Molecular Encapsulation of Histamine H₂-Receptor Antagonists by Cucurbit[7]Uril: An Experimental and Computational Study.

The histamine H₂-receptor antagonists cimetidine, famotidine and nizatidine are individually encapsulated by macrocyclic cucurbit[7]uril (CB[7]), with binding affinities of 6.57 (±0.19) × 10³ M(-1), 1.30 (±0.27) × 10⁴ M(-1) and 1.05 (±0.33) × 10⁵ M(-1), respectively. These 1:1 host-guest inclusion complexes have been experimentally examined by ¹H-NMR, UV-visible spectroscopic titrations (including Job plots), electrospray ionization mass spectrometry (ESI-MS), and isothermal titration calorimetry (ITC), as well as theoretically by molecular dynamics (MD) computation. This study may provide imp

Separation performance of cucurbit[8]uril and its coordination complex with cadmium (II) in capillary gas chromatography.

Here we report the investigation of using cucurbit[8]uril (CB8) and its coordination complex with cadmium (II) (CB8-Cd) as stationary phases for capillary gas chromatography (GC). The prepared capillary columns of CB8 and CB8-Cd stationary phases achieved column efficiency of 2200plates/m and 1508plates/m, respectively, and showed weak polarity based on the measured McReynolds constants. Their separation performance was investigated by GC separation of mixtures of different types while a commercial column was used for comparison. The CB8 stationary phase achieved high resolution for a wide ran

Supramolecular Organic Framework-Enabled AIE-Enhanced Electrochemiluminescence: Dual-Readout Ultrasensitive Determination of Microcystin-LR in Environmental Waters.

Accurate monitoring of microcystin-LR (MC-LR) in complex water matrices is critical for environmental safety but remains challenging due to the limited sensitivity and stability of traditional methods. Conventional electrochemiluminescence (ECL) emitters often suffer from aggregation-caused quenching (ACQ) in aqueous environments. To overcome this, we engineered a highly efficient aggregation-induced electrochemiluminescence (AIECL) emitter using a supramolecular confinement strategy. A shape-tunable supramolecular organic framework (SOF), tetrakis(1-carboxyethylpyridinium) tetraphenylethylene

Label-free fluorescence lateral flow biosensing platform based on host-guest recognition for on-site detection of amantadine in egg samples.

Rapid, accurate and convenient detection of illegally added drugs are of critical importance in food safety. Herein, based on the host-guest recognition strategy, we establish a novel label-free fluorescence lateral flow assay (LF-LFA) platform for detection of amantadine (AMD) in egg samples. Specially, two fluorescence reporters-BSA-β-CD@BER (formed via host-guest interaction) and BER-BSA (generated through AIE effect), are effectively designed. This platform can enable "turn-on" FL detection through host-guest interaction between cucurbit[7]uril (CB[7]) and AMD, as well as between CB[7] and

Programmable DNA probe-mediated nanopore biosensor for multiplex nucleic acid detection and its application in milk authenticity identification.

Simultaneous and sensitive detection of multiple nucleic acids in complex matrices has always been a significant challenge. Herein, we reported a novel nanopore biosensing strategy that achieved label-free, high-discrimination detection of multiplex nucleic acid targets by ingeniously integrating programmable DNA probe with click chemistry amplified strategy. In this platform, probe DNAs with alkynyl modification at different positions were clicked with azidoadamantane, which were then reacted with cucurbit[6]uril to produce host-guest complexes as signal tags. These reporters showed highly di

Improved Physical Stability of an Antibody-Drug Conjugate Using Host-Guest Chemistry.

Antibody-drug conjugates (ADCs) are an emerging class of biopharmaceutical products for oncology, with the cytotoxic pyrrolobenzodiazepine (PBD) family of "warheads" well-established in the clinic. While PBDs offer high potency, they are also characterized by their hydrophobicity, which can make formulation of the ADC challenging. Several approaches have been investigated to improve the physicochemical properties of PBD-containing ADCs, and herein a supramolecular approach was explored using cucurbit[8]uril (CB[8]). The ability of CB[8] to simultaneously encapsulate two guests was exploited to

Nanoscale Control of Amyloid Self-Assembly Using Protein Phase Transfer by Host-Guest Chemistry.

