Many male infertility cases have no apparent cause, being characterized as idiopathic. Both inflammation and obesity have long been associated with infertility. On one hand, inflammation, such as orchitis and male accessory gland infections (MAGIs), are regulated by inflammatory cytokines. The latter are also produced in the testis by Leydig and Sertoli cells, being associated with gap junctional communication at the blood-testis barrier. Furthermore, they regulate spermatogenesis through cell interaction, Toll-like receptors and production of reactive oxygen species. Additionally, they affect

The cytokines are an important substance involved in the immune reaction and maintenance of homeostasis. An imbalance in the cytokine network may lead to inflammation and autoimmune diseases such as rheumatoid arthritis (RA). RA is an autoimmune and systemic inflammatory disorder characterized by synovial inflammation, destruction of cartilage and bone and systemic manifestations. The pro-inflammatory cytokines such as tumor necrosis factor α (TNFα), interleukin-1 (IL-1), IL-6 and IL-17 induce the inflammation of the joints and destruction of bone and cartilage via activation of macrophages, f

Lymphangiogenesis is the process by which new lymphatic vessels grow in response to pathologic stimuli such as wound healing, inflammation, and tumor metastasis. It is well-recognized that growth factors and cytokines regulate lymphangiogenesis by promoting or inhibiting lymphatic endothelial cell (LEC) proliferation, migration and differentiation. Our group has shown that the expression of T-helper 2 (Th2) cytokines is markedly increased in lymphedema, and that these cytokines inhibit lymphatic function by increasing fibrosis and promoting changes in the extracellular matrix. However, while t

Th17 cells are implicated in human autoimmune diseases, such as rheumatoid arthritis (RA), although it has not been established whether this persistent destructive arthritis is driven by Th1 and/or Th17 cells. Interleukin-17A (IL-17A) contributes to the pathogenesis of arthritis as has been shown in several experimental arthritis models. Importantly, recent data from first clinical trials with anti-IL-17A antibody treatment in psoriatic arthritis patients and RA patients looks promising. This review summarizes the findings about the role of Th17 cells in arthritis and discusses the impact of t

Experimental studies indicate that the proinflammatory cytokines interleukin-1, interleukin-6, and tumor necrosis factor-alpha are important regulators of bone resorption and may play an important role in age- and estrogen deficiency-related bone loss. Although the observation of accelerated bone loss in patients with inflammatory disorders supports this mechanism, the role of cytokines in the etiology of osteoporosis has yet to be determined. Elucidation of this potential relationship could not only provide clinicians with an additional tool to identify patients at risk for osteoporosis, but

Cytokines are proteins produced by leukocytes and other cells that function as intercellular mediators acting on several target tissues, resulting in multiple biologic actions. Over the last decade, medical research has explored the interaction between cytokines and sleep disorders. The aim of this review is to illustrate recent advances in knowledge about the relationship between cytokines and disorders of excessive sleepiness. Cytokines may have an important role in mediating excessive daytime sleepiness in sleep loss or insomnia. Alterations of the immune system have also been associated wi

Cytokines are intercellular signal molecules involved in the immune pathogenesis of infectious diseases. Cytokine mediated pathological alterations are mainly attributable to an imbalance in cytokine production. Experimental and clinical studies have documented the fact that Salmonella infections induce a Th1 dependent immune response and interferon-gamma and interleukin-12 are central to its genesis. Salmonella endotoxins, flagellins and porins are among the chief mediators of cytokine release. Lectin binding proteins, Toll-like receptors, transcription factors and various cells also contribu

Cytokines are a heterogeneous group of polypeptides that were originally described to mediate activation of the immune system and inflammatory responses. Most of them have meanwhile been shown to be also produced by and act on the peripheral and central nervous system. There is ample evidence from experimental studies that proinflammatory cytokines induce or increase neuropathic pain as well as inflammatory pain. Direct actions of cytokines on afferent nerve fibers have been shown as well as actions involving further mediators such as prostaglandins or nerve growth factor. Inhibition of proinf

Cytokines (interleukins, chemokines, and some growth factors) play an important role in cancer, metabolic disorders, autoimmune disorders and inflammatory diseases, such as rheumatoid arthritis, asthma, Crohn's disease, psoriasis, multiple sclerosis and asthma. Cytokine-based drugs and anticytokine therapies are an increasingly important class of drugs in the treatment and management of many diseases. Interferons are being used to treat viral diseases and cancers. Anti-tissue necrosis factor therapies, such as Enbrel (etanercept; Immunex Corp) and Remicade (infliximab; Centocor) have demonstra

In many forms of cardiomyopathic left ventricular (LV) dysfunction, there is a rapid myocardial expression of pro-inflammatory cytokines such as interleukin 1, interleukin 6 and tumour necrosis factor-alpha (TNF-alpha) which mediate, via specific receptors, various processes such as gene expression, cell growth or apoptosis. In the initial stages of myocarditis, the myocardial expression of proinflammatory cytokines appears to be part of an inflammatory process. In many other conditions such as ischaemic cardiomyopathy and chronic LV pressure or volume overload, myocardial expression of proinf

