B-esterases characterisation in the digestive tract of the common octopus and the European cuttlefish and their in vitro responses to contaminants of environmental concern.

Cephalopods are a group of marine invertebrates that have received little attention as sentinel species in comparison to other molluscs, such as bivalves. Consequently, their physiological and biochemical xenobiotic metabolism responses are poorly understood. Here we undertake a comparative analysis of the enzymatic activities involved in detoxification reactions and neural transmission in the digestive tract of two commercial cephalopods: the Common octopus, Octopus vulgaris, and the European cuttlefish, Sepia officinalis. For methodological purposes, several common B-esterases (five carboxyl

Plerocercoids of Nybelinia surmenicola (Cestoda: Tentacularidae) in Squids, Todarodes pacificus, from East Sea, the Republic of Korea.

A visceral helminth of the squid, Todarodes pacificus, is reported from the East Sea, the Republic of Korea. Total 39 squid samples were purchased from a fish market in Jumunjin-eup, Gangneung-si (City) from August 2014 to July 2015 and were examined for helminth parasites with naked eyes and under a stereomicroscope after opening the abdominal cavity with a pair of scissors. Whitish larval worms were mainly found in the stomach and abdominal cavity of the squid. They were detected in 25 (64.1%) out of 39 squids examined, and the infection density was 7 larvae per infected squid. Spatula-shape

Thermal responses and climate change implications of spring and autumn spawning Patagonian squid (Doryteuthis gahi) embryos.

Ocean warming affects ectotherm physiological and phenological processes, potentially creating mismatches between early life stages and their prey. Seasonal spawning cohorts are thought to provide flexibility in responding to environmental variability, but if there is seasonal adaptation between these cohorts, then they may respond to ocean warming differently, affecting species resilience and potentially impacting the wider food web. We tested the response to warming of egg masses and paralarvae from two spawning cohorts (autumn and spring) of the Patagonian squid (Doryteuthis gahi). Treated

Central role of squid gene during oocyte development in the Hemiptera Rhodnius prolixus.

Oocyte polarity establishment is a conserved and crucial phenomenon for embryonic development. It relies on the precise spatial localization of maternal factors deposited during oocyte development, which is essential for establishing and maintaining cell polarity and subsequently specifying embryonic axes. The heterogeneous nuclear ribonucleoprotein (hnRNP) encoded by the squid (sqd) gene has been implicated in mRNA localization and embryonic axis establishment in Drosophila melanogaster. Comparative genomics allowed for the identification of a homologue in Rhodnius prolixus. In this study, we

A Putative Lipoprotein Mediates Cell-Cell Contact for Type VI Secretion System-Dependent Killing of Specific Competitors.

Interbacterial competition is prevalent in host-associated microbiota, where it can shape community structure and function, impacting host health in both positive and negative ways. However, the factors that permit bacteria to discriminate among their various neighbors for targeted elimination of competitors remain elusive. We identified a putative lipoprotein (TasL) in Vibrio species that mediates cell-cell attachment with a subset of target strains, allowing inhibitors to target specific competitors for elimination. Here, we describe this putative lipoprotein, which is associated with the br

Bacterial bioluminescence regulates expression of a host cryptochrome gene in the squid-Vibrio symbiosis.

The symbiosis between the squid Euprymna scolopes and its luminous symbiont, Vibrio fischeri, is characterized by daily transcriptional rhythms in both partners and daily fluctuations in symbiont luminescence. In this study, we sought to determine whether symbionts affect host transcriptional rhythms. We identified two transcripts in host tissues (E. scolopes cry1 [escry1] and escry2) that encode cryptochromes, proteins that influence circadian rhythms in other systems. Both genes cycled daily in the head of the squid, with a pattern similar to that of other animals, in which expression of cer

Gradients of strain and strain rate in the hollow muscular organs of soft-bodied animals.

The cylindrical shape of soft-bodied invertebrates is well suited to functions in skeletal support and locomotion, but may result in a previously unrecognized cost-large non-uniformities in muscle strain and strain rate among the circular muscle fibres of the body wall. We investigated such gradients of strain and strain rate in the mantle of eight long-finned squid Doryteuthis pealeii and two oval squid Sepioteuthis lessoniana. Transmural gradients of circumferential strain were present during all jets (n = 312); i.e. for a given change in the circumference of the outer surface of the mantle,

Developmental and transcriptomic responses of Hawaiian bobtail squid early stages to ocean warming and acidification.

Cephalopods play a central ecological role across all oceans and depths. However, under the current climate crisis, their physiology and behaviour are impacted, and we are beginning to comprehend the effects of environmental stressors at a molecular level. Here, we study the Hawaiian bobtail squid (Euprymna scolopes), known for its specific binary symbiosis with the bioluminescent bacterium Vibrio fischeri acquired post-hatching. We aim to understand the response (i.e. developmental and molecular) of E. scolopes after embryogenetic exposure to different conditions: (i) standard conditions (con

Development of Novel Squid Gladius Biomaterials for Cornea Tissue Engineering.