Amyloid fibrils have recently been highlighted for their diverse applications as functional nanomaterials in modern chemistry. However, tight control to obtain a targeted fibril length with low heterogeneity has not been achieved because of the complicated nature of amyloid fibrillation. Herein, we demonstrate that fibril assemblies can be homogeneously manipulated with desired lengths from ~40 nm to ~10 μm by a phase transfer of amyloid proteins based on host-guest chemistry. We suggest that host-guest interactions with cucurbit[6]uril induce a phase transfer of amyloid proteins (human insuli

Recent advances in supramolecular macrocycle-based artificial light-harvesting systems.

Artificial light-harvesting systems (ALHSs) inspired by the antenna function of natural photosynthesis provide molecular platforms for collecting excitation energy and directing it to emissive or reactive acceptors. In many supramolecular ALHSs, however, practical performance is limited by poorly defined donor-acceptor orientation, aggregation-caused quenching (ACQ), interfacial defects, and limited stability in aqueous or complex media. Supramolecular macrocycles-particularly pillar[n]arenes (PAs), cucurbit[n]urils (CBs), calixarenes (CAs), cyclodextrins (CDs), and supramolecular coordination

Visible-Light-Harvesting Supramolecular Nanoassembly of Tetrathienylethene Tetrapyridinium/Cucurbit[7]uril for Bacterial Biofilm Eradication.

Bacterial biofilms act as a protective "umbrella" that significantly impairs antibiotic penetration, thereby hindering the complete eradication of infections. The clinical translation of organic photosensitizers for antimicrobial photodynamic therapy (aPDT) is critically hindered by their intrinsic self-aggregation in aqueous media, which severely compromises reactive oxygen species (ROS) generation and impedes penetration into dense bacterial biofilms. Herein, we report a visible-light-harvesting photosensitizer, tetrathienylethene tetrapyridinium chloride (TTC), and construct a supramolecula

Matrix solid-phase dispersion based on cucurbit[7]uril-assisted dispersive liquid-liquid microextraction coupled with high performance liquid chromatography for the determination of benzimidazole fungicides from vegetables.

A new method involving matrix solid-phase dispersion (MSPD) and dispersive liquid-liquid microextraction (DLLME) was optimized with the aid of stoichiometry and applied to the extraction, purification, and determination of benzimidazole residues in vegetables. Carbendazim, thiabendazole, and thiophanate-methyl were selectively extracted from vegetables using cucurbit[7]uril as the MSPD extractant and transferred to an aqueous solution, then further enriched using DLLME with acetonitrile and chloroform as dispersive and extraction solvents, respectively. Photophysical Properties of Donor-Acceptor Stenhouse Adducts and Their Inclusion Complexes with Cyclodextrins and Cucurbit[7]uril.

Donor-acceptor Stenhouse adducts (DASAs) are a novel class of solvatochromic photoswitches with increasing importance in photochemistry. Known for their reversibility between open triene and closed cyclized states, these push-pull molecules are applicable in a suite of light-controlled applications. Recent works have sought to understand the DASA photoswitching mechanism and reactive state, as DASAs are vulnerable to irreversible "dark switching" in polar protic solvents. Despite the utility of fluorescence spectroscopy for providing information regarding the electronic structure of organic co

Host-Guest Complexation of Cucurbit[7]Uril and Cucurbit[8]Uril with the Antineoplastic and Multiple Sclerosis Agent Mitoxantrone (Novantrone).

The nature of interactions between the neutral/protonated mitoxantrone and the cucurbit[n]uril (n = 7, 8) host system was analyzed by employing density functional theory calculations. A comparison between the inclusion complexes of CB[7] and CB[8] shows various subtle differences in the complexation thermodynamics, given as changes in the Gibbs energy. Doubly and quadruply charged mitoxantrone (MX) molecules spontaneously form complexes in a water solvent, which are modeled using the polarizable continuum model approach. Both CB[7] and CB[8] complexes are stable as the geometry of the cavity a

On-line competitive host-guest interactions in a turn-on fluorometric method to amantadine determination in human serum and pharmaceutical formulations.

