BACKGROUND: Porcine deltacoronavirus (PDCoV) is a novel coronavirus that can cause diarrhea in nursing piglets. This study was aimed to investigate the roles of host differentially expressed genes on metabolic pathways in PDCoV infections.
RESULTS: Twenty thousand six hundred seventy-four differentially expressed mRNAs were identified in 5-day-old piglets responded to PDCoV experimental infections. Many of these genes were correlated to the basic metabolism, such as the peroxisome proliferator-activated receptor (PPAR) signaling pathway which plays a critical role in digestion. At the same ti

Porcine deltacoronavirus (PDCoV) is a significant enteric pathogen in swine, necessitating rapid and accessible diagnostic tools for effective control. In this study, we developed a streamlined, extraction-free Reverse Transcription Loop-Mediated Isothermal Amplification (RT-LAMP) assay integrated with a TaqMan probe for on-site PDCoV detection. By targeting the conserved N gene and modifying the loop primer (LB) as a fluorescence probe, a highly specific reaction system was established. The assay demonstrated high sensitivity, with a limit of detection (LOD) of 9.3 copies/μL for cDNA and 5.9

BACKGROUND: Porcine deltacoronavirus (PDCoV) is a newly emerging coronavirus that causes severe diarrhea and dehydration in newborn piglets, and can infect multiple animal species including humans, posing a substantial threat to global swine production and human health. To address these challenges, we developed a droplet digital PCR (ddPCR) method for detecting the N gene of PDCoV in clinical samples. RESULTS: Our ddPCR method exhibited good linearity, reproducibility, and repeatability. In comparison to conventional quantitative real-time PCR (qPCR), the ddPCR assay demonstrated exceptional s

Porcine deltacoronavirus (PDCoV) has emerged as a significant issue in multiple pork-producing countries. This study isolated a novel PDCoV strain, GNU-2105/KOR/2021, which caused a severe diarrhea outbreak with a high mortality rate among neonatal piglets in South Korea. The growth properties and sialic acid dependency of the GNU-2105 strain in cell culture were comparable to those of the 2016 domestic isolate, KNU-1607. Interestingly, phylogenetic analysis using the complete genome of GNU-2105 identified in 2021 demonstrated that this novel strain belongs to the US/South Korean/Japanese clad

The existing zoonotic coronaviruses (CoVs) and viral genetic variants are important microbiological pathogens that cause severe disease in humans and animals. Currently, no effective broad-spectrum antiviral drugs against existing and emerging CoVs are available. The CoV main protease (Mpro) plays an essential role in viral replication, making it an ideal target for drug development. However, the structure of the Deltacoronavirus Mpro is still unavailable. Porcine deltacoronavirus (PDCoV) is a novel CoV that belongs to the genus Deltacoronavirus and causes atrophic enteritis, severe diarrhea,

Porcine deltacoronavirus (PDCoV) is one of the most important enteropathogenic pathogens, and it causes enormous economic losses to the global commercial pork industry. PDCoV was initially reported in Hong Kong (China) in 2012 and subsequently emerged in swine herds with diarrhea in Ohio (USA) in 2014. Since then, it has spread to Canada, South Korea, mainland China, and several Southeast Asian countries. Information about the epidemiology, evolution, prevention, and control of PDCoV and its prevalence in China has not been comprehensively reported, especially in the last five years. This revi

Porcine deltacoronavirus (PDCoV) is a novel enteropathogenic coronavirus that causes watery diarrhea in piglets. Little is known regarding the alteration of the gut microbiota in PDCoV-induced diarrhea piglets. In this study, 5-day-old piglets were experimentally infected with PDCoV strain CH-01, and all piglets developed typical clinical disease, characterized by acute and severe watery diarrhea. Histologic lesions were limited to the villous epithelium of the duodenum and ileum. Gut microbiota profiles in the colon and feces of piglets inoculated with PDCoV were investigated using 16S rRNA s

Porcine deltacoronavirus (PDCoV) infection induces severe intestinal inflammation and acute diarrhea in piglets, resulting substantial economic losses. The NLR family pyrin domain containing 3 (NLRP3) inflammasome, a key component of the innate immune system, drives interleukin (IL)-1β maturation, and its excessive activation is closely linked to viral pathogenesis. However, its role in PDCoV infection remains unclear. Here, we report that PDCoV infection induces IL-1β maturation through NLRP3 inflammasome activation both in vivo and in vitro, and the M protein promotes NLRP3 inflammasome asse

Porcine deltacoronavirus (PDCoV) is an emerging swine enteropathogenic coronavirus that has the potential to infect humans. Histone deacetylase 6 (HDAC6) is a unique type IIb cytoplasmic deacetylase with both deacetylase activity and ubiquitin E3 ligase activity, which mediates a variety of cellular processes by deacetylating histone and nonhistone substrates. In this study, we found that ectopic expression of HDAC6 significantly inhibited PDCoV replication, while the reverse effects could be observed after treatment with an HDAC6-specific inhibitor (tubacin) or knockdown of HDAC6 expression b

