Structural basis for size-selective perception of chitin in plants.

Plants detect microbes through pattern recognition receptors that perceive conserved microbial surface motifs known as microbe-associated molecular patterns (MAMPs). Lysin motif (LysM) receptors recognize and mediate downstream responses to chitinous MAMPs. In this study, we elucidated the mechanism for the specific recognition of long-chain chitin oligomers and identified a hallmark bridge domain characteristic of the CHITIN PERCEPTION (CHIP) receptor class. Structural analysis of receptor-ligand complexes, biochemistry, and in planta functional studies of inhibitory nanobodies revealed the m

Effects of biomedical macromolecules on the structure of chitin liquid crystals.

The chitin whiskers (CHWs) form liquid crystals in colloidal suspensions due to their unique anisotropic structure, showing enormous potential for applications in optical materials, biomedical materials and other fields. Chitin liquid crystal (CLC) usually requires the introduction of a macromolecular system to form thin films or hydrogels to achieve the purpose of fixing the anisotropic liquid crystal texture. This study aims to systematically explore the interaction between macromolecules and CHWs, clarify the influence of macromolecules on the behavior of CLC. Herein, polyethylene glycol (P

Preparation and characterization of chitin beads as a wound dressing precursor.

Chitin was dissolved in N, N-dimethylacetamide/5% lithium chloride (DMAc/5%LiCl) to form a 0.5% chitin solution. Chitin beads were formed by dropping the 0.5% chitin solution into a nonsolvent coagulant, ethanol. The beads were left in ethanol for 24 h to permit hardening, consolidation, and removal of residual DMAc/5%LiCl solvent in order to give spherical chitin beads uniform size distribution. The ethanol-gelled chitin beads had an average diameter of 535 microm. The chitin beads were subsequently activated in 50% (w/v) NaOH solution and reacted with 1.9 M monochloroacetic acid/2-propanol s

Biomass-based mineralized chitin microsheets for high-efficiency radiative cooling.

The increasing frequency of extreme heat events under global climate change, combined with high energy consumption and CO2 emissions from conventional cooling systems, threatens both ecological sustainability and human health. While traditional passive radiative cooling materials offer a potential solution, they often lead to pollution and have complex designs with high costs. To address these limitations, we developed Mineralized Chitin-based Composite Microsheets (MCCM), a low-cost, eco-friendly radiative cooling material derived from shrimp shells. By leveraging the natural mineralized mult

Chitin-glucan improves important pathophysiological features of irritable bowel syndrome.

BACKGROUND: Irritable bowel syndrome (IBS) is one of the most frequent and debilitating conditions leading to gastroenterological referrals. However, recommended treatments remain limited, yielding only limited therapeutic gains. Chitin-glucan (CG) is a novel dietary prebiotic classically used in humans at a dosage of 1.5-3.0 g/d and is considered a safe food ingredient by the European Food Safety Authority. To provide an alternative approach to managing patients with IBS, we performed preclinical molecular, cellular, and animal studies to evaluate the role of chitin-glucan in the main pathoph

Overview of chitin dissolution, hydrogel formation and its biomedical applications.

Chitin hydrogel and hydrogel-based products are some of the frequently reported biomaterials for biomedical applications. Yet there is a void in understanding chitin's dissolution mechanism and its most suitable solvent system(s). Chitin is a natural polysaccharide polymer which can be dissolved in solvents such as calcium chloride- methanol, sodium hydroxide/urea (NaOH/urea), lithium chloride diacetamide (LiCl/DMAc), ionic liquids and deep eutectic solvents. Among the alkali/urea dissolution systems such as NaOH/urea, KOH/urea, LiOH/urea for dissolution of chitin we will be focussing on NaOH-

Preparation and properties of chitin/silk fibroin biocompatible composite fibers.

In the present world chitin is used enormously in various fields, such as biopharmaceuticals, medical and clinical bioproducts, food packaging, etc. However, its development has been curbed by the impaired performance and cumbersome dissolution process when chitin materials are dissolved and regenerated by physical or chemical methods. To further obtain the regenerated chitin fiber material with improved performance, silk fibroin was introduced into the chitin matrix material, and chitin/silk fibroin biocompatible composite fibers were obtained by formic acid/calcium chloride/ethanol ternary s

Early Application of Quaternized Chitin Derivatives Inhibits Hypertrophic Scar Formation.

Various treatments for hypertrophic scars (HS) are applied after wound re-epithelialization. However, the lack of early intervention within the wound bed leads to poor HS treatment outcomes. In this study, quaternized chitin (QC) derivatives with different degrees of deacetylation (7.4% and 78.9%) are synthesized and their effects on HS formation are evaluated in a rabbit ear scar model. Early application of QC alleviates scar hypertrophy without delayed wound healing. Fibroblast count, collagen content, and α-smooth muscle actin expression are decreased, while matrix metalloproteinase-1 is up

Biocompatible chitin-encapsulated CdS quantum dots: Fabrication and antibacterial screening.

