P-cresol and Indoxyl Sulfate Impair Osteogenic Differentiation by Triggering Mesenchymal Stem Cell Senescence.

Chronic kidney disease (CKD) patients obtained high levels of uremic toxins progressively develop several complications including bone fractures. Protein-bound uremic toxins especially p-cresol and indoxyl sulfate are hardly eliminated due to their high molecular weight. Thus, the abnormality of bone in CKD patient could be potentially resulted from the accumulation of uremic toxins. To determine whether protein-bound uremic toxins have an impact on osteogenesis, mesenchymal stem cells were treated with either p-cresol or indoxyl sulfate under in vitro osteogenic differentiation. The effects o

Contact sensitizing potential of pyrogallol and 5-amino-o-cresol in female BALB/c mice.

Hair dye components such as pyrogallol and cresol have been shown previously to promote allergic reactions such as rashes, dermal inflammation, irritation and dermatitis. The objective of this study was to determine the contact sensitization potential of pyrogallol (PYR) and 5-amino-o-cresol (AOC) when applied dermally to female BALB/c mice. Measurement of the contact hypersensitivity response was initially accomplished using the local lymph node assay. For PYR, significant increases in the proliferation of lymph node cells were observed at concentrations of 0.5% (w/v) and higher. For AOC, bor

Treatment of phenol and p-cresol in aqueous solution by adsorption using a carbonylated hypercrosslinked polymeric adsorbent.

Phenolic compounds are one of the most representative pollutants in industrial wastewater, and efficient removal and destruction of them have attracted significant concerns. In the present study, a carbonylated hypercrosslinked polymeric adsorbent HJ-1 was developed and its ability to remove phenol and p-cresol in aqueous solution was tested in comparison with the commercial Amberlite XAD-4 resin. The HJ-1 resin exhibited excellent applicability in the acidic and neutral pH, NaCl posed the positive effect, whereas Cd(2+) had the negative effect on the adsorption. The adsorption dynamics obeyed

Indole toxicity on removal of uremic toxin p-cresol, in-vitro study of Thauera aminoaromatica S2.

Protein-bound uremic toxins, such as indoxyl sulfate and p -cresyl sulfate, are major contributors to chronic kidney disease (CKD) complications and are poorly removed by dialysis due to strong albumin binding. Targeting their gut-derived microbial precursors offers a promising strategy to reduce systemic toxin load. Thauera aminoaromatica S2 is known to anaerobically degrade p -cresol, but its response to indole and its potential as an orally administered microbial therapy remain poorly characterized. Here, we investigated the activity of Thauera aminoaromatica S2 under exposure to both p -cr

p-Cresol and p-Cresyl Sulphate Boost Oxidative Stress: A Systematic Review of Recent Evidence.

Recent studies have emphasized the significant role of p-cresol and its conjugated form, p-cresyl sulphate (PCS), in enhancing oxidative stress, leading to potential detrimental effects on various biological systems. Both p-cresol and PCS contribute to increased production of reactive oxygen species (ROS), which can result in tissue damage, inflammation and a cascade of physiological abnormalities. Elevated p-cresol levels have been associated with greater clinical severity in autism spectrum disorder, correlating with more severe behavioural manifestations and a history of regression. This sy

C-H···Y hydrogen bonds in the complexes of p-cresol and p-cyanophenol with fluoroform and chloroform.

In this work, we present spectroscopic investigations of hydrogen bonded complexes of CHF3 and CHCl3 with p-cresol and p-cyanophenol. The systems were chosen as the potential candidates bound by C-H···Y type hydrogen bonds that are known to exhibit unconventional blue shifts in the C-H stretching frequency. The two phenol derivatives chosen offer multiple hydrogen bonding acceptor sites. They also differ from each other in regard to the electron-donating/withdrawing ability of the para substituents which could dictate the global minimum structure in each case. The complexes were formed using t

Fenton degradation of 4,6-dinitro-o-cresol with Fe(2+)-substituted ion-exchange resin.

The Fenton degradation of 4,6-dinitro-o-cresol (DNOC) was studied under different experimental conditions using Amberlyst 15 ion-exchange resin containing ferrous ion. DNOC was found to be effectively degraded under most conditions, and it was observed that, with the addition of HCl, the desorption of ferrous ion from the resin into the solution played a major role in this degradation. The total iron concentration in the reaction solution was found to increase with the addition of HCl, and a pseudo-first-order kinetic model was applied to the desorption of ferrous ion from the resin on the bas

4-Chloro-m-cresol test--a possible supplementary test for diagnosis of malignant hyperthermia susceptibility.

BACKGROUND: In vitro contracture test (IVCT) for diagnosis of MH in our laboratory has a sensitivity of 100% and a specificity of 93%. The results are equivocal in 10-15%, and supplementary tests may thus be required. We have tested the hypothesis that 4-chloro-m-cresol (4-cmc) may be useful for a supplementary test. METHODS: Muscle from 41 consecutive patients from 7 families undergoing diagnostic muscle biopsy with IVCT was exposed in vitro to increasing concentrations of 4-cmc (25, 50, 75, 100, 150, and 200 mumol l-1), and the force development recorded. Cresol Derivatives from Bacillus subtilis as Natural Oviposition Modulator of Culex quinquefasciatus: A Molecular Docking Approach.

