Measurement of menadione-mediated DNA damage in human lymphocytes using the comet assay.

The model quinone compound menadione has been used to study the effects of oxidative stress in mammalian cells, and to investigate the mechanism of action of the quinone nucleus which is present in many anti-cancer drugs. We have used the alkaline single cell gel electrophoresis assay (comet assay) to investigate the effects of low doses of this compound on isolated human lymphocytes. We found that concentrations of menadione as low as 1 microM were sufficient to induce strand breaks in these cells. Pre-incubation with the NAD(P)H quinone oxidoreductase inhibitor dicoumarol, enhanced the produ

Biomonitoring study of dry cleaning workers using cytogenetic tests and the comet assay.

Perchloroethylene (PCE) is the main solvent used in the dry cleaning industry worldwide. The aim of the present work was to evaluate the genotoxic potential of occupational exposure to PCE in dry cleaning workers. The study was carried out in 59 volunteers (30 workers, 29 controls). The genotoxic effect was evaluated by analyzing chromosome aberrations (CAs), and micronuclei (MN) and DNA damage (assessed by the comet assay) in peripheral blood lymphocytes. Environmental monitoring of exposure was carried out on personal breathing zone air samples collected during two consecutive working days b

Reducing risk of false positives in the in vivo comet assay and improving result reliability.

The risk of generating false positive in vivo comet assay results can be increased when procedural bias and/or technical variability is poorly controlled. This has been an ongoing concern since comet was first introduced into regulatory safety testing. But the proprietary nature of regulated studies and the 3Rs have limited the ability to conduct and publish the comparative in vivo studies necessary to determine the effect these factors can have on comet assay results when substances other than well characterized positive control compounds are evaluated in multiple tissues. That changed when H

DNA damage in lymphocytes of Indian rickshaw pullers as measured by the alkaline Comet assay.

Rickshaw pullers (RPs) engage in strenuous physical activity and are exposed to the air pollutants found in urban environments. Air pollutants and the reactive oxygen species generated by the physical activity both potentially can damage DNA. In the present study, the Comet assay, a sensitive tool for measuring DNA damage in single cells, was used to study genomic DNA damage in lymphocytes of Indian RPs. The study evaluated DNA damage in 118 healthy male volunteers, including 63 RPs whose work demanded high levels of physical activity for 7-9 hr/day, and 55 controls matched for age, habits, so

Development and validation of a modified comet assay to phenotypically assess nucleotide excision repair.

There is an increasing need for simple and reliable approaches to phenotypically assess DNA repair capacities. Therefore, a modification of the alkaline comet assay was developed to determine the ability of human lymphocyte extracts to perform the initial steps of the nucleotide excision repair (NER) process, i.e. damage recognition and incision. Gel-embedded nucleoids from A549 cells, pre-exposed to 1 microM benzo[a]pyrene-diol-epoxide, were incubated with cell extracts from frozen or freshly isolated lymphocytes. The rate at which incisions are introduced and the subsequent increase in tail

A quantitative comet assay: imaging and analysis of virus plaques formed with a liquid overlay.

Although the plaque assay defines a "gold-standard" for measuring virus infectivity, its reliance on plaque counting limits its sensitivity. When the assay is performed with a liquid overlay, instead of agar overlay, spontaneous flows can promote a uni-directional spread of infection, creating elongated regions of cytopathology that resemble comets. As a model system comet and plaque cultures of vesicular stomatitis virus (VSV) on baby hamster kidney (BHK-21) cells were compared. Host-cell monolayers were infected with VSV particles, incubated 15 h in the presence of liquid or agar overlays an

Modulation by flavonoids of the effects of a food mutagen in different thalassaemia genotypes in the Comet assay.

Thalassaemia is an inherited group of disorders caused by a reduction or total absence of one or more of the globin chains of the haemoglobin molecule. It has been shown that lymphocytes isolated from a sickle/beta thal double heterozygote-sickle phenotype patient showed increased sensitivity to the dietary food mutagen 3-amino-1-methyl-5H-pyridol(4,3-b)indole (Trp-P-2) when compared to the control. Furthermore, when a combination of Trp-P-2 with either quercitin or kaempferol was compared, the responses to Trp-P-2 were reduced to untreated control levels at the highest doses of quercitin and

Phenobarbital, oxazepam and Wyeth 14,643 cause DNA damage as measured by the Comet assay.

