Suicide represents an egregious threat to society despite major advancements in medicine, in part due to limited knowledge of the biological mechanisms of suicidal behavior. We apply a connectome predictive modeling machine learning approach to identify a reproducible brain network associated with suicidal ideation in the hopes of demonstrating possible targets for novel anti-suicidal therapeutics. Patients were recruited from an inpatient facility at The Menninger Clinic, in Houston, Texas (N = 261; 181 with active and specific suicidal ideation) and had a current major depressive episode and

Internet addiction symptomatology (IAS) is characterized by persistent and involuntary patterns of compulsive Internet use, leading to significant impairments in both physical and mental well-being. Here, a connectome-based predictive modeling approach was applied to decode IAS from whole-brain resting-state functional connectivity in healthy population. The findings showed that IAS could be predicted by the functional connectivity between prefrontal cortex with the cerebellum and limbic lobe and connections of the occipital lobe with the limbic lobe and insula lobe. The identified edges assoc

Recent advances in brain imaging have enabled non-invasive in vivo assessment of the fetal brain. Characterizing brain development in healthy fetuses provides baseline measures for identifying deviations in brain function in high-risk clinical groups. We examined 110 resting state MRI data sets from fetuses at 19 to 40 weeks' gestation. Using graph-theoretic techniques, we characterized global organizational features of the fetal functional connectome and their prenatal trajectories. Topological features related to network integration (i.e., global efficiency) and segregation (i.e., clustering

A better characterization of how an individual's brain is functionally organized will likely bring dramatic advances to many fields of study. Here we show a model-based approach toward characterizing resting state functional connectivity MRI (rs-fcMRI) that is capable of identifying a so-called "connectotype", or functional fingerprint in individual participants. The approach rests on a simple linear model that proposes the activity of a given brain region can be described by the weighted sum of its functional neighboring regions. The resulting coefficients correspond to a personalized model-b

This study investigated structural connectivity in Alternating Hemiplegia of Childhood (AHC) using advanced tractography and connectome analysis, as characterizing brain network alterations in this rare disorder may clarify its cognitive and motor impairments. This prospective monocentric study included 10 individuals with AHC (mean age 23.2 years) and 20 matched healthy controls. All participants underwent 3T MRI with 3D T1-weighted and high-angular resolution diffusion sequences. Brain structural networks were evaluated using global and nodal graph measures, modular organization, and network

Functional connectivity of the human brain changes through life. Here, we assemble task-free functional and structural magnetic resonance imaging data from 33,250 individuals at 32 weeks of postmenstrual age to 80 years from 132 global sites. We report critical inflection points in the nonlinear growth curves of the global mean and variance of the connectome, peaking in the late fourth and late third decades of life, respectively. After constructing a fine-grained, lifespan-wide suite of system-level brain atlases, we show distinct maturation timelines for functional segregation within differe

Following sensory deprivation, areas and networks in the brain may adapt and reorganize to compensate for the loss of input. These adaptations are manifestations of compensatory crossmodal plasticity, which has been documented in both human and animal models of deafness-including the domestic cat. Although there are abundant examples of structural plasticity in deaf felines from retrograde tracer-based studies, there is a lack of diffusion-based knowledge involving this model compared to the current breadth of human research. The purpose of this study was to explore white matter structural ada

There is strong evidence that the functional connectome is highly related to the white matter connectome in older children and adults, though little is known about structure-function relationships in early childhood. We investigated the development of cortical structure-function coupling in children longitudinally scanned at 1, 2, 4, and 6 years of age (N = 360) and in a comparison sample of adults (N = 89). We also applied a novel graph convolutional neural network-based deep learning model with a new loss function to better capture inter-subject heterogeneity and predict an individual's func

BACKGROUND: Despite increasing knowledge on the neuroimaging patterns of eating disorder (ED) symptoms in non-clinical populations, studies using whole-brain machine learning to identify connectome-based neuromarkers of ED symptomatology are absent. This study examined the association of connectivity within and between large-scale functional networks with specific symptomatic behaviors and cognitions using connectome-based predictive modeling (CPM).
METHODS: CPM with ten-fold cross-validation was carried out to probe functional networks that were predictive of ED-associated symptomatology, in

