Discrimination of multiple Dehalococcoides strains in a trichloroethene enrichment by quantification of their reductive dehalogenase genes.

While many anaerobic microbial communities are capable of reductively dechlorinating tetrachloroethene (PCE) and trichloroethene (TCE) to dichloroethene (DCE), vinyl chloride (VC), and finally ethene, the accumulation of the highly toxic intermediates, cis-DCE (cDCE) and VC, presents a challenge for bioremediation processes. Members of the genus Dehalococcoides are apparently solely responsible for dechlorination beyond DCE, but isolates of Dehalococcoides each metabolize only a subset of PCE dechlorination intermediates and the interactions among distinct Dehalococcoides strains that result i

Coupling Bioflocculation of Dehalococcoides mccartyi to High-Rate Reductive Dehalogenation of Chlorinated Ethenes.

Continuous bioreactors operated at low hydraulic retention times have rarely been explored for reductive dehalogenation of chlorinated ethenes. The inability to consistently develop such bioreactors affects the way growth approaches for Dehalococcoides mccartyi bioaugmentation cultures are envisioned. It also affects interpretation of results from in situ continuous treatment processes. We report bioreactor performance and dehalogenation kinetics of a D. mccartyi-containing consortium in an upflow bioreactor. When fed synthetic groundwater at 11-3.6 h HRT, the upflow bioreactor removed >99.7%

Bioaugmentation for chlorinated ethenes using Dehalococcoides sp.: comparison between batch and column experiments.

Batch and column experiments were performed to evaluate the transport, growth and dechlorination activity of Dehalococcoides sp. (DHC) during bioaugmentation for chlorinated ethenes. Batch experiments showed that the reductive dechlorination of trichloroethene (TCE), cis-1,2-dichloroethene (DCE), and vinyl chloride (VC), as well as growth of the DHC, were well described by the Monod kinetic model. The measured maximum utilization rate coefficients for TCE, DCE, and VC were 1.3x10(-12), 5.2x10(-13), and 1.4x10(-12)mmol Cl(-) (cellh)(-1), respectively. Results of the column experiments showed th

Temperature dependence of anaerobic TCE-dechlorination in a highly enriched Dehalococcoides-containing culture.

Temperature dependence of reductive trichloroethene (TCE) dechlorination was investigated in an enrichment culture (KB-1), using lactate or propionate as electron donors at a temperature interval from 4 to 60 degrees C. Dechlorination was complete to ethene at temperatures between 10 and 30 degrees C (lactate-amended) and between 15 and 30 degrees C (propionate-amended). Dechlorination stalled at cis-1,2-dichloroethene (cDCE) at 4 degrees C (lactate-amended), at and below 10 degrees C (propionate-amended), and at 40 degrees C with both electron donors. No dechlorination of TCE was observed at

Transcription and mass-spectroscopic proteomic studies of electron transport oxidoreductases in Dehalococcoides ethenogenes.

Besides 19 potential reductive dehalogenase genes, the genome of Dehalococcoides ethenogenes strain 195 contains over 60 genes annotated as encoding oxidoreductases, including five hydrogenase complexes and a formate dehydrogenase (Fdh). Using quantitative reverse transcriptase polymerase chain reaction, we found that genes encoding a periplasmic Hup hydrogenase and the Fdh were the most highly expressed in batch-grown pure cultures, in which the H2 partial pressure was >0.1 atm, and in butyrate/tetrachloroethene-mixed cultures, in which H2 partial pressures were 10(-4)-10(-5) atm. Unveiling the Differential Impacts of S-nZVI and nZVI on Microbial Dechlorination of Trichloroethene by Dehalococcoides.

Sulfidated nanoscale zerovalent iron (S-nZVI) is a promising material for the remediation of chlorinated hydrocarbons (CHCs) in groundwater, offering enhanced reactivity and selectivity over unmodified nanoscale zerovalent iron (nZVI). However, integrating S-nZVI with bioremediation using organohalide-respiring bacteria (OHRB) poses potential cytotoxicity challenges. This study investigated the differential impacts of S-nZVI and nZVI on the dechlorination activity of Dehalococcoides mccartyi strain 195 (Dhc 195), a representative OHRB. Key factors, including hydrogen evolution reaction (HER),

Enhanced resilience to oxygen exposure and toxicity of chlorinated solvents in immobilized Dehalococcoides mccartyi.