Mosquitoes rely heavily on olfactory cues for locating suitable oviposition sites, with microbial communities in aquatic habitats playing a crucial role in producing volatile organic compounds (VOCs) that influence mosquito behaviour. In this study, we isolated Bacillus subtilis DHB13 from the breeding habitat of Culex quinquefasciatus, a major vector of several human diseases. The partial 16S rRNA gene sequence of the isolate has been submitted to NCBI GenBank with the accession number PV698100. The identity and resistance profile of the strain was confirmed through biochemical and antibiotic

The Alkylation of Phenol with Methanol: The Influence of Acid-Base Properties of X Zeolite on the Selectivity of para- and meta-Cresol.

The vapor-phase alkylation of phenol with methanol was investigated on X zeolites and modified X zeolites. First, the difference of product distribution was tested between acid zeolite (HZSM-5, HX, HMCM-22, and Hβ) and basic zeolite X (KX and CsX). Then, X zeolites were modified with Li, K, Cs, Ca, Mg, La, and Ce ion exchange to adjust the acid-base properties of the zeolites. Finally, the type of acid sites and strength of acid-base sites on zeolite catalysts were determined by characterization techniques such as Py-IR and TPD, and the alkylation of phenol with methanol was tested. The result

Gut microbiota alterations promote traumatic stress susceptibility associated with p-cresol-induced dopaminergic dysfunctions.

Mounting evidence suggests a link between gut microbiota abnormalities and post-traumatic stress disorder (PTSD). However, whether and how the gut microbiota influences PTSD susceptibility is poorly understood. Here using the arousal-based individual screening model, we provide evidence for pre-trauma and post-trauma gut microbiota alterations in susceptible mice exhibiting persistent PTSD-related phenotypes. A more in-depth analysis revealed an increased abundance of bacteria affecting brain processes including myelination, and brain systems like the dopaminergic neurotransmission. Because do

Biodegradation of the pesticide 4,6-dinitro-ortho-cresol by microorganisms in batch cultures and in fixed-bed column reactors.

A mixed culture of microorganisms able to utilize 4,6-dinitro-ortho-cresol (DNOC) as the sole source of carbon, nitrogen and energy was isolated from soil contaminated with pesticides and from activated sludge. DNOC was decomposed aerobically in batch cultures as well as in fixed-bed column reactors. Between 65% and 84% of the substrate nitrogen was released as nitrate into the medium, and 61% of the carbon from uniformly 14C-labelled DNOC was recovered as 14CO2. The mixed microbial culture also decomposed 4-nitrophenol and 2,4-dinitrophenol but not 2,3-dinitrophenol, 2,6-dinitrophenol, 2,4-di

Nanomaterial-Based Enzymatic Biosensor for p-Cresol Detection as an Early Autism Spectrum Disorder Diagnosis Biomarker.

Neurodevelopmental disorders are cognitive and behavioral disorders that involve significant difficulties in the acquisition and execution of specific motor, intellectual, social or language functions, including disorders like autism spectrum disorder (ASD). To date, the diagnosis of ASD has been based on observation of daily functionality and behavioral characteristics of patients, and the standards for this diagnosis are complex, time-consuming and unpractical. Diagnosis based on biomarkers detection could be a good alternative. In this work, we propose an innovative electrochemical strategy

Parabacteroides distasonis regulates PPARɑ to alleviate colchicine-induced colonic injury through p-cresol sulfate.

Colchicine has been extensively utilized for treating gouty arthritis over many years, yet its dose-limiting gastrointestinal toxicity remains a significant clinical challenge. While diarrhoea and nausea are well recognised, the extent of colchicine-induced occult colonic injury-including its impact on the gut microbiota and mucosal homeostasis-is poorly defined. This study investigates colchicine-induced colonic injury (e.g., oxidative stress, mitochondrial dysfunction) in vivo and in vitro and also the imbalance of gut microbiota composition in mice. It revealed a significant decreased conte

The role of microbial-derived p-Cresol in autism spectrum disorder: A systematic review of the gut-brain axis.

BACKGROUND & AIMS: Autism Spectrum Disorder (ASD) is a complex neurodevelopmental condition increasingly associated with gut microbiota alterations. Among microbial metabolites, p-Cresol has emerged as a potential contributor to the pathophysiology of ASD. This systematic review aims to examine the evidence linking p-Cresol and its metabolites with ASD and explore their potential role within the gut-brain axis framework, a bidirectional communication system where gut microbiota influence brain function via immune, metabolic, and neural pathways (e.g., vagus nerve, microbial metabolites). A phenome-wide association study (PheWAS) to identify the health impacts of 4-cresol sulfate in the Nagahama Study.

Gut-microbiota derived metabolites are important regulators of host biology and metabolism. To understand the impacts of the microbial metabolite 4-cresol sulfate (4-CS) on four chronic diseases [type 2 diabetes mellitus, metabolic syndrome (MetS), non-alcoholic fatty liver disease, and chronic kidney disease (CKD)], we conducted association analyses of plasma 4-CS quantified by liquid chromatography coupled to mass spectrometry (LC-MS) in 3641 participants of the Nagahama study. Our results validated the elevation of 4-CS in CKD and identified a reducing trend in MetS. To delineate the holist

Microbial metabolite p-cresol inhibits gut hormone expression and regulates small intestinal transit in mice.

p-cresol is a metabolite produced by microbial metabolism of aromatic amino acid tyrosine. p-cresol and its conjugated forms, p-cresyl sulfate and p-cresyl glucuronide, are uremic toxins that correlate positively with chronic kidney disease and diabetes pathogenesis. However, how p-cresol affects gut hormones is unclear. Here, we expose immortalized GLUTag cells to increasing concentrations of p-cresol and found that p-cresol inhibited Gcg expression and reduced glucagon-like peptide-1 (GLP-1) secretion in vitro. In mice, administration of p-cresol in the drinking water for 2 weeks reduced the
