Emerging of Multidrug-Resistant Cronobacter sakazakii Isolated from Infant Supplementary Food in China.

Cronobacter is a foodborne pathogen associated with severe infections in restricted populations and particularly with high mortality in neonates and infants. The prevalence and antimicrobial resistance (AMR) phenotype of Cronobacter cultured from powdered infant formula and supplementary food were studied. The virulence factors, AMR genes, and genomic environments of the multidrug-resistant isolates were further studied. A total of 1,055 Cronobacter isolates were recovered from 12,105 samples of powdered infant formula and supplementary food collected from 29 provinces between 2018 and 2019 in

Antimicrobial Activity of Ferulic Acid Against Cronobacter sakazakii and Possible Mechanism of Action.

Cronobacter sakazakii is an opportunistic pathogen transmitted by food that affects mainly newborns, infants, and immune-compromised adults. In this study, the antibacterial activity of ferulic acid was tested against C. sakazakii strains. Minimum inhibitory concentration of ferulic acid against C. sakazakii strains was determined using the agar dilution method. Changes in intracellular pH, membrane potential and intracellular ATP concentration were measured to elucidate the possible antibacterial mechanism. Moreover, SYTO 9 nucleic acid staining was used to assess the effect of ferulic acid o

Characterization of outer membrane vesicles from a neonatal meningitic strain of Cronobacter sakazakii.

Cronobacter sakazakii is associated with severe and often fatal cases of infant meningitis and necrotizing enterocolitis. The form of meningitis differs from that due to Neisseria meningitidis and Streptococcus spp., in that it is highly invasive and destructive towards human brain cells. However, there is relatively little understanding of the cytopathogenic interaction of C. sakazakii with host cells which results in stimulation of an inflammatory immune response. The production of Cronobacter outer membrane vesicles (OMV) and their potential pathogenic functions have not yet been elucidated

My 40-Year History with Cronobacter/Enterobacter sakazakii - Lessons Learned, Myths Debunked, and Recommendations.

Much has been learned about organism in the Cronobacter/Enterobacter sakazakii complex since I first named and described Enterobacter sakazakii in 1980. However, there are still wide knowledge gaps. One of the most serious is that are still many uncertainties associated with assessing the public health risk posed by these bacteria, particularly in neonatal meningitis. Over the last few decades, Cronobacter contamination of commercial powdered infant formula products has apparently been reduced, but it is still an ongoing problem. The powdered infant formula industry still cannot produce powder

Hfq plays important roles in virulence and stress adaptation in Cronobacter sakazakii ATCC 29544.

Cronobacter spp. are opportunistic pathogens that cause neonatal meningitis and sepsis with high mortality in neonates. Despite the peril associated with Cronobacter infection, the mechanisms of pathogenesis are still being unraveled. Hfq, which is known as an RNA chaperone, participates in the interaction with bacterial small RNAs (sRNAs) to regulate posttranscriptionally the expression of various genes. Recent studies have demonstrated that Hfq contributes to the pathogenesis of numerous species of bacteria, and its roles are varied between bacterial species. Here, we tried to elucidate the

Case Report: Urinary tract isolation of Cronobacter sakazakii in an oncohaematological patient in Southern Italy.

BACKGROUND: Cronobacter sakazakii is an emerging Gram-negative opportunistic pathogen, mostly associated with severe neonatal infections. In adults, infections are rare and usually occur in immuno-compromised or elderly patients. Urinary tract infections caused by C. sakazakii in oncological adults are extremely uncommon. CASE: We report a 66-year-old Caucasian male with acute myeloid leukaemia (AML) with a history of bladder carcinoma. The patient was admitted to the Haematology ward with fever (38.5°C), dysuria, pyuria, abdominal pain, and diarrhoea. He was already undergoing treatment at o

Precision screening of DNAzymes and aptamers, and an ultrasensitive isothermal integrated biosensor for Cronobacter sakazakii detection.

Cronobacter sakazakii (C. sakazakii), an opportunistic pathogenic bacterium resistant to adverse environments, can induce meningitis and sepsis in infants with high mortality, which necessitates ultra-sensitive detection. In this study, the core binding region was derived by tailoring existing C. sakazakii aptamer, and novel aptamer screening libraries were designed based on this core sequences with high affinity and specificity. Additionally, we performed the first screening of C. sakazakii DNAzyme and explored its binding sites with the aid of molecular docking. Based on the aptamer, DNAzyme

Amino acid residues in the tail fiber differentiate the host specificity of Cronobacter sakazakii bacteriophage.

Cronobacter sakazakii is a Gram-negative pathogen that causes severe infections such as neonatal meningitis and sepsis. Bacteriophages (phages) rely on tail fibers for host recognition and infection, but the mechanisms of how phages recognize their bacterial hosts are not completely elucidated. In this study, two lytic C. sakazakii phages belonging to the Drexlerviridae family, CRES7 and CRES9, were isolated from sewage in South Korea. The genomes of both phages are almost the same, with only two nucleotide differences in the gene encoding a putative tail fiber, causing two amino acid differen

Subtractive genomics study for the identification of therapeutic targets against Cronobacter sakazakii: A threat to infants.

Cronobacter sakazakii is an opportunistic pathogen that has been associated with severe infection in neonates such as necrotizing enterocolitis (NEC), neonatal meningitis, and bacteremia. This pathogen can survive in a relatively dry environment, especially in powdered infant formula (PIF). Unfortunately, conventional drugs that were once effective against C. sakazakii are gradually losing their efficacy due to rising antibiotic resistance. In this study, a subtractive genomic approach was followed in order to identify potential therapeutic targets in the pathogen. The whole proteome of the pa

Membrane Proteins and Proteomics of Cronobacter sakazakii Cells: Reliable Method for Identification and Subcellular Localization.

Members of the genus Cronobacter are responsible for severe infections in infants and immunosuppressed individuals. Although several virulence factors have been described, many proteins involved in the pathogenesis of such infections have not yet been mapped. This study is the first to fractionate Cronobacter sakazakii cells into outer membrane, inner membrane, periplasmic, and cytosolic fractions as the basis for improved proteome mapping. A novel method was designed to prepare the fractionated samples for protein identification. The identification was performed via one-dimensional electropho

Exogenous α-linolenic acid acts as a negative regulator of Cronobacter sakazakii structural flagellar proteins.

AIM: This work aims to report on the mechanism of action by which Cronobacter sakazakii virulence is impacted by α-linolenic acid (ALA), a C18:3 fatty acid. METHODS AND RESULTS: To elucidate this, two concentrations of ALA (250 and 1000 µmol L-1) were added exogenously to C. sakazakii 29 544 in tryptic soy broth. Quantitative proteomic analysis using label-free mass spectrometry showed significantly different proteomic profiles of treated and control C. sakazakii samples. Across both ALA treatments, a total of 11 flagellar proteins were identified as reduced in abundance and showed a dose-dep

Cronobacter sakazakii is a foodborne pathogen linked to severe infections in infants and often associated with contaminated powdered infant formula. The RecA protein, a key player in DNA repair and recombination, also influences bacterial resilience and virulence. This study investigated the impact of recA deletion on the pathogenicity and environmental stress tolerance of C. sakazakii BAA-894. A recA knockout mutant displayed impaired growth, desiccation tolerance, and biofilm formation. In a rat model, the mutant demonstrated significantly reduced virulence evidenced by higher host survival
