Comparison of taurine chloramine and taurine bromamine effects on rheumatoid arthritis synoviocytes.

Fibroblast-like synoviocytes (FLS) participate in rheumatoid arthritis (RA) chronic synovitis by producing pro-inflammatory cytokines (IL-6, IL-8), growth factors (VEGF) and other inflammatory mediators (PGE2, NO). We have previously reported that Tau-Cl, generated by neutrophils, inhibits in vitro some of these pathogenic RA FLS functions. Taurine bromamine (Tau-Br) originates from eosinophils and neutrophils, and its immunoregulatory activities are poorly known. Therefore, we investigated the effects of Tau-Br on RA FLS functions and compared it to Tau-Cl anti-inflammatory action. When appli

Distinct modes of cell death induced by different reactive oxygen species: amino acyl chloramines mediate hypochlorous acid-induced apoptosis.

Oxidants derived from inflammatory phagocytes compose a key element of the host immune defense system and can kill mammalian cells by one of several different mechanisms. In this report, we compare mechanisms of cell death induced in human B lymphoma cells by the inflammatory oxidants superoxide, H(2)O(2), and HOCl. The results indicate that the mode of cell death induced depends on the nature of the oxidant involved and the medium in which the cells are treated. When human Burkitt's lymphoma cells are exposed to superoxide anion, generated as a flux from xanthine and xanthine oxidase, the cel

Influencing factors of disinfection byproducts formation during chloramination of Cyclops metabolite solutions.

Effects of reaction time, chlorine dosage, pH and temperature on the formation of disinfection byproducts (DBPs), were investigated during the chloramination of Cyclops metabolite solutions. The results showed that some species of DBPs like trichloromethane (TCM), dichloroacetic acid (DCAA) and trichloroacetic acid (TCAA) could accumulate to their respective stable values with a progressive elevation in reaction time and monochloramine concentration. And 1,1,1-2-trichloropropanone (1,1,1-TCP) content decreased correspondingly with a continuous increase of reaction time. The amounts of chloral

Trends in chlorine and chloramine gas exposures reported to United States poison centers.

INTRODUCTION: Chlorine and chloramine gas inhalation can occur when household cleaners are mixed. The increased emphasis on disinfecting practices during the COVID-19 pandemic may have contributed to an increase in chlorine and chloramine gas exposures in the United States, which has not been studied. METHODS: In a retrospective review, reported data on chlorine and chloramine gas exposures in the National Poison Data System were collected from January 1, 2015, to December 31, 2022. Data included demographics and exposure details, including location, dose, formulation, co-exposures, treatment

Generation of iodinated trihalomethanes during chloramination in the presence of solid copper corrosion products.

Copper water pipelines are widely used in water distribution systems, but the effects of solid copper corrosion products (CCPs) including CuO, Cu2O and Cu2(OH)2CO3 on the generation of iodinated trihalomethanes (I-THMs) during chloramination remain unknown. This study found that the formation of I-THMs during chloramination of humic acid (HA) was inhibited by the presence of CuO and Cu2O, but promoted with the addition of Cu2(OH)2CO3. The negative effect of CuO and Cu2O is mainly exerted by promoting the decay of both NH2Cl and HOI. Although Cu2(OH)2CO3 also accelerated the decomposition of NH

Tetracycline antibiotics as precursors of dichloroacetamide and other disinfection byproducts during chlorination and chloramination.

Pollution of natural water and even source water with pharmaceuticals is problematic worldwide and raises concern about the possibility of disinfection byproduct (DBP) formation during subsequent water treatment. In this study, the formation of DBPs, especially dichloroacetamide (DCAcAm), was investigated during chlorination and chloramination of tetracyclines, which are a class of broad-spectrum antibiotics. DBPs including DCAcAm were formed during chlorination and chloramination of tetracycline (TC). Although the concentrations and theoretical cytotoxicity of the DBPs formed from TC were aff

Inhibition of septic arthritis by local administration of taurine chloramine, a product of activated neutrophils.

OBJECTIVE: Taurine is an amino acid able to react with hypochlorous acid, produced endogenously by neutrophils, resulting in the more stable and less toxic taurine chloramine (Tau-Cl). Since Tau-Cl has been shown to down-regulate the production of proinflammatory mediators and to exert anti-bacterial properties, we investigated the efficacy of Tau-Cl treatment for infectious arthritis. METHODS: The murine model of hematogenous septic arthritis involved intravenous injection of a single dose of Staphylococcus aureus. Tau-Cl was administered by daily intraperitoneal injections. Assessment of the carcinogenic potential of chlorinated water: experimental studies of chlorine, chloramine, and trihalomethanes.

BACKGROUND: Water chlorination has been one of the major disease prevention treatments of this century. While epidemiologic studies suggest an association between cancer in humans and consumption of chlorination byproducts in drinking water, these studies have not been adequate to draw definite conclusions about the carcinogenic potential of the individual byproducts. PURPOSE: The purpose of this study was to investigate the carcinogenic potential of chlorinated or chloraminated drinking water and of four organic trihalomethane byproducts of chlorination (chloroform, bromodichloromethane, chl

Atopic allergy to chloramine-T and the demonstration of specific IgE antibodies by the radioallergosorbent test.

