Trk neurotrophin receptors in cholinergic neurons of patients with Alzheimer's disease.

Besides cortical pathology, Alzheimer's disease (AD) is characterized by a loss of cholinergic neurons in the basal forebrain but not in the caudate nucleus, putamen or mesencephalon. Since cholinergic neurons which degenerate in AD are sensitive to nerve growth factor (NGF), a link between NGF sensitivity and the vulnerability of cholinergic neurons has been suspected. Levels of NGF are not altered in patients with AD, however. Thus, cholinergic nerve cell death in AD could not result from a deficiency in NGF receptors. Using sequential immunohistochemistry with antibodies that recognize pref

Distribution of GABAergic and cholinergic neurons in the rat diagonal band.

GABAergic neurons are coextensive with cholinergic neurons in the medial septum-diagonal band complex. Serial sectioning, sequential staining and double immunofluorescence techniques employing antibodies to glutamate decarboxylase and choline acetyltransferase revealed the distribution of these transmitter-specific neurons in the rat. Morphologically, the two types of neurons appear similar, in that they are predominantly large multipolar cells, but they are characterized by different, overlapping distributions in the diagonal band. Glutamate decarboxylase-positive cells are scattered througho

Cholinergic modulation of neuronal responses to cholecystokinin in anesthetized rats.

The aim of this study was to investigate whether the effects of the neuropeptide cholecystokinin on neuronal firing can be changed by acetylcholine in various structures of the brain. Single unit activity was extracellularly recorded in rats anesthetized with urethane. The neurons were located in several nuclei of the thalamus, the basal ganglia and the cerebral cortex. Neurons responding to the sulfated octapeptide of cholecystokinin (CCK-8S) were mainly activated by the drug [Wilcoxon test (Wt) p < 0.0001, n=113]. Thalamic neurons could also increase the number of burst discharges (Wt p < 0.

Cholinergic responsiveness of neurons in the ventroposterior thalamus of the anesthetized rat.

Acetylcholine has been implicated as an important neurotransmitter in the mechanisms of thalamic activation. Cholinergic mechanisms are thought to directly underlie the high level of excitability observed in thalamic relay neurons during waking and rapid eye movement sleep. We sought to determine if the cholinergic responsiveness of neurons in the ventroposterior nuclei of the thalamus in rat is consistent with this view. Neurons in the chloral hydrate-anesthetized rat were studied with extracellular recording and microiontophoretic application of cholinergic agents. In most cases (63% of 63 c

The non-neuronal cholinergic system in the heart: A comprehensive review.

The autonomic influences on the heart have a ying-yang nature, albeit oversimplified, the interplay between the sympathetic and parasympathetic system (known as the cholinergic system) is often complex and remain poorly understood. Recently, the heart has been recognized to consist of neuronal and non-neuronal cholinergic system (NNCS). The existence of cardiac NNCS has been confirmed by the presence of cholinergic markers in the cardiomyocytes, which are crucial for synthesis (choline acetyltransferase, ChAT), storage (vesicular acetylcholine transporter, VAChT), reuptake of choline for synth

Non-neuronal cholinergic activity is potentiated in myasthenia gravis.

BACKGROUND: Non-neuronal acetylcholine (ACh) restricts autoimmune responses and attenuates inflammation by cholinergic anti-inflammation pathway. To date, the implication of ACh in myasthenia gravis (MG) remained unexplored. This study aimed to investigate the possible relationship between ACh levels, anti-muscle-specific tyrosine kinase (MuSK) antibody titers, main clinical features and outcomes of MG patients. METHODS: We successfully measured ACh levels in human peripheral blood mononuclear cells (PBMCs) from 125 MG patients and 50 matched healthy controls by using ultra-performance liquid

[The modulation of septal influences on the hippocampal neurons by cholinergic substances].

