Poly-ε-caprolactone/Chitosan and Chitosan Particles: Two Recombinant Antigen Delivery Systems for Intranasal Vaccination.

Several evidences converge on the idea that among the mucosal administration routes, the nasal mucosa is the most attractive site for the delivery of vaccines. Mucoadhesive particulate adjuvants should be able to increase the residence time of antigens in nasal cavity in order to increase their probability of being taken up by nasopharynx-associated lymphoid tissue (NALT) cells and subsequently to initiate the innate and adaptive immune response. Focusing on chitosan, a mucoadhesive biopolymer, we describe in this chapter a method to prepare antigen loaded chitosan nanoparticles and a second m

Characterization and biocompatibility of chitosan nanocomposites.

Chitosan nanocomposites were prepared from chitosan and gold nanoparticles (AuNPs) or silver nanoparticles (AgNPs) of ∼5 nm size. Transmission electron microscopy (TEM) showed the NPs in chitosan did not aggregate until higher concentrations (120-240 ppm). Atomic force microscopy (AFM) demonstrated that the nanocrystalline domains on chitosan surface were more evident upon addition of AuNPs (60 ppm) or AgNPs (120 ppm). Both nanocomposites showed greater elastic modulus, higher glass transition temperature (T(g)) and better cell proliferation than the pristine chitosan. Additionally, chitosan-A

Development of Antimicrobial Chitosan-Silver and Chitosan-Gold Nanocomposite Films by 3D Printing for Skin Application.

Silver (AgNP) and gold (AuNP) nanoparticles are notable in nanomedicine for their unique properties and strong antibacterial effects; however, formulating them into skin-applied systems that preserve bioactivity while controlling nanoparticle exposure remains challenging. Herein, semisolid extrusion 3D printing of chitosan nanocomposite hydrogels loaded with AgNPs or AuNPs was used to manufacture personalized topical films with antimicrobial performance and minimized skin penetration risk. AgNPs and AuNPs were synthesized with comparable nanoscale dimensions (11.83 ± 3.74 nm and 11.67 ± 1.96 n

Effects of Chitosan and N-Succinyl Chitosan on Metabolic Disorders Caused by Oral Administration of Olanzapine in Mice.

BACKGROUND: The issue of human mental health is gaining more and more attention nowadays. However, most mental disorders are treated with antipsychotic drugs that cause weight gain and metabolic disorders, which include olanzapine (OLZ). The search for and development of natural compounds for the prevention of obesity when taking antipsychotic drugs is an urgent task. The biopolymer chitosan (Chi) and its derivatives have lipid-lowering and anti-diabetic properties, which makes them potential therapeutic substances for use in the treatment of metabolic disorders. The purpose of this work was t

Beneficial Metabolic Effects of Chitosan and Chitosan Oligosaccharide on Epididymal WAT Browning and Thermogenesis in Obese Rats.

Many anti-obesity chemicals have been withdrawn from the market due to serious adverse reactions, and the researchers have turned their attention to low-toxic natural products. Previous studies have demonstrated that chitosan (CTS) and chitosan oligosaccharide (COS) were low-toxic natural products for the use of weight loss. However, it is still unclear whether CTS and COS have positive effects on the thermogenesis. In this study, CTS and COS significantly reduced the weight gain of rats without affecting food intake and effectively inhibited adipose tissue hypertrophy and hyperplasia. Consist

Protective nature of low molecular weight chitosan in a chitosan-Amphotericin B nanocomplex - A physicochemical study.

Chitosan, a biopolymer of immense potential, has been well-explored over the past decade in the biomedical field. Despite its numerous biological properties like anti-microbial, anti-inflammatory, anti-diabetic etc., limited studies have been conducted on its ability to protect therapeutic molecules in nano-formulations. Amphotericin B (AMP) is one such therapeutic molecule which requires protection as it possesses inherent limitations of poor water-solubility, susceptibility to oxidation- and/or light-induced degradation etc. Although AMP formulations have been quite successful in treating ch

Surface attachment of fucoidan through electrostatic interaction alleviates chitosan/chitosan oligosaccharide-induced reduction in protein digestion.

BACKGROUND: Chitosan (CTS) and chitosan oligosaccharides (COS) are widely used in the food industry due to their bioactive properties. However, their cationic nature enables interaction with whey protein isolate (WPI), affecting protein digestibility. Fucoidan (FUC), a naturally occurring anionic sulfated polysaccharide from brown algae, can form electrostatic complexes with CTS and COS to partially alleviate this effect; however, this process requires optimization. RESULTS: Chitosan-fucoidan (CTS-FUC) and chitosan oligosaccharide-fucoidan (COS-FUC) complexes were successfully prepared and ch

Electrospinning of hexanoyl chitosan/polylactide blends.

The electrospinning of hexanoyl chitosan (H-chitosan), polylactide (PLA) and their blends was performed using chloroform, dichloromethane or tetrahydrofuran as a spinning solvent. The as-spun PLA fibers appeared to be cross-sectionally round, with rough surface morphology, while the as-spun H-chitosan fibers were flat with smooth surface morphology. Electrospinning of the H-chitosan/PLA blend solutions in chloroform with the H-chitosan solution content of less than or equal to 50% (w/w) produced continuous fibers without the presence of beads, while that of the blend solutions in dichlorometha

Ag@MOF-loaded chitosan nanoparticle and polyvinyl alcohol/sodium alginate/chitosan bilayer dressing for wound healing applications.

In this paper, Ag-Metal-organic framework loaded chitosan nanoparticles (0.1%Ag@MOF/1.5%CSNPs) and polyvinyl alcohol/sodium alginate/chitosan (PACS) were used as the upper and lower layers to successfully prepare a bilayer composite dressing for wound healing. The performance of bilayer dressing was evaluated. The lower layer (PACS) had uniform pore size distribution, good water retention, swelling, water vapor permeability, and biocompatibility while PACS had almost no antibacterial activity. The upper layer (Ag@MOF/CSNPs) possessed excellent antibacterial activity and poor biocompatibility.
