Apolipoprotein E is a plasma protein that has an important role in transport and metabolism of lipids in serum as well as central nervous system. Among the 3 common alleles, the ε2 allele has the most stable structure followed by ε3 and ε4 in order. There is evidence for a deleterious role of ε4 allele by atherosclerosis and amyloid beta accumulation in brain and body. The presence and gene dose of ε4 allele are risk factors for late-onset Alzheimer's disease. Apolipoprotein E ε4 may have a role in the pathology of amyloid beta and tau and it has a strong relationship with the early onset of l

Amnestic mild cognitive impairment (aMCI) is associated with increased activation in the CA3-dentate region of hippocampus. Excess CA3 activity also occurs in aged rats with memory impairment. Therapies to counter such excess activity might include antiepileptics or agonists for GABA(A) α5 receptors, which regulate tonic inhibition. Use of GABA(A) α5 agonists may seem unexpected because GABA(A) α5 inverse agonists were developed as cognitive enhancers. We found that inverse agonists, while yielding benefit in normal young adult rats, are not effective in treating memory loss in aged rats. Inst

Survival from Alzheimer's disease (AD) and other dementias into late old age may be a useful phenotype for genetic studies of successful cognitive aging. To support molecular genetics studies for successful cognitive aging, we conducted a two-stage study to determine an optimal age phenotype for successful cognitive aging. First, risk of AD was evaluated, through informant interviews, in 4,794 parents and siblings of 976 elderly nondemented probands who were divided into three different proband age groups: those aged 60-74, 75-89, and 90+. Relatives of probands aged 90+ had a significantly low

INTRODUCTION: Loneliness in older adults is recognized as a psychosocial factor influencing cognitive performance. This study examines the impact of self-perceived loneliness both on actual cognitive performance and on individuals' self- perceived cognitive abilities.
METHODS: A sample of 510 healthy older adults (354 females) aged between 64 and 103 years (mean = 78.78±8.72) was studied. Participants' self-perceived loneliness was assessed on UCLA Loneliness Scale-3 (Hughes et al., 2004), their cognitive performance with the standardized cognitive screening GEMS (Global Examination of Mental

Aging is the primary cause of cognitive decline. Despite extensive study, the molecular mechanisms driving aging-associated cognitive decline remain unclear. Here, we describe a proteostasis-independent function of SEC61A1 and its involvement in aging-associated cognitive decline. SEC61A1 regulates ER-mitochondria contact sites, affecting mitochondrial DNA and RNA synthesis and subsequently leading to changes in innate immune signaling mediated by mitochondrial double-stranded RNA (mt-dsRNA). This pathway is activated in aged wild-type mice, Alzheimer's disease patients, and 5×FAD mice. Tissue

PURPOSE: Research in autism has historically focused on childhood, leaving cognitive aging in autistic adults poorly understood. This study investigated whether cognitive aging in autism follows similar or distinct patterns compared to non-autistic adults. We also explored how subjective executive function relates to objective performance across adulthood in these groups.
METHODS: Fifty-six autistic and 106 non-autistic adults (19-69 years) completed tasks assessing processing speed, verbal short-term and working memory, episodic memory (recall performance and semantic strategy use), and exec

Greater age deviation (AgeDev), in which a person's biological age is older than their chronological age, is associated with worse cognitive performance. Most findings, however, rely on lab-administered cognitive measures and do not account for time-ordered effects of practice. The present study leveraged a measurement burst dataset to examine six distinct features of cognitive performance (peak performance, change in peak performance, intraindividual variability, forgetting, retest-related gains), and explored whether individual differences in these features can be predicted by AgeDev. In a s

The study aim was to determine the contribution of dementia related pathologies to the association of conscientiousness with late-life cognitive health. At enrollment in 2 longitudinal clinical-pathologic cohort studies, 309 older individuals without cognitive impairment completed a standard conscientiousness measure. Annually thereafter, they completed a battery of 17 cognitive tests. On death, they underwent a uniform neuropathologic examination from which measures of neurofibrillary tangles, Lewy bodies, chronic gross cerebral infarction, and hippocampal sclerosis were derived. The relation

BackgroundIdentifying deficits in the public's memory aging knowledge is a promising avenue for improving our understanding of Alzheimer's disease (AD) risk and developing effective education and intervention strategies. Yet, it is important to contextualize knowledge within culturally relevant frameworks that may not only contribute to the extent of individuals' knowledge, but also to the reception of developed educational and intervention programs.ObjectiveOne prominent culture within the United States (U.S.) that has largely been neglected within AD research is the culture of honor, or thos

