Cellular sensitivity to native type IV collagen in primary biliary cirrhosis.

We examined 60 patients with various chronic liver diseases for cellular sensitivity to native human type I and IV collagens using an in vitro leucocyte migration inhibition test. Mononuclear cells from 7 (33%) of 21 patients with chronic active hepatitis, 14 (52%) of 27 patients with liver cirrhosis and 11 (92%) of 12 patients with primary biliary cirrhosis exhibited cellular sensitivity to type IV collagen, although cells from almost all patients responded to type I collagen. None except one for type I collagen of 25 normal controls showed positive response to both collagens. In chronic acti

Osteogenesis imperfecta type IV: evidence of abnormal triple helical structure of type I collagen.

Skin fibroblasts from a patient with mild osteogenesis imperfecta (OI) type IV synthesize two populations of type I procollagen molecules. One population contains pro alpha 1(I) and pro alpha 2(I) chains that migrate normally in sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and a second population contains only slower migrating pro alpha 1(I) and pro alpha 2(I) chains. The total amount of type I procollagen made by OI cells and the ratio of pro alpha 1(I):pro alpha 2(I) is normal. When labeled under conditions that inhibit post-translational modification of pro alpha cha

Immunohistochemical studies comparing the localization of type XV collagen in normal human skin and skin tumors with that of type IV collagen.

We investigated the localization of type XV collagen in normal human skin and skin tumors by immunohistochemical methods using a monoclonal antibody against the recombinant polypeptide of the non-collagenous region of the alpha1 chain of murine type XV collagen. Type XV collagen was localized in the dermo-epidermal, perivascular, and perineural basement membrane zones in normal skin. While this localization appeared to be similar to that of type IV collagen, detailed observation revealed that its localization was distinct in fact from that of type IV collagen which was thin and linear in appea

iPSC-derived type IV collagen α5-expressing kidney organoids model Alport syndrome.

Alport syndrome (AS) is a hereditary glomerulonephritis caused by COL4A3, COL4A4 or COL4A5 gene mutations and characterized by abnormalities of glomerular basement membranes (GBMs). Due to a lack of curative treatments, the condition proceeds to end-stage renal disease even in adolescents. Hampering drug discovery is the absence of effective in vitro methods for testing the restoration of normal GBMs. Here, we aimed to develop kidney organoid models from AS patient iPSCs for this purpose. We established iPSC-derived collagen α5(IV)-expressing kidney organoids and confirmed that kidney organoid

[Collagen type IV nephropathy: from thin basement membrane nephropathy to Alport syndrome].

INTRODUCTION: Collagen type IV nephropathy includes the Goodpasture syndrome, thin basement membrane nephropathy and the Alport syndrome. Mutations in the coding Col(IV)A3/A4 and Col(IV)A5 genes are probable causes of the latter two. Thin basement membrane nephropathy is mostly familial and has an autosomal dominant inheritance, at least 40% of the families have hematuria that co-segregates with the Col(IV)A3 and/or Col(IV)A4 loci. 85% of Alport syndrome cases are transmitted as an X-linked semidominant form due to Col(IV)A5 mutations. About 14% of Alport syndrome cases exhibit autosomal reces

Role of distinct type IV collagen networks in glomerular development and function.

BACKGROUND: In X-linked Alport syndrome, mutations in the COL4A5 gene encoding the alpha 5 chain of type IV collagen result in progressive renal failure. This nephropathy appears to relate to the arrest of a switch from an alpha 1/alpha 2 to an alpha 3/alpha 4/alpha 5 network of type IV collagen in the developing glomerular basement membrane (GBM; Kalluri et al, J Clin Invest 99:2470, 1997). METHODS: We examined the role of this switch in glomerular development and function using a canine model of X-linked nephritis with a COL4A5 mutation. The electron microscopic appearance and the expressio

Amniotic fluid laminin and type IV collagen in normal and pathological pregnancies.

Specific radioimmunoassays for the fragment P1 of human laminin and the 7-S collagen domain of human type IV collagen were used to quantify these basement membrane proteins in second trimester amniotic fluid samples from 21 normal and 41 pathological pregnancies, the latter group being defined by elevated amniotic fluid alpha-foetoprotein (AFP) or abnormal foetal karyotype, or both. The mean laminin P1 concentration in the normal 15 to 18-wk pregnancies was 36 micrograms/l (range 10-77) and that of the 7-S collagen was 46 micrograms/l (range 7-152). Lung collagen in type IV Ehlers-Danlos syndrome: ultrastructural and biochemical studies.

