N-acetylaspartylglutamate inhibits forskolin-stimulated cyclic AMP levels via a metabotropic glutamate receptor in cultured cerebellar granule cells.

The neuronal dipeptide N-acetylaspartylglutamate (NAAG) fulfills several of the criteria for classification as a neurotransmitter including localization in synaptic vesicles, calcium-dependent release after neuronal depolarization, and low potency activation of N-methyl-D-aspartate receptors. In the present study, the influence of NAAG on metabotropic receptor activation in cerebellar granule cells was examined in cell culture. Stimulation of granule cell adenylate cyclase with forskolin increased cyclic AMP (cAMP) several hundredfold above basal levels within 10 min in a concentration-depende

Agonists at metabotropic glutamate receptors presynaptically inhibit EPSCs in neonatal rat hippocampus.

1. The effects of metabotropic glutamate receptor agonists on excitatory synaptic transmission in the CA1 region of rat hippocampal slices (11-30 days) were studied using extracellular and whole-cell patch-clamp recording techniques. 2. Trans-1-amino-1,3-cyclopentanedicarboxylic acid (trans-ACPD; 25-100 microM) reversibly depressed excitatory postsynaptic currents (EPSCs) without affecting presynaptic fibre excitability or EPSC reversal potential. 3. Ibotenate (25 microM) or L-glutamate (250 microM), in the presence of the N-methyl-D-aspartate (NMDA) receptor antagonist, D-2-amino-5-phosphonov

Keratinocyte-derived exosomal miR-31-5p reduces vemurafenib sensitivity in melanoma cells.

BACKGROUND: Resistance to BRAFV600E inhibitors (e.g., vemurafenib) remains a major challenge in malignant melanoma treatment. Keratinocytes influence melanoma cell sensitivity to vemurafenib, but the role of keratinocyte-derived exosomes in melanoma cell sensitivity to vemurafenib remains unclear. OBJECTIVE: In this study, we aimed to investigate the effects and mechanisms of keratinocyte-derived exosomes on melanoma cell growth and sensitivity to vemurafenib. METHODS: Exosomes were extracted from HaCaT keratinocytes. The growth of melanoma cell line A375 and UACC62 was assessed in vitro and

Molecular Umbrella as a Nanocarrier for Antifungals.

A molecular umbrella composed of two O-sulfated cholic acid residues was applied for the construction of conjugates with cispentacin, containing a "trimethyl lock" (TML) or o-dithiobenzylcarbamoyl moiety as a cleavable linker. Three out of five conjugates demonstrated antifungal in vitro activity against C. albicans and C. glabrata but not against C. krusei, with MIC90 values in the 0.22-0.99 mM range and were not hemolytic. Antifungal activity of the most active conjugate 24c, containing the TML-pimelate linker, was comparable to that of intact cispentacin. A structural analogue of 24c, conta

Behavioral activity of 1S,3R-ACPD, an agonist of metabotropic glutamate receptors.

The influence of intracerebroventricular (i.c.v.) injections of 1-aminocyclopentane-1,3-dicarboxylic acid (1S,3R-ACPD), an agonist of metabotropic glutamate receptors, on the activity of the central nervous system was examined in rats. 1S,3R-ACPD at the i.c.v. doses of 100 and 200 nmole significantly decreased apomorphine-induced stereotypy, and at a dose 200 nmole significantly diminished catalepsy caused by haloperidol, two behavioral symptoms controlled mainly by central dopaminergic system. The tested compound had no influence on the locomotor and exploratory activity of rats estimated in

Antagonism of presynaptically mediated depressant responses and cyclic AMP-coupled metabotropic glutamate receptors.

The depression of monosynaptic excitation of neonatal rat motoneurones by (1S,3S)- and (1S,3R)-1-aminocyclopentane-1,3-dicarboxylate (ACPD) and by L-2-amino-4-phosphonobutyrate (L-AP4), which is probably presynaptically mediated, is antagonized by (+/-)- and (+)-alpha-methyl-4-carboxyphenylglycine (MCPG). The same phenylglycine derivatives also antagonize the depression of forskolin-stimulated cyclic AMP synthesis effected by the two ACPD stereoisomers and by L-AP4. These results support previous suggestions that presynaptic depression is mediated by metabotropic glutamate receptors negatively

Activation of metabotropic glutamate receptors induces an outward current which is potentiated by methylxanthines in rat cerebellar Purkinje cells.

The responses of slice-cultured Purkinje cells to trans-DL-1-amino-1,3-cyclopentanedicarboxylic acid (t-ACPD) were examined by intracellular recording techniques and fura-2 microfluorometry. Bath-application of t-ACPD (100 microM, 30 s), a selective agonist of metabotropic glutamate receptors (mGluRs), to Purkinje cells voltage-clamped near their resting potential -65 to -60 mV) consistently induced a transient inward current, followed by a slower outward current (Iout). This outward current was characterized by a linear current-voltage relationship in the range from -130 to -60 mV and accompa

Nature of inhibition of renal aspartate reabsorption in experimental Fanconi syndrome.

Administration of heavy metals or maleic acid induces a condition resembling the Fanconi syndrome. In the case of metals, an apparently uncompetitive inhibition of reabsorption of amino acids was observed 2 days or longer after injection into rabbits (E. C. Foulkes and S. Blanck, Toxicol. Appl. Pharmacol. 64, 103-107, 1982). Transport of aspartate is now shown to be similarly inhibited by maleic acid and cephaloridin. Because maleic acid, as well as p-chloromercuribenzoate (PCMB) and Cd (in presence of mercaptoethanol), exerts an apparent uncompetitive effect within minutes of injection, the i

N6-methyladenosine RNA landscape in the aged mouse hearts.

