A Comprehensive QSAR Study on Antileishmanial and Antitrypanosomal Cinnamate Ester Analogues.

Parasitic infections like leishmaniasis and trypanosomiasis remain as a worldwide concern to public health. Improvement of the currently available drug discovery pipelines for those diseases is therefore mandatory. We have recently reported on the antileishmanial and antitrypanosomal activity of a set of cinnamate esters where we identified several compounds with interesting activity against L. donovani and T. brucei rhodesiense. For a better understanding of such compounds' anti-infective activity, analyses of the underlying structure-activity relationships, especially from a quantitative poi

Isolation of Cinnamic Acid Derivatives from the Bulbs of Allium tripedale.

BACKGROUND: Allium genus with 750 species is the most diverse genus in the Amaryllidaceae family. Historically, Allium species have been used as medicinal plants, especially for prevention and treatment of cardiovascular diseases and considered as valuable sources of phytonutrients. Phytochemical investigation of Allium tripedale, locally called "Anashq," which is an edible plant of the "Zagros" region (west of Iran) was conducted in the present study. MATERIALS AND METHODS: Air-dried bulbs of the plant were extracted in a four-step extraction method with increasing polarity using hexane, chl

Reversal of ethanol-induced hepatotoxicity by cinnamic and syringic acids in mice.

Ethanol was used to induce acute hepatotoxicity in mice. Effects of cinnamic acid (CA) and syringic acid (SA) post-intake for hepatic recovery from alcoholic injury was investigated. Ethanol treated mice were supplied by CA or SA at 40 or 80 mg/kg BW/day for 5 days. Results showed that ethanol stimulated protein expression of CYP2E1, p47phox, gp91phox, cyclooxygenase-2 and nuclear factor kappa B in liver. CA or SA post-intake restricted hepatic expression of these molecules. Ethanol suppressed nuclear factor erythroid 2-related factor (Nrf2) expression, and CA or SA enhanced Nrf2 expression in

Do 'cinnamon-sensitive' patients react to cinnamate UV filters?

BACKGROUND: Use of sunscreens has increased dramatically worldwide, and some sunscreen chemicals may be allergens. Ultraviolet (UV) filters are added to various cosmetic products. Cinnamate UV filters are structurally related to cinnamon-related fragrances. OBJECTIVE: The purpose of this study was to determine if 'cinnamon-sensitive' patients show positive photopatch tests to cinnamate UV filters and, therefore, should avoid these UV filters. METHOD: We photopatch tested cinnamon-sensitive patients (n = 18) with cinnamon, cinnamon-related fragrances, Myroxylon pereirae, and two cinnamate UV

Synthesis and biological evaluation of sulfur-containing cinnamate and salicylate derivatives.

UV irradiation induced formation of reactive oxygen radical species and matrix metalloproteinases (MMPs) are thought to be involved in photo-damage to the skin. MMP-1 is the major collagenolytic enzyme responsible for collagen destruction in skin tissue. To develop new anti-photoaging agents, a series of 2,2'-dithiocinnamate derivatives and 2,2'-dithio or 2-thiobenzoate derivatives were designed and synthesized. The biological activities of the synthesized compounds were assayed for ABTS [2,2'-azinobis-(3-ethyl-benzo-thiazoline-6-sulfonic acid)] radical scavenging activity, MMP-1 inhibitory ac

The effect of quenching agents on contact urticaria caused by cinnamic aldehyde.

Inhibition (quenching) of contact sensitization by cinnamic aldehyde (CA) reportedly occurs from eugenol (E) and d-limonene (d-L). Experimentally the former inhibited nonimmunologic contact urticaria (NICU) from CA in seven of eleven test subjects, which prompted a search for possible mechanisms, including chemical interaction, altered absorption, and anti-inflammatory activity, and competitive inhibition. Mixtures of CA and E and CA and d-L showed no chemical changes or intermolecular bonding. Absorption was not increased by cellophane tape stripping, and neither E nor d-L inhibited urticaria

The metabolism of cinnamic acid by healthy and phenylketonuric adults: a kinetic study.

The enzyme phenylalanine ammonia lyase taken orally has been found to reduce the rise in blood phenylalanine that normally occurs following a protein meal. Therefore the enzyme has a potential use in the management of the genetic disease phenylketonuria. The enzyme mediates the conversion of phenylalanine to cinnamic acid and its possible clinical future has necessitated a more detailed study of the product of its reaction. Cinnamic acid is a compound of low toxicity which is converted in the mammalian body primarily to hippuric acid. We have examined the kinetics of this process in a healthy

Cinnamic aldehyde in toothpaste. 1. Clinical aspects and patch tests.