Members of Dehalococcoides mccartyi (Dhc) are strictly anaerobic and play crucial roles in the restoration of many industrial sites impacted by chlorinated solvents, such as tetrachloroethene (PCE) and trichloroethene (TCE). In situ bioremediation with Dhc involves intricate procedures intended to minimize oxygen intrusion, and achieving optimal dechlorination performance in aquifers near the dense non-aqueous phase liquids source zone is challenging. Here, we respectively embedded Dhc strain 195 and the biomass of a Dhc-containing, PCE-dechlorinating consortium in poly(vinyl alcohol)-alginate

Biological effects of tourmaline treatment on Dehalococcoides spp. during the reductive dechlorination of trichloroethylene.

In the present study, to develop the application of biostimulation for the in situ remediation of trichloroethylene (TCE) in contaminated groundwater/soil, a mixed culture containing Dehalococcoides spp. was employed to investigate the biological effects of the polarized mineral tourmaline on the dechlorination performance, community structure, cell proliferation and expression of two model gene (tceA and vcrA) coding for reductive dehalogenases (Rdase). It was observed that tourmaline could speed up the biological dechlorination of TCE by promoting the growth and metabolism of the bacteria, i

Mechanistic insights into chloroethene dechlorination by Dehalococcoides mccartyi strain CWV2: A multi-omics perspective.

This study provides an in-depth investigation of the novel Dehalococcoides mccartyi (Dhc) strain CWV2, isolated from Taiwan, for its effectiveness in dechlorinating various chloroethenes, including PCE, TCE, DCEs, and VC, to ethene. Through multi-omics analyses, including genomic, transcriptomic, translatomic and proteomic profiling, we uncovered the mechanisms behind TCE dechlorination by strain CWV2. The genomic analysis identified key reductive dehalogenase (RDase) genes, pceA and vcrA, which enhance our understanding of the versatile dechlorination pathways in Dhc. Ribosome profiling provi

Bioenhanced back diffusion and population dynamics of Dehalococcoides mccartyi strains in heterogeneous porous media.

Diffusion, sorption-desorption, and biodegradation influence chlorinated solvent storage in, and release (mass flux) from, low-permeability media. Although bioenhanced dissolution of non-aqueous phase liquids has been well-documented, less attention has been directed towards biologically-mediated enhanced diffusion from low-permeability media. This process was investigated using a heterogeneous aquifer cell, packed with 20-30 mesh Ottawa sand and lenses of varying permeability (1.0 × 10-12-1.2 × 10-11 m2) and organic carbon (OC) content (<0.1%-2%), underlain by trichloroethene (TCE)-saturated

Genomic Characterization of Dehalococcoides mccartyi Strain JNA That Reductively Dechlorinates Tetrachloroethene and Polychlorinated Biphenyls.

Dehalococcoides mccartyi strain JNA detoxifies highly chlorinated polychlorinated biphenyl (PCB) mixtures via 85 distinct dechlorination reactions, suggesting that it has great potential for PCB bioremediation. However, its genomic and functional gene information remain unknown due to extremely slow growth of strain JNA with PCBs. In this study, we used tetracholorethene (PCE) as an alternative electron acceptor to grow sufficient biomass of strain JNA for subsequent genome sequencing and functional gene identification. Analysis of the assembled draft genome (1 462 509 bp) revealed the presenc

Chloroethene degradation and expression of Dehalococcoides dehalogenase genes in cultures originating from Yangtze sediments.

The anaerobic Dehalococcoides spp. is the only microorganism known to completely dechlorinate the hazardous compounds tetrachloroethene (PCE) or trichloroethene (TCE) via dichloroethene (DCE) and vinyl chloride (VC) to the terminal product, ethene. In this study, growth of Dehalococcoides spp. (DHC) and the expression of DHC dehalogenase genes were demonstrated for Yangtze enrichment cultures. Reductive dechlorination of chloroethenes occurred in Yangtze sediment without the addition of any external auxiliary substrates. All Yangtze enrichment cultures completely dechlorinated PCE and cis-DCE

Dehalococcoides abundance and alternate electron acceptor effects on large, flow-through trichloroethene dechlorinating columns.

