Citrulline evaluation in bowel transplantation.

INTRODUCTION: The purpose of this study was to evaluate the correlation of plasma citrulline and rejection episodes in intestinal transplantation. METHODS: From January 2007 until present, we performed citrulline assays on our small bowel patients. We investigated the correlation of these assays with the rejection status of the patients. The rejection status of the graft was defined based on graft biopsies. RESULTS: Of 5195 citrulline samples, average serum citrulline levels decreased significantly when the patients presented a rejection episode. We found the following: no rejection, 17.38 m

An amperometric l-citrulline biosensor.

Plasma l-citrulline is widely recognized as a clinically important biomarker, primarily serving as an indicator of intestinal function; it is used to assess and monitor the severity of intestinal failure and short bowel syndrome, as well as to diagnose congenital citrullinemia associated with urea-cycle disorders. However, its rapid and simple quantification remains challenging. To our knowledge, this is the first report of an amperometric biosensor for l-citrulline. In this work, the biosensor was developed based on mediated electron transfer (MET)-type bioelectrocatalysis of l-citrulline deh

Immune recognition of citrullinated epitopes.

Conversion of arginine into citrulline is a post-translational modification that is observed in normal physiological processes. However, abnormal citrullination can provoke autoimmunity by generating altered self-epitopes that are specifically targeted by autoantibodies and T cells. In this review we discuss the recognition of citrullinated antigens in human autoimmune diseases and the role that this modification plays in increasing antigenic diversity and circumventing tolerance mechanisms. Early published work demonstrated that citrullinated proteins are specifically targeted by autoantibodi

Differential histone tail citrullination by protein arginine deiminases observed via NMR spectroscopy.

Citrullination is a charge-modifying post-translational modification whereby proteinogenic arginine is converted to the non-coded amino acid citrulline by calcium-activated protein arginine deiminases (PADs; EC 3.5.3.15). The five known PAD enzymes in humans (PADs 1, 2, 3, 4, and 6) are differentially expressed and have distinct protein targets, including histones. While some PAD histone citrullination sites are known, a comprehensive investigation of all histone tail arginines targeted by catalytically active PADs 1-4 is lacking. Here, we sought to identify PAD citrullination sites in histone

Cellular Consequences, Citrullination Substrates, and Antigenicity of Targeting PAD4 to Cell Surfaces.

Protein arginine deiminase-4 (PAD4) catalyzes hydrolysis of arginine to citrulline in proteins that promote widespread cellular changes that can induce innate immunity and promote cancer. Hyperactivity of PAD4 leads to a form of cell death called NETosis, releasing PAD4 to the extracellular space to promote various autoimmune diseases through the generation of anticitrulline protein antibodies (ACPAs). Little is known about the specific citrullinated substrates that lead to autoimmunity, but there is growing evidence that PAD4 is localized to the cell surface in response to inflammation. Here,

NLRP3 inflammasome promotes PAD2/PAD4 release and protein citrullination in rheumatoid arthritis.

Anti-citrullinated protein antibodies (ACPAs), a hallmark of rheumatoid arthritis (RA), arise from citrullinated proteins presenting immunogenic neoepitopes that break self-tolerance. Citrullination is a post-translational modification catalyzed by peptidylarginine deiminases (PADs). Among five isoforms, PAD2 and PAD4 expressed in immune cells are most closely associated with RA pathogenesis. Although typically intracellular, PAD2 and PAD4 are detectable extracellularly in RA, yet the mechanisms governing their release remain unclear. Here, we show that NLRP3 inflammasome activation triggers P

NEMO (IKKγ) citrullination in microglia promotes neuroinflammation in the post-ischemic brain.

Peptidylarginine deiminase (PAD) catalyzes the post-translational conversion of positively charged arginine residues into neutral citrulline. This modification, known as citrullination, is implicated in various pathological conditions, including those affecting the central nervous system. This study investigated the pro-inflammatory role of PAD in microglia using a middle cerebral artery occlusion (MCAO) animal model of ischemic stroke. Pharmacological inhibition of PAD via BB-Cl-amidine (BBCA; a pan-PAD inhibitor) exerted robust anti-inflammatory and neuroprotective effects in the post-ischem

PAD2-mediated citrullination of MBP and early myelin destabilization in multiple sclerosis.

Proteins affect most physiological functions in cells and play a key role in maintaining cellular homeostasis. Protein function is tuned at multiple levels, including synthesis and degradation, intracellular localization, and posttranslational modifications (PTMs). PTMs are a key chemical modification for regulating protein biological activity, including protein-folding relevant shifts in electrical charge, protein-protein interactions, and hydrogen bond formation. Such chemical changes can also result in protein unfolding or denaturation. PTMs can target proteins across multiple cellular comp

CXCL1-Triggered PAD4 Cytoplasmic Translocation Enhances Neutrophil Adhesion through Citrullination of PDIA1.

