Ecological applications : a publication of the Ecological Society of America2026eng
Wildfires are a natural disturbance in dry ponderosa pine forest ecosystems, and are increasing in frequency and severity across western North America. The effects of wildfire on biotic and abiotic conditions have important implications for higher trophic levels, including native bees. We sought to quantify the responses of flowering plants to fire age and severity and determine the effects of these shifts on pollinator richness through time. We sequenced pollen from pan and blue vane traps deployed every 2 weeks from April to August in 2022 and 2023 to quantify shifts in foraging resources fo

Lasioseius foliatisetus n. sp. is described based on the morphology and molecular markers of adult females collected in litter/soil samples of the Caatinga and Pantanal, extensive Brazilian biomes. This new species can be distinguished from other Lasioseius species mainly by having fixed cheliceral digit with three teeth in addition to apical tooth, most dorsal shield setae leaf-shaped, and ventrianal shield with seven pairs of setae (including Jv5) in addition to the circumanal setae. The determined nucleotide sequences of the CytB gene and of ITSS of Lasioseius foliatisetus n. sp. are the fi

The family Paramyxoviridae consists of large enveloped RNA viruses infecting mammals, birds, reptiles and fish. Many paramyxoviruses are host-specific and several, such as measles virus, mumps virus, Nipah virus, Hendra virus and several parainfluenza viruses, are pathogenic for humans. The transmission of paramyxoviruses is horizontal, mainly through airborne routes; no vectors are known. This is a summary of the current International Committee on Taxonomy of Viruses (ICTV) Report on the family Paramyxoviridae. which is available at ictv.global/report/paramyxoviridae.

Background: Orchids of the Masdevallia genus are characterized by their beautiful esthetic qualities. However, they are vulnerable to habitat destruction, illegal harvesting, tourism and climate change. These extinction threats have led to the listing of all Masdevallia species in Appendix II of CITES (Convention on International Trade in Endangered Species of Wild Fauna and Flora) and their selection by the IUCN (International Union for Conservation of Nature). Orchids are sold in various forms, e.g., stems or tubers, rendering them impossible to identify based on morphology alone. DNA barcod

Anopheles (Cellia) dirus Peyton & Harrison and Anopheles baimaii Sallum & Peyton are sibling species within the Dirus complex belonging to the Leucosphyrus group, and have been incriminated as primary vectors of malaria in Thailand. In the present study, DNA barcoding and geometric morphometrics were used to distinguish between An. dirus and An. baimaii in the international border areas, Trat Province, eastern Thailand. Our results revealed that DNA barcoding based on the cytochrome c oxidase subunit I gene could not be used to distinguish An. dirus from An. baimaii. The overlapping values bet

Assessing and conserving marine biodiversity remain critical global challenges, particularly in highly disturbed coastal regions. The use of environmental DNA (eDNA)-metabarcoding has revolutionized biodiversity assessment and management; however, the prevalence of both false positives and negatives continues to be a significant concern. To address these technical errors, we tested two potential methodological improvements in the highly disturbed Guangdong-Hong Kong-Macao Greater Bay Area: (1) the use of random whole-genome amplification (WGA) to reduce false negatives derived from low eDNA co

Environmental DNA (eDNA) integrated with metabarcoding is a promising and powerful tool for species composition and biodiversity assessment in aquatic ecosystems and is increasingly applied to evaluate fish diversity. To date, however, no standardized eDNA-based protocol has been established to monitor fish diversity. In this study, we investigated and compared two filtration methods and three DNA extraction methods using three filtration water volumes to determine a suitable approach for eDNA-based fish diversity monitoring in the Pearl River Estuary (PRE), a highly anthropogenically disturbe

Plankton samples have been routinely collected and preserved in formalin in many laboratories and museums for more than 100 years. Recently, attention has turned to use DNA information from formalin-fixed samples to examine changes in plankton diversity over time. However, no molecular ecological studies have evaluated the impact of formalin fixation on the genetic composition of the plankton community structure. Here, we developed a method for extracting DNA from archived formalin-preserved plankton samples to determine their community structure by a DNA metabarcoding approach. We found that

We present a detailed morphological description and a DNA barcoding of Parabrachiella platensis n. sp. collected from Mugil liza Valenciennes in Samborombon Bay (Buenos Aires, Argentina). This new species was compared with two Parabrachiella species parasitic on mugilids: Parabrachiella exilis (Shiino, 1956) and Parabrachiella mugilis (Kabata, Raibaut et Ben Hassine, 1971). Parabrachiella platensis n. sp. differs from those species in the shape of posterior processes, the anal slit with two pairs of bipartite papillae, the size of cephalothorax, the trunk, the maxilla, the microhabitat on the