Trichloroethene (TCE) in groundwater is a major health concern and biostimulation/bioaugmentation-based strategies have been evaluated to achieve complete reductive dechlorination with varying success. Different carbon sources were hypothesized to stimulate different extents of TCE reductive dechlorination. Ecological conditions that developed different dechlorination stages were investigated by quantitating Dehalococcoides 16S rRNA (Dhc) and reductive dehalogenase gene abundance, and by describing biogeochemical properties of laboratory columns in response to this biostimulation. DNA extraction-free quantification of Dehalococcoides spp. in groundwater using a hand-held device.

Nucleic acid amplification of biomarkers is increasingly used to measure microbial activity and predict remedial performance in sites with trichloroethene (TCE) contamination. Field-based genetic quantification of microorganisms associated with bioremediation may help increase accuracy that is diminished through transport and processing of groundwater samples. Sterivex cartridges and a previously undescribed mechanism for eluting biomass was used to concentrate cells. DNA extraction-free loop mediated isothermal amplification (LAMP) was monitored in real-time with a point of use device (termed

Versatility in corrinoid salvaging and remodeling pathways supports corrinoid-dependent metabolism in Dehalococcoides mccartyi.

Corrinoids are cobalt-containing molecules that function as enzyme cofactors in a wide variety of organisms but are produced solely by a subset of prokaryotes. Specific corrinoids are identified by the structure of their axial ligands. The lower axial ligand of a corrinoid can be a benzimidazole, purine, or phenolic compound. Though it is known that many organisms obtain corrinoids from the environment, the variety of corrinoids that can serve as cofactors for any one organism is largely unstudied. Here, we examine the range of corrinoids that function as cofactors for corrinoid-dependent meta

Comparative genomics of two newly isolated Dehalococcoides strains and an enrichment using a genus microarray.

Comparative genomics of Dehalococcoides strains and an enrichment were performed using a microarray targeting genes from all available sequenced genomes of the Dehalococcoides genus. The microarray was designed with 4305 probe sets to target 98.6% of the open-reading frames from strains 195, CBDB1, BAV1 and VS. The microarrays were validated and applied to query the genomes of two recently isolated Dehalococcoides strains, ANAS1 and ANAS2, and their enrichment source (ANAS) to understand the genome-physiology relationships. Strains ANAS1 and ANAS2 can both couple the reduction of trichloroethe

Dechlorination of PCE in the presence of Fe0 enhanced by a mixed culture containing two Dehalococcoides strains.

A mixed culture capable of supplying its energy requirements by the oxidation of zero-valent iron (Fe0) and concomitant reduction of chlorinated ethenes was established. The culture contained Dehalococcoides species as determined by polymerase chain reaction (PCR) with genus specific primers. The use of a newly designed ARDRA procedure and subsequent sequencing revealed the presence of two Dehalococcoides strains, one closely related to Dehalococcoides ethenogenes strain 195, a bacterium respiring with chlorinated ethenes, and one closely related to strain CBDB1 a chlorobenzene and dioxin deha

Addition of Shewanella oneidensis MR-1 to the Dehalococcoides-containing culture enhances the trichloroethene dechlorination.

Dehalococcoides is able to completely dehalogenate tetrachloroethene (PCE) and trichloroethene (TCE) to ethene (ETH). However, the dechlorination efficiency of Dehalococcoides is low and result in the accumulation of toxic intermediates. In this study, Shewanella oneidensis MR-1 (S. oneidensis MR-1) was added to the Dehalococcoides-containing culture and the complete TCE to ETH dechlorination was shortened from 24 days to 16 days. A modeling study and implications of competition between Dehalococcoides ethenogenes and other tetrachloroethene-respiring bacteria.

Bioaugmentation with cultures containing Dehalococcoides strains that dechlorinate cis-1,2-dichloroethene (cDCE) to ethene is often recommended at sites where indigenous populations dechlorinate tetrachloroethene (PCE) to cDCE. In these cases, Dehalococcoides populations may have to compete with other dehalorespirers for chlorinated ethenes and/or electron donors. A continuous-flow stirred tank reactor model was used to describe the substrate interactions in three conceptual models of competition between PCE-respiring populations under natural attenuation and engineered bioremediation scenario