A patient with type IV Ehlers-Danlos syndrome developed recurrent pneumothoraces. At operation for poudrage of the pleura, an apical bulla was resected. Biochemical analysis of the lung tissue revealed increased solubilization of collagen by acetic acid extraction and by pepsin proteolysis. The relative proportion of type III collagen in the pepsin-solubilized collagen fraction was markedly decreased. Fibroblasts cultured from the patient's lung produced less type III procollagen relative to type I procollagen than control lung fibroblasts. Electron microscopic examination of the lung tissue s

Immunohistochemical localization of type IV collagen and laminin (alpha1) in denture stomatitis.

Chronic clinical irritation of the palatal mucosa by dentures involves a series of histological changes in epithelial and connective tissues, inflammatory cells and the vasculature. No single change is pathognomic of this inflammatory process. The rupture of basement membrane associated with the development of denture stomatitis often marks an important stage. This study investigated modifications in basement membrane organisation, especially the distribution of type IV collagen and a specific laminin chain (alpha1), during denture stomatitis. Biopsies of palatal mucosa were obtained from 12 p

Rat amnion type IV collagen composition and metabolism: implications for membrane breakdown.

We report here that rat amnion type IV collagens are composed primarily of alpha1(IV) and alpha2(IV) chains. Amnion basement membrane collagens were more sensitive to degradation by collagenases than were adult rat kidney basement membrane collagens, which are enriched in alpha3(IV), alpha4(IV), and alpha6(IV) chains. Amnion type IV collagen content per unit of protein was markedly reduced by Day 21 of pregnancy, the day of delivery. Increased amnion levels of matrix metalloproteinase (MMP)-2 and MMP-9, gelatinases that degrade type IV collagen, were found by Day 21, suggesting that collagen b

Decreased type IV collagen expression by human decidual tissues in spontaneous abortion.

To provide some insight into the etiology of spontaneous abortion, the expression of type IV collagen was investigated in human decidual tissues obtained after spontaneous abortion (n = 17) and normal pregnancy (n = 22). Indirect immunofluorescent staining was performed for type I, III, and IV collagen as well as laminin, and Northern blot analysis was conducted to assess the expression of messenger ribonucleic acid for the alpha 1(IV) chain. Immunohistochemical analysis did not reveal any significant differences between normal pregnancy and spontaneous abortion with respect to interstitial co

Autosomal recessive Alport syndrome: immunohistochemical study of type IV collagen chain distribution.

Alport syndrome (AS) is an hereditary disease of basement membrane collagen. It is mainly transmitted as a dominant X-linked trait and caused by mutations in the COL4A5 gene encoding the alpha 5 chain of type IV collagen. However, autosomal recessive AS due to mutations in the COL4A3 or COL4A4 genes could represent up to 15% of AS. Using the immunofluorescence technique, we analyzed the distribution of the different chains of type IV collagen in renal (12 specimens) and skin (4 specimens) basement membranes of 12 AS patients belonging to 11 unrelated kindreds in which autosomal recessive inher

Characterization of antibodies to basement membrane (type IV) collagen in immunohistological studies.

Antisera were produced in rabbits and guinea pigs against basement membrane (type IV) collagens which were extracted from a mouse tumor with acetic acid and from human placenta after limited digestion with pepsin. The antisera were specific for type IV collagen and did not crossreact with collagens type I, II and III from interstitial connective tissue, with collagen type V (AB2) from placenta and with a non-collagenous protein (laminin) obtained from basement membranes. Purified antibodies against both human and type IV collagen reacted in indirect immunofluorescence tests with the mouse tumo

The Ultrastructural Localization of Type II, IV, and VI Collagens at the Vitreoretinal Interface.

BACKGROUND: The vitreoretinal interface is the border of the cortical vitreous and the inner surface of the retina. The adhesion of the cortical vitreous to the ILM, namely vitreoretinal adhesion, involves a series of complex molecular adhesion mechanisms and has been considered as an important pathogenic factor in many vitreoretinal diseases. The presence of type VI collagen at the vitreoretinal interface and its possible interaction with collagens and glycoproteins indicates that type VI collagen may contribute to the vitreoretinal adhesion. PURPOSE: To clarify the ultrastructural location
