In Silico and In Vitro Assessment of Portuguese Oyster (Crassostrea angulata) Proteins as Precursor of Bioactive Peptides.

In this study, the potential bioactivities of Portuguese oyster (Crassostrea angulata) proteins were predicted through in silico analyses and confirmed by in vitro tests. C. angulata proteins were characterized by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and identified by proteomics techniques. Hydrolysis simulation by BIOPEP-UWM database revealed that pepsin (pH > 2) can theoretically release greatest amount of bioactive peptides from C. angulata proteins, predominantly angiotensin I-converting enzyme (ACE) and dipeptidyl peptidase IV (DPP-IV) inhibitory peptides,

Transcriptomic response and hydrocarbon accumulation in the eastern oyster (Crassostrea virginica) exposed to crude oil.

The adverse effect of crude oil on marine invertebrates is well known. To have a better understanding of its effects on marine invertebrates, Crassostrea virginica was exposed to different concentrations (50, 100 and 200 μg/L) of a mixture of super-light and light crude oil for two weeks, evaluating the transcriptomic response of the digestive gland using RNA-Seq and their accumulation in soft tissues. A total of 33,469,374 reads were assembled, which resulted in 61,356 genome assemblies ('Genes'). Trinotate was used for transcript annotation. At the end of this process, 86,409 transcripts wer

A review of the effects of bromate on aquatic organisms and toxicity of bromate to oyster (Crassostrea gigas) embryos.

A critical literature review has been undertaken of the effects that bromate (BrO3-) has on aquatic organisms. Chronic (multigeneration) studies using four marine phytoplankton species indicate a stimulation of cell division at concentrations up to 13.6 mg BrO3- liter-1, such that the EC50 is > 13.6 mg BrO3- liter-1 (exposure period unspecified). No published data were available for any freshwater algal or phytoplankton species. For freshwater invertebrates, the available data are limited to acute studies using two species: the water flea, Daphnia magna, with a 48-h LC50 of 179 mg BrO3- liter-

Recognition and function analysis of MKK6 in immune reaction against Vibrio Parahaemolyticus in Crassostrea hongkongensis.

MKK6 is a key protein kinase that regulates inflammatory response under pathogen infection by activating MAPK signal pathway. However, the function of MKK6 remained unclear within oysters. Our research recognized a novel KK6 gene from C. hongkongensis, which is named ChMKK6. The cDNA of ChMKK6 gene was 2,760 bp in length, containing an ORF of 1,032 bp encoding 344 amino acids, with a conserved S-TKc region and a DVD region. As illustrated by amino acid alignment and phylogenetic tree, ChMKK6 shared the maximal (99.0%) identity showing MKK6 of Crassostrea gigas. As indicated by tissue expressio

Identification and validation of core microbes for the formation of the characteristic flavor of fermented oysters (Crassostrea gigas).

Self-fermented oyster homogenates were prepared to investigate core microbes and their correlations with flavor formation mechanisms. Five bacterial and four fungal genera were identified. Correlation analysis showed that Saccharomyces cerevisiae, Kazachstania, and L. pentosus were core species for the flavor of fermented products. Four core microbes were selected for inoculation into homogenates. Twelve key aroma compounds with odor activity values >1 were identified by gas chromatography-mass spectrometry. L. plantarum and S. cerevisiae were beneficial for producing key aroma compounds such

Gut Microbiota and Transcriptomics Reveal the Effect of Human Norovirus Bioaccumulation on Oysters (Crassostrea gigas).

Human norovirus (HuNoV) is a major foodborne pathogen that causes acute viral gastroenteritis, and oysters are one of the main carriers of HuNoV transmission. While progress has been made toward understanding the pattern of oyster-bioaccumulated HuNoV, the response of oysters to HuNoV bioaccumulation, including changes in gene expression and gut microbiota, is unclear. In this study, histo-blood group antigen (HBGA)-like molecule expression and gene regulation features and the HuNoV-microbiome interactions of oysters during HuNoV bioaccumulation were characterized. A hexokinase from the oyster Crassostrea gigas is involved in immune recognition as a pattern recognition receptor.

Hexokinase (HK) is generally recognized as a crucial enzyme participating in glycolysis. In the present study, a HK (named CgHK) was identified as a potential pattern recognition receptor (PRR) from the Pacific oyster Crassostrea gigas. The open reading frame (ORF) of CgHK was of 1395 bp, encoding a peptide of 464 amino acids with one Hexokinase_1 domain and one Hexokinase_2 domain. The predicted amino acid sequence of CgHK shared 17%-29% similarities with that of other known HKs. The mRNA transcripts of CgHK were constitutively detected in all the examined tissues, with relative high expressi

D1 dopamine receptor is involved in shell formation in larvae of Pacific oyster Crassostrea gigas.

Dopamine (DA), a significant member of catecholamines, is reported to induce biomineralization of calcium carbonate vaterite microspheres via dopamine receptor (DR) in bivalves, implying the modulation of dopaminergic system on shell formation during larval development. In this research, a homologue of D1 type DR (CgD1DR-1) was identified from oyster Crassostrea gigas, whose full length cDNA was 1197 bp. It was widely expressed in various tissues of C. gigas, with the significantly higher levels in hepatopancreas, mantle, muscle and gill. During developmental stages, the mRNA transcripts of Cg

Inheritance and Variation of Genomic DNA Methylation in Diploid and Triploid Pacific Oyster (Crassostrea gigas).

