Drug metabolism and disposition: the biological fate of chemicals1998eng
Human subjects were exposed by inhalation to 250, 500, and 1000 ppm 1,1-dichloro-1-fluoroethane (HCFC-141b) for 4 hr, and urine samples were collected from 0-4, 4-12, and 12-24 hr for metabolite analysis. 19F nuclear magnetic resonance spectroscopic analysis of urine samples from exposed subjects showed that 2,2-dichloro-2-fluoroethyl glucuronide and dichlorofluoroacetic acid were the major and minor metabolites, respectively, of HCFC-141b. Semivolatile organic compounds (SOC) occur in the troposphere either adsorbed at aerosol particles or in the free gas phase, depending on temperature, vapor pressure of the compound, and total particular surface. In order to estimate the abiotic degradation of such compounds, the OH reaction rate constant must be known, which cannot be measured directly in the gas phase at the relevant temperature due to experimental difficulties. This case report presents two males with drug-induced liver injury acquired from working at a glass factory dealing with silica and 2,2-dichloro-1,1,1-trifluoroethane (HCFC-123). Within one month of work, both patients presented with fever, icterus with liver dysfunction, and eosinophilia. A simple method for extracting and purifying lipids from rat liver in a single step using nontoxic solvents is described. The method consists homogenizing the pulverized tissue with a mixture of trichlorotrifluoroethane (Cl2CF-CCIF2) and isopropyl alcohol (1 : 1, v/v). A physiologically based pharmacokinetic model was used to determine the in vivo metabolic constants of the candidate chlorofluorocarbon replacement 1,1-dichloro-1-fluoroethane (HCFC-141b). Rats were exposed by inhalation to HCFC-141b concentrations ranging from 1,000 to 10,000 ppm. Uptake studies of HCFC-141b in the rat indicated the involvement of saturable and first-order components. The chlorofluorocarbon substitute 1,1-dichloro-2,2,2-trifluoroethane (HCFC-123) is a structural analog of halothane. Both are oxidatively metabolized by CYP2EI, producing a reactive trifluoroacyl acid chloride intermediate and have been shown to cause acute liver necrosis in the guinea pig. With halothane, liver injury has been associated with the degree of reactive intermediate binding to hepatic protein. This injury can be potentiated by prior glutathione (GSH) depletion. Thus, the combination of GSH depletion and HCFC-123 exposure was evaluated for its hepatotoxic potential in this species.

The interaction of Freon 113 (1,1,2-trichloro-1,2,2-trifluoroethane) and hypoxia on the heart conduction system was studied using electrocardiogram monitoring of isolated perfused rat hearts. Freon 113 (0.2 mM) alone elicited significant atrioventricular conduction delay (p less than 0.05) and heart rate decrease (p less than 0.01), which were significantly enhanced by hypoxia (75% oxygen decrease), for instance, resulting in 2:1 AV block. The data suggest that arrhythmogenicity of Freon 113 on the heart conduction system may be enhanced synergistically by hypoxia.

We use recent aircraft measurements of a comprehensive suite of anthropogenic halocarbons, carbon monoxide (CO), and related tracers to place new constraints on North American halocarbon emissions and quantify their global warming potential. Using a chemical transport model (GEOS-Chem) we find that the ensemble of observations are consistent with our prior best estimate of the U.S. anthropogenic CO source, but suggest a 30% underestimate of Mexican emissions. Two cases of atrial fibrillation and one case of sudden death occurred in workers exposed to trifluorotrichloroethane (CFC 113) as a solvent/degreasing agent. This agent and related halogenated hydrocarbons are widely used in industry as solvents and degreasing agents, and have been previously linked to ventricular arrhythmias and cardiac sudden death when inhaled in excessive concentrations. We suggest that occupational overexposure to halogenated hydrocarbons should be considered a potential precipitant for atrial as well as ventricular arrhythmias.
