Dietary cholesterol-induced enhancement of hepatic biotransformation rate in male rats.

Male rats were fed a cholesterol-free diet for 5 weeks, followed by a 2% cholesterol diet for 4 weeks. Another group of rats was continuously fed a cholesterol-free diet. A third group was fed standard pelllets during the whole experiment. Hepatic microsomal protein and cholesterol contents and drug-metabolizing enzyme activities were measured. The cholesterol-rich diet increased microsomal protein content and this increase disappeared after trypsin digestion of microsomal membranes. Microsomal cholesterol content was enhanced three-fold by cholesterol feeding. Cytochrome P-450 concentration,

The Impact of Dietary Cholesterol on Low-Density Lipoprotein: Lessons in Absorption and Overconsumption.

This case describes a 58-year-old woman with past medical history of ulcerative colitis, hyperlipidemia, and radiological evidence of atherosclerosis without prior cardiovascular disease who presented for management of hyperlipidemia. At baseline, her lipid panel in 2015 noted a calculated low-density lipoprotein (LDL-C) of 125 mg/dL (3.2 mmol/L). Over the course of the next 5 years, she developed severe LDL elevations to >400 mg/dL (>10.3 mmol/L) following the addition of 1600 mg dietary cholesterol daily achieved through 9 eggs. Following cessation of this intake she had dramatic improvement

Changes in apolipoprotein E expression in response to dietary and pharmacological modulation of cholesterol.

Apolipoprotein E (ApoE) influences the risk of late onset Alzheimer's disease (AD) in an isoform-dependent manner, such that the presence of the apoE epsilon4 allele increases the risk of AD while the presence of the apoE epsilon2 allele appears to be protective. Although a number of ApoE functions are isoform dependent and may underlie the "risk factor" activity of AD, its ability to bind amyloid beta peptides and influence their clearance and/or deposition has gained strong experimental support. Evidence suggests that in addition to genotype, increased ApoE transcription can contribute to AD

Effect of dietary cholesterol oxidation products on the plasma clearance of chylomicrons in the rat.

Oxidized cholesterols in the diet have been shown to exacerbate arterial cholesterol deposition and the development of atherosclerosis in animal models. Dietary oxidized cholesterols are absorbed through the intestine and incorporated into lymph chylomicrons. The aim of this study was to investigate the effect of oxidized cholesterols on the metabolism of nascent chylomicrons in vivo. It was shown that oxidized cholesterols markedly delay the clearance of chylomicrons from plasma compared to rats given TG alone. However, there was no difference in the clearance of chylomicrons containing oxidi

Effects of dietary phytosterols on cholesterol metabolism and atherosclerosis: clinical and experimental evidence.

Although plant sterols (phytosterols) and cholesterol have similar chemical structures, they differ markedly in their synthesis, intestinal absorption, and metabolic fate. Phytosterols inhibit intestinal cholesterol absorption, thereby lowering plasma total and low-density lipoprotein (LDL) cholesterol levels. In 16 recently published human studies that used phytosterols to reduce plasma cholesterol levels in a total of 590 subjects, phytosterol therapy was accompanied by an average 10% reduction in total cholesterol and 13% reduction in LDL cholesterol levels. Phytosterols may also affect oth

Effects of Dietary Cholesterol Regulation on Spermatogenesis of Gobiocypris rarus Rare Minnow.

Cholesterol is an important component of cell membranes, and also a precursor for the synthesis of sex hormones, playing an important role in reproduction. However, few studies have focused on cholesterol and reproductive health. To investigate the toxic effects of different cholesterol levels on the spermatogenesis of rare minnows, we regulate the cholesterol content in fish by feeding them a high-cholesterol diet and cholesterol inhibitor pravastatin, and cholesterol levels, sex hormone (T and 11KT) levels, testis histology, sperm morphology and function, and the expression of genes related

The effects of dietary cholesterol on metabolism and daily torpor patterns in Siberian hamsters.

The concentrations of fatty acids in the diet influence torpor in numerous species of mammals. Much less is known, however, about the potential effects of other types of dietary lipids. One study demonstrated that increasing dietary cholesterol levels during fall feeding increased torpor bout length and also decreased minimum body temperatures during hibernation by chipmunks. Another hibernation study with ground squirrels revealed that the cholesterol contents of both the cerebral cortex and the microsomes were significantly greater during arousal episodes than during torpor bouts, suggesting

The effects of dietary cholesterol-lowering on psychological symptoms: a randomised controlled study.

