White spots amidst the gold: ultrastructural and histological aspects of the chronic inflammatory response of goldfish with ichthyophthiriasis.

Ichthyophthirius multifiliis, the causative agent of white spot disease, is a ciliated protozoan parasite that infects freshwater fish and induces high mortality. Outbreaks occur both in natural and production sites. The aim of the present study was to describe the lesions caused by chronic infection by I. multifiliis in goldfish (Carassius auratus) from an ornamental fish farm, highlighting important ultrastructural aspects of this protozoan. Damaged skin and gills, collected from fish with white or ulcerative skin lesions, were routinely processed for histological analysis and transmission e

Characterization and functional analysis of a tandem-repeat galectin-9 in large yellow croaker Larimichthys crocea.

Galectins are a family of endogenous lectins with β-galactosides affinity, playing significant roles in the innate immunity of vertebrates and invertebrates. In this report, a new galectin-9 cDNA was identified and characterized in large yellow croaker Larimichthys crocea (designated as LcGal-9). The complete cDNA sequence of LcGal-9 was 1795 bp, with an open reading frame (ORF) of 1032 bp encoding 343 amino acids. The putative LcGal-9 protein contained two carbohydrate recognition domains (CRDs) connected by a linker peptide, with each carrying two conserved β-galactoside binding motifs H-NPR

Recombinant fusion subunit vaccine provides enhanced immune protection against Cryptocaryon irritans infection in grouper.

Cryptocaryon irritans causes severe losses in marine aquaculture, yet effective vaccines remain limited. In this study, we developed a novel fusion vaccine composed of an immobilization antigen (IA8) and the 26S proteasome, and evaluated its immunogenicity and protective efficacy in Epinephelus coioides. The recombinant fusion protein was successfully expressed in Escherichia. coli and exhibited strong immunoreactivity. Following parasite challenge, the fusion vaccine conferred the highest protection, reducing parasite abundance by 81.2%, compared with 44.6% and 22.6% for IA8 and 26S proteasom

Alternative Splicing Events and Differently Expressed Genes During Peak Mortality in Large Yellow Croaker (Larimichthys crocea) Infected with Scuticociliate.

Large yellow croaker (Larimichthys crocea) is facing various threats from bacterial, viral, and parasitic diseases, especially scuticociliate. Scuticociliate is a facultative parasite causing high mortality in various marine fishes. In this study, an artificial scuticociliate infection model was successfully established for large yellow croaker. Comparative transcriptome analysis was performed on gill tissues collected from control fish and fish at the peak of mortality following exposure to the parasite to investigate the underlying molecular mechanisms of host-parasite interactions. A total

Transcriptomic variation of locally-infected skin of Epinephelus coioides reveals the mucosal immune mechanism against Cryptocaryon irritans.

Fish skin is the largest immunologically active mucosal organ, providing first-line defense against external pathogens. However, the skin-associated immune mechanisms of fish are still unclear. Cryptocaryon irritans is an obligate ectoparasitic ciliated protozoan that infects almost all marine fish, and is believed to be an excellent pathogen model to study fish mucosal immunity. In this study, a de novo transcriptome assembly of Epinephelus coioides skin post C. irritans tail-infection was performed for the first time using the Illumina HiSeq™ 2500 system. Comparative analyses of infected ski

Stage-Specific Expression and Perinuclear Enrichment of γ-Tubulin During Tomont Development and Theront Morphogenesis in Cryptocaryon irritans.

Cryptocaryon irritans is a lethal parasitic ciliate of marine fishes whose tomont stage is marked by repeated nuclear division and the morphogenesis of infective theronts, both dependent on dynamic microtubule reorganization. To investigate cytoskeleton-associated regulation during this proliferative process, we cloned and characterized the γ-tubulin gene (Ci-TUBG) and γ-tubulin complex protein 3 gene (Ci-TUBGCP3) from C. irritans. Sequence analysis showed that Ci-TUBG contains a canonical dual tubulin domain architecture and is highly conserved relative to homologs from Ichthyophthirius multi

Screening of immunogenic antigens from scuticociliate in Takifugu rubripes, and the development and evaluation of subunit vaccines.