The global rise in dementia necessitates scalable cognitive assessments that can evolve to serve both clinical and research applications. We present the Oxford Cognitive Testing Portal (OCTAL), a remote, browser-based platform providing performance metrics for memory, attention, visuospatial and executive function domains. Four validation studies (N = 1664) confirmed cross-cultural applicability, lifespan sensitivity and clinical utility. Task performance was equivalent in English- and Chinese-speaking younger adults and mapped domain-specific ageing trajectories in mid- to late-adulthood. In

BACKGROUND: Cognitive–exercise dual-task training has been shown to enhance cognitive function through mechanisms such as suppression of chronic inflammation, reduction of oxidative stress, and enhancement of synaptic plasticity. However, the precise mechanisms underlying the ability of dual-task training to delay aging-related cognitive decline remain incompletely understood. METHODS: Aged male C57BL/6J mice were subjected to a 12-week intervention program consisting of cognitive training, exercise, or cognitive–exercise dual-task training. Cognitive and physical function were assessed using

Introduction: This study aims to investigate the relationship between olfactory identification (OI) and cognitive impairment by examining OI abilities across various stages of cognitive deterioration. Methods: A total of 264 participants were divided into three groups based on cognitive status: cognitively healthy, subjective cognitive, and mild cognitive impairment. All participants were assessed using the Sniffin' Sticks Olfactory Identification test and a comprehensive neuropsychological test battery. Results: Our results highlight the main effects of age and cognitive status on OI scores.

BACKGROUND: Aging changes whole-body anthropometry. Accelerated anthropometric aging can be operationalized as the age gap between the anthropometric-predicted age and the chronological age (AAG). What disease conditions affect AAG and subsequent functional deficits remains unclear.
METHODS: In 302 Baltimore Longitudinal Study of Aging participants (mean age=71.7 years, 58% women, 30% Black), we examined the associations between AAG and disease conditions using multivariable linear regression, adjusted for age, sex, and race, and further tested whether AAG would mediate the association betwee

Among the many changes associated with aging, inflammation in the central nervous system (CNS) and throughout the body likely contributes to the constellation of health-related maladies associated with aging. Genetics, lifestyle factors, and environmental experiences shape the trajectory of aging-associated inflammation, including the developmental timing, frequency, and intensity of alcohol consumption. This chapter posits that neuroinflammatory processes form a critical link between alcohol exposure and the trajectory of healthy aging, at least in part through direct or indirect interactions

Word-finding difficulty (WFD) is a common cognitive complaint in aging, manifesting both in natural speech and in controlled laboratory tests. Various theories of cognitive aging have addressed WFD, and understanding its underlying mechanisms can help to clarify whether it has diagnostic value for neurodegenerative disease. Two influential "information-universal" theories attribute it to rather broad changes in cognition. The processing speed theory posits a general slowdown of all cognitive processes, while the inhibitory deficit hypothesis (IDH) predicts a specific problem in suppressing irr

Adults with Down syndrome (DS) experience high risk for Alzheimer's disease (AD), but there is variability in the timing of transition from a cognitively stable state to prodromal AD and dementia. The present study examined the association between a modifiable lifestyle factor, employment complexity, and cognitive decline across two time points in adults with DS. Employment complexity, defined as the degree of problem-solving or critical thinking required for employment activities, was operationalized using the Dictionary of Occupational Titles, a system which classifies occupations based on t

Dementia is a growing public health concern, and African Americans and Latinos are disproportionately affected compared to White Americans. Improving cognitive health outcomes and reducing disparities requires a diverse, interdisciplinary workforce. The US Centers for Disease Control and Prevention's Healthy Brain Research Network (HBRN) Scholars Program trained racially/ethnically and gender-diverse scholars through mentored, collaborative research. Entry, exit, and alumni surveys and a Scholar Spotlight Series queried motivation, goals, acquired skills, accomplishments, program impact, and s

While several cognitive domains have been widely investigated in the field of aging, the age-related effects on tool use are still an open issue and hardly any studies on tool use and aging is available. A significant body of literature has indicated that tool use skills might be supported by at least two different types of knowledge, namely, mechanical knowledge and semantic knowledge. However, neither the contribution of these kinds of knowledge to familiar tool use, nor the effects of aging on mechanical and semantic knowledge have been explored in normal aging. The aim of the present study
