Albinism and auditory function in the laboratory mouse. II. Effects of acoustic priming and cross-fostering.

Albino (c/c) and nonalbino (+/c and +/+) congenic C57BL/6J mice were examined for the effects of acoustic prestimulation (priming) on audiogenic seizures. While no genotypic-specific effects were noticed 1 day after priming, major effects were observed in separate groups of mice tested 5 days after priming. The c/c mice were most susceptible to audiogenic seizures, and no differences were observed between +/c and +/+ mice. While cross-fostering did not change this relationship, it provided protection from acoustic trauma is considered inconclusive because of the interaction of innate- and prim

Within- and cross-language repetition priming in verb generation: evidence from non-proficient Chinese-English bilinguals.

The present study aims to investigate the repetition priming within and across languages in verbs among non-proficient Chinese-English bilinguals, using a verb generation task. The impact of levels of processing on repetition priming was also examined. A total of 160 non-proficient Chinese-English bilinguals participated in the experiments, generating English verbs (Experiment 1) or Chinese verbs (Experiment 2) that corresponded to the same set of 50 translation-equivalent nouns across the two languages. The results revealed significant within-language repetition priming in both deep- and shal

Cross-priming amplification targeting the 18S rRNA gene for the rapid diagnosis of Babesia bovis infection.

Babesia bovis is a known causative agent of bovine babesiosis and is widely distributed across China. Rapid detection and accurate identification of B. bovis is essential for follow-up management and epidemiological investigations. In this study, a cross-priming amplification combined with vertical flow (CPA-VF) assay was developed. The detection limit of the CPA-VF assay targeting the 18S rRNA gene was 320 fg per reaction at 61 °C for 60 min. No cross-reactions were observed with other piroplasms infective to cattle. Furthermore, 36 blood samples from experimentally-infected animals were accu

Detection of Vibrio cholerae by isothermal cross-priming amplification combined with nucleic acid detection strip analysis.

Vibrio cholerae is a water- and food-borne human pathogen, and V. cholerae serotypes O1 and O139 have attracted attention because of their severe pathogenesis. However, non-O1, non-O139 cholera vibrios (NCVs) were also recently recognized as having virulence properties. In this study, we developed a cross-priming amplification (CPA) method for the detection of all serotypes of V. cholerae. The specificity of the CPA method was tested using a panel of 60 different bacterial strains. All of the V. cholerae strains showed positive results, and 41 other types of bacteria gave negative results. The

Covalent conjugation of TLR9 agonists to phospholipids enhances mRNA-LNP delivery efficiency and dendritic cell cross-priming.

Current mRNA-LNP systems face challenges in efficient co-delivery of nucleic acid adjuvants, limiting their capacity to elicit robust cellular immunity. To overcome the dissociation limitations of physically admixed CpG in conventional lipid nanoparticles, we developed a covalent conjugation platform that integrates TLR9 agonists with phospholipids via amide bonding. We synthesized a DSPE-CpG conjugate, which exhibited high loading efficiency (97.1%) onto mRNA-LNPs. The immune activation efficacy was evaluated in models encoding SARS-CoV-2 spike protein and HBV antigens. The DSPE-CpG-mRNA LNP

Impairment of stromal-epithelial regenerative cross-talk in Hirschsprung disease primes for the progression to enterocolitis.

Hirschsprung disease (HSCR) is a congenital condition characterized by the improper migration of enteric neural crest cells, leading to aganglionosis most commonly in the rectosigmoid colon. This severe and life-threatening disorder often results in the development of Hirschsprung-associated enterocolitis (HAEC), which can occur either before or after surgical resection of the affected bowel segment. Using colonic tissue from patients with HSCR alongside the well-established endothelin receptor B knockout mouse model, we investigated epithelial regeneration dynamics and stromal-epithelial cros

Cross-clade HIV-1 neutralizing antibodies induced with V3-scaffold protein immunogens following priming with gp120 DNA.

The V3 epitope is a known target for HIV-1 neutralizing antibodies (NAbs), and V3-scaffold fusion proteins used as boosting immunogens after gp120 DNA priming were previously shown to induce NAbs in rabbits. Here, we evaluated whether the breadth and potency of the NAb response could be improved when boosted with rationally designed V3-scaffold immunogens. Rabbits were primed with codon-optimized clade C gp120 DNA and boosted with one of five V3-cholera toxin B fusion proteins (V3-CTBs) or with double combinations of these. The inserts in these immunogens were designed to display V3 epitopes s

The interrelationship of frailty, multimorbidity and disability in Parkinson's disease: PRIME-UK cross-sectional study.

BACKGROUND: The prevalence of Parkinson's disease rises with age and so patients may also be living with multimorbidity, two or more long-term conditions, and frailty, a loss of physiological reserve. However, these individuals are typically under-represented in clinical research. The aim was to describe the prevalence and interrelationship of frailty, multimorbidity, disability, sarcopenia and polypharmacy in a representative sample of people with parkinsonism recruited to the PRIME-UK cross-sectional study. METHODS: In this single-centre cross-sectional study of people with parkinsonism, we

Cross-Segmental Transfer Effects of Lower Limb Cross-Training Priming on Upper Extremity Functional Recovery in Pediatric Unilateral Cerebral Palsy: A Randomized Controlled Trial.