In recent years, scuticociliatosis has emerged as a major parasitic disease in the aquaculture of Takifugu rubripes, inflicting significant economic losses on the pufferfish farming industry. The conventional approach of using chemical drugs to prevent and control parasitic diseases poses substantial risks to the environment and food safety. Consequently, the utilization of vaccines for the prevention and treatment of aquatic animal diseases is currently regarded as the mainstream direction for future aquatic animal disease prevention and control. In this study, utilizing the laboratory-establ

Characterization and expression analysis of six interleukin-17 receptor genes in grouper (Epinephelus coioides) after Cryptocaryon irritans infection.

Interleukin-17 receptors (IL17Rs) mediate the activation of several downstream signal pathways to induce inflammatory response and contribute to the pathology of many autoimmune diseases. In this study, six IL17Rs (IL17RA1, RA2, RB, RC, RD and RE) were cloned and characterized from Epinephelus coioides, an orange-spotted grouper. Multiple sequence alignment and structural analysis revealed that all members of IL17Rs were low in sequence identity with each other. But their structures were conservative in grouper, which contain signal peptide, extracellular FNIII domain (IL17RA1/RA2/RB) or IL-17

Effects of a Synthetic Isoquinoline Derivative Against Ichthyophthirius multifiliis In Vivo and In Vitro in Grass Carp (Ctenopharyngodon idella).

White spot disease, caused by the parasitic Ichthyophthirius multifiliis, induces mortality exceeding 80% in intensive aquaculture systems, resulting in global annual losses of >$1 billion. Current chemical controls (e.g., formaldehyde) face environmental persistence and drug resistance challenges. This study developed a synthetic isoquinoline derivative, BHTCA (s)-2-tert-butoxycarbonyl-7-hydroxy-1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid), as a biodegradable alternative against I. multifiliis in grass carp (Ctenopharyngodon idella). Removal of two pathogenic scuticociliates Miamiensis avidus and Miamiensis sp. using cells or culture filtrates of the dinoflagellate Alexandrium andersonii.

Scuticociliatosis, which is caused by parasitic protistan pathogens known as scuticociliates, is one of the most serious diseases in marine aquaculture worldwide. Thus, elimination of these ciliates is a primary concern for scientists and managers in the aquaculture industry. To date, formalin and other toxic chemicals have been used as anti-scuticociliate agents, but issues regarding their secondary effects often arise. Consequently, development of safer methods is necessary. To find out a safe method of controlling scuticociliate populations in aqua-tanks or small-scale natural environments,

Experimental Scuticociliatosis in Japanese flounder (Paralichthys olivaceus) infected with Miamiensis avidus: pathological study on the possible neural routes of invasion and dissemination of the scuticociliate inside the fish body.

Japanese flounder (Paralichthys olivaceus) were experimentally infected with the highly pathogenic scuticociliate Miamiensis avidus (syn. Philasterides dicentrarachi) using the immersion method to clarify/identify the possible neural routes of entry and possible ways of dissemination of the scuticociliate in the fish body. Scuticociliates were observed on the skin and gills right from day 0-1 post-infection, muscle tissue on day 2 post-infection, reached the brain, and spinal cord on day 3 post-infection, and systemic infection was prominent afterwards. Brain lesions were observed in most of t

5-HTR1D and 5-HTR2B regulated Cryptocaryon irritans-induced inflammatory response of large yellow croaker Larimichthys crocea.

In mammals, 5-hydroxytryptamine receptors (5-HTRs) are expressed on various immune cells, and regulate the host immune response. However, the immunomodulatory function of 5-HTR in fish still remains poorly understood. In this study, Lc_5-HTR1D and Lc_5-HTR2B genes were cloned from large yellow croaker (Larimichthys crocea), showing highly conserved structural characteristics with other vertebrates orthologs. Lc_5-HTR1D and Lc_5-HTR2B mRNAs exhibited high expression not only in the brain, but also in the immune-related tissues (gill and head kidney). Following Cryptocaryon irritans infection, t

Cadmium effects on Ichthyophthirius: evidence for metal-sequestration in fish tissues following administration of recombinant vaccines.

