Determination of triptolide in root extracts of Tripterygium wilfordii by solid-phase extraction and reverse-phase high-performance liquid chromatography.

Extracts of Tripterygium wilfordii roots have a long history of use in traditional Chinese medicine and have shown great promise in recent clinical trials as a treatment for rheumatoid arthritis. The major active component of Tripterygium root extracts is the diterpenoid triptolide. This paper describes a method for the determination of triptolide in root extracts that is suitable for the analysis of many small samples simultaneously. Extracts are applied to aminopropyl solid-phase extraction (SPE) tubes that are then eluted with dichloromethane-methanol (49:1, v/v). The eluate is chromatograp

Neurohypophyseal hormone-like peptides in the ovine pineal gland using reverse-phase liquid chromatography and radioimmunoassay.

A method is described for the determination of the neurohormone contents of ovine pineal tissue by radioimmunoassay (RIA) after successive fractionation on gel filtration in formic acid and reverse-phase liquid chromatography (HPLC). This method gives a good resolution for the neurohormones vasopressin, vasotocin and oxytocin, without a significant interference of aspecific cross-reacting of peptides with the RIA. An acid extract from ovine pineal tissue was found to contain amounts of immunoreactive AVP- and OXT-like peptides, whereas an AVT-like peptide was not detectable over background lev

Mobile-Phase Contributions to Analyte Retention and Selectivity in Reversed-Phase Liquid Chromatography: 2. Solute-Specific Effects.

Analyte retention in reversed-phase liquid chromatography decreases with increasing solute polarity of a compound, whereby the parameters of the water-organic solvent (W-OS) mobile phase can influence the retention order of the compounds (selectivity). Through molecular dynamics simulations in a slit-pore model of a silica-based, endcapped, C18 stationary phase equilibrated with W-methanol and W-acetonitrile mobile phases, we investigate how the system discriminates between small, neutral compounds with low to moderate solute polarity at the molecular level. The experimental retention behavior

Imidazolium embedded C8 based stationary phase for simultaneous reversed-phase/hydrophilic interaction mixed-mode chromatography.

A new imidazolium embedded C8 based stationary phase (SIL-MPS-VOL) was facilely prepared by two steps and characterized by Fourier transform infrared spectrometry and thermogravimetric analysis. Due to the introduction of quaternary imidazolium group to the traditional C8 stationary phase, the developed SIL-MPS-VOL column demonstrated both reversed-phase liquid chromatography (RPLC) and hydrophilic interaction liquid chromatography (HILIC) retention mechanisms. A series of hydrophobic and hydrophilic test samples, including benzene homologues, anilines, positional isomers, nucleosides and nucl

Phenyl-bonded stationary phases--the influence of polar functional groups on retention and selectivity in reversed-phase liquid chromatography.

The chromatographic properties of four phenyl-bonded phases with different structures were studied. The columns used were packed with a stationary phase containing a phenyl ring attached to the silica surface using different types of linkage molecules. As a basic characteristic of the bonded phases, the hydrophobicity and silanol activity (polarity) were investigated. The presence of the polar amino and amide groups in the structure of the bonded ligand strongly influences the polarity of the bonded phase. Columns were compared according to methylene selectivity using a series of benzene homol

Study of overload for basic compounds in reversed-phase high performance liquid chromatography as a function of mobile phase pH.

The retention and overloading properties for eight basic solutes and two quaternary ammonium compounds were studied over the pH range 2.7-10.0 using phosphate and carbonate buffers. At low pH, a hybrid inorganic-organic silica-ODS phase (XTerra RP-18, 15 cm x 0.46 cm) showed substantial loss in efficiency when sample masses exceeded about 0.5 microg; these results were similar to those obtained previously on pure silica ODS and wholly polymeric phases, suggesting a common overloading mechanism. At pH 7-8.5, substantial improvements in loading capacity were obtained on XTerra due apparently to

Dimethyl carbonate as a green mobile phase modifier for reversed phase liquid chromatography of polar aromatic and polyaromatic hydrocarbons.

Linear alkyl carbonate solvents such as dimethyl carbonate (DMC) are considered to be environmentally safe with a low toxicology risk. The modest viscosity of 0.625 mPa·s and the UV cutoff wavelength of about 215 nm makes DMC a potential alternative aprotic HPLC modifier solvent to acetonitrile. The main issue is the limited solubility of DMC in water, about 14 %. A polarity parameter (P') for DMC of 3.4 ± 0.1 was determined by ratioing Hildebrand solubility parameters for three alcohols and DMC to the corresponding alcohol P' values. A test mixture of polar aromatic compounds (benzene sulfona

Reversed phase liquid chromatography for the enantioseparation of local anaesthetics in polysaccharide-based stationary phases. Application to biodegradability studies.

