Hypersensitivity of Intrinsically Photosensitive Retinal Ganglion Cells in Migraine Induces Cortical Spreading Depression.

Migraine is a complex disorder characterized by episodes of moderate-to-severe, often unilateral headaches and generally accompanied by nausea, vomiting, and increased sensitivity to light (photophobia), sound (phonophobia), and smell (hyperosmia). Photophobia is considered the most bothersome symptom of migraine attacks. Although the underlying mechanism remains unclear, the intrinsically photosensitive retinal ganglion cells (ipRGCs) are considered to be involved in photophobia associated with migraine. In this study, we investigated the association between the sensitivity of ipRGCs and migr

Photothrombotic infarction triggers multiple episodes of cortical spreading depression in distant brain regions.

The purposes of this study were to determine whether cortical spreading depression occurs outside of the infarct produced by photothrombotic vascular occlusion, and also the direction of spreading. Focal cerebral thrombotic infarction was produced by irradiating the exposed skull of anesthetized rats with green light (560 nm) following systemic injection of rose bengal dye. At proximal sites (approximately 2 mm anterior to the infarct border), transient, severe hyperemic episodes (THEs) lasting 1-2 min were intermittently recorded. THE frequency was greatest in the first hour and declined over

Recurrent migraine aura-like symptoms in an elderly woman: symptomatic cortical spreading depression?

Cortical spreading depression (CSD) has been directly observed in humans with malignant stroke, traumatic brain injury and subarachnoid haemorrhage and is also considered to be the correlate of migraine aura. We report on a 76-year-old woman with new-onset episodes of headache, paraesthesia, hemiparesis and dysarthria, in whom a small cortical subarachnoid haemorrhage was diagnosed with MRI. Repeated diffusion-weighted MRI scans shortly after transient focal neurological episodes as well as diagnostic workup were normal, which makes recurrent transient ischaemic attacks unlikely. Chronic paracetamol treatment increases alterations in cerebral vessels in cortical spreading depression model.

Recently, a number of non-beneficial effects of chronic treatment with paracetamol (APAP) have been reported in several systems, including circulatory system. In this study, the effects of acute (1 hour) and chronic (30 days) APAP treatments on cerebral microvessels in a cortical spreading depression (CSD) migraine animal model were investigated. Rats were divided into control, CSD only, and APAP treatment with or without CSD groups. A single dose (200 mg/kg body weight) or once-daily APAP treatment over 30 days was intraperitoneally injected into the acute and chronic APAP treated groups, res

[Memory confinement versus generalization decrement on retrograde amnesia produced by cortical spreading depression].

An examination of memory confinement vs. generalization decrement under unilateral cortical spreading depression was undertaken using a licking suppression to a tone previously paired with a shock. In Experiment I, rats that were trained and tested under normal cortical conditions showed an impairment of memory when KCl-induced depression was given to an unilateral hemisphere within 15 min after the training. In Experiment II, where rats were trained under normal cortical conditions followed by unilateral depression and tested under unilateral cortical depression, the degree of impairment was

The effect of P2X7 antagonism on subcortical spread of optogenetically-triggered cortical spreading depression and neuroinflammation.

Migraine is a neurological disorder characterized by episodes of severe headache. Cortical spreading depression (CSD), the electrophysiological equivalent of migraine aura, results in opening of pannexin 1 megachannels that release ATP and triggers parenchymal neuroinflammatory signaling cascade in the cortex. Migraine symptoms suggesting subcortical dysfunction bring subcortical spread of CSD under the light. Here, we investigated the role of purinergic P2X7 receptors on the subcortical spread of CSD and its consequent neuroinflammation using a potent and selective P2X7R antagonist, JNJ-47965

Cortical spreading depression and multi-territorial cortical infarction following traumatic subdural hematoma: A case report and literature review.

BACKGROUND: Cortical spreading depression (CSD) has been recognized recently as a potential driver of secondary brain injury following traumatic brain injury and subdural hematoma (SDH). It can propagate in metabolically compromised cortical tissue, leading to the spread of ischemia and delayed infarction. CASE DESCRIPTION: The case is an 87-year-old man with atrial fibrillation on long-term anticoagulation who sustained an unwitnessed fall at home and was only found a day later. On presentation, he was alert but suffering from a profound left-sided hemiparesis, as well as significant dysarth

Cortical spreading depression aggravates early brain injury in a mouse model of subarachnoid hemorrhage.

Cortical spreading depression (CSD) has been observed during the early phase of subarachnoid hemorrhage (SAH). However, the effect of CSD on the cerebral blood flow (CBF) and cerebral oxyhemoglobin (CHbO) during the early phase of SAH has not yet been assessed directly. We, therefore, used laser speckle imaging and optical intrinsic sinal imaging to record CBF and CHbO during CSD and cerebral cortex perfusion (CCP) at 24 hours after CSD in a mouse model of SAH. SAH was induced by blood injection into the prechiasmatic cistern. When CSD occurred, the change trend of CBF and CHbO in Sham group a

Transient loss of interhemispheric functional connectivity following unilateral cortical spreading depression in awake rats.

OBJECTIVE: Growing evidence shows a critical role of network disturbances in the pathogenesis of migraine. Unilateral pattern of neurological symptoms of aura suggests disruption of interhemispheric interactions during the early phase of a migraine attack. Using local field potentials data from the visual and motor cortices, this study explored effects of unilateral cortical spreading depression, the likely pathophysiological mechanism of migraine aura, on interhemispheric functional connectivity in freely behaving rats. METHODS: Temporal evolution of the functional connectivity was evaluated

Sex-dependent cognitive and behavioral outcomes induced by repetitive cortical spreading depression in mice.

