Metabarcoding Reveals Diversity of Potentially Toxic Algae in Papeete Port (Tahiti).

Harmful algae are transported in various compartments of maritime vessels, making ports with heavy maritime traffic potential hotspots for their introduction and spread. In this study, we investigate the port of Papeete (Tahiti, French Polynesia), a key hub for numerous South Pacific shipping routes. Using metabarcoding on DNA extracted from water samples (environmental DNA, eDNA) we identified 21 species of harmful algae comprising to Bacillariophyceae (4), Dinophyceae (14), and Haptophyta (3 species). Three of those species are directly associated with fish mortality events without recognize

Cell death upon H(2)O(2) induction in the unicellular green alga Micrasterias.

In the present study, we investigated whether the unicellular green alga Micrasterias denticulata is capable of executing programmed cell death (PCD) upon experimental induction, and which morphological, molecular and physiological hallmarks characterise this. This is particularly interesting as unicellular freshwater green algae growing in shallow bog ponds are exposed to extreme environmental conditions, and the capacity to perform PCD may be an important strategy to guarantee survival of the population. The theoretically 'immortal' alga Micrasterias is an ideal object for such investigation

Ultrasensitive okadaic acid detection via DNA hydrogel-gated organic photoelectrochemical transistor biosensing platform.

The burgeoning organic photoelectrochemical transistor (OPECT) biosensor has ascended as a promising technology for the detection and analysis of biomolecules. In this endeavor, a novel DNA hydrogel ultramicro-change gated OPECT aptasensor was innovatively conceived. Specifically, Sn3O4/CdS QDs photosensitive material was introduced onto the working electrode, which served for interface and light modulation and exhibited a high current gain. Unlike conventional biomolecular interfaces, a targeted DNA hydrogel layer was established at the interface through the actions of a catalyst and initiato

Zooplankton may serve as transmission vectors for viruses infecting algal blooms in the ocean.

Marine viruses are recognized as a major driving force regulating phytoplankton community composition and nutrient cycling in the oceans. Yet, little is known about mechanisms that influence viral dispersal in aquatic systems, other than physical processes, and that lead to the rapid demise of large-scale algal blooms in the oceans. Here, we show that copepods, abundant migrating crustaceans that graze on phytoplankton, as well as other zooplankton can accumulate and mediate the transmission of viruses infecting Emiliania huxleyi, a bloom-forming coccolithophore that plays an important role in

Combined cytotoxicity and genotoxicity of a marine toxin and seafood contaminant metal ions (chromium and cadmium).

Algal bloom with consequent production of marine toxins contaminating bivalves is increasing in costal regions worldwide because of sea water quality worsening. Contamination of seafood by diarrheic shellfish poisoning toxins (DSP) together with metals is frequently reported, a phenomenon not fully explained yet. In this context, metal ions were assayed in clams collected from the banned area of Boughrara, Tunisia, contaminated by Gymnodinium and other algae such as Dinophysis sp, accumulated by these bivalves. The presence of toxic metals ions such as Chromium (Cr) and Cadmium (Cd) in meat, s

Aequorivita gen. nov., a member of the family Flavobacteriaceae isolated from terrestrial and marine Antarctic habitats.

Several strains isolated from Antarctic winter sea water, sea-ice algal assemblages and quartz stone subliths were found to belong to a novel 16S rDNA sequence cluster within the family Flavobacteriaceae (Cytophaga-Flavobacterium-Bacteroides division). The strains were gram-negative, non-motile, psychrotolerant, strictly aerobic, chemoheterotrophic rod-shaped cells that contained orange or yellow carotenoid pigments and required yeast extract when grown in defined mineral-salts media. The requirement for sodium ions varied between strains. Results of DNA-DNA hybridization analysis were used to

Strong and widespread cycloheximide resistance in Stichococcus-like eukaryotic algal taxa.

