Influence of cytokinins, auxins and polyamines on in vitro mass multiplication of cotton (Gossypium hirsutum L. cv. SVPR2).

In the present investigation, the influence of different forms of cytokinins, auxins and polyamines were tested for mass multiplication and regeneration of cotton. Initially, for the identification of effective concentration for multiple shoot induction, various concentrations of BAP, Kin and 2iP along with IAA and NAA were tested. Among tested concentrations, media fortified with MS salts; B5 vitamins; 30 g/l, glucose; 2.0 mg/l, 2iP; 2.0 mg/l, IAA and 0.7 % agar showed best response for multiplication of shoot tip explants (20 shoots per shoot tip explants). In nodal explants, maximum of 18.6

The plant longevity gene AHL15 delays leaf senescence by repressing ORESARA1 and cytokinin degradation.

The Arabidopsis thaliana (Arabidopsis) AT-HOOK MOTIF NUCLEAR LOCALIZED 15 (AHL15) gene is associated with various longevity phenotypes and extends the life span of plants when overexpressed. In this study, we show that, in addition to previously described longevity phenotypes, constitutive overexpression of AHL15 in Arabidopsis delays leaf senescence, whereas ahl15 loss of function accelerates this process. Dexamethasone-induced nuclear localization of AHL15-GR during dark-triggered senescence results in a stay-green phenotype and represses the expression of several early senescence-associated

A Constitutively Active Cytokinin Receptor Variant Increases Cambial Activity and Stem Growth in Poplar.

The cambial meristem is responsible for bark and wood formation in woody plants. The activity of the cambial meristem is controlled by various factors; one of them is the plant hormone cytokinin. Here, we have explored different approaches to genetically engineering cambial activity in poplar plants by the ectopic expression of a cytokinin biosynthesis gene with enhanced activity (named ROCK4) or of a gene encoding a constitutively active cytokinin receptor variant (ROCK3). Both genes are derived from Arabidopsis thaliana and were expressed in poplar trees under the control of their own promot

GmRAV1 regulates regeneration of roots and adventitious buds by the cytokinin signaling pathway in Arabidopsis and soybean.

The division and differentiation of cells are the basis of growth and development. Cytokinin plays an active role in cell growth division and differentiation. The Related to ABI3/VP1 (RAV) family comprises transcription factors in plants and all contain both AP2- and B3-like domains. In this study, GmRAV1 (Glycine max), which belongs to the AP2/ERF transcription factor family, was isolated and functionally characterized. Subcellular localization showed that GmRAV1 was localized to the nucleus and quantitative real-time polymerase chain reaction (qRT-PCR) analysis indicated that GmRAV1 was indu

Senescence-induced expression of cytokinin reverses pistil abortion during maize flower development.

Maize is a monoecious species that produces imperfect (unisexual), highly derived flowers called florets. Within the spikelet, the basic repeating unit of the maize inflorescence, the spikelet meristem gives rise to an upper and a lower floret. Although initially bisexual, floret unisexuality is established through selective organ elimination. In addition, the lower floret of each ear spikelet is aborted early in its development, leaving the upper floret to mature as the only pistillate floret. Expression from the cytokinin-synthesizing isopentenyl transferase (IPT) enzyme under the control of

Comparative Effects of Cytokinins and Spermidine on In Vitro Organogenesis of Ruscus hypoglossum L. and Rooting of Regenerated Shoots.

Ruscus hypoglossum L. is an evergreen ornamental species valued as cut foliage, but its commercial production is constrained by slow vegetative propagation and limited availability of uniform planting material. This study aimed to develop an efficient in vitro propagation protocol for R. hypoglossum by evaluating the effects of different cytokinins and spermidine on shoot organogenesis and subsequent rooting. Thidiazuron (TDZ), zeatin, kinetin, and spermidine were applied at 0.5 and 1.0 mg L-1 to rhizome explants derived from in vitro-grown seedlings. Explant length, shoot and cladode number,

Auxin and Cytokinins-Mediated Regulation of Adventitious Shoot Regeneration via the RpARR12/RpARF19-RpWOX4-RpLBD4 Pathway in Robinia pseudoacacia.