The Himalayan region is recognized as one of the world's major biodiversity hotspots due to its remarkable altitudinal variation, high species richness, and exceptional endemism. Despite its ecological significance, the endemic flora of the western Himalayas remains insufficiently explored at the molecular level. This study aimed to evaluate the performance of multiple nuclear and chloroplast DNA barcode loci (ITS2, rbcL, trnH-psbA, trnA, trnV, rpoB, ycf3, and rbcLa) for the identification and phylogenetic assessment of 32 endemic and threatened plant species representing 31 genera and 24 fami

DNA metabarcoding of freshwater macroinvertebrate samples can provide rapid and accurate identification of species diversity. Processing samples where macroinvertebrates are not separated from net contents can substantially reduce time and cost. We devised and tested a standardised method for processing combined kick/sweep samples using DNA metabarcoding to provide accurate estimates of macroinvertebrate species diversity. Kick and sweep samples were collected from 10 sites across Greater Melbourne known to vary in catchment land use, macroinvertebrate abundance and diversity as well as the ty

Demosponge classification is notoriously challenged by the paucity of informative morphological characters with sufficient complexity to discriminate between apomorphies and convergences. Molecular data, preferably from type material, helps shed light on phylogenetic relationships. In the following, we review, based on the results of DNA barcoding of type (and other) material, the classification of several demosponge species and genera with either eminent or previously uncertain classification. We report that the aster-bearing genus Leptosastra Topsent, 1904, is a poecilosclerid, unlike Clathr

Gut nematodes influence animal health and fitness, with effects shaped by their abundance and community composition. While closely related host species tend to harbour similar nematodes, the relative roles of host ecology and phylogeny in structuring nematode communities remain unclear. Here, we assess how host holobiont traits-body size, diet, space use, potentially pathogenic gut microbes and overall gut microbiome composition-predict gut nematode abundance and composition, while controlling for host phylogenetic relatedness. We jointly analyse DNA metabarcoding data on nematodes, microbiome

Estimating the number of insect species on Earth is a daunting challenge. The current consensus estimate-about six million species-is likely far too low, as we will show. Our estimate of the global number of insect species rests on a sample of more than 1,600,000 DNA-barcoded insect specimens representing 53,945 species from 15 "core" Malaise traps deployed in dry forest, cloud forest, and rainforest ecosystems of the Área de Conservación Guanacaste (ACG) in Costa Rica. Even this massive sample fails to reveal the full extent of ACG insect species richness. To estimate total ACG insect richnes

Using DNA techniques for prey identification is an emerging approach for enhancing the precision and accuracy of trophic information. We evaluated the effectiveness of DNA-based prey identification in conjunction with visual stomach content analysis of commercially important groundfish in the nearshore Gulf of Maine, with a focus on distinguishing consumed anadromous (Alosa spp.) and closely related marine prey (Atlantic herring Clupea harengus, Atlantic menhaden Brevoortia tyrannus). DNA barcoding of 179 consumed prey specimens provided species- or genus-level identification in 122 cases (68.

Environmental RNA-based metabarcoding has emerged as a powerful tool for biodiversity assessment across diverse disciplines. During field sampling and sample processing, collected water samples often require short-term storage. However, the effects of different storage conditions on biodiversity recovery remain largely unexplored. Here we evaluated the impacts of various storage temperatures (4, 10, 20 °C, and air temperature) and durations (1-72 h) on fish biodiversity recovery from eRNA samples. Our findings revealed that taxon richness declined significantly with increasing storage time and

Salinity has been identified as a primary factor structuring community composition in global diversity assessments of free-living bacteria in non-extreme environments. It remains unknown whether the same is true for protists. Moreover, changes in alpha diversity of both bacteria and protists across the brackish salinity gradient remain elusive. Here, we analyzed protist and bacterial diversity using DNA metabarcoding across 398 surface water samples synchronized with national monitoring of the Baltic Sea and the Kattegat-Skagerrak. We show that salinity, unlike other environmental factors, had

Advances in high-throughput DNA sequencing technologies have accelerated research on aquatic microorganisms, their diversity, and ecological functions. This has also significantly advanced research on fungi that shape aquatic ecosystems and food webs. Here, we present the FunAqua dataset, which describes global fungal biodiversity in aquatic ecosystems based on eukaryotic long-read metabarcoding data from 1,076 sediment and 1,299 filtered water samples collected across 1,452 sampling sites in 87 countries. We targeted the internal transcribed spacer (ITS) region. In total, 50,152 fungal OTUs w

Ribes janczewskii is a rare and valuable plant known for its resistance to spring frosts, pests, and diseases. It is used in hybridization to develop resistant currant varieties but is on the verge of extinction, listed in Kazakhstan Red Book. This study developed a micropropagation and slow-growth storage protocol for conservation. Genotypes were identified through DNA barcode analysis (rbcL, ITS, and matK) and sequences uploaded to the National Center for Biotechnology Information database. Genetic diversity was assessed using iPBS primers, generating 98 fragments with 88-94% polymorphic ban