This study was initiated following the serendipitous discovery of a unialgal culture of a Stichococcus-like green alga (Chlorophyta) newly isolated from soil collected on Signy Island (maritime Antarctica) in growth medium supplemented with 100 µg/mL cycloheximide (CHX, a widely used antibiotic active against most eukaryotes). In order to test the generality of CHX resistance in taxa originally identified as members of Stichococcus (the detailed taxonomic relationships within this group of algae have been updated since our study took place), six strains were studied: two strains isolated from

Sequence-Based Characterization of Microalgal Microbiomes: Impact of DNA Extraction Protocol on Yield and Community Composition.

The bacterial communities associated with microalgae are vital for the growth and health of the host, and engineering algal microbiomes can enhance the fitness of the algae. Characterization of these microbiomes mostly relies on sequencing of DNA, which can be extracted with an array of protocols that potentially impact DNA quantity and quality and thus potentially affect subsequent analyses of microbiome composition. Here, we extracted DNA from Isochrysis galbana, Tetraselmis suecica, and Conticribra weissflogii microbiomes using four different protocols. DNA yield and quality was greatly imp

Cytologic, histologic, microbiologic, and electron microscopic characterization of a canine Prototheca wickerhamii infection.

An adult dog was presented for chronic cough and a recent development of ulcerated, erythematous nares with nasal discharge. Cytology of enlarged peripheral lymph nodes revealed many intracellular and extracellular organisms. These round or rarely oval organisms measured approximately 5-9 µm in diameter and frequently contained several globular structures, ranging from deeply basophilic to magenta. A thin, clear halo was present. Smaller 1-2 µm, magenta forms were also observed. Fungal culture yielded small, wet, raised, irregularly shaped, white to pale tan colonies. Microbiologic staining of

Optimizing DNA extraction and quantitative PCR protocols for the efficient and precise quantification of the toxic dinoflagellate Karenia brevis in water and marine sediments.

In southwest Florida, Harmful Algal Blooms (HABs) caused by the dinoflagellate Karenia brevis occur nearly annually. Monitoring efforts typically depend on the enumeration of K. brevis, identified using morphological characters via light microscopy. Although this technique remains the standard, it requires taxonomic expertise, is not high-throughput, and may be biased towards cells displaying typical morphology. To overcome these limitations, we explore the use of a modified qPCR assay that targets the rbcL gene for monitoring K. brevis in water samples and marine sediments. We tested seven DN

Pyrogallic acid triggers a cascade from suppression of photosynthesis and proliferation to PCD‑like in Microcystis aeruginosa.

Pyrogallic acid (PA) is an effective allelochemical against Microcystis aeruginosa. However, it remains unclear whether its inhibitory specificity is derived from a regulated programmed cell death (PCD)-like cascade. A time-resolved analysis (8-96 h) of M. aeruginosa responses confirmed that 2.0 mg·L-1 PA induced sequential photosynthetic inhibition and proliferative arrest. These early physiological dysfunctions initiated a coherent, time-ordered ROS-NO-Ca2+ signaling cascade, subsequently activating caspase‑3‑like protease and triggering typical PCD‑like events, including DNA fragmentation a

Seasonal distribution of Gambierdiscus spp. in Wakasa Bay, the Sea of Japan, and antagonistic relationships with epiphytic pennate diatoms.

The occurrence of the ciguatera fish poisoning (CFP) causative Gambierdiscus spp. was confirmed in the Sea of Japan for the first time in 2009. This paper reports seasonal distribution of Gambierdiscus spp. and epiphytic diatoms in the Sea of Japan. Monitoring results suggested an antagonistic interaction in abundances between epiphytic diatoms and the dinoflagellate Gambierdiscus spp. Allelopathic effects of diatoms were considered to be involved in the competitive phenomenon. Therefore it is hypothesized that cell densities of epiphytic pennate diatoms on macroalgae are a novel determinant a

Ribonucleotide Misincorporation and Reverse Transcriptase Activities of Plasmodium falciparum Apicoplast DNA Polymerase.