Chloramine-T is a small molecular oxidizing agent that has been widely used as a disinfectant since the beginning of this century. It is generally used in a 5% solution but it is also supplied in powder form. Sporadic case reports of immediate-type sensitization to this agent associated with symptoms of asthma, rhinitis and urticaria have appeared during recent decades. In one of the reports, specific IgE antibodies in sera of four patients who developed asthmatic symptoms after exposure to chloramine-T were demonstrated using a radioimmuno-assay. Three cases of bronchial asthma in workers who

Comparison of Bolton-Hunter and chloramine-T techniques for the radioiodination of prostatic acid phosphatase.

A double antibody, semi-automated radioimmunoassay for serum prostatic acid phosphatase (PAP) is described, which uses the 125I-labelled N-succinimidyl-3-(4-hydroxyphenyl) propionate ester of PAP. This type of label has substantially higher immunoreactivity than that prepared using the chloramine-T method of radioiodination. Chromatographic purification of either label on Ultrogel ACA44 further improved immunoreactivity. The lowest detection limit (0 . 35 micrograms/l) was achieved with the chromatographically purified ester label.

Inorganic chloramines: a critical review of the toxicological and epidemiological evidence as a basis for occupational exposure limit setting.

Inorganic chloramines are not commercially available, but monochloramine is produced in situ for disinfection or for use in chemical synthesis. Inorganic chloramines are also formed when free chlorine reacts with nitrogen containing substances, e.g. ammonia and urea, present in chlorinated water sources. Occupational exposure may, therefore, occur in e.g. swimming pool facilities and the food processing industry. Monochloramine is soluble and stable in water and the dominating inorganic chloramine in chlorinated water sources. No clinical effects were seen in healthy volunteers given monochlor

Algae impacted drinking water: Does switching to chloramination produce safer drinking water?

Harmful algal (cyanobacterial) blooms (HABs) are increasing throughout the world. HABs can be a direct source of toxins in freshwater sources, and associated algal organic matter (AOM) can act as precursors for the formation of disinfection by-products (DBPs) in drinking water. This study investigated the impacts of algae on DBP formation using treatment with chloramine, which has become a popular disinfectant in the U.S. and in several other countries because it can significantly lower the levels of regulated DBPs formed. Controlled laboratory chloraminations were conducted using live field-c

Characterization of Carbonyl Disinfection By-Products During Ozonation, Chlorination, and Chloramination of Dissolved Organic Matters.

Carbonyl compounds are an important class of by-products that are generated in disinfection reactions. These chemicals are ingredients contributing to toxicology in the drinking water system, the compositions and structures of which are worthy of attention. In this study, a chemical derivatization method based on simultaneous light/heavy isotope labeling was established for general recognition of carbonyl compounds and carbonyl disinfection by-products (DBPs) as per the humic substance reference standard (Suwannee river fulvic acid II, SRFA) before and after ozonation, chlorination, and chlora

Ultraviolet-induced effects on chloramine and cyanogen chloride formation from chlorination of amino acids.

Ultraviolet (UV)-based treatment is commonly used to augment chlorination in swimming pools. However, the effects of combined application of UV254/chlorine on disinfection byproduct (DBP) formation are incompletely defined. To examine this issue, experiments were conducted with amino acids (l-arginine, l-histidine, and glycine) that are representative of those introduced to swimming pools via human body fluids. For each precursor, stepwise experiments were conducted with chlorination and UV254 exposure, with/without post-chlorination. Net formation and decomposition of chloramines and cyanogen

Taurine chloramine inhibits osteoclastogenesis and splenic lymphocyte proliferation in mice with collagen-induced arthritis.

Taurine chloramine (TauCl) is produced by activated neutrophils in the inflammatory joint cavities of rheumatoid arthritis and is known to have anti-inflammatory and anti-arthritic effects. However, the mechanisms underlying the anti-arthritic effect of TauCl have not been elucidated. Here, we investigated that mechanism using a collagen-induced arthritis (CIA) mouse model. DBA/1J mice were immunized with bovine type II collagen to induce CIA and were given daily subcutaneous injections of TauCl from the day of first collagen immunization. Severity of arthritis was scored by paw swelling and t

Immunological specificity of chloramine-T-induced IgE antibodies in serum from a sensitized worker.

A procedure for the preparation of chloramine-T (CT) conjugates used to assay IgE antibodies was developed using response surface methodology and serum from a subject occupationally exposed to the substance. The conjugates, synthesized by reacting CT with human serum albumin (HSA) and other protein carriers, were used as antigens in a radio-allergosorbent test (RAST). Human serum albumin was found to be a suitable carrier, although other protein carriers also gave specific IgE-binding of a similar extent. The CT-HSA conjugates used in the RAST were characterized by high performance liquid chro

Physiologic and biologic behavior of autologous canine 131L-fibrinogen labeled by a chloramine-T method.

Autologous canine 131I-fibrinogen labeled by a chloramine-T method was examined to determine whether the fibrinogen had been altered by the iodination procedure as suggested by other investigators. Before and after radioiodination, clottability of the fibrinogen was always equal to or greater than 89 percent. Gel chromatography studies revealed that labeled fibrinogen behaved as unlabeled fibrinogen and no high molecular weight material was found. Gamma camera scintigraphy reflected the chromatography results, as iodinated material did not appear in the liver or spleen until 48 to 72 hours. Th