OBJECTIVE: Cardiac aging is a major risk factor for the development of cardiovascular diseases. Although evidence suggests an association between N6-methyladenosine (m6A) modification and numerous cardiovascular diseases, its role in cardiac aging remains unclear. This study was conducted to elucidate the role of m6A modification in cardiac aging and the molecular mechanisms involved. METHODS: Global methylation levels and the expression of major m6A regulators were compared between young and aged hearts. Transcriptome-wide m6A landscape analysis was conducted using methylated RNA immunopreci

Hepatoprotective effects of S-adenosyl-L-methionine against alcohol- and cytochrome P450 2E1-induced liver injury.

S-adenosyl-L-methionine (SAM) acts as a methyl donor for methylation reactions and participates in the synthesis of glutathione. SAM is also a key metabolite that regulates hepatocyte growth, differentiation and death. Hepatic SAM levels are decreased in animal models of alcohol liver injury and in patients with alcohol liver disease or viral cirrhosis. This review describes the protection by SAM against alcohol and cytochrome P450 2E1-dependent cytotoxicity both in vitro and in vivo and evaluates mechanisms for this protection.

ACPD-mediated slow-onset potentiation is associated with cell death in the rat CA1 region in vivo.

Slow-onset potentiation of synaptic transmission in the hippocampus in vitro and in vivo is induced by application of the metabotropic glutamate receptor (mGluR) agonist, 1S,3R-amino cyclopentane 2,3-dicarboxylic acid (ACPD). This study investigated the cellular response in the CA1 region of freely moving rats to ACPD application. Drugs were applied via the lateral cerebral ventricle, and measurements were obtained from the CA1 region via permanently implanted electrodes. ACPD (20 nmol/5 microl) produced a dose-dependent slow-onset potentiation in the CA1 region which lasted over 4 h. Histolog

Metabotropic glutamate receptor agonist ACPD inhibits some, but not all, muscarinic-sensitive K+ conductances in basolateral amygdaloid neurons.

Muscarinic agonists produce membrane depolarization and losses of spike frequency accommodation and the slow afterhyperpolarization (AHP) when applied to neurons of the basolateral amygdala (BLA). Underlying these changes are the muscarinic-induced inhibitions of several K+ conductances, including the voltage-activated M-current (IM), a slowly decaying Ca(2+)-activated current (IAHP), a voltage-insensitive leak current (ILeak), and the hyperpolarization-activated inward rectifier current (IIR). Similar depolarizations and losses of the slow AHP have been observed in other neuronal cell types f

Ablation of TRPV1 Elevates Nocturnal Blood Pressure in Western Diet-fed Mice.

BACKGROUND: This study tested the hypothesis that genetically ablation of transient receptor potential vanilloid type 1 (TRPV1) exacerbates impairment of baroreflex in mice fed a western diet (WD) and leads to distinct diurnal and nocturnal blood pressure patterns. METHODS: TRPV1 gene knockout (TRPV1-/-) and wild-type (WT) mice were given a WD or normal diet (CON) for 4 months. RESULTS: Capsaicin, a selective TRPV1 agonist, increased ipsilateral afferent renal nerve activity in WT but not TRPV1-/- mice. The sensitivity of renal sympathetic nerve activity and heart rate responses to barorefle

Competitive antagonism at metabotropic glutamate receptors by (S)-4-carboxyphenylglycine and (RS)-alpha-methyl-4-carboxyphenylglycine.

Intrastriatal and intrasubthalamic stimulation of metabotropic glutamate receptors: a behavioral and Fos immunohistochemical study.

Prior work has shown that intrastriatal injection of the metabotropic glutamate receptor agonist 1S,3R-ACPD results in pronounced contralateral rotation, and the basis for this effect is thought to be increased activity of dopaminergic nigrostriatal neurons. We tested this hypothesis by determining the expression of Fos-like immunoreactivity after intrastriatal injection of 1S,3R-ACPD. Intense Fos-like immunoreactivity was noted in the globus pallidus, entopeduncular nucleus, subthalamic nucleus and substantia nigra pars reticulata. Properties of a slow nonselective cation conductance modulated by neurotensin and other neurotransmitters in midbrain dopaminergic neurons.

1. A widespread mechanism of slow excitation throughout the nervous system involves overlapping changes in nonselective ion conductance and K+ conductance. We used whole cell patch-clamp recording to characterize such a nonselective conductance induced by neurotensin (NT) and other neurotransmitters in immunocytochemically identified dopaminergic neurons cultured from the rat ventral tegmental area (VTA). 2. The NT-induced inward current consisted of an initial peak and later "hump." The response was blocked reversibly by the nonpeptide NT-receptor antagonist SR48692, suggesting that it result

Protective effect of the metabotropic glutamate receptor agonist, DCG-IV, against excitotoxic neuronal death.

(2S,1'R,2'R,3'R)-2-(2,3-Dicarboxycyclopropyl)glycine (DCG-IV), a potent agonist of subtypes 2 and 3 of metabotropic glutamate receptors (mGluR2 or 3), protected cultured cortical neurons against excitotoxicity induced either by a brief exposure to N-methyl-D-aspartate (NMDA) or a prolonged exposure to kainate. As a neuroprotective agent, DCG-IV was much more potent than the mixed agonists 1S,3R-aminocyclopentane-1,3-dicarboxylic acid (1S,3R-ACPD) or (2S,1'S,2'S)-2-(carboxycyclopropyl)glycine (L-CCG-I), suggesting a neuroprotective role for mGluR2 or 3 against excitotoxic neuronal death.