Plasmodium falciparumis the most common and harmful causative agent of malaria worldwide. As a member of the phylum Apicomplexa, P. falciparum is characterized by the presence of a unique and essential organelle called the apicoplast. Reminiscent of an algal chloroplast, the apicoplast possesses its own genome, which is maintained by a single apicoplast DNA polymerase (apPol). Ribonucleotides misincorporated into the genome are among the most common lesions encountered by DNA polymerases, and the ability to replicate past these lesions varies widely among characterized enzymes. Here, we have i

Evaluation of genotoxic responses of Chaetoceros tenuissimus and Skeletonema costatum to water accommodated fraction of petroleum hydrocarbons as biomarker of exposure.

Genotoxic responses towards chronic exposure of Chaetoceros tenuissimus and Skeletonema costatum to water accommodated fraction of petroleum hydrocarbons (WAF-P) were evaluated as biomarkers of petroleum hydrocarbons pollution. The DNA damage caused by water accommodated fraction of petroleum hydrocarbons was assessed in terms of the DNA integrity measured by alkaline unwinding assay. The comparative study of the growth pattern of C. tenuissimus with respect to DNA integrity and the DNA strand breaks in different concentrations of WAF-P showed sufficient tolerance. However, its toxicity increa

Antiviral Mx proteins have an ancient origin and widespread distribution among eukaryotes.

Mx proteins, first identified in mammals, encode potent antiviral activity against a wide range of viruses. Mx proteins arose within the Dynamin superfamily of proteins (DSP), which mediate critical cellular processes, such as endocytosis and mitochondrial, plastid, and peroxisomal dynamics. Despite their crucial role, the evolutionary origins of Mx proteins are poorly understood. Through comprehensive phylogenomic analyses with progressively expanded taxonomic sampling, we demonstrate that Mx proteins predate the interferon signaling system in vertebrates. Our analyses find an ancient monophy

Epibiotic green alga (Ulva spp.) on the skin of turbot (Scophthalmus maximus) with chronic aeromoniasis.

Chronic aeromoniasis in turbot (Scophthalmus maximus), caused by Aeromonas salmonicida, provokes cutaneous lesions that occasionally expose dermal tubercles, which may act as unconventional ecological niches. A storm in February 2024 caused severe damage at a turbot farm in Galicia (Northwestern Spain), leaving various tanks uncovered and subsequently exposing fish with chronic aeromoniasis to several days of intense solar irradiation. Afterward, farm staff observed green tufts over the animals. Two affected turbots were submitted for necropsy. Morphological features identified the epibiont as

Multi-dimensional epigenomic dynamics converge on H3K4-mediated regulation of low-CO2 adaptation in Nannochloropsis oceanica.

Despite their ecological and biotechnological importance, the extent to which microalgae are regulated by epigenetic mechanisms has remained poorly understood. In the model industrial microalga Nannochloropsis oceanica, by comprehensive, multi-dimensional epigenomic analyses, this study uncovers an epigenetic regulatory network responsive to CO2 levels. This network involves intricate interactions among DNA methylation, histone modifications, dynamic nucleosome positioning, and three-dimensional chromatin organization during adaptation to low-CO2 conditions. Although DNA methylation is minimal

Potential role of viruses in white plague coral disease.

White plague (WP)-like diseases of tropical corals are implicated in reef decline worldwide, although their etiological cause is generally unknown. Studies thus far have focused on bacterial or eukaryotic pathogens as the source of these diseases; no studies have examined the role of viruses. Using a combination of transmission electron microscopy (TEM) and 454 pyrosequencing, we compared 24 viral metagenomes generated from Montastraea annularis corals showing signs of WP-like disease and/or bleaching, control conspecific corals, and adjacent seawater. TEM was used for visual inspection of dis
