Ion Homeostasis in Chloroplasts under Salinity and Mineral Deficiency: II. Solute Distribution between Chloroplasts and Extrachloroplastic Space under Excess or Deficiency of Sulfate, Phosphate, or Magnesium.

Spinach (Spinacia oleracea var "Yates") plants grown hydroponically were exposed to an excess or deficiency of various mineral ions. Solutes were measured in leaf extracts and in isolated intact chloroplasts. Under phosphate (120 millimoles per liter NaH(2) PO(4)), sulfate (200 millimolar per liter (Na(2) SO(4)), or magnesium excess (150 millimolar per liter MgCl(2)), concentrations of these ions in leaf extracts increased, but in chloroplasts, concentrations of all ions remained constant. Concentrations of quarternary ammonium compounds in chloroplasts increased. Under mild phosphate or magne

The chloroplast-localised metalloprotease EGY1 and Mg-dechelatase SGR1 control chloroplast development and senescence by modulating photosynthetic functions.

The chloroplast plays an essential role in the production of biochemical energy and metabolism, as well as in plant development, including senescence. We describe a mutant with unusual progression of senescence and identify it as a new allele of ethylene-dependent gravitropism-deficient and yellow-green 1 (egy1-5). EGY1 encodes a chloroplast-localized metalloprotease that regulates chlorophyll metabolism. The single egy1-5 mutant exhibits various defects in chloroplast function, including abnormal accumulation of photosynthetic complexes and defects in photosynthetic parameters, as well as alt

Chloroplast Genome Sequence of Snap Bean (Phaseolus vulgaris L.) and Comparative Analyses with Other Legume Chloroplast Genomes.

Common bean (Phaseolus vulgaris L.) is an economically important grain legume for the human diet and sustainable agricultural development. Snap bean, a vegetable-type common bean cultivated for edible immature pods, serves as a crucial dietary source of essential vitamins, minerals and proteins. However, its evolutionary and domestication history remains poorly characterized. Here, we assembled and characterized the complete chloroplast genome of snap bean using PacBio HiFi sequencing technology. The chloroplast genome formed a single circular molecule of 150,248 bp, consisting of a pair of in

Chloroplast DNA deletions associated with wheat plants regenerated from pollen: possible basis for maternal inheritance of chloroplasts.

Albinism in plants regenerated from pollen by anther culture may result from the same mechanisms as postulated to be responsible for maternal inheritance of chloroplasts. Consistent with this view, Southern blotting was used to show that large regions of the chloroplast genome (over 80% in one case) were deleted in the major ctDNA molecules present in eight albino wheat plants regenerated from separate pollen calluses. A wide variety of deleted ctDNA molecules was present in these plants. Most albino plants appeared to contain a heterogeneous population of ctDNA molecules. Albino plant 6 conta

Photoreduction of sulfur dioxide by spinach leaves and isolated spinach chloroplasts.

Labeled sulfur dioxide was found to be extensively absorbed by spinach (Spinacea oleracea L.) leaves. Labeled sulfides detected in leaf blades following fumigations with sulfur dioxide in light indicated that photoreduction of sulfur dioxide had occurred. Measurable proportions of this labeled sulfur was localized within the chloroplast fraction. Suspensions of isolated chloroplasts supplied with labeled sulfur dioxide contained labeled sulfides following a 30-minute illumination period in water-cooled reaction vessels. With reference to recent studies of the chloroplast sulfur reduction pathw

Cytosine deaminase as a negative selectable marker for the microalgal chloroplast: a strategy for the isolation of nuclear mutations that affect chloroplast gene expression.

Negative selectable markers are useful tools for forward-genetic screens aimed at identifying trans-acting factors that are required for expression of specific genes. Transgenic lines harbouring the marker fused to a gene element, such as a promoter, may be mutagenized to isolate loss-of-function mutants able to survive under selection. Such a strategy allows the molecular dissection of factors that are essential for expression of the gene. Expression of individual chloroplast genes in plants and algae typically requires one or more nuclear-encoded factors that act at the post-transcriptional

Altered cytokinin metabolism affects cytokinin, auxin, and abscisic acid contents in leaves and chloroplasts, and chloroplast ultrastructure in transgenic tobacco.

Cytokinins (CKs) are involved in the regulation of plant development including plastid differentiation and function. Partial location of CK biosynthetic pathways in plastids suggests the importance of CKs for chloroplast development. The impact of genetically modified CK metabolism on endogenous CK, indole-3-acetic acid, and abscisic acid contents in leaves and isolated intact chloroplasts of Nicotiana tabacum was determined by liquid chromatography/mass spectrometry and two-dimensional high-performance liquid chromatography, and alterations in chloroplast ultrastructure by electron microscopy

The complete chloroplast genome of Artocarpus gongshanensis (Moraceae).