Although phenobarbital, oxazepam and Wyeth 14,643 are carcinogens that do not form DNA adducts, they induce mutations in the Big Blue transgenic mouse model. The mutations produced by these compounds were predominantly G-->T and G-->C transversions that we suspect arose from oxidative damage to DNA. To test this, we employed the single cell electrophoresis (Comet) assay that detects alkali-labile lesions in cells sustaining DNA damage. Human myeloid leukemia K562 cells were treated with non-cytotoxic doses of the above compounds for 3 h, then placed on slides containing low melting point agaro

Measurement of DNA damage and repair capacity as a function of age using the comet assay.

A large number of studies indicate that DNA damage and mutation increase with age in human cells and tissues (1). Age-related degenerative disorders in which DNA damage has been invoked include heart disease and neurodegenerative conditions such as Alzheimer's disease, amyotrophic lateral sclerosis, or Parkinson's disease (2, 3). Patients with deficiencies in DNA repair, including xeroderma pigmentosum (XP) (4) and ataxia-telangiectasia (A-T) (5) show characteristic patterns of neurodegeneration (as opposed to a failure of normal development). The implication is that failure of repair can lead

The bromodeoxyuridine comet assay: detection of maturation of recently replicated DNA in individual cells.

The single-cell gel electrophoresis (Comet) assay is a relatively simple method of measuring DNA single strand breaks and alkali-labile sites in individual cells. We have combined this with bromodeoxyuridine (BrdUrd) labeling of DNA and immunolocalization of the BrdUrd to assess DNA replicative integrity on a single-cell basis. We show that the existence of strand discontinuities in recently replicated domains of DNA, caused during semiconservative replication or exacerbated by the arrest of replicative polymerases at UV irradiation- or chemical-induced lesions, can be detected in individual c

A comparative genotoxicity study of agrochemicals: nuclear abnormalities, comet assay, and gene expression alterations.

Agrochemicals (AGs) are known for their ability to have a negative impact on the health of non-target species, despite the fact that they are meant to protect agricultural plants from harmful pests. Catla catla (Hamilton, 1822) gill cells (ICG) were exposed to four AGs: insecticide (Imidacloprid (IMI)), fungicide (Curzate (CZ)), herbicide (pyrazosulfuron ethyl (PE)), and fertilizer micronutrients (MN) with sublethal concentrations 1/20th, 1/10th, and 1/5th of IC50, described here as low dose (LD), medium dose (MD), and high dose (HD), respectively. A significant dose-dependent increase in the

Radiation-induced apoptosis measured in TK6 human B lymphoblast cells using the comet assay.

The comet assay, a sensitive method of measuring DNA strand breaks in individual cells, is also capable of identifying apoptotic cells which contain highly fragmented DNA. This method requires embedding cells in agarose, lysing cells to remove proteins, and providing a brief exposure to an electric field to allow broken pieces of DNA to migrate. TK6 human B lymphoblast cells undergo fragmentation which is dependent on both time after irradiation and radiation dose. While some TK6 cells undergo apoptosis within 2 h after irradiation, the fragmentation rate increases approximately 10 h after exp

Optimizing the Comet Assay-Based In Vitro DNA Repair Assay for Placental Tissue: A Pilot Study with Pre-Eclamptic Patients.

The comet assay-based in vitro DNA repair assay has become a common tool for quantifying base excision repair (BER) activity in human lymphocytes or cultured cells. Here, we optimized the protocol for studying BER in human placental tissue because the placenta is a non-invasive tissue for biomonitoring of early-life exposures, and it can be used to investigate molecular mechanisms associated with prenatal disorders. The optimal protein concentration of placental protein extracts for optimal damage recognition and incision was 2 mg protein/mL. The addition of aphidicolin did not lead to reduced
