Precision Design in Cancer Treatment: Cucurbitacin E Modulates AMPK, PGK1, and PKM2 to Optimize Radiation Efficacy in Melanoma.

INTRODUCTION: Cucurbitacin E (CE), a naturally occurring triterpenoid derived from Cucurbitaceae plants, exhibits potent anticancer activity, particularly against melanoma. Previous studies indicate that CE suppresses melanoma progression by activating AMP-activated protein kinase (AMPK), thereby disrupting metabolic pathways and inducing apoptosis. Despite these findings, the potential synergistic interaction between CE and γ-irradiation in melanoma cell death remains unexplored. OBJECTIVE: This study investigates the combined effects of CE and γ-irradiation on apoptosis in A375 melanoma cel

The neuroprotective effect of Cucurbitacin B against Aβ and tau toxicities requires functional HDAC6 and stress granule pathways.

Alzheimer's disease (AD) is characterized by proteostasis collapse driven by amyloid-β (Aβ) plaques and tau tangles. Dysregulation of stress granule (SG) dynamics and aberrant histone deacetylase 6 (HDAC6) activity are emerging as pivotal pathogenic mechanisms promoting neurodegeneration. Here, we identify that Cucurbitacin B (CB), a natural triterpenoid, acts as a potent SG inducer that confers broad-spectrum neuroprotection. Mechanistically, we demonstrate a novel "recruit-and-sequester" model: CB promotes the assembly of HDAC6-recuited SGs, thereby physically sequestering HDAC6 and function

The CmABF1-CmBt/CmBr Module Regulates Cucurbitacin B Biosynthesis to Promote Cold Tolerance of Oriental Melon Seedlings.

Low temperature is a common abiotic stress in the early spring and winter cultivation of oriental melons, which seriously affects their normal growth and development, and plants can activate the defence mechanism related to reactive oxygen species through secondary metabolic pathways. Cucurbitacins are a defensive secondary metabolite that produces a bitter taste in Cucurbitaceae plants. However, the molecular regulatory mechanism between low temperature and biosynthesis of cucurbitacins, as well as whether cucurbitacins will enhance the plant's resistance to cold stress, has not been studied