Woody plants are fast-growing and ecologically important, with widespread applications in urban landscaping and ecological restoration. However, limited understanding of the molecular mechanisms underlying adventitious shoot regeneration has hindered advances in tissue culture and genetic improvement of woody species. Here, we present a comprehensive framework for elucidating adventitious shoot regeneration in Robinia pseudoacacia. Histological analyses delineated key developmental stages, while RNA-sequencing and hormone profiling highlighted the critical roles of auxin and cytokinins. WGCNA

Cytokinin activity of N6-benzyladenine derivatives assayed by interaction with the receptors in planta, in vitro, and in silico.

Biological effects of hormones in both plants and animals are based on high-affinity interaction with cognate receptors resulting in their activation. The signal of cytokinins, classical plant hormones, is perceived in Arabidopsis by three homologous membrane receptors: AHK2, AHK3, and CRE1/AHK4. To study the cytokinin-receptor interaction, we used 25 derivatives of potent cytokinin N6-benzyladenine (BA) with substituents in the purine heterocycle and/or in the side chain. The study was focused primarily on individual cytokinin receptors from Arabidopsis. The main in planta assay system was ba

The Compact Root Architecture 2 systemic pathway is required for the repression of cytokinins and miR399 accumulation in Medicago truncatula N-limited plants.

Legume plants can acquire mineral nitrogen (N) either through their roots or via a symbiotic interaction with N-fixing rhizobia bacteria housed in root nodules. To identify shoot-to-root systemic signals acting in Medicago truncatula plants at N deficit or N satiety, plants were grown in a split-root experimental design in which either high or low N was provided to half of the root system, allowing the analysis of systemic pathways independently of any local N response. Among the plant hormone families analyzed, the cytokinin trans-zeatin accumulated in plants at N satiety. Cytokinin applicati

Cytokinin represses NH4+ efflux by suppressing GH3.5 transcription through an ARR10/12-dependent pathway.

Ammonium (NH4+) is an essential nitrogen source for plants, yet its excessive accumulation triggers toxicity, a process typically coupled to futile NH4+ cycling in roots cells. While the cytokinin-ARR10/12 signaling pathway is known to regulate vacuolar NH4+ sequestration, it is not known whether it also coordinates NH4+ efflux across the root-cell plasma membrane to maintain intracellular homeostasis. Here, we identify the auxin-amido synthetase GH3.5 as a direct transcriptional target of ARR10, using a combination of yeast one-hybrid (Y1H) assays, dual-luciferase reporter assays, and CUT&RUN

Thidiazuron, a phenyl-urea cytokinin, inhibits ergosterol synthesis and attenuates biofilm formation of Candida albicans.

Candida albicans is a common human fungal pathogen that colonizes mucosa and develops biofilm in the oral cavity that causes oral candidiasis. It has been reported that cytochrome P450 enzyme (CYP51), a vital part of the ergosterol synthesis cascade, is associated with Candida infections and its biofilm formation. Thidiazuron, a phenyl-urea cytokinin, exhibits anti-senescence and elicitor activity against fungal infection in plants. However, how Thidiazuron impacts C. albicans biofilm formation is still uncertain. Here, we aimed to investigate the effects of a Thidiazuron against the growth an

Cytokinin response induces immunity and fungal pathogen resistance, and modulates trafficking of the PRR LeEIX2 in tomato.

Plant immunity is often defined by the immunity hormones: salicylic acid (SA), jasmonic acid (JA), and ethylene (ET). These hormones are well known for differentially regulating defence responses against pathogens. In recent years, the involvement of other plant growth hormones such as auxin, gibberellic acid, abscisic acid, and cytokinins (CKs) in biotic stresses has been recognized. Previous reports have indicated that endogenous and exogenous CK treatment can result in pathogen resistance. We show here that CK induces systemic immunity in tomato (Solanum lycopersicum), modulating cellular t

Genome-wide characterization of histidine kinase genes in Aciculosporium take and their potential roles in cytokinin sensing.