To evaluate the functional significance of the cholinergic component of the septo-hippocampal input the effects of electrical stimulation of the medial septal area (MS--DB) on the hippocampal neurons were studied in chronic waking rabbits. Initial inhibitory effects 40-140 msec long were observed in 54% of neurons in intact rabbits. Other neurons usually responded by a diffuse excitation. Initial inhibitory effects absolutely dominated (90%) in the animals with the septum basally undercut. Increase of the endogenous acetylcholine level by physostigmine completely or partially blocked the effec

The distribution of cholinergic neurons in the central nervous system of turtles.

The distribution of cholinergic neurons was studied in painted and red-eared turtles using antisera against choline acetyltransferase (ChAT). The results showed that the organization of cholinergic systems in the central nervous system of turtles was remarkably similar to that previously described in diverse other species. For example, the present studies revealed the presence of cholinergic neurons in the striatum, the basal telencephalon, the isthmo-tectal system, and a region of cells near the cerebellar peduncle. The motoneurons of the III, IV, V, VI, VII, X and XII cranial nerves and the

[Topography of cholinergic neurons in the brain stem of the human fetus].

Cholinacetyltransferase (ChAT) activity has been studied in 56 nuclei of the cerebral trunk in human fetuses at the age of 6-8 lunar months. Cytoplasmic and synaptic ChAT activity has been revealed and three types of neurons for cholinergic synaptic transmission has been distinguished. There are only cholinergic-noncholinoceptive neurons in five macrocellular nuclei of the cranial nerves. In 25 nuclei (paravicellular, reticular, pigmented, sensitive nuclei of the cranial nerves, nuclei of the funiculi posterior and some other switching centres) there are only noncholinergic-cholinoceptive neur

[Hypogenic cholinergic neuronal route in the central nervous system].

Because of the extensive overlapping within the brain of arousing and hypnogenic neurons the method of electrical stimulation has proved unsuitable for mapping out specific hypnogenic structures. On the other hand, the method of localized chemical stimulation achieved by introducing minute crystals of chemical substances permits the dissection of adjacent neurons with different functions. The aim of this study was to map out the central hypnogenic pathways introducing minute crystals of acetylcholine, eserine or atropine in different brain regions through cannulae permanently implanted in the

Effects of grafts containing cholinergic and/or serotonergic neurons on cholinergic, serotonergic and noradrenergic markers in the denervated rat hippocampus.

Long-Evans female rats sustained aspirative lesions of the septohippocampal pathways and, 2 weeks later, received intrahippocampal suspension grafts prepared from the regions including either the medial septum and the diagonal band of Broca (group S), or the mesencephalic raphe (group R), or from both these regions together (group S + R). Sham-operated (group SHAM) and lesion-only (group LES) rats were used as controls. Six months after grafting, high affinity synaptosomal uptake of choline (HACU) and serotonin (HASU), choline acetyltransferase (ChAT) activity and, using HPLC, the content of s

Cholinergic depolarization recruits a persistent Ca2+ current in Aplysia bag cell neurons.

Many behaviors and types of information storage are mediated by lengthy changes in neuronal activity. In bag cell neurons of the hermaphroditic sea snail Aplysia californica, a transient cholinergic synaptic input triggers an ∼30-min afterdischarge. This causes these neuroendocrine cells to release egg laying hormone and elicit reproductive behavior. When acetylcholine is pressure-ejected onto a current-clamped bag cell neuron, the evoked depolarization is far longer than the current evoked by acetylcholine under voltage clamp, suggesting recruitment of another conductance. Our earlier studies

Lesion of cholinergic neurons in nucleus basalis enhances response to general anesthetics.

Acetylcholine in the brain has been associated with consciousness and general anesthesia effects. We tested the hypothesis that the integrity of the nucleus basalis magnocellularis (NBM) affects the response to general anesthetics. Cholinergic neurons in NBM were selectively lesioned by bilateral infusion of 192IgG-saporin in adult, male Long-Evans rats, and control rats were infused with saline. Depletion of choline-acetyltransferase (ChAT)-immunoreactive cells in the NBM and decrease in optical density of acetylcholinesterase (AChE) staining in the frontal and visual cortices confirmed a sig

Mechanisms of cell death in cholinergic basal forebrain neurons in chronic alcoholics.

