Cyclodextrins in action: Modulating candida albicans biofilm formation and morphology.

The primary objective of this research was to explore the bioactive potential of cyclodextrins in attenuating biofilm formation in C. albicans. The concentration-dependent effects of both native cyclodextrins and specific derivatives were studied at concentrations ranging from 0.1 to 12.5 mM, to determine the mechanisms by which the extent of biofilm formation can be reduced. Besides, the efficiency of various combinations of cyclodextrins and farnesol, as an antifungal substance, were examined. The present study revealed both stimulatory and inhibitory effects of cyclodextrins on biofilm form

Unprecedented Cubic Mesomorphic Behaviour of Crown-Ether Functionalized Amphiphilic Cyclodextrins.

Amphiphilic supramolecular materials based on biodegradable cyclodextrins (CDs) have been known to self-assemble into different types of thermotropic liquid crystals, including smectic and hexagonal columnar mesophases. Previous studies on amphiphilic CDs bearing 14 aliphatic chains at the secondary face and 7 oligoethylene glycol (OEG) chains at the primary face showed that the stability of the mesophase can be rationally tuned through implementation of terminal functional groups to the OEG chains. Here, we report the syntheses of first examples of crown ether-functionalized amphiphilic cyclo

Anomalous Properties of Cyclodextrins and Their Complexes in Aqueous Solutions.

Cyclodextrins (CDs) are cyclic oligosaccharides that emerged as industrial excipients in the early 1970s and are currently found in at least 130 marketed pharmaceutical products, in addition to numerous other consumer products. Although CDs have been the subject of close to 100,000 publications since their discovery, and although their structure and properties appear to be trivial, CDs are constantly surprising investigators by their unique physicochemical properties. In aqueous solutions, CDs are solubilizing complexing agents of poorly soluble drugs while they can also act as organic cosolve

Poly(ε-caprolactone) nanowebs functionalized with α- and γ-cyclodextrins.

The effects of alpha- and gamma-cyclodextrins (α- and γ-CDs) on the thermal and crystal nucleation behavior of electrospun poly(ε-caprolactone) (PCL) nanofibers have been investigated. PCL/CD composite nanofibers were obtained for the first time by electrospinning the mixture from chloroform/N,N-dimethylformamide (60:40). Scanning electron microscopy analyses indicated that neat PCL nanofibers have an average diameter of 400 nm, which increases with the addition of CDs. The presence of CDs on or in the electrospun PCL fibers in the electrospun mats was investigated using Fourier transform infr

Regioselective phosphorylation of branched cyclodextrins with cyclo-mono-mu-imidotriphosphate.

The phosphorylation of the branched cyclodextrins, mono-6-O-(alpha-D-glucopyranosyl)cyclomaltohexaose, mono-6-O-(alpha-D-maltosyl)cyclomaltohexaose, mono-6-O-(alpha-D-glucopyranosyl)cyclomaltoheptaose, and mono-6-O-(alpha-D-maltosyl)cyclomaltoheptaose, in aqueous solution by sodium cyclo-mono-mu-imidotriphosphate (cMITP) was examined. In these reactions, only the 2-OH group of a single alpha-D-glucopyranosyl residue of the cyclodextrin ring was phosphorylated, in a maximum yield of 67%. A possible mechanism for the phosphorylation is discussed.

The Impact of Cyclodextrins on the Physiology of Candida boidinii: Exploring New Opportunities in the Cyclodextrin Application.

Cyclodextrins, commonly used as excipients in antifungal formulations to improve the physicochemical properties and availability of the host molecules, have not been systematically studied for their effects and bioactivity without a complex active substance. This paper evaluates the effects of various cyclodextrins on the physiology of the test organism Candida boidinii. The research examines their impact on yeast growth, viability, biofilm formation and morphological changes. Native ACD, BCD, randomly methylated α- and β-CD and quaternary ammonium α-CD and β-CD were investigated in the 0.5-12

Cyclodextrins in chronic pain: Membrane cholesterol remodeling and neuroimmune signaling.

BACKGROUND: Chronic pain remains challenging to treat, with current therapies offering incomplete relief and significant side effects. This narrative review examines cyclodextrins (CDs) beyond their conventional excipient role, focusing on their emerging function as membrane-active modulators of cholesterol-dependent signaling. METHOD/SCOPE: We integrate preclinical evidence linking CD-associated cholesterol remodeling to altered lipid raft organization, nociceptive ion channel regulation (e.g., TRPV1, TRPA1), and neuroimmune pathways implicated in selected chronic pain conditions. Cyclodextrins: Synthesis, Pharmaceutical Applications, Drug Delivery Systems and Safety Perspectives.

