CXCL12-induced rescue of cortical dendritic spines and cognitive flexibility.

Synaptodendritic pruning is a common cause of cognitive decline in neurological disorders, including HIV-associated neurocognitive disorders (HAND). HAND persists in treated patients as a result of chronic inflammation and low-level expression of viral proteins, though the mechanisms involved in synaptic damage are unclear. Here, we report that the chemokine CXCL12 recoups both cognitive performance and synaptodendritic health in a rodent model of HAND, which recapitulates the neuroinflammatory state of virally controlled individuals and the associated structural/functional deficiencies. CXCL1

In Medio Stat Virtus: Moderate Cognitive Flexibility as a Key to Affective Flexibility Responses in Long-Term HRV.

This study examines the relationship between cognitive and affective flexibility, two critical aspects of adaptability. Cognitive flexibility involves switching between activities as rules change, assessed through task-switching or neuropsychological tests and questionnaires. Affective flexibility, meanwhile, refers to shifting between emotional and non-emotional tasks or states. Although similar conceptually, prior research shows inconsistent findings on their link, especially regarding physiological and self-reported measures. Affective flexibility was operationalized as changes in heart rat

Altered Insula Functional Connectivity Correlates to Cognitive Flexibility in Insomnia.

This study aimed to investigate the impaired cognitive flexibility and its underlying neural mechanisms in insomnia. By combining resting-state fMRI and the Cognitive Flexibility Inventory (CFI), we examined the associations between insomnia severity, spontaneous brain activity (the fractional amplitude of low-frequency fluctuations, fALFF) and functional connectivity (FC) with total cognitive flexibility scores. Behavioral results showed that insomnia severity significantly affected the control sub-dimension of cognitive flexibility. The fALFF analyses indicated that the right insula (Ins) wa

How to Improve Cognitive Flexibility: Evidence From Noninvasive Neuromodulation Techniques.

BACKGROUND: Cognitive flexibility (CF) is a core component of executive function that enables humans to adaptively process and respond effectively to diverse and dynamic contextual needs. The execution of CF-related tasks depends on the collective interaction of various neural circuits in essential brain regions, such as the prefrontal, anterior cingulate, and posterior parietal cortices, which support the harmonic and smooth transition of thoughts and perspectives. Therefore, clarifying the mechanism of CF and introducing non-invasive neuromodulatory methods that enhance CF provides a way for

Cognitive flexibility in a Tanganyikan bower-building cichlid, Aulonocranus dewindti.

Cognitive flexibility, the ability to modify one's decision rules to adapt to a new situation, has been extensively studied in many species. In fish, though, data on cognitive flexibility are scarce, especially in the wild. We studied a lekking species of cichlid fish in Lake Tanganyika, Aulonocranus dewindti. Males create sand bowers as spawning sites and maintain them by removing any objects falling into it. In the first part of our experiment, we investigated the existence of spontaneous decision rules for the maintenance of the bowers. We showed that if a snail shell and a stone are placed

Drug reinforcement impairs cognitive flexibility by inhibiting striatal cholinergic neurons.

Addictive substance use impairs cognitive flexibility, with unclear underlying mechanisms. The reinforcement of substance use is mediated by the striatal direct-pathway medium spiny neurons (dMSNs) that project to the substantia nigra pars reticulata (SNr). Cognitive flexibility is mediated by striatal cholinergic interneurons (CINs), which receive extensive striatal inhibition. Here, we hypothesized that increased dMSN activity induced by substance use inhibits CINs, reducing cognitive flexibility. We found that cocaine administration in rodents caused long-lasting potentiation of local inhib

The zinc finger/RING domain protein Unkempt regulates cognitive flexibility.

Correct orchestration of nervous system development is a profound challenge that involves coordination of complex molecular and cellular processes. Mechanistic target of rapamycin (mTOR) signaling is a key regulator of nervous system development and synaptic function. The mTOR kinase is a hub for sensing inputs including growth factor signaling, nutrients and energy levels. Activation of mTOR signaling causes diseases with severe neurological manifestations, such as tuberous sclerosis complex and focal cortical dysplasia. However, the molecular mechanisms by which mTOR signaling regulates nerv

Tsc1-mTORC1 signaling controls striatal dopamine release and cognitive flexibility.