Porcine deltacoronavirus (PDCoV) is an economically important enteropathogen of swine with worldwide distribution. PDCoV primarily infects the small intestine instead of the large intestine in vivo However, the underlying mechanism of PDCoV tropism to different intestinal segments remains poorly understood as a result of the lack of a suitable in vitro intestinal model that recapitulates the cellular diversity and complex functions of the gastrointestinal tract. Here, we established the PDCoV infection model of crypt-derived enteroids from different intestinal segments. Enteroids were suscepti

Porcine deltacoronavirus (PDCoV) is a porcine intestinal coronavirus that causes severe diarrhea in piglets and is extremely harmful to the pig industry. Rapid and accurate detection of antibody levels is necessary for effective prevention of the disease. We previously demonstrated that PDCoV and porcine epidemic diarrhea (PEDV) N protein have serum cross-reactivity. Therefore, in this study, we first constructed and expressed five recombinant truncated PDCoVN proteins ( N1-N5). Through the reactivity of these proteins to porcine PDCoV positive serum and porcine PEDV positive serum and the est

In 2024, an outbreak of piglet diarrhea occurred in Guangxi Zhuang Autonomous Region, China. RT-PCR assay of clinical samples confirmed that porcine deltacoronavirus (PDCoV) was the primary pathogen. In this study, the PDCoV GX/2024 strain was successfully isolated in PK-15 cells with optimized trypsin treatment conditions. The cytopathic effect (CPE) of this strain gradually intensified during serial passages. After plaque purification, the strain achieved stable proliferation with high titer, reaching a peak titer of 107·2 TCID50/mL, which indicated its potential as an inactivated vaccine ca

Coronaviruses (CoVs) comprise a group of important human and animal pathogens that threaten public health because of their interspecies transmission potential to humans. However, virus-like particles (VLPs) constitute versatile tools in CoVs vaccine development due to their favorable immunological characteristics. Here, we engineered the VLPs composed of the spike (S), membrane (M), and envelope (E) structural proteins of the Porcine deltacoronavirus (PDCoV) and examined their immune responses in mice. Neutralization assays and flow Cytometry demonstrated that PDCoV VLPs induced highly robust

Porcine deltacoronavirus (PDCoV) is an emerging enteric pathogen that causes substantial economic losses in the swine industry worldwide. The PDCoV NS6 protein is an accessory protein that plays a pivotal role in the viral life cycle and immune evasion. However, the functions of NS6 and its role in PDCoV pathogenesis remain largely unknown. In this study, we prepared a monoclonal antibody (mAb) 5-A11 that specifically recognizes the PDCoV NS6 protein. The mAb 5-A11 exhibited high specificity for PDCoV, with no cross-reactivity with several major porcine pathogenic viruses. Furthermore, the epi

Porcine deltacoronavirus (PDCoV), an emerging enteropathogenic coronavirus, not only causes diarrhea in piglets but also possesses the potential to infect humans. To better understand host-virus genetic dependencies and find potential therapeutic targets for PDCoV, we used a porcine single-guide RNA (sgRNA) lentivirus library to screen host factors related to PDCoV infection in LLC-PK1 cells. The solute carrier family 35 member A1 (SLC35A1), a key molecule in the sialic acid (SA) synthesis pathway, was identified as a host factor required for PDCoV infection. A knockout of SLC35A1 caused decre

Determination of the mechanisms of interspecies transmission is of great significance for the prevention of epidemic diseases caused by emerging coronaviruses (CoVs). Recently, porcine deltacoronavirus (PDCoV) was shown to exhibit broad host cell range mediated by surface expression of aminopeptidase N (APN), and humans have been reported to be at risk of PDCoV infection. In the present study, we first demonstrated overexpression of APN orthologues from various species, including mice and felines, in the APN-deficient swine small intestine epithelial cells permitted PDCoV infection, confirming

Porcine deltacoronavirus (PDCoV) is a highly transmissible intestinal pathogen that causes mild to severe clinical symptoms, such as anorexia, vomiting, and watery diarrhea in pigs. By comparing the genetic sequences of the spike glycoprotein between historical and current Taiwanese PDCoV strains, we identified a novel PDCoV variant that displaced the PDCoV responsible for the 2015 epidemic. This PDCoV variant belongs to a young population within the US lineage, and infected pigs carry high concentrations of the virus. It also has several critical point mutations and an amino acid insertion at

Porcine deltacoronavirus (PDCoV) can cause diarrhea and dehydration in newborn piglets. Here, we developed a double antibody sandwich quantitative enzyme-linked immunosorbent assay (DAS-ELISA) for detection of PDCoV by using a specific monoclonal antibody against the PDCoV N protein and an anti-PDCoV rabbit polyclonal antibody. Using DAS-ELISA, the detection limit of recombinant PDCoV N protein and virus titer were approximately 0.5 ng/mL and 103.0 TCID50/mL, respectively. A total of 59 intestinal and 205 fecal samples were screened for the presence of PDCoV by using DAS-ELISA and reverse tran