The implantation process is currently considered the most relevant limiting factor for successful pregnancy. It is evident that molecular interactions at the embryo-maternal interface at the time of implantation are crucial in order to understand the mechanisms of embryonic implantation. The principal aim of this study is to demonstrate the existence of specific communication pathways between the human embryo and endometrium. The molecular interactions appears to be initiated by the endometrium in the presence of an implanting blastocyst and is mediated through embryonic cytokines (specificall

Cytokines are peptide regulatory factors which are involved in initiating, amplifying, perpetuating and resolving the immune and inflammatory responses. Each may have several actions, but can be broadly categorized into pro- and anti-inflammatory cytokines. The cytokine profile depends on the nature of the antigen, how it is presented to the host, and which cell types are involved in the response. Elucidation of the cytokine cascade will improve understanding of the mechanisms of inflammation, and may allow the identification of therapeutic targets in chronic inflammatory diseases of unknown a

OBJECTIVE: Cytokines take place in immunologic and inflammatory reactions. The aim of this study was to describe the roles of various cytokines in the pathogenesis of Behçet's disease (BD).
METHODS: Serum levels of interleukin-1 beta (IL-1 beta), interleukin-2 (IL-2), soluble IL-2 receptor (sIL-2R), interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-alpha) were studied in 24 patients with active BD and 15 healthy adults as controls, by ELISA technique. Patients were separated into 2 groups according to the number of active organ systems (Group I: < 3 vs Group II: > or = 3).

Cytokines play a crucial role in autoimmune thyroid disease (ATD) through various mechanisms. They are produced in the thyroid by intrathyroidal inflammatory cells, in particular lymphocytes, as well as by the thyroid follicular cells (TFC) themselves and may thus act in a cascade to enhance the autoimmune process (Fig. 1). Cytokines upregulate the inflammatory reaction through stimulation of both T and B cells, resulting in antibody production and tissue injury. In addition, intrathyroidal cytokines induce immunological changes in TFC including enhancement of both major histocompatibility com

Cytokines are soluble factors that play a critical role in mediating cell to cell interactions within skeletal tissues. These effects are mediated by paracrine, autocrine, and juxtacrine mechanisms. There are also examples in which the cytokines can function in an endocrine fashion. The regulatory functions of the cytokines are performed throughout life, beginning with bone growth and development and continuing in the mature organism, in which bone remodeling is regulated. The cytokines can be grouped into distinct families based on their principal biologic activity and cell target. However, t

The strong resemblance between the clinical manifestations of hemolytic transfusion reactions and sepsis, in which cytokine production has a central role, suggests that similar pathophysiologic mechanisms are involved. There is an expanding body of clinical and experimental evidence that cytokines, especially interleukin-1, tumor necrosis factor, interleukin-6, and interleukin-8, are principle mediators of immune responses to erythrocyte incompatibility. Recent studies have further suggested that the monocyte chemotactic and activating factor, monocyte chemoattractant protein-1, and the anti-i

Cytokines are multifunctional signaling peptide molecules which regulate a plethora of cellular activities in the immune system. Cytokines provide a means of communication between the immune system and its non-immune neighbours. Several clinical entities have been recognized as targets for clinical applications including interaction with malignant cell growth, host defence against infectious agents, negative regulation of autoagressive disorders and regulation of tissue and cell regeneration. Interferon-alpha (IFN-alpha) can effectively regulate malignant growth in hairy-cell leukaemia, the fi

Cytokines are secreted by several cell types in the bone microenvironment. These peptides act on bone cells by a paracrine or autocrine mechanism and play an important role, although not completely clarified, in the regulation of bone remodeling. Postmenopausal osteoporosis could be due to a local overproduction of some osteoclast-stimulating cytokines in response to estrogen deficiency. During chronic inflammatory joint diseases, such as rheumatoid arthritis, synovial cells produce large amounts of cytokines leading to increased local bone resorption and juxta-articular bone destructions. Numerous abnormalities in neonatal host-defense mechanisms have been documented over the past decade. Profound disturbances in myeloid progenitor proliferation, bone marrow neutrophil storage pools, a tendency to peripheral neutropenia and significant in vitro abnormalities of mature neutrophil cell effector function all predispose the neonate to a high mortality rate during bacterial sepsis. The recent use of hematopoietic CSFs and other cytokines to enhance host defense mechanisms in the adult has suggested a role for this new form of immunotherapy in the newborn. Several studies over the pa

Elements of the immune system must take their place alongside other potential mechanisms of psoriasis, such as psoriatic epidermal keratinocytic hyperproliferation, endothelial cell and fibroblast activation and proliferation, abnormal lipid regulation, and transmembrane signalling abnormalities. These data provide support for the concept that cellular immunologic processes are active in a manner that further promotes the patho-physiologic events observed in psoriasis. Thus, therapies useful for psoriasis may have activity on immunologic processes in addition to more traditional mechanistic co
