Assessing the Impact of Partial Decellularization on Tracheal Chondrocytes and Extracellular Matrix in Airway Reconstruction.

OBJECTIVE: Partially decellularized tracheal grafts (PDTG) are potential candidates for tracheal replacement as they support neotissue formation without stenosis or rejection. However, the effects of partial decellularization (PD) on extracellular matrix (ECM) and chondrocytes are not currently understood, limiting PDTG translatability for clinical use. We aim to quantify the impact of PD on trachea using mouse and rabbit models. STUDY DESIGN: An animal model. SETTING: Research Institute affiliated with a Tertiary Pediatric Hospital. METHODS: PDTG and syngeneic tracheal grafts (STG) were im

Immunogenicity of decellularized extracellular matrix scaffolds: a bottleneck in tissue engineering and regenerative medicine.

Tissue-engineered decellularized extracellular matrix (ECM) scaffolds hold great potential to address the donor shortage as well as immunologic rejection attributed to cells in conventional tissue/organ transplantation. Decellularization, as the key process in manufacturing ECM scaffolds, removes immunogen cell materials and significantly alleviates the immunogenicity and biocompatibility of derived scaffolds. However, the application of these bioscaffolds still confronts major immunologic challenges. This review discusses the interplay between damage-associated molecular patterns (DAMPs) and

Decellularized extracellular matrix microparticles as a vehicle for cellular delivery in a model of anastomosis healing.

Extracellular matrix (ECM) materials from animal and human sources have become important materials for soft tissue repair. Microparticles of ECM materials have increased surface area and exposed binding sites compared to sheet materials. Decellularized porcine peritoneum was mechanically dissociated into 200 µm microparticles, seeded with fibroblasts and cultured in a low gravity rotating bioreactor. The cells avidly attached and maintained excellent viability on the microparticles. When the seeded microparticles were placed in a collagen gel, the cells quickly migrated off the microparticles

Decellularization and cell seeding of whole liver biologic scaffolds composed of extracellular matrix.

The definitive treatment for patients with end-stage liver disease is orthotropic transplantation. However, this option is limited by the disparity between the number of patients needing transplantation and the number of available livers. This issue is becoming more severe as the population ages and as the number of new cases of end-stage liver failure increases. Patients fortunate enough to receive a transplant are required to receive immunosuppressive therapy and must live with the associated morbidity. Whole organ engineering of the liver may offer a solution to this liver donor shortfall.

From biological scaffold to multifunctional bioplatform: research progress and applications of decellularized extracellular matrix.

Decellularized extracellular matrix (dECM) is a biomaterial generated by the selective removal of cellular components from native tissues while preserving the three-dimensional ultrastructure and endogenous biochemical cues of the extracellular matrix. Owing to its favorable biocompatibility, low immunogenicity, and capacity to mimic tissue-specific microenvironments, dECM has been increasingly explored across a wide range of biomedical applications. Its role has expanded from a conventional scaffold for tissue repair to a multifunctional biomaterial platform integrating 3D bioprinting bioinks

Effect of Storage Conditions on Rheological Properties of Decellularized Extracellular Matrix-Based Injectable Hydrogels.

Decellularized extracellular matrix-based scaffolds have gained significant attention for tissue regeneration applications. However, there is a limited understanding of how storage conditions influence the rheological and biological properties of decellularized matrix-based injectable formulations. This study investigates the effects of different storage conditions (4°C, -80°C, and freeze-dried) on the rheological properties of a decellularized porcine peripheral nerve-derived extracellular matrix hydrogel. Our results demonstrated that, compared to storage at 4°C, formulations stored at -80°C

Flowable and microporous granular grafts made from decellularized extracellular matrix microgels for tissue repair.

Decellularized extracellular matrix (dECM) scaffolds are widely used in tissue engineering, primarily due to their inherent bioactivity and cell-instructive properties. ECM scaffolds are conventionally prepared as monolithic and rigid sheet-like constructs, and while these form factors are generally sutured at the defect site it can be challenging to achieve even filling of the defect volume, where flowable scaffolds may be beneficial. Although infusible and injectable dECM formulations have been developed, optimizing for high mechanical strength and a porous microstructure that permits integr

Composition and Function of Decellularized Human Lung Extracellular Matrix from Congenital Pulmonary Airway Malformation.