Cyclodextrins (CDs) are cyclic oligosaccharides composed of α-(1,4)-linked glucopyranose units that have emerged as multifunctional and versatile pharmaceutical excipients. One of the major challenges in modern drug development is that nearly half of newly discovered drug molecules exhibit poor aqueous solubility, which adversely affects formulation development, bioavailability, and therapeutic efficacy. CDs address this limitation by forming non-covalent inclusion and non-inclusion complexes, thereby enhancing drug solubility, stability, dissolution rate, and overall biopharmaceutical perform

Binding of voriconazole aptamers and modulation of binding by cyclodextrins.

Voriconazole is a broad-spectrum antifungal drug widely used to treat life-threatening fungal infections, including invasive aspergillosis and severe candidiasis. Despite its efficacy, large inter- and intra-patient pharmacokinetic variability and frequent drug-drug interactions can lead to subtherapeutic or toxic plasma concentrations, making therapeutic drug monitoring essential. Recently, several aptamer selections for voriconazole have been reported, including two independent selections from our laboratory that yielded multiple aptamer families. In this work, we systematically characterize

Cyclodextrins as Active Therapeutic Agents: Beyond Their Role as Excipients.

Cyclodextrins (CDs) have traditionally been recognized as excipients that enhance solubility and stability of drugs. However, growing evidence shows that CDs themselves can act as active therapeutic agents. Their unique supramolecular properties enable them to interact with biological membranes, mobilize cholesterol, and modulate immune responses. This review highlights four therapeutic areas where CDs demonstrate particular promise. First, in gene and mRNA therapy, cationic CD derivatives form nanoparticles that protect nucleic acids, promote endosomal escape, and achieve targeted delivery. S

Exploring In Vitro Biological Cellular Responses of Pegylated β-Cyclodextrins.

βCDPEG5 and βCDPEG2 are two derivatives comprising seven PEG linear chains of 5 and 2 kDa, respectively, conjugated to βCD. As βCDPEGs display different physicochemical properties than their precursors, they could also trigger distinct cellular responses. To investigate the biological behavior of βCDPEGs in comparison to their parent compounds, we performed broad toxicological assays on RAW 264.7 macrophages, MC3T3-E1 osteoblasts, and MDCK cells. By analyzing ROS and NO2- overproduction in macrophages, we found that βCDPEGs induced a moderate stress response without affecting cell viability. A

Self-Assembly of Cyclodextrins and Their Complexes in Aqueous Solutions.

Cyclodextrins (CDs) are enabling pharmaceutical excipients that can be found in numerous pharmaceutical products worldwide. Because of their favorable toxicologic profiles, CDs are often used in toxicologic and phase I assessments of new drug candidates. However, at relatively high concentrations, CDs can spontaneously self-assemble to form visible microparticles in aqueous mediums and formation of such visible particles may cause product rejections. Formation of subvisible CD aggregates are also known to affect analytical results during product development. How and why these CD aggregates for

Studying supramolecular assemblies by ESEEM spectroscopy: inclusion complexes of cyclodextrins.

Electron spin-echo envelope modulation (ESEEM) spectroscopy was used to investigate intramolecular and intermolecular complexes of cyclodextrins (CDs) with a nitroxide group. The interaction with solvent molecules (D(2)O) was followed through the (2)H modulation depth. Competition experiments with adamantane-type guests were used to confirm complexation. The shielding of the nitroxide group from solvent upon inclusion into a CD cavity made this technique more sensitive to complexation than cw EPR spectroscopy. ESEEM analysis of a series of CDs mono and bis spin-labeled on the primary rim of th

[Substituted cyclodextrins as chelating reagents for ethers, thioethers and yperite].

The complexation of mustard gas Cl(CH(2))(2)S(CH(2))(2) Cl, HD, yperite) and of ethers and thioethers derivatives by cyclodextrins: natural alpha-cyclodextrin (ACD) and substituted B-cyclodextrins was studied by NMR. A 1/1 stoechiometry was found in all cases, while affinity constants were found relatively weak (from 5 M(-1) to 100 M(-1)). However, these results show that chelation of HD by cyclodextrins can be reasonably expected, especially if chemical modifications provide stronger affinity constants.

An enzyme-mediated controlled release system for curcumin based on cyclodextrin/cyclodextrin degrading enzyme.

In this study, an enzyme-triggered system based on β-cyclodextrin (β-CD) has been developed to achieve controlled release of hydrophobic drugs in the presence of maltogenic amylase (MAase). The inclusion complex formation of curcumin (CUR), as a model anticancer compound, with β-CD was characterized by fluorescence and Fourier transform infrared (FTIR) spectroscopy. CUR was loaded into β-CD with an encapsulation efficiency of approximately 30 %. The in vitro profiles of CUR release from β-CD showed that 100 % of the drug was released after one hour incubation in the presence of MAase with cycl