A competitive assay between the antiviral Amantadine and the dye Thionine for the Cucurbit[8]uril cavity was carried out in a flow injection analysis system for the indirect fluorescence detection of Amantadine. Both, Cucurbit[7]uril and Cucurbit[8]uril Thionine complexes were evaluated for the competitive assay. The use of a 12-port injection valve allows the on-line reaction in the flow system. Once optimized all the experimental variables, the methodology developed allows the detection of Amantadine at the 0.16µM level with excellent accuracy (Er ≤ 8.2%) and reproducibility (RSD ≤ 6.3%) for

Host-Guest Tethered DNA Transducer: ATP Fueled Release of a Protein Inhibitor from Cucurbit[7]uril.

Host-guest complexes are emerging as powerful components in functional systems with applications ranging from materials to biomedicine. In particular, CB7 based host-guest complexes have received much attention for the controlled release of drugs due to the remarkable ability of CB7 toward binding input molecules in water with high affinity leading to displacement of CB7 from included pharmacophores (or from drug loaded porous particles). However, the release of bound guests from CB7 in response to endogenous biological molecules remains limited since the input biomolecule needs to have the ap

Host-guest assembly of squaraine dye in cucurbit[8]uril: its implication in fluorescent probe for mercury ions.

The binding interactions between SQ2 squaraine dye and cucurbit[8]uril (CB8) effectively removes the aggregation of SQ2 in aqueous solution by forming a 1 : 1 inclusion complex. The resulting SQ2.CB8 complex exhibits highly selective fluorescence quenching by Hg(2+) ions, as a result of the synergetic binding between CB8, SQ2 and metal cation.

Sodium-Potassium Competition Controls Xenon Uptake in Cucurbiturils.

The remarkable specificity of 129Xe magnetic resonance has motivated efforts to expand its application to medical diagnostics and to develop Xe-cage guest-host complexes compatible with in vivo conditions. Cucurbit[n]urils (CB[n]) are among the most promising host molecules for direct biocompatible Xe encapsulation. Building on earlier observations that alkali cations influence the binding of Xe to CB[n], this study provides a comprehensive experimental and computational analysis of how the ratio of Na+ to K+ in the solution governs Xe uptake by CB[6] and CB[5]. Using one- and two-dimensional

The study of the deposition process of bambus[6]uril on the porous TiNi surface using various physical methods.

This study investigates new composite porous titanium nickelide-based materials, which have been obtained for the first time by functionalizing their surface with a promising macrocyclic compound-bambus[6]uril. Such functionalization aims to enhance the hemocompatibility and cytocompatibility of bone implants, addressing issues of uncontrolled drug release and poor implant integration. The surface modification was carried out by applying bambus[6]uril in the form of a dispersion using various physical methods such as immersion, vacuum treatment, and ultrasonic exposure. The obtained materials

Red aqueous room-temperature phosphorescence modulated by anion-π and intermolecular electronic coupling interactions.

Aqueous room temperature phosphorescence (aRTP) from purely organic materials has been intriguing but challenging. In this article, we demonstrated that the red aRTP emission of 2Br-NDI, a water-soluble 4,9-dibromonaphthalene diimide derivative as a chloride salt, could be modulated by anion-π and intermolecular electronic coupling interactions in water. Specifically, the rarely reported stabilization of anion-π interactions in water between Cl- and the 2Br-NDI core was experimentally evidenced by an anion-π induced long-lived emission (λ Anion-π) of 2Br-NDI, acting as a competitive decay path

A Fluorescently Ratiometric Natural Probe for Selective Detection of Sulfur Dioxide Derivative and Host-Guest Supramolecular Regulation.

Natural sanguinarine (SG) was first used as a fluorescent probe to develop a novel ratiometric sensor for selective HSO3- detection. The nucleophilic addition reaction of HSO3- occurs at the C=N+ group of SG, and subsequent breakage of the conjugated π cycle leads to a decrease in the SG iminium fluorescence that is accompanied by an increase in the alkanolamine fluorescence. Therefore, a ratiometric fluorescence method with a large wavelength shift can be established for HSO3- detection. Furthermore, cucurbit[8]uril was used as an efficient host to encapsulate SG for an improved selectivity f