Background: Congenital pulmonary airway malformation (CPAM) is a rare developmental disorder characterized by cystic lung lesions, yet its extracellular matrix (ECM) composition remains poorly understood. This study employed decellularization and data-independent acquisition (DIA) proteomics to compare ECM profiles between cystic (CPAM) and histologically normal non-diseased (ND) regions from the lungs of four patients. Results: The decellularized scaffolds retained their native architecture with minimal residual DNA (<50 ng/mg). Proteomic analysis revealed 431 differentially expressed protein

Bilayer book- like decellularized extracellular matrix scaffold with bioactive coatings for rotator cuff repair.

Repairing the tendon-bone interface after rotator cuff injury and regulating the local immune microenvironment present significant clinical challenges, necessitating the development of bioactive materials capable of simultaneous interface repair and inflammation modulation. In this study, we developed a book-like scaffold (dtECM-TAZn/Mg) with bilayers of bioactive coatings to target the bone microenvironment and promote rotator cuff repair. Fabricated from decellularized tendon extracellular matrix (dtECM) to preserve native collagen fiber alignment, the scaffold underwent covalent cross-linki

3D bioprinting of corneal decellularized extracellular matrix: GelMA composite hydrogel for corneal stroma engineering.

Millions of individuals across the world suffer from corneal stromal diseases that impair vision. Fortunately, three-dimensional (3D) bioprinting technology which has revolutionized the field of regenerative tissue engineering makes it feasible to create personalized corneas. In this study, an artificial cornea with a high degree of precision, smoothness, and programmable curvature was prepared by using digital light processing (DLP) 3D bioprinting in one piece with no support structure, and the construct was then confirmed by optical coherence tomography (OCT). On the basis of this approach,

Injectable Decellularized Extracellular Matrix-Based Bio-Ink with Excellent Biocompatibility for Scarless Urethra Repair.

In recent years, decellularized extracellular matrices (dECM) derived from organs have attracted much attention from researchers due to their excellent biocompatibility, capacity to promote cell proliferation and migration, as well as pro-vascularization. However, their inferior mechanical properties, slow cross-linking, weak strengths, and poor supporting properties remain their inevitable challenges. In our study, we fabricated a novel dECM hydrogel with better crosslinking strength and speed, stronger support properties, and better mechanical properties. The hydrogel, which we named gelatin

Regional and disease-specific glycosaminoglycan composition and function in decellularized human lung extracellular matrix.

Decellularized lung scaffolds and hydrogels are increasingly being utilized in ex vivo lung bioengineering. However, the lung is a regionally heterogenous organ with proximal and distal airway and vascular compartments of different structures and functions that may be altered as part of disease pathogenesis. We previously described decellularized normal whole human lung extracellular matrix (ECM) glycosaminoglycan (GAG) composition and functional ability to bind matrix-associated growth factors. We now determine differential GAG composition and function in airway, vascular, and alveolar-enrich

Enhancing organoid culture: harnessing the potential of decellularized extracellular matrix hydrogels for mimicking microenvironments.

Over the past decade, organoids have emerged as a prevalent and promising research tool, mirroring the physiological architecture of the human body. However, as the field advances, the traditional use of animal or tumor-derived extracellular matrix (ECM) as scaffolds has become increasingly inadequate. This shift has led to a focus on developing synthetic scaffolds, particularly hydrogels, that more accurately mimic three-dimensional (3D) tissue structures and dynamics in vitro. The ECM-cell interaction is crucial for organoid growth, necessitating hydrogels that meet organoid-specific require

Decellularization and antibody staining of mouse tissues to map native extracellular matrix structures in 3D.

The extracellular matrix (ECM) is a major regulator of homeostasis and disease, yet the 3D structure of the ECM remains poorly understood because of limitations in ECM visualization. We recently developed an ECM-specialized method termed in situ decellularization of tissues (ISDoT) to isolate native 3D ECM scaffolds from whole organs in which ECM structure and composition are preserved. Here, we present detailed surgical instructions to facilitate decellularization of 33 different mouse tissues and details of validated antibodies that enable the visualization of 35 mouse ECM proteins. Extracellular matrix from decellularized porcine organs as scaffolds for insulin-secreting cells and pancreatic islets.

Extracellular Matrix (ECM) from different organs has been used to cultivate several cell types. ECM produced by organ decellularization contains collagens, fibronectin, glycosaminoglycans (GAGs), laminins and other components essential in providing structural support and biochemical cues for cells to attach, function, and proliferate. The organ from which ECM is extracted and produced is hypothesized to play a vital role in cell responses upon recellularization. To investigate this hypothesis, five porcine organs (bladders, kidneys, livers, lungs, and pancreas) were decellularized by a deterge