A comprehensive study on the chiral separation of bupivacaine, mepivacaine, prilocaine and propanocaine with eight commercial polysaccharide-based chiral stationary phases (CSPs) in reversed phase conditions compatible with MS detection is performed. Methanol and acetonitrile are used as organic modifiers. Retention and resolution values obtained for each compound in the different CSPs and mobile phases are compared. The polysaccharide-based CSPs tested present different enantioselectivity towards the analytes. From the results, the experimental conditions for determining the enantiomers of bu

Loss of bonded phase in reversed-phase liquid chromatography in acidic eluents and practical ways to improve column stability.

Silica-based, reversed-phase liquid chromatographic (RPLC) stationary phases are very widely used to separate basic compounds in acidic eluents due to their high efficiency, good mechanical strength, and the versatile selectivity offered by different functional groups and the chemistry on the silica surface. However, the stability in acid of most silica-based stationary phases is poor, especially at elevated temperatures, due to hydrolysis of the siloxane bonds, which hold silanes on the silica substrate. This hydrolysis is commonly believed to be solely the result of catalysis by protons. How

Reversed-phase ion-pair partion chromatography of biogenic catecholamines and their alpha-methyl homologues with tributylphosphate as stationary phase.

Ion-pair partition chromatography is applied to the separation of the biogenic catecholamines and their alpha-methyl homologues. A useful selectivity has been obtained using an adduct-forming organic stationary phase (tributylphosphate). The retention of the compounds can be regulated easily by means of the concentration of the counter-ion (the perchlorate ion) in the mobile phase. The selectivity for separation of amines from amino acids can be influenced by changing the pH of the aqueous phase. The phase system shows a good long-term stability and reproducibility with respect to the capacity

Development of a green design reverse phase liquid chromatography method for determining the ionization profile of parabens.

The most popular method for determining chromatographic separation selectivity is an optimization method based on the use of different solvents. This article presents a sustainable chromatographic framework using reverse phase liquid chromatography (RPLC) to determine the ionization behavior of methylparaben, ethylparaben, propylparaben, and butylparaben, which are widely used in hydroorganic mixtures. In this study, retention-mobile phase pH data in methanol-water (40-55% v/v) and ethanol-water (30-45% v/v) mixtures were analyzed using a nonlinear regression (NLREG) approach to calculate the

Simultaneous Analysis of Cyproconazole and Tebuconazole in Suspension Concentrate Formulation by Reverse-Phase High-Performance Liquid Chromatography.

A novel and validated reverse-phase high-performance liquid chromatography (RP-HPLC) approach was established for the concurrent measurement of cyproconazole (CYP) isomers and tebuconazole (TBZ) in suspension concentrate (SC) agrochemical formulations. The approach employed a C18 column with a gradient elution of 0.1% formic acid in water and methanol, attaining baseline resolution of CYP isomer-1, isomer-2 and TBZ without requiring chiral columns or sample pretreatment. Linearity was confirmed within the range of 80-120% of the target concentration, with a R2 value of 0.999. Accuracy varied f

[Deep eutectic solvents modified reversed-phase liquid chromatography for the separation of natural products].

The composition of natural products is usually very complex. During the separation of natural products, high loading capacity and satisfactory chromatographic resolution are often hardly achieved. To address this limitation, various modifiers were added in the mobile phase to alter the chromatographic behavior. Here, deep eutectic solvents (DESs) were chosen to accomplish this objective on C18 column. Changes in retention factor, overloaded peak shape and adsorption capacity were systematically investigated and interpreted. The application potential of DESs in the separation of natural product

Investigation of the influence of magnetic fields on the separation of selected substances in reversed-phase column chromatography.

In this study, the effects of the induced magnetic field on the separation of drugs with different structures and polarities were investigated. Herein, column chromatography within an induced magnetic field was introduced to examine the influence of magnetic fields on materials and their interactions. For this purpose, the drugs were categorized into three series based on polarity: tramadol (logP 1.34) and heroin (logP 1.58) as the polar series; cocaine (logP 2.3) and noscapine (logP 2.58) as the moderate polarity series; and papaverine (logP 3.00) and ketamine (logP 3.21) as the non-polar ser
