Mild osmotic stress offers photoprotection in Chlamydomonas reinhardtii under high light.

The exposure of autotrophs to high light intensities significantly impacts their photosynthetic performance. When combined with unpredictable climate changes, the lethality of these effects is exacerbated and, often surpassing the organisms' threshold for tolerance. In this regard, our study centres on examining the mitigating effects of mild osmotic stress induced by 2% Polyethylene Glycol (PEG) in conjunction with high-light conditions, using Chlamydomonas reinhardtii as a model system. Cells were cultivated under low PEG-induced osmotic stress at various light intensities, and their respons

Characteristics of N6-methyladenosine Modification During Sexual Reproduction of Chlamydomonas reinhardtii.

The unicellular green alga Chlamydomonas reinhardtii (hereafter Chlamydomonas) possesses both plant and animal attributes, and it is an ideal model organism for studying fundamental processes such as photosynthesis, sexual reproduction, and life cycle. N6-methyladenosine (m6A) is the most prevalent mRNA modification, and it plays important roles during sexual reproduction in animals and plants. However, the pattern and function of m6A modification during the sexual reproduction of Chlamydomonas remain unknown. Here, we performed transcriptome and methylated RNA immunoprecipitation sequencing (

Interspecific competition between Microcystis aeruginosa and Chlamydomonas microsphaera stressed by tetracyclines.

The extensive use of tetracyclines in human and veterinary medicine causes contamination in the environment that could contribute to the spread of antibiotic-resistant bacteria or competition between species of phytoplankton. In this study, Microcystis aeruginosa (a bloom-forming cyanobacterium) and Chlamydomonas microsphaera (common green alga) were selected to test the effects of different concentrations of tetracyclines (tetracycline and oxytetracycline) in monoculture and co-culture. The results showed that compared with monoculture, the cell growth of C. microsphaera decreased significant

Genetically Programmed Changes in Photosynthetic Cofactor Metabolism in Copper-deficient Chlamydomonas.

Genetic and genomic studies indicate that copper deficiency triggers changes in the expression of genes encoding key enzymes in various chloroplast-localized lipid/pigment biosynthetic pathways. Among these are CGL78 involved in chlorophyll biosynthesis and HPPD1, encoding 4-hydroxyphenylpyruvate dioxygenase catalyzing the committed step of plastoquinone and tocopherol biosyntheses. Copper deficiency in wild-type cells does not change the chlorophyll content, but a survey of chlorophyll protein accumulation in this situation revealed increased accumulation of LHCSR3, which is blocked at the le

CrGNAT gene regulates excess copper accumulation and tolerance in Chlamydomonas reinhardtii.

Excess copper (Cu) in environment affects the growth and metabolism of plants and green algae. However, the molecular mechanism for regulating plant tolerance to excess Cu is not fully understood. Here, we report a gene CrGNAT enconding an acetyltransferase in Chlamydomonas reinhardtii and identified its role in regulating tolerance to Cu toxicity. Expression of CrGNAT was significantly induced by 75-400μM Cu. The top induction occurred at 100μM. Transgenic algae overexpressing CrGNAT (35S::CrGNAT) in C. reinhardtii showed high tolerance to excess Cu, with improved cell population, chlorophyll

Chlorophyll-deficient mutants of Chlamydomonas reinhardtii that accumulate magnesium protoporphyrin IX.

Two Chlamydomonas reinhardtii mutants defective in CHLM encoding Mg-protoporphyrin IX methyltransferase (MgPMT) were identified. The mutants, one with a missense mutation (chlM-1) and a second mutant with a splicing defect (chlM-2), do not accumulate chlorophyll, are yellow in the dark and dim light, and their growth is inhibited at higher light intensities. They accumulate Mg-protoporphyrin IX (MgProto), the substrate of MgPMT and this may be the cause for their light sensitivity. In the dark, both mutants showed a drastic reduction in the amounts of core proteins of photosystems I and II and

Adenosine 3':5'-Cyclic Monophosphate in Chlamydomonas reinhardtii: Isolation and Characterization.