Cornea tissue engineering is strictly dependent on the development of biomaterials that fulfill the strict biocompatibility, biomechanical, and optical requirements of this organ. In this work, we generated novel biomaterials from the squid gladius (SG), and their application in cornea tissue engineering was evaluated. Results revealed that the native SG (N-SG) was biocompatible in laboratory animals, although a local inflammatory reaction was driven by the material. Cellularized biomaterials (C-SG) demonstrated that the SG provides an adequate substrate for cell attachment and growth, and cor

Arginylation in a Partially Purified Fraction of 150 k xg Supernatants of Axoplasm and Injured Vertebrate Nerves.

Transfer RNA-mediated posttranslational protein modification by arginine has been demonstrated in vitro in axoplasm extruded from the giant axons of squid and in injured and regenerating vertebrate nerves. In nerve and axoplasm, the highest activity is found in a fraction of a 150,000 g supernatant containing high molecular weight protein/RNA complexes but lacking molecules of <5 kDa. Arginylation (and protein modification by other amino acids) is not found in more purified, reconstituted fractions. The data are interpreted as indicating that it is critical to recover the reaction components i

Synthesis and characterization of novel Spirulina protein isolate (SPI)-based hydrogels through dual-crosslinking with genipin/Zn2.

Spirulina protein isolate (SPI) is a bio-polymer that can be used in the design and synthesis of hydrogels due to its biodegradability and non-toxicity. Herein, dual-crosslinked hydrogels were synthesized based on spirulina protein isolate through covalent crosslinking with genipin and metal coordination with Zn2+, and used as delivery systems. From the analysis of colorimetric indices, a high correlation between color changes and the reaction of cross-linkers with amine group structure was observed. By increasing genipin concentration in the range of 5-15 mM, the degree of cross-linking incre

Behavioural responses to infrasonic particle acceleration in cuttlefish.

Attacks by aquatic predators generate frontal water disturbances characterised by low-frequency gradients in pressure and particle motion. Low-frequency hearing is highly developed in cephalopods. Thus, we examined behavioural responses in juvenile cuttlefish to infrasonic accelerations mimicking main aspects of the hydrodynamic signals created by predators. In the experimental set-up, animals and their surrounding water moved as a unit to minimise lateral line activation and to allow examination of the contribution by the inner ear. Behavioural responses were tested in light versus darkness a

Determination of arsenic speciation of arsenic in squid and its human consumption risk.

Arsenic species were quantified to further explore the biotransformation of arsenic and better explain the potential health risks. In this research, the levels of six arsenic species (AsB, AsC, MMA, DMA, As3+, and As5+) were determined in 160 squid samples by HPLC-ICP-MS. AsB was the most predominant speciation in squid tissues. The arsenic species, including AsB, AsC, and MMA, were found in all samples, followed by DMA (49.38%), As3+ (21.25%), and As5+ (2.5%). Results indicated squid samples from offshore had higher levels of AsB, AsC, MMA, and DMA. Pearson's correlation analysis found correl

Effect of ultrasound combined with papain treatment on the tenderness of equatorial squid (Dosidicus gigas): in terms of myofibrillar protein modification.

Ultrasonic combined papain treatment was effective in tenderizing equatorial squid. When the ultrasonic power was 200 W combined with papain treatment, the shear force decreased by 23.35 %, and the tenderness of squid was the highest. Structural analysis of myofibrillar proteins showed that the myofibrillar proteins treated with ultrasound combined with papain treatment was decomposed to the highest extent at an ultrasound power of 200 W, when the particle size was the smallest and the α-helix content was reduced. Ultrasonic power of 300 W combined with papain treatment can also improve the te

Two unusual cases of a foreign body in the oral cavity caused by eating raw squid.

Foreign bodies are often encountered by otolaryngologists, but the oral cavity is considered a place where foreign bodies are rare because of its structural and functional features. We here present 2 cases with a foreign body in the oral cavity resulting from eating raw squid. In one of these 2 cases endoscopic examination revealed a gastric foreign body. The foreign bodies in the oral cavity were removed using forceps, together with surrounding mucous membrane. These foreign bodies were identified as sperm bulbs and discharging tube of sogittated calamary (Todarodes pacificus Steenstrup). Lar

Regulation of intracellular calcium in squid axons.

Internal dialysis and metallochromic indicators were used to determine the free calcium concentration and calcium buffering properties of squid axoplasm. The free calcium concentration in fresh unloaded squid axons is about 30 to 50 nM. About 6% of the calcium content (ca. 50 mumol/kg axoplasm) of a fresh squid axon is held in a metabolically labile, presumably mitochondrial, component. A morphological consequence of this finding is that there should be no accumulation of calcium in mitochondria of fresh squid axons unless there is a large component of nonlabile calcium. The physiological impl