Histidine kinases (HKs) are critical receptors within two-component regulatory systems, helping fungi perceive environmental stimuli. However, the functions of the HKs gene family in Aciculosporium take, which mediates cytokinin (CKs) imbalance during bamboo witches' broom disease, and the mechanisms by which HKs respond to CKs and environmental stimuli remain unelucidated. This study identified 10 AtaHKs genes, categorizing them into 8 groups. Phylogenetic analysis across multiple species suggested that Group I AtaHKs were closely linked to plant pathogenicity. The promoter regions of AtaHKs

ABA Signaling De-Repression Coordinates With Cytokinin to Drive Large Petal Formation in Brassica napus.

Petal size is a critical agronomic trait influencing pollination efficiency and yield in field crops. However, the cellular basis and upstream hormonal interplay governing petal size in Brassica napus remain unclear. In this study, utilizing a natural variation population of "Zhongshuang 11", we investigated biological mechanisms underlying petal development through integrated phenotypic screening, paraffin section-based cytological analysis, targeted hormone metabolomics, and transcriptome. The results revealed that the single-flower area of large petals (BEP; 13.0 mm2) was significantly larg

Hydrogen sulfide antagonizes cytokinin to change root system architecture through persulfidation of CKX2 in Arabidopsis.

Hydrogen sulfide (H2S) is an endogenous gaseous signaling molecule, which has been shown to play an important role in plant growth and development by coupling with various phytohormones. However, the relationship between H2S and cytokinin (CTK) and the mechanisms by which H2S and CTK affect root growth remain poorly understood. Endogenous CTK was analyzed by UHPLC-ESI-MS/MS. Persulfidation of cytokinin oxidase/dehydrogenases (CKXs) was analyzed by mass spectrometry (MS). ckx2/CKX2wild-type (WT), OE CKX2 and ckx2/CKX2Cys(C)62alanine(A) transgenic lines were isolated with the ckx2 background. Sex ratio of mirid populations shifts in response to hostplant co-infestation or altered cytokinin signaling .

Herbivore species sharing a host plant often compete. In this study, we show that host plant-mediated interaction between two insect herbivores - a generalist and a specialist - results in a sex ratio shift of the specialist's offspring. We studied demographic parameters of the specialist Tupiocoris notatus (Hemiptera: Miridae) when co-infesting the host plant Nicotiana attenuata (Solanaceae) with the generalist leafhopper Empoasca sp. (Hemiptera: Cicadellidae). We show that the usually female-biased sex ratio of T. notatus shifts toward a higher male proportion in the offspring on plants co-i

Tobacco susceptibility to Potato virus Y(NTN) infection is affected by grafting and endogenous cytokinin content.

Faster or stronger response to pathogen occurs if plants undergo prior priming. Cytokinins seem to be also involved in plant priming and in response to pathogens. Susceptibility to Potato virus Y(NTN) (PVY(NTN)) was studied in transgenic cytokinin overproducing (Pssu-ipt) tobacco and compared with nontransgenic plants. Since cytokinin overproduction inhibits development of plant roots and grafting overcomes this limitation, both types were grown as rooted and/or grafted plants to check also the effect of grafting. Control rooted tobacco (C), the most susceptible to PVY(NTN), showed always symp

Agroinfiltration by cytokinin-producing Agrobacterium sp. strain GV3101 primes defense responses in Nicotiana tabacum.

Transient infiltrations in tobacco are commonly used in plant studies, but the host response to different disarmed Agrobacterium strains is not fully understood. The present study shows that pretreatment with disarmed Agrobacterium tumefaciens GV3101 primes the defense response to subsequent infection by Pseudomonas syringae in Nicotiana tabacum. The presence of a trans-zeatin synthase (tzs) gene in strain GV3101 may be partly responsible for the priming response, as the tzs-deficient Agrobacterium sp. strain LBA4404 only weakly imparts such responses. Besides inducing the expression of defens

The genes involved in cytokinin biosynthesis in Erwinia herbicola pv. gypsophilae: characterization and role in gall formation.

A locus conferring cytokinin production was previously isolated from the gall-forming bacterium Erwinia herbicola pv. gypsophilae. This locus resided in a cluster with the genes specifying indole-3-acetic acid production on the pathogenicity-associated plasmid pPATH (A. Lichter, S. Manulis, O. Sagee, Y. Gafni, J. Gray, R. Meilen, R. O. Morris, and I. Barash, Mol. Plant Microbe Interact., 8:114-121, 1995).
