The claustrum is a telencephalic gray matter nucleus that is richly interconnected with the neocortex. This structure subserves top-down executive functions that require frontal cortical control of posterior cortical regions. However, functional anatomical support for the claustrum allowing for long-range intercortical communication is lacking. To test this, we performed a channelrhodopsin-assisted long-circuit mapping strategy in mouse brain slices. We find that anterior cingulate cortex input to the claustrum is transiently amplified by claustrum neurons that, in turn, project to parietal as

Dementia and visual hallucinations are common complications of Parkinson's disease (PD), yet their patho-anatomical bases are poorly defined. We studied alpha-synuclein (alphaSyn), tau and amyloid-beta (Abeta) pathology in the claustrum of 20 PD cases without dementia, 12 PD cases with dementia (PDD) and 7 cases with dementia with Lewy bodies (DLB). alphaSyn positivity was observed in 75% of PD cases without dementia and in 100% of PDD and DLB cases. Abeta was observed in the claustrum in 25% of PD, 58% of PDD and 100% of DLB cases. Tau was negligible in all cases restricting further analysis.

We experienced an 8-year-old-boy with non-herpetic acute limbic encephalitis (NHALE), who developed headache, convulsion, consciousness disturbance, and ataxia following cold like symptoms. Disturbance of short term memory and a change of character were recognized. Myoclonic seizures and generalized tonic clonic convulsions developed, that responded to antiepileptic agents. Although other symptoms resolved spontaneously, short term memory disturbance persisted. Brain MRI demonstrated the lesion involving the bilateral claustrum and right hippocampus. Three months later, the lesion in the claus

Methamphetamine (METH) abstinence anxiety is a common METH abstinence symptom, which drives METH relapse. Our previous study found that claustrum (CL) is involved in coding abstinence anxiety-like behaviors caused by adolescent cocaine experiences. Here, we aim to investigate the role of CL in the development of METH abstinence-induced anxiety, especially exploring the related glutamatergic projections to CL. We found that 14 d of METH abstinence heightened anxiety-like behaviors in male mice, accompanied by hyperactivation of glutamatergic neurons (GNs) in the CL, as well as in several brain

Aberrant cognitive network activity and cognitive deficits are established features of chronic pain. However, the nature of cognitive network alterations associated with chronic pain and their underlying mechanisms require elucidation. Here, we report that the claustrum, a subcortical nucleus implicated in cognitive network modulation, is activated by acute painful stimulation and pain-predictive cues in healthy participants. Moreover, we discover pathological activity of the claustrum and a region near the posterior inferior frontal sulcus of the right dorsolateral prefrontal cortex (piDLPFC)

Pathological changes in the claustrum and its main parts (dorsal, temporal, orbital and paramygdalar) were studied on ten brains of patients with Alzheimer's disease and five control brains. The brains after fixation in formalin were embedded in paraffin, coronal-8-micron-thick serial sections were cut and stained either with cresyl violet or with immunocytochemical methods for amyloid and tangles. Morphometrical studies were performed in all parts of the claustrum along its whole extension. The claustrum in control brains was free of neurofibrillary and amyloid pathology except for one case w

Cognitive impairment is a major component of Alcohol Use Disorder. Optimal cognitive performance requires anterior cingulate input activation of the claustrum, a subcortical nucleus that orchestrates cortical activity. Yet the impact of chronic alcohol exposure on the ability for the anterior cingulate cortex to drive activity of claustrum projection neuron subtypes is unknown. In adult male and female mice, we found that the majority of non-burst firing Type 1 claustrum projection neurons did not express the vesicular glutamate transporter 2 (VGLUT2), while the majority of burst firing Type 2

Itch, an uncomfortable sensation that induces scratching, is suppressed by activation of kappa opioid receptors (KORs). KOR agonists, such as Nalfurafine, offer potent central antipruritic effects, yet the underlying mechanisms are not fully understood. In this study, we report the important role of glutamatergic neurons in the claustrum (CLA) in itch modulation. Chemogenetic inhibition of these neurons significantly reduces histamine-induced scratching behavior, whereas their activation heightens scratching behavior. These CLAGlu neurons connect to the mediodorsal thalamus (MD), forming a pat

Studies on gene expression in the developing claustrum of the mouse have clarified the relationships and identity of the claustrum proper and related endopiriform nuclei. The cells of the claustrum primordium express Nr4a2; they are formed in combination with the Nr4a2-labeled subplate cells in the lateral pallium at the site of the future insular cortex. The insular cortex cells, which are born later and which are Nr4a2-negative, migrate through the claustrum toward the pial surface to form layers (2-6a) of the insular cortex. The claustrum is made up of distinct deep (subplate-like) and supe

