Identification of plasma biomarkers in lipid metabolism for accurate prediction of Alzheimer's disease.

Metabolomics may reveal non-invasive biomarkers for early diagnosis in Alzheimer's disease (AD) and provide new insights into the disease mechanisms to develop effective treatments. Here, we comprehensively analyzed the blood plasma metabolomes from a Chinese cohort of 447 individuals, including 188 AD, 181 MCI (mild cognitive impairment), and 78 NC (normal control). Differential analysis identified altered metabolites, followed by forward feature selection to prioritize a panel of key metabolites, and construction of a diagnostic model using logistic regression. Key metabolite-enriched pathwa

Integrated serum metabolomics and MRI biomarkers for MASLD: a practical framework for noninvasive staging and monitoring.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) affects nearly one in four adults globally, yet early diagnosis remains challenging due to the limitations of current diagnostic modalities. While liver biopsy is the reference standard, its invasiveness limits its utility for population screening. Similarly, single-modality noninvasive tests often fail to distinguish inflammation from fibrosis or detect early metabolic injury. This review synthesized recent evidence (2020-2025) regarding the integration of serum metabolomic profiling with quantitative magnetic resona

The FEMA GRAS assessment of aromatic substituted secondary alcohols, ketones, and related esters used as flavor ingredients.

This publication is the 11th in a series of safety evaluations performed by the Expert Panel of the Flavor and Extract Manufacturers Association (FEMA). In 1993, the Panel initiated a comprehensive program to re-evaluate the safety of more than 1700 GRAS flavoring substances under conditions of intended use. The list of GRAS substances has now grown to more than 2100 substances. Elements that are fundamental to the safety evaluation of flavor ingredients include exposure, structural analogy, metabolism, pharmacokinetics and toxicology. Flavor ingredients are evaluated individually and in the c

Evolutionary metabolic landscape from preneoplasia to invasive lung adenocarcinoma.

Metabolic reprogramming evolves during cancer initiation and progression. However, thorough understanding of metabolic evolution from preneoplasia to lung adenocarcinoma (LUAD) is still limited. Here, we perform large-scale targeted metabolomics on resected lesions and plasma obtained from invasive LUAD and its precursors, and decipher the metabolic trajectories from atypical adenomatous hyperplasia (AAH) to adenocarcinoma in situ (AIS), minimally invasive adenocarcinoma (MIA) and invasive adenocarcinoma (IAC), revealing that perturbed metabolic pathways emerge early in premalignant lesions. Targeted plasma metabolomics reveals potential biomarkers of the elderly with mild cognitive impairment in Qingdao rural area.

INTRODUCTION: Previous research has suggested a link between the onset of Alzheimer's disease (AD) and metabolic disorder; however, the findings have been inconsistent. To date, the majority of metabolomics studies have focused on AD, resulting in a relative paucity of research on early-stage conditions such as mild cognitive impairment (MCI) underexplored. In this study, we employed a comprehensive platform for the early screening of individuals with MCI using high-throughput targeted metabolomics. METHOD: We included 171 participants including 124 individuals with MCI and 47 healthy subject

A Case Report of a 61-Year-Old Woman With Jaundice and Cholelithiasis Presenting With Autoimmune Hepatitis.

Autoimmune hepatitis (AIH) is a complex disease with a chronic cell-mediated immunologic process against healthy liver cells. The clinical presentation can vary since the exact cause of AIH is multifactorial. Here, we report a case of a 61-year-old woman with a past medical history of post-traumatic stress disorder and hypothyroidism who presented clinically with diffuse abdominal distention, nausea, vomiting, jaundice, rash on the torso and legs, and tea-burnt orange urine. The patient underwent an initial workup with a complete blood count (CBC), comprehensive metabolic panel (CMP), and gamm

Use of remdesivir in the presence of elevated LFTs for the treatment of severe COVID-19 infection.

An 82-year-old man with an extensive medical history presented to the emergency room with complaints of generalised weakness and cough. He tested positive for COVID-19 10 days prior to presenting to the emergency room. Although his symptoms started a week prior to diagnosis, his weakness increased, warranting emergency response. A comprehensive metabolic panel was drawn from the patient on admission, indicating markedly high liver function tests (LFTs) ≥20 times above the upper limit of normal. On day 1 of admission, the decision was still made to start remdesivir (5-day course) due to decompe

Stage-specific lipidomic signatures as biomarkers in progression of cervical cancer.

Background and objectives Lipidomics has advanced cancer research by uncovering biomarkers that reveal key mechanisms of tumourigenesis. By analysing endpoint lipid metabolites influenced by genetic and environmental factors, the lipidome closely reflects a cell's functional state. Mass spectrometry, strengthened by modern bioinformatics and biostatistics, now enables precise detection of cancer-related lipid changes, motivating us to adopt this approach. Methods We have applied advanced mass spectrometry and machine-learning methods to develop a novel tool for cervical cancer screening, the s

Targeted metabolomics in PCOS mice identifies hippuric acid as a therapeutic metabolite.

Polycystic ovary syndrome (PCOS) represents a complex endocrine disorder with profound detrimental effects on women's reproductive and metabolic health, yet the absence of specific pharmacotherapeutic interventions has resulted in a significant therapeutic gap. To elucidate the metabolic alterations in PCOS, we conducted a comprehensive targeted metabolomics analysis of plasma samples from healthy control mice and PCOS model mice, examining a panel of 197 metabolites spanning diverse metabolic classes. Our analysis revealed substantial metabolic differences between the groups, with organic aci

Characterization of the UDP glucuronosyltransferase activity of human liver microsomes genotyped for the UGT1A1*28 polymorphism.

The UGT1A1*28 polymorphism is known to correlate with altered clearance of bilirubin (Gilbert syndrome) and drugs such as 7-ethyl-10-[4-(1-piperidino)-1-piperidino] carbonyloxy camptothecin (CPT-11). Although this polymorphism is clinically relevant and leads to significant drug-related toxicity of CPT-11, in vitro tools to allow prediction of how it will affect the clearance of new chemical entities have not been completely developed. To allow a more complete assessment of whether new chemical entities will be affected by the UGT1A1*28 polymorphism, a panel of microsomes was prepared from 15