Tau immunoreactivity was examined in post mortem tissue from patients in three groups: neurologically-asymptomatic and neuropathologically normal alcoholics, alcoholics with Wernicke's Encephalopathy (WE) and age matched non-alcoholic controls. Tau-positive granular and fibrillary inclusions were frequently observed within the magnocellular neurons of the cholinergic nucleus basalis, within occasional nucleus basalis neurons in non-WE alcoholics, but not in controls. Tau immunoreactivity was not however observed in cortical, brainstem, diencephalic or non-cholinergic forebrain structures. Pero

Dopamine 2 receptor ablation from cholinergic neurons attenuates L-DOPA induced dyskinesias.

INTRODUCTION: L-DOPA-induced dyskinesia (LID) formation requires prominent dopamine oscillations over hours, across years of treatment. Striatal cholinergic interneurons (CIN) have been implicated in both the facilitation and attenuation of LID, but how CIN sense and respond to striatal dopamine levels in this context remains incompletely understood. CIN express both the inhibitory, Gαi-coupled dopamine D2 receptor and the facilitatory, Gαs-coupled dopamine D5 receptor. Although systemic ablation of D5 exacerbates LID, the contribution of CIN-specific D2 expression to LID has not been studied.

Methyl-CpG binding-protein 2 function in cholinergic neurons mediates cardiac arrhythmogenesis.

Sudden unexpected death occurs in one quarter of deaths in Rett Syndrome (RTT), a neurodevelopmental disorder caused by mutations in Methyl-CpG-binding protein 2 (MECP2). People with RTT show a variety of autonomic nervous system (ANS) abnormalities and mouse models show similar problems including QTc interval prolongation and hypothermia. To explore the role of cardiac problems in sudden death in RTT, we characterized cardiac rhythm in mice lacking Mecp2 function. Male and female mutant mice exhibited spontaneous cardiac rhythm abnormalities including bradycardic events, sinus pauses, atriove

Role of tegmental cholinergic neurons in dopaminergic activation, antimuscarinic psychosis and schizophrenia.

Cholinergic neurons of the pedunculopontine nucleus (Ch5) and laterodorsal tegmental nucleus (Ch6) monosynaptically activate dopamine neurons of the substantia nigra, zona compacta (A9), and ventral tegmental area (A10) via muscarinic and nicotinic receptors. Ch5 cells and Ch6 cells are inhibited by local injections of muscarinic agonists, suggesting the presence of autoreceptors. This review advances the hypothesis that the psychotogenic effects of antimuscarinics are triggered by disinhibition of Ch5 and Ch6 cells via their autoreceptors, and that these effects are distinct from the memory-b

Evidence for cholinergic synapses between dissociated rat sympathetic neurons in cell culture.

Sympathetic principal neurons were dissociated from superior cervical ganglia of new-born rats, and grown in cell culture. In electrophysiological experiments two types of excitatory synapses were found. One, which was relatively rare, was shown to operate by electrical transmission. The other, the predominant type, had several characteristics of chemical transmission, and pharmacological evidence indicated it was cholinergic.

Transmitter and ion channel profiles of saccadic omnipause neurons and cholinergic non-omnipause neurons in human nucleus raphe interpositus.

BACKGROUND: Omnipause neurons (OPN) are glycinergic neurons that tonically inhibit burst neurons between saccades. In primates, OPNs are located bilaterally around the midline at the level of the traversing rootlets of the abducens nerve in the pontine brainstem forming the nucleus raphe interpositus (RIP). Healthy OPNs are previously characterized by dense perineuronal net (PN) ensheathment, parvalbumin (PAV) and voltage-gated potassium channel Kv1.1 and Kv3.1 expression. MOTIVATION: The ion channel and transmitter profile of OPNs in human has not been established. The further characterizati