Visual response properties of neurons in the dorsocaudal claustrum were studied in two cats reared with convergent strabismus and three cats reared with monocular lid-suture. In the normal claustrum, most cells respond about equally well to stimulation of either eye (Sherk and LeVay, '81). In the strabismic animals, there was a partial breakdown of binocularity: most cells remained binocular but were influenced more strongly by one eye (usually the contralateral eye) than the other. The loss of binocularity was less extreme in the claustrum than in area 17 of the same animals. In the monocular

The differential mechanisms between proteinopathies and neurodegeneration in Alzheimer's disease (AD) and dementia with Lewy bodies (DLB) remain unclear. To address this issue, we conducted a voxel-based morphometry and cerebrospinal fluid biomarker (α-synuclein, Aβ42, t-Tau and p-Tau181) level correlation study in patients with DLB, AD and mixed cases (AD + DLB). Cerebrospinal fluid samples obtained by lumbar puncture and whole-brain T1-weighted images were collected in the AlphaLewyMA cohort. Within the cohort, 65 DLB patients, 18 AD patients, 24 AD + DLB patients and 16 neurological control

COVID-19-related encephalopathy is a complication of COVID-19 that affects the brain. It can present with symptoms such as altered mental status, ranging from mild confusion to deep coma. It is a major and devastating complication of the SARS-CoV-2 virus. COVID-19-related neurological symptoms may be associated with cytokine release induced by the virus, and tocilizumab targeting IL-6 receptor may provide significant benefits. We report a case of COVID-19-related encephalopathy with cerebrospinal fluid analysis revealing significantly elevated levels of IL-6. Brain magnetic resonance imaging s

OBJECT: The goal in this study was to examine the microsurgical and tractographic anatomy of the claustrum and its projection fibers, and to analyze the functional and surgical implications of the findings.
METHODS: Fifteen formalin-fixed human brain hemispheres were dissected using the Klingler fiber dissection technique, with the aid of an operating microscope at x 6-40 magnification. Magnetic resonance imaging studies of 5 normal brains were analyzed using diffusion tensor (DT) imaging-based tractography software.
RESULTS: Both the claustrum and external capsule have 2 parts: dorsal and v

The cytoarchitecture of the claustrum in the hedgehog (Erinaceus europaeus) brain, the morphology of its neurons, and the efferent connections with cortical and subcortical structures were studied with the Nissl and Klüver-Barrera, the Golgi, and the horseradish peroxidase methods. It was found that the claustrum is a well developed nucleus in the hedgehog telencephalon and, as in other mammals, is divided into dorsal and ventral parts. In Golgi-stained sections, spiny multipolar cells are the predominant neurons of both the dorsal and the ventral claustrum and are projection neurons. Aspiny m

The claustrum is a thin gray matter structure in each brain hemisphere, characterized by exceptionally high connectivity with nearly all brain regions. Despite extensive animal studies on its anatomy and function and growing evidence of claustral deficits in neuropsychiatric disorders, its specific roles in normal and abnormal human brain function remain largely unknown. This is primarily due to its thin and complex morphology, which limits accurate anatomical delineation and neural activity isolation in conventional in vivo neuroimaging. To facilitate future neuroimaging studies, we developed

BACKGROUND: Febrile infection‑related epilepsy syndrome (FIRES) is a severe acute inflammation‑associated encephalopathy in children, characterized by high mortality and poor long‑term outcomes. FIRES with claustrum lesions (FIRES‑C) represents a distinct subtype. However, the relationship between its neuroimaging heterogeneity and electrophysiological phenotype remains unclear, and the association between sleep spindles and acute‑phase severity has not been systematically investigated. This study aimed to characterize the electroclinical features of FIRES‑C to facilitate improvements in diagn

Methamphetamine (METH) addiction involves escalating intake with strong cue reactivity, and high relapse risk, yet its neural mechanism remains unclear. Using c-Fos mapping and machine learning, we identified the claustrum (CLA), a subcortical region reciprocally connected with the anterior cingulate cortex (ACC), as key mediators of both METH taking and seeking in self-administering male rats. Chemogenetic inhibition of CLA suppressed both drug consumption and cue-induced reinstatement, while ACC inhibition selectively reduced drug-seeking. Circuit tracing and manipulation revealed that the C

We have investigated the hippocampal connectivity of the marmoset presubiculum (PreS) and reported that major connections of PreS in the rat were conserved in the marmoset. Moreover, our results indicated the presence of several additional projections that were almost absent in the rat brain, but abundant in the marmoset, such as direct projections from CA1 to PreS. However, little is known about the connectivity between the frontal brain regions and PreS or hippocampal formation. Therefore, we investigated the distribution of cells of the origins and terminals of the presubicular and hippocam