Although cinnamon is known to cause dermatitis in bakers and confectioners, it has only rarely been reported as causing trouble in food or cosmetics. A newly-formulated 'spicy' toothpaste containing cinnamon as a flavouring agent was the cause of oral symptoms in eight patients referred to clinics in Buckinghamshire and in Malmö and in a further eight patients discovered subsequently. Similar symptoms and patch test results were observed independently at both centres. Positive reactions were obtained with 1% cinnamic aldehyde in 15 out of 16 patients tested. Only one patient reacted to a stand

Discovery of novel cinnamic acid derivatives with anti-Helicobacter pylori mechanism.

With the efficacy of standard anti-Helicobacter pylori (Hp) regimens declining due to increasing resistance and adverse effects, the development of safer alternatives is imperative. Inspired by the bioactivity of cinnamic acid, a series of cinnamic acid derivatives was designed and screened for their activity against Hp. Among the synthesized derivatives, compound 23, bearing a dinitro group, exhibited potent anti-Hp activity with a minimum inhibitory concentration (MIC) of 4 µM, demonstrating a two-fold greater potency than metronidazole. Meanwhile, the quinoline-containing derivative (compou

Cinnamic acid attenuates Klebsiella pneumoniae virulence by suppressing capsule biosynthesis.

The widespread dissemination and serious clinical consequences of hypervirulent Klebsiella pneumoniae (hvKP), an encapsuled gram-negative pathogenic bacterium characterized by its specific virulence factor hypermucoviscosity (HMV), is becoming a concerning global public threat. In contrast to the common health care-associated infection of classical K. pneumoniae (cKp), hvKP is more virulent and capable of causing community-acquired invasive infections in healthy individuals, which mainly relies on the increased production of capsular polysaccharides (CPS) termed hypercapsule, and capsule-assoc

Cinnamic Acid Derivatives: Recent Discoveries and Development Strategies for Alzheimer's Disease.

Alzheimer's Disease (AD) is a progressive neurodegenerative disorder that leads to cognitive decline and memory impairment. It is characterized by the accumulation of Amyloid-beta (Aβ) plaques, the abnormal phosphorylation of tau protein forming neurofibrillary tangles, and is often accompanied by neuroinflammation and oxidative stress, which contribute to neuronal loss and brain atrophy. At present, clinical anti-AD drugs are mostly single-target, improving the cognitive ability of AD patients, but failing to effectively slow down the progression of AD. Therefore, research on effective multi-

Discovery of Cinnamic Acid Derivatives as Potent Anti-H. pylori Agents.

Antibiotics are currently used for the treatment of Helicobacter pylori (H. pylori), which is confirmed to be the major cause of gastric disorders. However, the long-term consumption of antibiotics has already caused antibiotic resistance and side effects in vivo. Therefore, there is an emerging need for searching for safe and effective anti-H. pylori agents. Inspired by the excellent bioactivities of cinnamic acid, a series of cinnamic acid derivatives (compounds 1-30) were synthesized and determined for H. pylori inhibition. The initial screening revealed that compound 23, a 2,4-dinitro cinn

Structure-Activity Relationships of Cinnamate Ester Analogues as Potent Antiprotozoal Agents.

Protozoal infections are still a global health problem, threatening the lives of millions of people around the world, mainly in impoverished tropical and sub-tropical regions. Thus, in view of the lack of efficient therapies and increasing resistances against existing drugs, this study describes the antiprotozoal potential of synthetic cinnamate ester analogues and their structure-activity relationships. In general, Leishmania donovani and Trypanosoma brucei were quite susceptible to the compounds in a structure-dependent manner. Detailed analysis revealed a key role of the substitution patter

The Neuroprotective Effects of Cinnamic Aldehyde in an MPTP Mouse Model of Parkinson's Disease.

Cinnamic aldehyde (CA), a key flavor compound in cinnamon essential oil, has been identified as an anti-oxidant, anti-angiogenic, and anti-inflammatory material. Recently, the neuroprotective effects of CA have been reported in various neurodegenerative disorders, including Parkinson's disease (PD). In neurons, autophagy is tightly regulated, and consequently, the dysregulation of autophagy may induce neurodegenerative disorders. In the present study, we found that the selective dopaminergic neuronal death in the substantia nigra of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) mouse mod

Novasomal Gel for Enhanced Dermal Delivery and Antibacterial Efficacy of Cinnamic Acid.

While bacterial skin infections are highly prevalent worldwide, their eradication with conventional topical medications remains highly challenging. Cinnamic acid (CA) is a naturally occurring molecule with interesting antibacterial properties, but its efficacy is hindered by poor aqueous solubility and skin permeability. To overcome these challenges, CA was encapsulated within novasomes, which are multilamellar vesicles composed of fatty acids, cholesterol, and nonionic surfactants. The novasomes were optimized using a 23 factorial design and the optimized formulation was incorporated in a car