BACKGROUND: Migraine with aura is clinically linked to cognitive impairment, yet sex differences and underlying mechanisms are unclear. Cortical spreading depression (CSD) is widely regarded as the electrophysiological correlate of migraine aura. Therefore, we utilized a mouse model of repetitive CSD to model recurrent aura events and investigate how this phenomenon modulates behavior and cognition in a sex-dependent manner. METHODS: CSD was repeatedly induced in both male and female mice via epidural KCl application, using six consecutive stimuli at 20-minute intervals. Behavioral tests were

Small-molecule CGRP antagonist atogepant does not affect cortical spreading depression susceptibility in rats.

BACKGROUND: Atogepant is a small-molecule non-competitive calcitonin gene-related peptide (CGRP) receptor antagonist efficacious in preventing episodic migraine. Cortical spreading depolarization (CSD) is a slowly propagating depolarization wave underlying migraine aura. We investigated whether the efficacy of atogepant in migraine is mediated by CSD suppression. METHOD: We used Sprague-Dawley rats (n = 28 males, 28 females) to determine the electrical stimulation intensity threshold for CSD induction and the frequency of CSDs induced by continuous topical 1 M KCl application sequentially in

Somatosensory migraine auras evoked by bihemispheric cortical spreading depression events in human parietal cortex.

Cortical spreading depression (CSD) is associated with pronounced alterations in cerebral blood flow. These alterations can be captured using high-field functional magnetic resonance imaging (fMRI). While compelling clinical and experimental data suggest that CSD is involved in the pathogenesis of migraine aura, the mechanistic intricacies remain poorly understood. Here, we use visual stimulus-induced blood oxygen level-dependent (BOLD) fMRI responses to characterize spatiotemporal alterations in cerebral blood flow during spontaneous attacks with migraine aura. Six adult participants diagnose

Increased susceptibility to cortical spreading depression in an animal model of medication-overuse headache.

OBJECTIVE: The objective of this article is to evaluate electrically evoked thresholds for cortical spreading depression (CSD) and stress-induced activation of trigeminal afferents in a rat model of medication-overuse headache (MOH). METHODS: Sumatriptan or saline was delivered subcutaneously by osmotic minipump for six days to Sprague-Dawley rats. Two weeks after pump removal, animals were anesthetized and recording/stimulating electrodes implanted. The animals were pretreated with vehicle or topiramate followed by graded electrical stimulation within the visual cortex. CSD events were ident

Lateralized effects of vagus nerve stimulation on cortical spreading depression: insights from a mouse model.

OBJECTIVES: Vagus nerve stimulation (VNS) is increasingly recognized as a therapeutic approach for neurological disorders, such as epilepsy, migraine, and stroke rehabilitation. However, clinical outcomes vary considerably among individuals, underscoring the importance of elucidating the distinct effects of left versus right VNS. In this study, we aimed to investigate how lateralized VNS influences cortical spreading depression (CSD) in mice. MATERIALS AND METHODS: Left or right VNS and sciatic nerve stimulation (SNS) were delivered directly by electrodes placed around the vagus or sciatic ne

Transient Global Amnesia: An Electrophysiological Disorder Based on Cortical Spreading Depression-Transient Global Amnesia Model.

Transient global amnesia (TGA) is a benign memory disorder with etiologies that have been debated for a long time. The prevalence of stressful events before a TGA attack makes it hard to overlook these precipitating factors, given that stress has the potential to organically effect the brain. Cortical spreading depression (CSD) was proposed as a possible cause decades ago. Being a regional phenomenon, CSD seems to affect every aspect of the micro-mechanism in maintaining the homeostasis of the central nervous system (CNS). Corresponding evidence regarding hemodynamic and morphological changes

[The Effect of Cortical Spreading Depression Wave on EEG Spectral Power Anaesthesed and Conscious Rats].

EEG power changes in anaesthetized and conscious rats were studied (under repeated experiments) in wide frequency band (0.1-200 Hz) during cortical spreading depression wave (SD). In anaesthetized rats the decrease of EEG spectral power was shown through all diapasons under consideration. The most pronounced decay of the EEG power was marked in the 30-40 Hz band (27.3 ± 18.5, p = 2.46 x 10-(11)). In other frequency ranges the power decrease was less but its significance remained high. In conscious rats the simultaneous decay of the EEG power from 20 to 100 Hz range was also the most informativ

Impaired cerebrovascular reactivity after cortical spreading depression in rats: Restoration by nitric oxide or cGMP.

We investigated the role of the NO/cGMP system in the vasodilatory response to hypercapnia after cortical spreading depression (CSD) in barbiturate anesthetized rats in vivo. Regional cerebral blood flow (rCBF) was measured by laser Doppler flowmetry (LDF). Hypercapnia (arterial pCO2 50-60 mm Hg) increased rCBF by 2.8+/-1.0%/mm Hg (n = 34). Fifteen minutes after CSD, resting rCBF was reduced to 87%, and rCBF response to hypercapnia was abolished (p < 0.001, n = 28). Within 1 h after CSD, only little restoration of vascular reactivity occurred. Topical application of the NO-donors S-nitroso-N-a

Anti-migraine compounds fail to modulate the propagation of cortical spreading depression in the cat.

Leao's cortical spreading depression (SD) is often cited as the pathophysiological substrate for the neurological symptoms of migraine with aura. If this is the case it might be expected that drugs useful as anti-migraine agents, particularly those useful in prophylaxis, may alter or prevent SD. Indeep it has been suggested that the anti-migraine compound dihydroergotamine (DHE) blocks or reduces the speed of propagation of SD in the rabbit. In this study we attempted to further investigate the effects of DHE and other anti-migraine drugs on SD by measuring cortical blood flow with laser Doppl
