Anaerobic bacteria as a delivery system for cancer gene therapy: in vitro activation of 5-fluorocytosine by genetically engineered clostridia.

Certain species of anaerobic bacteria have been shown to localise and germinate specifically in the hypoxic regions of tumours, resulting in tumour lysis. We propose an innovative approach to cancer gene therapy in which genetically engineered anaerobic bacteria of the genus Clostridium are used to achieve tumour-specific gene delivery. Our strategy involves enzyme/prodrug therapy, in which the Escherichia coli enzyme cytosine deaminase is used to convert the non-toxic prodrug 5-fluorocytosine to the active chemotherapeutic agent 5-fluorouracil. The E. coli gene encoding cytosine deaminase has

Wood pulp as an immobilization matrix for the continuous production of isopropanol and butanol.

The study was focused on developing a continuous method to produce an alcohol mixture suitable to be used as a gasoline supplement. The immobilized column reactor with wood pulp fibers was successfully used for the continuous production of butanol and isopropanol using Clostridium beijerinckii DSM 6423. A sugar mixture (glucose, mannose, galactose, arabinose and xylose) representing lignocellulose hydrolysate was used as a substrate for the production of solvents. The effect of dilution rate on solvent production was studied during continuous operation. The maximum total solvent concentration

Production of isopropanol by metabolically engineered Escherichia coli.

A genetically engineered strain of Escherichia coli JM109 harboring the isopropanol-producing pathway consisting of five genes encoding four enzymes, thiolase, coenzyme A (CoA) transferase, acetoacetate decarboxylase from Clostridium acetobutylicum ATCC 824, and primary-secondary alcohol dehydrogenase from C. beijerinckii NRRL B593, produced up to 227 mM of isopropanol from glucose under aerobic fed-batch culture conditions. Acetate production by the engineered strain was approximately one sixth that produced by a control E. coli strain bearing an expression vector without the clostridial gene

Biobutanol production from rice straw using a redox-modulated fungal-bacterial consortium: Transcriptional regulation and process integration.

Producing butanol from lignocellulosic biomass is a key strategy for addressing climate change and the energy crisis. Consolidated bioprocess (CBP), which combines enzyme production, hydrolysis, and fermentation into a single step, offers an effective way to lower the production costs of biobutanol. Despite the crucial role of fungi in cellulolytic decomposition, developing efficient fungal-bacterial consortia for butanol production remains challenging due to the fundamental conflict between the aerobic nature of fungi and the strict anaerobicity of solventogenic clostridia. In this study, a m

Consolidated bioprocessing performance of bacterial consortium EMSD5 on hemicellulose for isopropanol production.

Consolidated bioprocessing (CBP) of lignocellulose by bacterial consortium for isopropanol production is considered as the most promising strategy. However, low utilization of xylan caused by the complex sidechain structure remains inhibit the conversion of full-biomass. In this study, isopropanol production from different lignocelluloses by the consortium EMSD5 through CBP was performed. A total of 7.00 g/L of isopropanol was obtained from corncob by optimizing fermentation conditions. Isopropanol production by EMSD5 was mainly based on utilizing xylan in corncob and isopropanol titer was inc

An artificial coculture fermentation system for industrial propanol production.

Converting plant biomass into biofuels and biochemicals via microbial fermentation has received considerable attention in the quest for finding renewable energies and materials. Most approaches have so far relied on cultivating a single microbial strain, tailored for a specific purpose. However, this contrasts to how nature works, where microbial communities rather than single species perform all tasks. In artificial coculture systems, metabolic synergies are rationally designed by carefully selecting and simultaneously growing different microbes, taking advantage of the broader metabolic spac

Systematic analysis of cotranscriptional RNA folding using transcription elongation complex display.

RNA can fold into structures that mediate diverse cellular functions. Understanding how RNA primary sequence directs the formation of functional structures requires methods that can comprehensively assess how changes in an RNA sequence affect its structure and function. Here we have developed a platform for performing high-throughput cotranscriptional RNA biochemical assays, called Transcription Elongation Complex display (TECdisplay). TECdisplay measures RNA function by fractionating a TEC library based on the activity of cotranscriptionally displayed nascent RNA. In this way, RNA function is

Isopropanol-butanol-ethanol production by cell-immobilized vacuum fermentation.

The Isopropanol-Butanol-Ethanol productivity by solventogenic clostridia can increase when cells are immobilized on low-cost, renewable fibrous materials; however, butanol inhibition imposes the need for dilute sugar solutions (less than40 g/L). To alleviate this problem, the in-situ vacuum product recovery technique was applied to recover IBE in repeated-batch cultivation of Clostridium beijerinckii DSM 6423 immobilized on sugarcane bagasse. Five repeated batch cycles were conducted in a 7-L bioreactor containing P2 medium (∼60 g/L glucose) and bagasse packed in 3D-printed concentric annular

Metabolic engineering of Clostridium acetobutylicum for the enhanced production of isopropanol-butanol-ethanol fuel mixture.