The relationship of plasma cholesterol-reducing interventions to emotional states, such as depression and hostility, remains a topic of debate. The present study employed a randomised, controlled design, and was conducted at a clinical research center to test the effect of dietary cholesterol-lowering on psychological symptoms. Ten women and eight men were randomly assigned to one of two counterbalanced diet cycles (low-fat versus high-fat diet; isocaloric; 6 weeks each; separated by a washout period). Analyses for repeated measures revealed that the low-fat diet significantly reduced total, L

Dietary cholesterol enhances pulmonary eosinophilic inflammation in a murine model of asthma.

BACKGROUND: Epidemiological studies have suggested that the dietary pattern may be associated with the prevalence of asthma. We previously reported that an increased intake of foods of animal origin was associated with the occurrence of allergic rhinitis and asthma in adolescents. Here we examined the effect of dietary cholesterol in a murine model of allergic pulmonary inflammation. METHODS: Weanling C57BL/6 mice were fed a control diet containing 0.02% cholesterol or a diet supplemented with 1% or 2% cholesterol. Four weeks later the mice were sensitized with intraperitoneal ovalbumin (OVA)

Randomized trial of the effects of cholesterol-lowering dietary treatment on psychological function.

PURPOSE: Epidemiological studies have suggested that cholesterol lowering could affect psychological functioning. This study was designed to test whether cholesterol-lowering diets adversely affect mood and cognitive function.5.2 mM [198 mg/dL]) to either a low-fat diet, a Mediterranean diet, or a waiting-list control. Cholesterol levels, psychological well-being (depression, anxiety, hostility), and cognitive function were assessed at baseline, 6 weeks, and 12 weeks. RESULTS: Total serum cholesterol levels fell significantly more in the intervention groups (8.2% reduction) than in the contro

Effect of dietary chromium on glucose tolerance and serum cholesterol in guinea pigs.

The effects of feeding three levels of dietary chromium (Cr) to non-pregnant guinea pigs, guinea pigs during pregnancy and lactation and F-1 offspring guinea pigs on weight gain, glucose tolerance, glucose peak time value and serum cholesterol concentration has been investigated. Dietary levels of Cr were: Basal Diet (B) = 0.125 ppm; B supplemented with 0.5 ppm Cr (S1 Diet) and B supplemented with 50 ppm Cr (S2 Diet). All groups had similar weight gain patterns and daily feed intake levels. Parent generation guinea pigs fed the B diet consumed less than 10 mug Cr/kg body weight/day while F-U g

Molecular mechanisms of chronic dietary cholesterol-driven remodeling of LDLR protein fate.

The hepatic low-density lipoprotein receptor (LDLR) mediates the clearance of circulating low-density lipoprotein (LDL) and is a key determinant of cholesterol homeostasis. However, how nutritional cues such as chronic dietary cholesterol regulate LDLR intracellular trafficking and protein turnover through specific molecular switches remains poorly understood. Dr. Alan Saltiel's team at the University of California San Diego discovered a cholesterol stress-response pathway mediated by Ral GTPases, revealing a new mechanism by which dietary cholesterol reshapes LDLR protein fate and thereby aff

Ezetimibe Normalizes Dietary Cholesterol-Induced Exacerbation of Liver Injury in Alcohol-Fed Mice.

Interactions between alcohol and nutrition play an important role in the development and progression of alcohol-associated liver disease (ALD). Although dietary cholesterol was shown to exacerbate fatty liver and liver injury in alcohol-fed mice, findings regarding the combined effect of dietary cholesterol and heavy alcohol drinking on cholesterol homeostasis remain controversial. Ezetimibe has been widely used as a cholesterol-lowering drug in hypercholesterolemic subjects. It is not fully understood whether ezetimibe blunts the adverse effect of cholesterol on lipid and biliary bile acid me

High dietary fat and cholesterol exacerbates chronic vitamin C deficiency in guinea pigs.

Vitamin C deficiency - or hypovitaminosis C defined as a plasma concentration below 23 μm - is estimated to affect hundreds of millions of people in the Western world, in particular subpopulations of low socio-economic status that tend to eat diets of poor nutritional value. Recent studies by us have shown that vitamin C deficiency may result in impaired brain development. Thus, the aim of the present study was to investigate if a poor diet high in fat and cholesterol affects the vitamin C status of guinea pigs kept on either sufficient or deficient levels of dietary ascorbate (Asc) for up to

Interaction between TCF7L2 polymorphism and dietary fat intake on high density lipoprotein cholesterol.

Recent evidence suggests that lifestyle factors influence the association between the Melanocortin 4 receptor (MC4R) and Transcription Factor 7-Like 2 (TCF7L2) gene variants and cardio-metabolic traits in several populations; however, the available research is limited among the Asian Indian population. Hence, the present study examined whether the association between the MC4R single nucleotide polymorphism (SNP) (rs17782313) and two SNPs of the TCF7L2 gene (rs12255372 and rs7903146) and cardio-metabolic traits is modified by dietary factors and physical activity. This cross sectional study inc