The chloroplast genome of Artocarpus gongshanensis S. K. Wu ex C. Y. Wu & S. S. Chang 1989, a threatened species endemic to China, was sequenced for the first time using Illumina NovaSeq 6000. The complete chloroplast genome assembly reveals a quadripartite structure with a total length of 160,174 bp, consisting of a large single-copy (LSC) region (89,311 bp) and a small single-copy (SSC) region (19,489 bp), separated by two inverted repeat (IR) regions (25,687 bp each). Phylogenetic reconstruction based on chloroplast genome data strongly supports that A. gongshanensis forms a sister clade wi

Erythromycin resistance and the chloroplast ribosome in Chlamydomonas reinhardi.

Five classes of erythromycin-resistant mutants of Chlamydomonas reinhardi have been identified. Each class corresponds to a different genetic locus, three nuclear and two chloroplast. The three nuclear loci appear to be unlinked, while Conde et al. (1975) have shown that the two chloroplast loci are linked, but not allelic. Mutants in each class have a unique pattern of cross-resistance in vivo to other antibiotics (lincomycin, carbomycin, and cleocin) that affect chloroplast protein synthesis. The chloroplast ribosomes from each class have a distinctive erythromycin-binding reaction in vitro.

Functional Nitrogenase Cofactor Maturase NifB in Mitochondria and Chloroplasts of Nicotiana benthamiana.

Engineering plants to synthesize nitrogenase and assimilate atmospheric N2 will reduce crop dependency on industrial N fertilizers. This technology can be achieved by expressing prokaryotic nitrogen fixation gene products for the assembly of a functional nitrogenase in plants. NifB is a critical nitrogenase component since it catalyzes the first committed step in the biosynthesis of all types of nitrogenase active-site cofactors. Here, we used a library of 30 distinct nifB sequences originating from different phyla and ecological niches to restore diazotrophic growth of an Azotobacter vineland

Moss Chloroplasts Are Surrounded by a Peptidoglycan Wall Containing D-Amino Acids.

It is believed that the plastids in green plants lost peptidoglycan (i.e., a bacterial cell wall-containing d-amino acids) during their evolution from an endosymbiotic cyanobacterium. Although wall-like structures could not be detected in the plastids of green plants, the moss Physcomitrella patens has the genes required to generate peptidoglycan (Mur genes), and knocking out these genes causes defects in chloroplast division. Here, we generated P patens knockout lines (∆Pp-ddl) for a homolog of the bacterial peptidoglycan-synthetic gene encoding d-Ala:d-Ala ligase. ∆Pp-ddl had a macrochloropl

Light Mediated Changes in the Chloroplasts of the Liverwort, Sphaerocarpos donnellii Aust.

Dark-grown plants of Sphaerocarpos, incubated in a liquid medium containing sucrose and mineral salts, have a much lower chlorophyll and nitrogen content than do light-grown plants. Two minutes of red light per 12 hours is about two-thirds as effective in increasing chlorophyll and nitrogen content as is continuous white light. These red light-induced increases are mediated by phytochrome, as they are reversible by alternating exposures to red and far-red light. They appear to be related to differences in the ultrastructure of the chloroplasts. Plastids from dark-grown plants are full of starc

The complete chloroplast genome of Heterostemma pingtaoi (Apocynaceae).

Heterostemma pingtaoi, a species endemic to Hainan Island, China, was discovered and formally described in the family Apocynaceae in 2010. However, genetic and evolutionary information on the genus Heterostemma remains poorly understood. Here, we report the complete chloroplast (cp) genome sequence of H. pingtaoi. The genome displays the typical quadripartite structure characteristic of dicotyledonous plant cp genomes, with a total length of 162,681 bp and a GC content of 38.25%. The cp genome encodes 131 genes, including 84 protein-coding genes, 8 rRNA genes, and 39 tRNA genes. Phylogenetic a

Copper Delivery to Chloroplast Proteins and its Regulation.

Copper is required for photosynthesis in chloroplasts of plants because it is a cofactor of plastocyanin, an essential electron carrier in the thylakoid lumen. Other chloroplast copper proteins are copper/zinc superoxide dismutase and polyphenol oxidase, but these proteins seem to be dispensable under conditions of low copper supply when transcripts for these proteins undergo microRNA-mediated down regulation. Two ATP-driven copper transporters function in tandem to deliver copper to chloroplast compartments. This review seeks to summarize the mechanisms of copper delivery to chloroplast prote

Cd2+ transport and storage in the chloroplast of Euglena gracilis.

Euglena gracilis lacks a plant-like vacuole and, when grown in Cd2+-containing medium, 60% of the accumulated Cd2+ is located inside the chloroplast. Hence, the biochemical mechanisms involved in Cd2+ accumulation in chloroplast were examined. Percoll-purified chloroplasts showed a temperature-sensitive uptake of the free 109Cd2+ ion. Kinetics of the uptake initial rate was resolved in two components, one hyperbolic and saturable (Vmax 11 nmol 109Cd2+ min(-1) mg protein (-1), Km 13 microM) and the other, linear and non-saturable. 109Cd2+ uptake was not affected by metabolic inhibitors or illum