In the mature brain, structural and functional 'fingerprints' of brain connectivity can be used to identify the uniqueness of an individual. However, whether the characteristics that make a given brain distinguishable from others already exist at birth remains unknown. Here, we used neuroimaging data from the developing Human Connectome Project (dHCP) of preterm born neonates who were scanned twice during the perinatal period to assess the developing brain fingerprint. We found that 62% of the participants could be identified based on the congruence of the later structural connectome to the in

Although cortical thinning and atrophy are well documented in aging and neurodegenerative disease, the cortical structural features associated with preserved or superior cognition across typical adulthood remain incompletely characterized. We examined associations between cortical thickness and cognitive performance in a large, typically aging cohort and tested whether they vary by age and cognitive domain. We analyzed cross-sectional data from cognitively healthy adults in the Human Connectome Project-Aging/Aging Adult Brain Connectome, spanning middle (36-59 years), early older (60-79), and

INTRODUCTION: Mapping individual differences is crucial to improve personalized medicine approaches in Alzheimer's disease (AD), which is characterized by strong inter-individual variability in the accumulation patterns of tau and amyloid beta pathology.
METHODS: We assess the progression of AD across the disease continuum by building individual molecular connectomes using longitudinal positron emission tomography (PET) data.
RESULTS: We demonstrate that these connectomes constitute a unique fingerprint, capable of identifying a single individual from a large group of subjects. Alterations i

Mitochondria are integral to the metabolism and cell biology of a neuron. Electron microscopy images of fly brain volumes, taken for connectomics, can be analyzed for mitochondria as well as the cells and synapses already reported. Here, from the Drosophila Hemibrain connectome dataset, we extract, classify, and measure approximately 6 million mitochondria, with the majority located among more than 20 thousand neurons and over 5500 cell types. Each mitochondrion is annotated with its location, orientation, voxel size, and appearance (dark and dense, light and sparse, or medium), and each synap

INTRODUCTION: Brain function emerges from coordinated activity across anatomically connected regions, where structural connectivity (SC)-the network of white matter pathways-provides the physical substrate for functional connectivity (FC), defined as the correlated activity between brain areas. While structural and functional networks exhibit substantial overlap, their relationship involves complex, indirect mechanisms, including the dynamic interplay of direct and indirect pathways. To systematically untangle how structural architecture shapes functional patterns, this work aims to establish

BACKGROUND: Methamphetamine use disorder (MUD) is characterized by prominent psychological craving and its relapsing nature. Previous studies have linked trait impulsivity and abstinence duration to drug use, but the neural substrates of drug cue-induced craving and its relationship with these traits remain unclear in MUD.
METHODS: We acquired high-density resting-state electroencephalography (EEG) after participants watched a five-minute video demonstrating methamphetamine use. Combining precise source imaging to reconstruct brain activities with connectome predictive modeling (CPM), we buil

UNLABELLED: GABA B receptors (GABABRs) are heterodimeric seven-transmembrane receptors that interact with a range of proteins and form large protein complexes on cholesterol-rich membrane microdomains. As the brain ages, membrane cholesterol levels exhibit alterations, although it remains unclear how these changes impact protein-protein interactions and downstream signaling. Herein, we studied the structural bases for the interaction between GABABR and the KCC2 transporter, including their protein expression and distribution, and we compared data between young and aged rat cerebella. Also, we

Neural consequences of social disparities are not yet rigorously investigated. How socioeconomic conditions influence children's connectome development remains unknown. This paper endeavors to gauge how precisely the connectome structure of the brain can predict an individual's social environment, thereby inversely assessing how social influences are engraved in the neural development of the Adolescent brain. Utilizing Adolescent Brain and Cognition Development (ABCD) data (9099 children residing in the United States), we found that social conditions both at the household and neighborhood leve