Chitin-encapsulated cadmium sulfide quantum dots (CdS@CTN QDs) were successfully synthesized from chitin and Cd(NO3)2 precursor using the colloidal chemistry method, toward the development of biocompatible and biodegradable QDs for biomedical applications. CdS@CTN QDs exhibited the nanocrystalline cubic CdS encapsulated by α-chitin. The average particle size of CdS@CTN QDs was estimated using empirical Henglein model to be 3.9 nm, while their crystallite size was predicted using Scherrer equation to be 4.3 nm, slightly larger compared to 3-mercaptopropionic acid-capped CdS QDs (3.2 and 3.6 nm,

CuS@Corn Stalk/Chitin Composite Hydrogel for Photodegradation and Antibacterial.

Copper sulfide nanoparticles (CuS NPs) have recently attracted extensive attention in various fields due to their excellent optical and electrical properties. However, CuS NPs are easy to agglomerate in their preparation on account of the high surface activity. In this study, uniform dispersion of CuS NPs were fabricated with corn stalk as a template and stabilizer, further CuS@corn stalk/chitin composite hydrogel was obtained by crosslinking with chitin. The results reveal that the CuS NPs were evenly dispersed into the composite hydrogels with a three-dimensional network structure, which wer

Sorption studies on heavy metal removal using chitin/bentonite biocomposite.

Contamination of water by toxic heavy metals due to urbanization is a world-wide environmental problem, which changes chemical and biological properties of both surface and ground water. The heavy metals render the water unsuitable for drinking and are also highly toxic to human beings. Removal of heavy metals is therefore essential. Thus, in my present work batch adsorption studies have been used to remove the Cr(VI) from aqueous solution using chitin composite. The data obtained from batch method at optimized conditions have been subjected to Freundlich and Langmuir isotherm studies. The dat

Multifunctional chitin nanogels for simultaneous drug delivery, bioimaging, and biosensing.

In this work, we developed biodegradable chitin nanogels (CNGs) by controlled regeneration method. For multifunctionalization, we have conjugated CNGs with MPA-capped-CdTe-QDs (QD-CNGs) for the in vitro cellular localization studies. In addition, the Bovine Serum Albumin (BSA) was loaded on to QD-CNGs (BSA-QD-CNGs). The CNGs, QD-CNGs, and BSA-QD-CNGs were well-characterized by SEM and AFM, which shows that the nanogels are in the range of <100 nm. These were further characterized by FT-IR and Cyclic Voltametry. The cytocompatibility assay showed that the nanogels are nontoxic to L929, NIH-3T3,

Structural and chemical properties of insect's chitin-containing extracellular matrices.

Insect's body barriers rely on specialized extracellular matrices that protect against harmful environmental influences. The outer barrier is the cuticle, which is composed of chitin, cuticle proteins and lipids. The peritrophic matrix (PM) serves as an inner barrier lining the midgut epithelium. It is composed of chitin fibers that are organized by PM proteins. While cuticle and PM proteins have received considerable attention in the past, supramolecular organization and physicochemical properties of the chitin component - particularly of the PM - remain poorly understood. Here, we combine sy

Curvature of the Drosophila corneal lens depends on localized chitin secretion.

The Drosophila corneal lens is an apical extracellular matrix structure with a biconvex shape that enables it to focus light onto the underlying photoreceptors. Here, we investigated how this shape is influenced by the source of one of its major components, the polysaccharide chitin. Knocking down the chitin synthase Krotzkopf verkehrt strongly reduced the thickness and curvature of the corneal lens. Conversely, enhancing chitin export by overexpressing Rebuf expanded and distorted the corneal lens. We found that the cone and primary pigment cells in the center of each ommatidium produce most

Elevated chitin content reduces the susceptibility of Candida species to caspofungin.

The echinocandin antifungal drugs inhibit synthesis of the major fungal cell wall polysaccharide β(1,3)-glucan. Echinocandins have good efficacy against Candida albicans but reduced activity against other Candida species, in particular Candida parapsilosis and Candida guilliermondii. Treatment of Candida albicans with a sub-MIC level of caspofungin has been reported to cause a compensatory increase in chitin content and to select for sporadic echinocandin-resistant FKS1 point mutants that also have elevated cell wall chitin. Here we show that elevated chitin in response to caspofungin is a com

Anti-ulcer effects of chitin and chitosan, healthy foods, in rats.

In this study, we compared the effects of low molecular weight (LMW) chitosan (MW: 25,000-50,000), high molecular weight (HMW) chitosan (MW: 500,000-1000,000) and chitin on ethanol-induced gastric mucosal injury and on the healing of acetic acid-induced gastric ulcers in rats. Oral administration of LMW chitosan (250, 500 and 1000 mg/kg) dose-dependently prevented ethanol-induced gastric mucosal injury. Repeated oral administration of LMW chitosan (100, 200 and 400 mg/kg twice daily) also dose-dependently accelerated the gastric ulcer healing. However, the effects of HMW chitosan and chitin on
