmium (Cd) or copper (Cu

Effects of chlorpyrifos on macroinvertebrate communities in coastal stream mesocosms.

This study measured the effects of a single pulse of chlorpyrifos at nominal concentrations of 1 and 10 microg/l on the macroinvertebrate community structure of a coastal stream mesocosm system. Analysis of data using Principal Response Curves (PRC) and Monte Carlo tests showed significant changes in the treated stream mesocosms relative to that of the controls. These changes in the macroinvertebrate assemblages occurred within 6 h, and persisted for at least 124 days after dosing. Significant community-level effects were detected at the lowest concentration on days 2 and 16 post-dosing, givin

Widespread IgE-mediated hypersensitivity in Northern Sudan to the chironomid Cladotanytarsus lewisi ('green nimitti').

Hypersensitivity to Chironomidae (non-biting midges) has been a problem in Northern Sudan since about 1927 and is probably due to the working of dams which have produced lake-like conditions on parts of the Blue and main Niles where breeding has evidently increased. Studies were undertaken to determine whether this hypersensitivity is mediated by IgE. Sixteen Sudanese, with bronchial asthma associated with exposure to the chironomid, Cladotanytarsus lewisi ('green nimitti'), were investigated. All patients gave a positive immediate-type skin reaction to an extract of the midge and the majority

Interspecies bacterial communication produces a delicate balance between Vibrio cholerae and the chironomid egg mass microbiome.

The evolution of antimicrobial resistance in bacterial pathogens is considered by the World Health Organization to be one of the ten most concerning public health threats facing humanity (World Health Organization, 2020). Bacterial diseases previously controllable by antibiotics are resurging and treatment options are dwindling. Cholera is one such disease. Human pathogenic strains of Vibrio cholerae cause as many as 4 million cases of disease resulting in over 100,000 deaths each year (Ali et al. 2015) and multidrug-resistant V. cholerae is now established where pandemic cholera persists. Life cycle responses of the midge Chironomus riparius to compounds with different modes of action.

Compounds with different modes of action may affect life cycles of biota differently. The aim of the present study was therefore to investigate the impact of four chemicals with different modes of action, including the essential metal copper, the nonessential metal cadmium, the organometal tributyltin, and the polycyclic aromatic compound phenanthrene, on chronic lethal and sublethal life-cycle effect parameters of the nonbiting midge Chironomus riparius, applying a 28-day sediment toxicity test. Tributyltin and cadmium delayed emergence significantly over a wide range of sublethal concentrati

The larval alimentary canal of the Antarctic insect, Belgica antarctica.

On the Antarctica continent the wingless midge, Belgica antarctica (Diptera, Chironomidae) occurs further south than any other insect. The digestive tract of the larval stage of Belgica that inhabits this extreme environment and feeds in detritus of penguin rookeries has been described for the first time. Ingested food passes through a foregut lumen and into a stomodeal valve representing an intussusception of the foregut into the midgut. A sharp discontinuity in microvillar length occurs at an interface separating relatively long microvilli of the stomodeal midgut region, the site where perit

Vibrio cholerae Invasion Dynamics of the Chironomid Host Are Strongly Influenced by Aquatic Cell Density and Can Vary by Strain.

Cholera has been a human scourge since the early 1800s and remains a global public health challenge, caused by the toxigenic strains of the bacterium Vibrio cholerae. In its aquatic reservoirs, V. cholerae has been shown to live in association with various arthropod hosts, including the chironomids, a diverse insect family commonly found in wet and semiwet habitats. The association between V. cholerae and chironomids may shield the bacterium from environmental stressors and amplify its dissemination. However, the interaction dynamics between V. cholerae and chironomids remain largely unknown.

Chironomid larvae enhance phosphorus burial in lake sediments: Insights from long-term and short-term experiments.

Tube-dwelling macrozoobenthos can affect lake ecosystems in myriad ways, including changes in nutrient fluxes across the sediment-water interface. The pumping activity of chironomid larvae reinforces the transport of solutes between sediment and water. The transport of oxygen into the area surrounding the burrows generates oxidized compounds such as iron(oxy)hydroxides, which results in an additional phosphorus (P) sorption capacity similar to that of oxidized sediment surfaces. In the present study, the effect of the oxidized burrow walls of Chironomus plumosus on P binding capacity and P bin

Mitochondrial DNA suggests a single maternal origin for the widespread triploid parthenogenetic pest species, Paratanytarsus grimmii, but microsatellite variation shows local endemism.

Parthenogenesis is common among invasive pest species, with many parthenogenetic species also showing polyploidy. Parthenogenetic polyploid species often have multiple hybrid origins and the potential to rapidly spread over vast geographical areas. In this study, we examine patterns of mitochondrial and microsatellite variation in a widespread triploid parthenogenetic chironomid pest species, Paratanytarsus grimmii. Based on samples from five countries, including Australia, England, Germany, Japan, and Canada, we found extremely low mitochondrial diversity (<0.14%), with most individuals shari

Habitat availability mediates chironomid density-dependent oviposition.

Knowledge of density-dependent processes and how they are mediated by environmental factors is critically important for understanding population and community ecology of insects, as well as for mitigating harmful insect-borne diseases. Here, we tested whether the oviposition of chironomids (Diptera: Chironomidae; non-biting midges), known to carry the Cholera pathogen Vibrio cholerae, is density dependent and if it is mediated by habitat availability. We used two multiple choice experiments in habitat-limited and habitat-unlimited environments and performed isodar analysis on counts of egg bat

Effects of cadmium and tributyltin on development and reproduction of the non-biting midge Chironomus riparius (Diptera): baseline experiments for future multi-generation studies.

In this study the effects of tributyltin (TBT) and cadmium (Cd) were tested on the freshwater arthropod Chironomus riparius (Diptera) in life-cycle experiments. To this end, the OECD guideline 218 was extended with reproduction relevant parameters (e.g. number of fertile egg masses per female). Based on these reproduction data the number of larvae for next generation and the population growth rate were calculated. Experiments were performed using environmentally relevant concentrations of the test substances in the sediment (nominal ranges: 50-200 micro g Sn/kg and 0.2-2.1 mg Cd/kg, on a dry w