We are developing Tetrahymena thermophila as a delivery system for recombinant vaccines against parasitic protozoa, including the common fish parasite, Ichthyophthirius multifiliis. T. thermophila cell lines expressing I. multifiliis genes under the control of a cadmium-inducible metallothionein gene promoter conferred strong protection against a lethal parasite challenge when administered parenterally to naive fish. Nevertheless, given that heavy metals can be toxic to parasites, a question arose as to whether protection resulted from Cd residues carried over with the vaccine, rather than acq

A safe antiparasitic extract from Psoralea corylifolia for Tetrahymeniasis control.

Tetrahymeniasis is a ciliate disease that presents significant economic challenges for the aquaculture industry. Previous research has shown promising control effects on Tetrahymena pyriformis using flavonoids from Psoralea corylifolia (P. corylifolia), but their high cost hinders practical application. This study aims to find an affordable and safe alternative antiparasitic extract derived from P. corylifolia. Initially, six different solvent extracts from P. corylifolia were compared for in vitro antiparasitic activity and toxicity, with ethyl acetate (EtOAc) extract selected for in vivo tes

Comparative analysis of the expression patterns of IL-1β, IL-11, and IL-34 in golden pompano (Trachinotus ovatus) following different pathogens challenge.

Interleukins (ILs) are a subgroup of cytokines, which are molecules involved in the intercellular regulation of the immune system. These cytokines have been extensively studied in mammalian models, but systematic analyses of fish are limited. In the current study, 3 IL genes from golden pompano (Trachinotus ovatus) were characterized. The IL-1β protein contains IL-1 family signature motif, and four long helices (αA - αD) in IL-11 and IL-34, which were well conserved. All 3 ILs clustered phylogenetically with their respective IL relatives in mammalian and other teleost species. Under normal phy

Immune regulation differences in Large yellow croaker with varied resistance to Cryptocaryon irritans infection.

Large yellow croaker (Larimichthys crocea) is one of the main marine aquaculture species in China, but has faced considerable losses due to Cryptocaryon irritans infection. In this study, we successfully established a C. irritans-susceptible population of large yellow croaker by genomic selection technology. We then compared the immune genetic mechanisms of this susceptible population with those of a large yellow croaker population from eastern Fujian in response to C. irritans infection. GWAS identified 44 significant SNPs across 11 QTL regions on different chromosomes associated with C. irri

Identification and expression analysis of three XCR1-like receptors from Epinephelus coioides after Cryptocaryon irritans infection.

The unique receptor XCR1 of the XC subfamily of chemokines is specially expressed in CD8α-like dendritic cells. This receptor has one ligand in mice (XCL1) and two ligands in humans (XCL1 and XCL2). In mammals, the XCR1-XCL1 complex performs a vital role in regulating the localization and function of T cells, dendritic cells, and other cell types. In this study, three XCR1-like receptors (EcXCR1, EcXCR1L, and EcCCR12) were identified from a transcriptome database of orange-spotted grouper. The open reading frames (ORFs) of EcXCR1, EcXCR1L, and EcCCR12 predictably encode 337, 348, and 358 amino

Discovery of shikonin as a safe and potent oral candidate for the sustainable management of ichthyophthiriasis.

Ichthyophthirius multifiliis remains a devastating ciliate parasite in global aquaculture, yet sustainable therapies are critically scarce. While many plant-derived compounds display potent in vitro efficacy, their practical application is frequently hindered by the failure of immersion treatment to reach tissue-embedded parasites. To overcome this challenge, following a screening of 20 natural compounds, we identified shikonin (SK) as a highly promising orally bioavailable candidate with a novel mechanism. In vitro, SK demonstrated potent efficacy with EC50 values of 0.24, 0.37, and 4.05 mg/L
