A click chemistry mediated in vivo activity probe for dimethylarginine dimethylaminohydrolase.

Asymmetric N(omega),N(omega)-dimethyl-l-arginine (ADMA) is an endogenously produced inhibitor of human nitric oxide synthase and an emerging biomarker for cardiovascular disease. Concentrations of ADMA are controlled by two isoforms of its catabolic enzyme dimethylarginine dimethylaminohydrolase (DDAH), the dysregulation of which has been studied as a mediating factor for endothelial dysfunction. A two-part, click-chemistry mediated activity-based probe, N-but-3-ynyl-2-chloroacetamidine, is shown to label myc-tagged DDAH-1 expressed in HEK 293T cells, but not an inactive mutant or inhibited en

Bio-orthogonal functionalization of bacterial cellulose combining metabolic glycoengineering and click chemistry.

Bacterial cellulose possesses excellent biocompatibility and mechanical strength but lacks the bioactivity needed for many biomedical and healthcare applications. To address this limitation, we develop a metabolic glycoengineering-click chemistry strategy that enables in situ incorporation of azide groups into bacterial cellulose, followed by mild and selective conjugation of alkyne-bearing functional molecules. This approach avoids harsh chemical treatments, preserves the native properties of bacterial cellulose, and supports stable attachment of diverse bioactive agents, including antibacter

Organelle-Resolved Tetrazine-trans-Cyclooctene Click Chemistry for Cargo Delivery and Release.

Accurate delivery of small molecules to specific cell compartments or organelles offers great potential for bioimaging and targeted therapy but remains challenging to predict and control. In this study, we evaluate a modular strategy for organelle-directed delivery based on organelle-targeting groups combined with tetrazine-trans-cyclooctene bioorthogonal chemistry. We demonstrate that organelle-targeting probes bearing tetrazine or trans-cyclooctene can efficiently undergo inverse-electron-demand Diels-Alder reactions within various cellular compartments, including the plasma membrane, mitoch

ImmunoPET for mesothelin positive tissues using bio-orthogonal in-vivo click chemistry.

Mesothelin is a membrane bound antigen overexpressed in a wide array of cancers including ovarian, pancreatic, lung, and triple negative breast cancers. Here a full-length IgG antibody developed against mesothelin, called MESO-HSS1 was conjugated for immunoPET imaging and assessed in-vivo in a H956 mesothelin positive model. Furthermore, random lysine conjugation as well as a site selective conjugation method were compared in two pretargeting models for overall tumor uptake alongside non target organs and tumor to organ ratios. Overall, [89Zr]Zr-DFO-MESO-HSS1 was found to be a suitable immunoP

Development of Novel Immobilized Copper-Ligand Complex for Click Chemistry of Biomolecules.

Copper-catalyzed azide-alkyne cycloaddition click (CuAAC) reaction is widely used to synthesize drug candidates and other biomolecule classes. Homogeneous catalysts, which consist of copper coordinated to a ligand framework, have been optimized for high yield and specificity of the CuAAC reaction, but CuAAC reaction with these catalysts requires the addition of a reducing agent and basic conditions, which can complicate some of the desired syntheses. Additionally, removing copper from the synthesized CuAAC-containing biomolecule is necessary for biological applications but inconvenient and req

Engineered macrophage-derived exosomes via click chemistry for the treatment of osteomyelitis.

Osteomyelitis is a severe bone condition caused by bacterial infection that can lead to lifelong disabilities or fatal sepsis. Given that the infection is persistent and penetrates deep into the bone tissue, targeting and rapidly treating osteomyelitis remain a significant challenge. Herein, a triblock targeting peptide featuring ROS-cleavable linkage/antibacterial/bone-targeting unit was grafted onto the macrophage-derived exosomes (RAB-EXO). In vitro, the effective eradication of osteomyelitis pathogens MRSA/E. coli and induction of M2 macrophage differentiation were triggered by RAB-EXO. In

Covalent Linkage of BODIPY-Photosensitizers to Anderson-Type Polyoxometalates Using CLICK Chemistry.

The covalent attachment of molecular photosensitizers (PS) to polyoxometalates (POMs) opens new pathways to PS-POM dyads for light-driven charge-transfer and charge-storage. Here, we report a synthetic route for the covalent linkage of BODIPY-dyes to Anderson-type polyoxomolybdates by using CLICK chemistry (i. e. copper-catalyzed azide-alkyne cycloaddition, CuAAC). Photophysical properties of the dyad were investigated by combined experimental and theoretical methods and highlight the role of both sub-components for the charge-separation properties. The study demonstrates how CLICK chemistry c

Optical Properties of New Third-Order Nonlinear Materials Modified by Click Chemistry.

A high-yielding click reaction was used to synthesize a series of highly conjugated, symmetrical, as well as asymmetrical compounds with a benzene core. Cyclic voltammetry and ultraviolet/visible absorption spectroscopy were carried out, and proved that the side groups of the benzene derivatives played an important role in the energy gaps, and affected the third-order non-linear optical response. The maximum absorption wavelength of the series of benzene derivatives showed an obvious red-shift. Moreover, the addition of resilient electron-withdrawing groups significantly narrowed the energy le

Alternative approaches utilizing click chemistry to develop next-generation analogs of solithromycin.

The marked rise in bacterial drug resistance has created an urgent need for novel antibacterials belonging to new drug classes and ideally possessing new mechanisms of action. The superior biological activity of solithromycin against streptococci and other bacteria causative of community-acquired pneumonia pathogens, compared to telithromycin and other macrolides encouraged us to extensively explore this class of antibiotics. We, thus, present the design and synthesis of a novel series of solithromycin analogs. Three main strategies were pursued in structure-activity relationship studies cover

Click and Release Chemistry for Activity-Based Purification of β-Lactam Targets.

β-Lactams, the cornerstone of antibiotherapy, inhibit multiple and partially redundant targets referred to as transpeptidases or penicillin-binding proteins. These enzymes catalyze the essential cross-linking step of the polymerization of cell wall peptidoglycan. The understanding of the mechanisms of action of β-lactams and of resistance to these drugs requires the development of reliable methods to characterize their targets. Here, we describe an activity-based purification method of β-lactam targets based on click and release chemistry. We synthesized alkyne-carbapenems with suitable proper

An Organometallic Cu20 Nanocluster: Synthesis, Characterization, Immobilization on Silica, and "Click" Chemistry.

The development of atomically precise nanoclusters (APNCs) protected by organometallic ligands, such as acetylides and hydrides, is an emerging area of nanoscience. In principle, these organometallic APNCs should not require harsh pretreatment for activation toward catalysis, such as calcination, which can lead to sintering. Herein, we report the synthesis of the mixed-valent organometallic copper APNC, [Cu20(CCPh)12(OAc)6)] (1), via reduction of Cu(OAc) with Ph2SiH2 in the presence of phenylacetylene. This cluster is a rare example of a two-electron copper superatom, and the first to feature