Tuberous Sclerosis Complex (TSC) is a neurodevelopmental disorder caused by mutations in TSC1 or TSC2, which encode proteins that negatively regulate mTOR complex 1 (mTORC1). TSC is associated with significant cognitive, psychiatric, and behavioral problems, collectively termed TSC-Associated Neuropsychiatric Disorders (TAND), and the cell types responsible for these manifestations are largely unknown. Here we use cell type-specific Tsc1 deletion to test whether dopamine neurons, which modulate cognitive, motivational, and affective behaviors, are involved in TAND. We show that loss of Tsc1 an

The acquisition of robust and flexible cognitive skills.

The authors introduce a model of skill acquisition that incorporates elements of both traditional models and models based on embedded cognition by striking a balance between top-down and bottom-up control. A knowledge representation is used in which pre- and postconditions are attached to actions. This model captures improved performance due to learning not only in terms of shorter solution times and lower error rates during the task but also in an increased flexibility to solve similar problems and robustness against unexpected events. In 3 experiments using a complex aviation task, the autho

Distinct neural oscillations predict individual differences in feedback-guided cognitive flexibility.

Cognitive flexibility is the ability to keep or update contextual cognitive representations. The underlying neural mechanisms and how it is influenced by individual sensitivity to feedback are not known. We measured cognitive flexibility under uncertainty with the Wisconsin Card Sorting Test and brain activity with magnetoencephalography. Cognitive flexibility had large interindividual variability as indexed by the learning speed. Using a behavioral sequential learning model, we show that this variability in cognitive flexibility is predicted by individual sensitivity to feedback and anticipat

Mineralocorticoid receptor signaling in GABAergic neurons regulates stress-induced cognitive flexibility.

The mineralocorticoid receptor (MR) plays a pivotal role in modulating the neuroendocrine stress response and cognitive function. While recent evidence highlights the importance of MRs in glutamatergic neurons in regulating anxiety-like behavior, the specific contribution of MRs within inhibitory networks remains incompletely understood. To address this gap, we generated a mouse model with targeted ablation of MR in forebrain GABAergic neurons (MRDlx). Comprehensive behavioral profiling revealed a profound, state-dependent cognitive phenotype in male MRDlx mice. Under non-stressful baseline co

Frontostriatal Circuits Alterations Associated with Cognitive Flexibility Deterioration in Huntington's Disease.

BACKGROUND: Recent resting-state functional magnetic resonance imaging studies have reported abnormal functional connectivity (FC) in the prefrontal cortex (PFC)-striatum circuit in patients with premanifest Huntington's disease (HD). However, there is a lack of evidence showing persistence of abnormal frontostriatal FC and its relation to cognitive flexibility performance in patients with clinically manifest HD. OBJECTIVE: The aim of this study was to evaluate the resting-state FC integrity of the frontostriatal circuit and its relation to cognitive flexibility in HD patients and healthy con

Catecholaminergic modulation of indices of cognitive flexibility: A pharmaco-tDCS study.

BACKGROUND: Dopaminergic activity within the dorsolateral prefrontal cortex (dlPFC) has been implicated in the control of cognitive flexibility. Much of the evidence for a causative relationship between cognitive flexibility and dopamine has come from animal studies, whilst human data have largely been correlational. OBJECTIVE/HYPOTHESIS: The current study examines whether changes in dopamine levels through tyrosine administration and suppression of dlPFC activity via cathodal tDCS could be causally related to cognitive flexibility as measured by task switching and reversal learning. Neural basis of impaired cognitive flexibility in patients with anorexia nervosa.

BACKGROUND: Impaired cognitive flexibility in anorexia nervosa (AN) causes clinical problems and makes the disease hard to treat, but its neural basis has yet to be fully elucidated. The purpose of this study was to evaluate the brain activity of individuals with AN while performing a task requiring cognitive flexibility on the Wisconsin Card Sorting Test (WCST), which is one of the most frequently used neurocognitive measures of cognitive flexibility and problem-solving ability. METHODS: Participants were 15 female AN patients and 15 age- and intelligence quotient-matched healthy control wom

Effects of milnacipran on cognitive flexibility following chronic stress in rats.

Cognitive dysfunction is a component of affective disorders, including depression. Chronic stress is a risk factor for depression, and we have shown that exposing rats to chronic unpredictable stress (CUS) induces a deficit of cognitive flexibility, the ability to modify behavior based on feedback from a changing environment. Deficits of cognitive flexibility, measured by extra-dimensional set-shifting on the Attentional Set-shifting Test (AST), are consistent with dysregulation of prefrontal cortical function, also characteristic of depression. We have shown that increasing norepinephrine in