Butanol is considered as a superior biofuel, which is conventionally produced by clostridial acetone-butanol-ethanol (ABE) fermentation. Among ABE, only butanol and ethanol can be used as fuel alternatives. Coproduction of acetone thus causes lower yield of fuel alcohols. Thus, this study aimed at developing an improved Clostridium acetobutylicum strain possessing enhanced fuel alcohol production capability. For this, we previously developed a hyper ABE producing BKM19 strain was further engineered to convert acetone into isopropanol. The BKM19 strain was transformed with the plasmid pIPA100 c

Integrating genomics, phenotypes, and volatile profiling with cheese slurry trials reveals strain-dependent clostridial spoilage originating from silage.

Silage is a primary source of Clostridium spores entering the dairy chain; however, the contributions of spoilage beyond Clostridium tyrobutyricum remain undercharacterized. Here, we integrated whole-genome sequencing with fermentation phenotypes and controlled cheese slurry trials, including volatile profiling measured directly in inoculated slurries, to define strain- and species-specific spoilage modes originating from silage. 15 isolates recovered from silage were confirmed by 16S rRNA sequencing and whole-genome sequencing and analyzed alongside 10 ATCC reference strains for phylogenomic

Performance improvements and increased ruminal microbial interactions in Angus heifers via supplementation with native rumen bacteria during high-grain challenge.

Feedlot cattle may be subjected to digestive disorders, including ruminal acidosis, due to high concentration of grain in their diet. Therefore, novel feeding strategies are required to maximize animal performance and mitigate economic losses in the operation. This study employed a two-period crossover design to assess the effect of direct ruminal administration of native rumen microorganisms (NRM) inoculation on cattle that underwent a high-grain challenge. The NRM inoculation consisted of six microorganisms (1.70 M CFU /day/animal) isolated from the rumen of healthy feedlot cattle: Succinivi

Pontic morphology as local risk factor in root decay and periodontal disease.

INTRODUCTION: In absence of treatment, old single edentulous spaces are subjected to progressively decrease in length due to tilting and rotation of the teeth situated mesial and distal. Lately when these spaces are prosthetically restored with a bridge and these teeth are becoming abutments, sanitizing of the prosthetic restoration is achieved with difficulty, the space between the pontic and the gingival surface becoming an area rich in pathogens that can cause various complications on the abutments. AIM: Considering these aspects, in this study we wanted to highlight the presence and the t

Unveiling rhizobacterial diversity in rice-maize systems in response to penoxsulam-pendimethalin application.

BACKGROUND: The rice-maize system is crucial for Asia for improving productivity and resilience. The conventional puddled transplanting (CT) of rice, while prevalent, is resource-intensive and environmentally unsustainable. Zero-tillage direct-seeded rice (ZT-DSR) provides a conservation-oriented alternative, but greater weed pressure demands efficient herbicide applications. The ready-mix penoxsulam (PEN) and pendimethalin (PND) are widely used soil-applied pre-emergence herbicides, but their combined effects on soil microbial communities under different crop establishment practices remain un

Improving sustainable isopropanol production in engineered Escherichia coli W via oxygen limitation.

BACKGROUND: Due to ecological concerns, alternative supply lines for fuel and bulk chemicals such as isopropanol are increasingly pursued. By implementing the formation pathways from natural producers like Clostridium beijerinckii and Clostridium aurantibutyricum, isopropanol can be produced in Escherichia coli. However, developing an industrially and economically feasible microbial production process requires a robust and efficient process strategy. Therefore, this study explores microaerobic conditions in combination with lactose and sour whey as sustainable carbon source as a basis for larg

Metabolic engineering of isopropyl alcohol-producing Escherichia coli strains with 13 C-metabolic flux analysis.

Metabolic engineering of isopropyl alcohol (IPA)-producing Escherichia coli strains was conducted along with 13 C-metabolic flux analysis (MFA). A metabolically engineered E. coli strain expressing the adc gene derived from Clostridium acetobutylicum and the IPADH gene from C. beijerinckii did not produce IPA during its exponential growth phase in the aerobic batch culture. 13 C-MFA was carried out, and revealed a deficiency in NADPH regeneration for IPA production in growth phase. Based on these findings, we used nitrogen-starved culture conditions to reduce NADPH consumption for biomass synt

Direct Production of 2-Butanol from Glucose by Recombinant Klebsiella pneumoniae Strains.

2-Butanol is a promising biofuel due to its favorable properties and lower microbial toxicity compared to other butanol isomers. However, microbial production remains challenging due to the absence of a native biochemical pathway for directly converting sugars into 2-butanol. To achieve this goal, glucose should be directed through the 2,3-butanediol (2,3-BD) pathway, involving α-acetolactate synthase, α-acetolactate decarboxylase, and butanediol dehydrogenase for the formation of meso-2,3-BD, followed by diol dehydratase-catalyzed conversion of meso-2,3-BD to butanone and alcohol dehydrogenas

Pathologic complete response to enfortumab vedotin plus pembrolizumab in node-positive urothelial carcinoma: a case report.

BACKGROUND: Biofuels obtained from first-generation (1G) sugars-starch streams have been proven unsustainable as their constant consumption is not only significantly costly for commercial scale production systems, but it could potentially lead to problems associated with extortionate food items for human usage. In this regard, biofuels' production in alkali-thermophilic environs from second-generation (2G) bio-waste would not only be markedly feasible, but these extreme conditions might be able to sustain aseptic fermentations without spending much for sterilization. RESULTS: Present investig