Background: Upper extremity deficits in unilateral cerebral palsy (UCP) severely restrict daily autonomy. Although movement-based priming is known to stimulate neuroplasticity, the distal transfer of lower extremity (LE) training to augment paretic upper limb (UL) function remains largely uninvestigated. This randomized controlled trial evaluated whether a 6-week LE cross-training (CT) priming regimen enhances UL functional restoration in pediatric UCP. Methods: Thirty-six children (6-8 years) were randomized to a conventional physical therapy cohort (n = 18) or an experimental CT cohort (n =

A role for the transcription factor RelB in IFN-alpha production and in IFN-alpha-stimulated cross-priming.

Chimeric mice generated with bone marrow from RelB-deficient (-/-), RelB-heterozygous (+/-) and wild-type (+/+) mice were used to determine how total or partial absence of the transcription factor RelB in haematopoietic cells affects the immune response generated after lymphocytic choriomeningitis virus (LCMV) infection. In RelB(-/-) chimeras, early virus replication was enhanced and LCMV clearance was impaired. Although plasmacytoid dendritic cell numbers were similar, serum interferon (IFN)-alpha levels in RelB(-/-) and RelB(+/-) chimeras were markedly lower than in RelB(+/+) chimeras during

Cross-Modality Treatment Generalization in Sentence Comprehension in Aphasia: Evidence From Structural Priming and Webcam-Based Eye-Tracking.

BACKGROUND: Acquired neurogenic communication disorders such as aphasia are associated with sentence production and comprehension difficulties. However, sentence-level treatments for aphasia do not consistently yield generalized gains across modalities, from production to comprehension or vice versa. This limits the effectiveness of current aphasia treatments. In this study, we present structural priming, the phenomenon where repeated grammatical structures are used more frequently and are easier to process, as a potentially effective treatment for sentence-level deficits in aphasia, and one w

Cytomegalovirus inhibitors of programmed cell death restrict antigen cross-presentation in the priming of antiviral CD8 T cells.

CD8 T cells are the predominant effector cells of adaptive immunity in preventing cytomegalovirus (CMV) multiple-organ disease caused by cytopathogenic tissue infection. The mechanism by which CMV-specific, naïve CD8 T cells become primed and clonally expand is of fundamental importance for our understanding of CMV immune control. For CD8 T-cell priming, two pathways have been identified: direct antigen presentation by infected professional antigen-presenting cells (pAPCs) and antigen cross-presentation by uninfected pAPCs that take up antigenic material derived from infected tissue cells. Priming of Salmonella enterica serovar typhi-specific CD8(+) T cells by suicide dendritic cell cross-presentation in humans.

BACKGROUND: The emergence of antibiotic-resistant strains of Salmonella enterica serovar Typhi (S. Typhi), the etiologic agent of typhoid fever, has aggravated an already important public health problem and added new urgency to the development of more effective typhoid vaccines. To this end it is critical to better understand the induction of immunity to S. Typhi. CD8(+) T cells are likely to play an important role in host defense against S. Typhi by several effector mechanisms, including killing of infected cells and IFN-gamma secretion. However, how S. Typhi regulates the development of spec

Tetanus-diphtheria vaccine can prime SARS-CoV-2 cross-reactive T cells.

Vaccines containing tetanus-diphtheria antigens have been postulated to induce cross-reactive immunity to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), which could protect against coronavirus disease (COVID-19). In this work, we investigated the capacity of Tetanus-diphtheria (Td) vaccine to prime existing T cell immunity to SARS-CoV-2. To that end, we first collected known SARS-CoV-2 specific CD8+ T cell epitopes targeted during the course of SARS-CoV-2 infection in humans and identified as potentially cross-reactive with Td vaccine those sharing similarity with tetanus-diphth

CD91 and Its Ligand gp96 Confer Cross-Priming Capabilities to Multiple APCs during Immune Responses to Nascent, Emerging Tumors.

During cancer immunosurveillance, dendritic cells (DC) play a central role in orchestrating T-cell responses against emerging tumors. Capture of miniscule amounts of antigen along with tumor-initiated costimulatory signals can drive maturation of DCs. Expression of CD91 on DCs is essential in cross-priming of T-cell responses in the context of nascent tumors. Multiple DC and macrophage subsets express CD91 and engage tumor-derived gp96 to initiate antitumor immune responses, yet the specific CD91+ antigen-presenting cells (APC) that are required for T-cell cross-priming during cancer immunosur

Seed priming with H2S and Ca2+ trigger signal memory that induces cross-adaptation against nickel stress in zucchini seedlings.

In this study, the effect of seed priming with sodium hydro sulfide (NaHS) and CaCl2 as well as the possible relationship between them in inducing post-germinative cross-adaptation in zucchini seedlings (cv Courgette d'Italie) were investigated. Results showed that Ni toxicity reduced plant growth and photosynthetic pigments, decreased the content of ascorbate (AsA) and total thiols, increased hydrogen peroxide (H2O2) content and electrolyte leakage (EL), up-regulated the transcription levels of Ca2+-dependent protein kinase (CDPK) and phytochelatin (PCs) genes and elevated H2S content in leav