AIMS: Vascular inflammation is critical for the development and progression of atherosclerosis. Previously, we reported that neutrophils adhere to the vascular endothelium in low-density lipoprotein receptor null mice fed a high-fat diet through hypercitrullination of histone H3 by peptidylarginine deiminase 4 (PAD4) in neutrophils. However, the involvement of PAD4 and citrullination of proteins other than histone H3 in neutrophil adhesion is not well known. In this study, we investigated the function of PAD4 and identified citrullinated proteins during vascular inflammation. METHODS: We peff

Peptidyl Arginine Deiminase 2 (PADI2)-Mediated Arginine Citrullination Modulates Transcription in Cancer.

Protein arginine deimination leading to the non-coded amino acid citrulline remains a key question in the field of post-translational modifications ever since its discovery by Rogers and Simmonds in 1958. Citrullination is catalyzed by a family of enzymes called peptidyl arginine deiminases (PADIs). Initially, increased citrullination was associated with autoimmune diseases, including rheumatoid arthritis and multiple sclerosis, as well as other neurological disorders and multiple types of cancer. During the last decade, research efforts have focused on how citrullination contributes to diseas

Citrullination-dependent differential presentation of a self-peptide by HLA-B27 subtypes.

Inflammatory processes are accompanied by the posttranslational modification of certain arginine residues within proteins to yield citrulline, although it is largely unknown how this modification influences antigen presentation. We employed crystallographic and functional studies to investigate whether the exchange of arginine to citrulline affects the display of a peptide by two human major histocompatibility antigen class I subtypes, HLA-B(*)2705 and HLA-B(*)2709. Both differ only in residue 116 within the peptide binding groove despite their differential association with ankylosing spondyli

Probing tau citrullination in Alzheimer's disease brains and mouse models of tauopathy.

Alzheimer's disease (AD) includes a defining hallmark that correlates most closely to cognitive decline, namely misfolded tau protein. However, the "upstream" etiology and downstream clinical manifestations of tauopathies are quite diverse. Tau deposition elicits different pathological phenotypes and outcomes depending on the tau strain, proteoforms, and regional susceptibility. Posttranslational modifications (PTM) can alter tau structure, function, networks, and its pathological sequelae. We uncovered tau citrullination on multiple epitopes caused by peptidyl arginine deiminase (PAD) enzymes

GAPDH citrullination as a molecular signature of neurodegeneration, assessed in prion diseases.

BackgroundGlyceraldehyde 3-phosphate dehydrogenase (GAPDH) is a well-known glycolytic enzyme that plays multiple roles in energy metabolism, cell growth, and cell death, and functions as a housekeeping protein. GAPDH has been identified as a potential target of citrullination, a calcium-dependent post-translational modification mediated by peptidylarginine deiminase (PAD), in which arginine residues are converted to citrulline. This modification has been implicated in various human pathologies.ObjectiveTo investigate the function and pathological relevance of citrullinated GAPDH.MethodsWe gene

Anti-cyclic citrullinated peptide positivity in non-rheumatoid arthritis disease samples: citrulline-dependent or not?

BACKGROUND: Antibodies directed against citrullinated proteins (eg anti-cyclic citrullinated peptide (CCP)) have excellent diagnostic and good prognostic potential for rheumatoid arthritis. Type 1 autoimmune hepatitis (AIH-1) is a chronic liver disease characterised by a variety of serum autoantibodies. Recently, in a large group of patients with AIH-1 without clear rheumatoid arthritis overlap, a relatively high percentage (9%) of anti-CCP2 positivity was scored. OBJECTIVES: To characterise the citrulline-dependence of the observed anti-CCP2 positivity in AIH-1 sera as well as in other group

Pathogenic Role of Circulating Citrullinated Antigens and Anti-Cyclic Monoclonal Citrullinated Peptide Antibodies in Rheumatoid Arthritis.

We examined whether it is possible to directly detect citrullinated antigens in the serum of rheumatoid arthritis (RA) patients using a monoclonal antibody (mAb) designed to be specific for citrullinated peptides. In order to confirm the potential of the mAb as a direct arthritis-inducing substance through experimental model of RA, a monoclonal antibody (mAb) 12G1 was generated using by immunization of mice with a challenging cyclic citrullinated peptide. Immunohistochemical analysis of RA-affected synovial tissue showed that our mAb 12G1 could indeed detect citrullinated proteins in target ti

Citrullinated Antigens with Multiple Citrulline Similar Motif in the Diagnosis of Rheumatoid Arthritis: A Preliminary Single-Center Study.

The presence of anti-citrullinated protein antibodies (ACPAs) in the serum is one of the immunological features of rheumatoid arthritis (RA). Anti-cyclic citrullinated peptide (CCP) assay has been widely used in clinic for the diagnosis of RA. However, up to 40% of RA patients are anti-CCP negative and the diagnostic sensitivity in this population needs to be improved for better clinical management. In this study, peptides with Multiple Citrulline Similar Motif (MCSM) were synthesized and a new ELISA system, which we called RA_CP, was developed to detect citrullinated antigens with MCSM presen