DNA methylation is an important epigenetic mechanism that could be responsive to environmental changes indicating a potential role in natural selection and adaption. In order to evaluate an evolutionary role of DNA methylation, it is essential to first gain a better insight into inheritability. To address this question, this study investigated DNA methylation variation from parents to offspring in the Pacific oyster Crassostrea gigas using fluorescent-labeled methylation-sensitive amplified polymorphism (F-MSAP) analysis. Most of parental methylated loci were stably transmitted to offspring se

Reduced Vitellogenin Expression is Associated with Impaired Oogenesis in Female Triploid Pacific Oysters (Crassostrea gigas).

Triploidy is widely applied in oyster aquaculture because of its association with reduced gametogenesis, yet female triploid Pacific oyster (Crassostrea gigas) exhibit pronounced variability in ovarian development. The mechanisms underlying ovarian impairment in triploid females remain poorly understood. In this study, we investigated the role of vitellogenin (VTG) in oogenesis and gonadal development by comparing diploid females with β triploid females exhibiting severe gonadal arrest. Quantitative PCR and in situ hybridization analyses revealed that the expression of VTG was significantly re

Transcriptome profiling of triploid Crassostrea gigas gills indicates the host immune mechanism against bacterial infection.

As an important member of global aquaculture, oysters (Crassostrea gigas) have significant economic value. With the development of commercial aquaculture, the frequent occurrence of diseases caused by Vibrio alginolyticus has become a hindrance to high-density aquaculture. Gill tissue, as an important component of immune system of the oysters, plays the key point in the face of invasion by foreign substances. Compared to the diploid oyster, the triploid oyster presents a higher growth rate and lower growth investment, making it a more ideal model for studying oyster immune defense. In this stu

Identification and Genome Analysis of Vibrio coralliilyticus Causing Mortality of Pacific Oyster (Crassostrea gigas) Larvae.

Vibrio coralliilyticus is known as a coral pathogen that also infects marine bivalve larvae worldwide. It is considered to be one of the major constraints in artificial marine bivalve seed production as it causes mortality. In this study, we first isolated and characterized a high virulent of V. coralliilyticus designated as SNUTY-1 that was the cause of Pacific oyster larvae mortality in Korea. In the pathogenicity test, exposure to 2.14 × 105 CFU/mL for 24 h caused mortality to 88.65 ± 2.4% of the tested healthy Pacific oyster larvae. SNUTY-1 showed anti-microbial resistance to β-lactams, su

How annual course of photoperiod shapes seasonal behavior of diploid and triploid oysters, Crassostrea gigas.

In this work, we study if ploidy (i.e. number of copies of chromosomes) in the oyster Crassostrea gigas may introduce differences in behavior and in its synchronization by the annual photoperiod. To answer to the question about the effect of the seasonal course of the photoperiod on the behavior of C. gigas according to its ploidy, we quantified valve activity by HFNI valvometry in situ for 1 year in both diploid and triploid oysters. Chronobiological analyses of daily, tidal and lunar rhythms were performed according the annual change of the photoperiod. In parallel, growth and gametogenesis

Preparation of Hypoallergenic Enzymatic Hydrolysates from Crassostrea angulata with In Vitro Immune Tolerance-Inducing Potential.

Oyster allergy has emerged as a worldwide food safety concern, and enzymatic hydrolysis is an effective strategy for preparing hypoallergenic oyster products. In this study, crude extract from Crassostrea angulata was hydrolyzed with trypsin, Neutrase, and Flavorzyme. The IgE/IgG-binding activity of the hydrolysates was compared, structures and epitopes were explored, and immune tolerance-inducing potential was evaluated using a T-cell proliferation assay. Under optimized conditions (0.5 h reaction; 1.2% trypsin, 0.8% Neutrase, and 0.4% Flavorzyme), the hydrolysates exhibited markedly reduced

Male triploid oysters of Crassostrea gigas exhibit defects in mitosis and meiosis during early spermatogenesis.

The Pacific oyster, Crassostrea gigas, is a successive irregular hermaphrodite mollusk which has an annual breeding cycle. Oysters are naturally diploid organisms, but triploid oysters have been developed for use in shellfish aquaculture, with the aim of obtaining sterile animals with commercial value. However, studies have shown that some triploid oysters are partially able to undergo gametogenesis, with numerous proliferating cells closed to diploids (3n alpha) or a partial one with an accumulation of locked germ cells (3n beta). The aim of our study therefore was to understand the regulatio

Growth Variation in the Pacific Oyster (Crassostrea gigas) Is Associated with Sequential Transcriptional Programs of Tissue Differentiation.

Growth variation is one of the most economically important traits in oyster aquaculture, yet the transcriptional programs associated with divergent growth performance remain incompletely understood. In this study, three growth cohorts of the Pacific oyster (Crassostrea gigas) originating from the same production batch were examined to characterize coordinated differences in tissue-associated transcriptional programs across contrasting growth cohorts. A total of 25 RNA-seq libraries were generated from slow-growth (SG) whole-body samples and mantle and gill tissues of medium-growth (MG) and fas