Chlamydomonas reinhardtii contains a factor that can replace adenosine 3':5'-cyclic monophosphate (cAMP) in the stimulation of rabbit-muscle protein kinase. The factor cochromatographs and coelectrophoreses with authentic cAMP, and is inactivated by beef heart cyclic nucleotide phosphodiesterase. When C. reinhardtii is exposed to aminophylline (theophylline(2) ethylenediamine), the concentration of the factor in the cells increases within 1 hr, from about 25 pmol of cAMP equivalents per g dry weight to more than 250 pmol. Cyclic nucleotide phosphodiesterase activity is present in crude extract

SAGA1 and SAGA2 localize the starch sheath to the pyrenoid in Chlamydomonas reinhardtii.

Most algae enhance their CO2 assimilation by concentrating CO2 within the pyrenoid, a biomolecular condensate of the CO2-fixing enzyme Rubisco. Many pyrenoids are surrounded by a starch sheath thought to slow the escape of CO2 from the pyrenoid, but how the starch sheath is localized to the pyrenoid remains poorly understood. Here, in the model alga Chlamydomonas reinhardtii, we find that the protein SAGA2 is necessary for early pyrenoid starch sheath biogenesis and works redundantly with its homolog, SAGA1, to localize the starch sheath to the pyrenoid. SAGA2 and SAGA1 were enriched in differ

Intracellular distribution and speciation of indium in the green microalgae Chlamydomonas reinhardtii.

The interactions between indium and aquatic organisms are poorly documented despite indium increasing use. This study aimed to improve current knowledge of indium effects and intracellular fate in microalgae. The model green algae Chlamydomonas reinhardtii were thus exposed for 96 h to total indium concentrations ranging from 0.08 to 36.5 µM and examined for their growth and subcellular parameters as well as for their indium subcellular distribution and speciation using differential ultracentrifugation and size-exclusion chromatography coupled with inductively coupled plasma mass spectrometry

Architecture and evolution of subtelomeres in the unicellular green alga Chlamydomonas reinhardtii.

In most eukaryotes, subtelomeres are dynamic genomic regions populated by multi-copy sequences of different origins, which can promote segmental duplications and chromosomal rearrangements. However, their repetitive nature has complicated the efforts to sequence them, analyse their structure and infer how they evolved. Here, we use recent genome assemblies of Chlamydomonas reinhardtii based on long-read sequencing to comprehensively describe the subtelomere architecture of the 17 chromosomes of this model unicellular green alga. We identify three main repeated elements present at subtelomeres,

[Effect of microwaves on Chlamydomonas actinochloris culture in the stationary phase of growth].

Effects of the microwave radiation on the culture of Chlamydomonas actinochloris green flagellar alga in the stationary phase of growth are studied. After exposure to radiation at the maximum dose of 125 J/g, the cell functional state worsened but all the studied parameters were restored in 20 days and in the long run found to be even better than the control indices. The data are compared with the similar ones obtained earlier for the lag phase culture. The studied sample is found to be more resistant to the irradiation than the previous one.

Effects of chromium on photosynthetic and photoreceptive apparatus of the alga Chlamydomonas reinhardtii.

Chromium is a highly toxic non-essential metal for microorganisms and plants. Due to its widespread industrial use, chromium (Cr) has become a serious pollutant in diverse environmental settings. The presence of Cr leads to the selection of algal populations able to tolerate high levels of Cr compounds. The diverse Cr-resistance mechanisms displayed by microorganisms include biosorption, diminished accumulation, precipitation, reduction of Cr(6+) to Cr(3+), and chromate efflux. In this paper we describe the effects of Cr(6+) (the more toxic species) on the photosynthetic and photoreceptive app

Comparative kinetics and inhibition of a carbonic anhydrase from Chlamydomonas reinhardii.

A native carbonic anhydrase (CA) from Chlamydomonas reinhardii exhibited an apparent molecular weight of 165,000. Denaturation under reducing conditions yielded a single polypeptide, mol. wt 42,000, while the same treatment under nonreducing conditions yielded higher molecular weight aggregates at least two of which could be reactivated to exhibit enzyme activity. In CO2 hydration the enzyme has a Km value of 3.4 mM and a relatively high turnover number. It is very sensitive to inhibition by sulfonamide and moderately sensitive to anion inhibition. In general, the C. reinhardii enzyme resemble

The effect of manganese on chloroplast structure and photosynthetic ability of Chlamydomonas reinhardi.

Manganese deficiency was induced in mixotrophically-grown cultures of the green alga Chlamydomonas reinhardi and the effects of this deficiency on photosynthetic activity and cell structure were investigated. Manganese-deficient cells are unable to carry out, at normal rates, photosynthetic reactions involving system II of the photosynthetic electron transport chain. Reactions requiring only system I are not inhibited. Normal system II activity returns within 2 hr in the light following addition of manganous ions to deficient cells.Significant structural alterations for deficient cells were ob

Differential antioxidant pathways displayed by Chlamydomonas reinhardtii exposed to selected NSAIDs.

The chemically diverse group of nonsteroidal anti-inflammatory drugs (NSAIDs) is a significant source of aquatic pollution, inducing oxidative stress in algae. The aim of this study was to evaluate the antioxidant response of Chlamydomonas reinhardtii exposed to four NSAIDs differing in toxicity and structure: flufenamic acid (FFA), nabumetone (NBT), ibuprofen (IBU), and naproxen (NPX). This study demonstrated that all pharmaceuticals significantly increased H₂O₂ production, confirming redox imbalance in cells. The antioxidant defense also showed compartment- and compound-specific signatures d

Photoreceptor-induced LHL4 protects the photosystem II monomer in Chlamydomonas reinhardtii.

Photosynthesis, the fundamental process using light energy to convert carbon dioxide to organic matter, is vital for life on Earth. It relies on capturing light through light-harvesting complexes (LHC) in photosystem I (PSI) and PSII and on the conversion of light energy into chemical energy. Composition and organization of PSI and PSII core complexes are well conserved across evolution. PSII is particularly sensitive to photodamage but benefits from a large diversity of photoprotective mechanisms, finely tuned to handle the dynamic and ever-changing light conditions. Light Harvesting Complex

CO2-Dependent Promotion of Photosynthesis Drives Metabolic Photoacclimation in Chlamydomonas reinhardtii.

Light and inorganic carbon (Ci) drive photosynthesis, which fuels cellular maintenance, energy storage, and growth in photosynthetic organisms. Despite its pivotal role, how primary metabolism adjusts to contrasting light and Ci availability in algae remains elusive. Here, we characterized bioenergetics and profiled primary metabolites of photoautotrophic Chlamydomonas reinhardtii cultures grown under constant low/sub-saturating (LL) or high/saturating (HL) light with 2% (CO2) or ambient 0.04% (Amb) CO2. HL-Amb cells suffered photoinhibition and limitation of photosystem I electron flow at the

Interplay between CO2 and light governs carbon partitioning in Chlamydomonas reinhardtii.

Increasing CO2 availability is a common practice at the industrial level to trigger biomass productivity in microalgae cultures. Still, the consequences of high CO2 availability in microalgal cells exposed to relatively high light require further investigation. Here, the photosynthetic, physiologic, and metabolic responses of the green microalga model Chlamydomonas reinhardtii were investigated in high or low CO2 availability conditions: high CO2 enabled higher biomass yields only if sufficient light energy was provided. Moreover, cells grown in high light and high CO2 availability were charac

Monitoring Autophagic Flux in the Model Single-Celled Microalga Chlamydomonas reinhardtii.

Autophagy is a catabolic process by which eukaryotic cells degrade and recycle unnecessary or damaged intracellular components to maintain cellular homeostasis and to cope with stress. The development of specific tools to monitor autophagy in microalgae and plants has been fundamental to investigate this catabolic pathway in photosynthetic organisms. The protein ATG8 is a widely used molecular marker of autophagy in all eukaryotes, including the model microalga Chlamydomonas reinhardtii. The drug concanamycin A, a specific inhibitor of vacuolar ATPase, has also been extensively used to block a

Enhanced Lipid Production in Chlamydomonas reinhardtii Caused by Severe Iron Deficiency.

Microalgae are used as a source of lipids for the production of biofuels. Most algae produce neutral lipids under stress conditions. Here, lipid accumulation by the unicellular alga Chlamydomonas reinhardtii was examined during cultivation under iron-limiting conditions. Severe iron stress caused the cells to accumulate a significant amount of lipid, specifically triacylglycerols (TAGs), by compromising the growth. Semi-quantitative measurements by Fourier transform infrared (FTIR) spectroscopy showed an increase in both carbohydrate and lipid content in iron-stressed C. reinhardtii cells comp
