Crystalline structure of pulverized dental calculus induces cell death in oral epithelial cells.

BACKGROUND AND OBJECTIVE: Dental calculus is a mineralized deposit attached to the tooth surface. We have shown that cellular uptake of dental calculus triggers nucleotide-binding oligomerization domain-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome activation, leading to the processing of the interleukin-1β precursor into its mature form in mouse and human phagocytes. The activation of the NLRP3 inflammasome also induced a lytic form of programmed cell death, pyroptosis, in these cells. However, the effects of dental calculus on other cell types in periodontal tissue have

Multimodal analysis of physicochemical changes in dental enamel under plaque and calculus coverage.

OBJECTIVE: This study aimed to compare changes in enamel morphology, mineral composition, crystal structure, and mechanical properties under naturally formed dental plaque at different maturation stages versus dental calculus, using an intraoral in situ device and multimodal characterization. DESIGN: Six groups were analyzed (n = 5/group): a blank control group (fully bone-impacted mandibular third molars), a calculus group (extracted molars with naturally adherent dental calculus), and plaque groups formed in vivo for 1, 7, 14, and 28 days using an intraoral in situ device. Detection of hepatitis C virus in saliva before and after scaling of dental calculus.

Dentists and dental health care workers are at risk of contracting hepatitis C virus (HCV) through dental treatment, since HCV RNA was reported to be easily detectable in the saliva of patients with chronic HCV liver disease. We tested for the presence of HCV RNA in saliva before and after removal of dental calculus, and in splashes on the chin-length face shields of dentists following treatment of six patients with HCV chronic liver diseases. We used a sensitive reverse transcription polymerase chain reaction (RT-PCR) method, to estimate exposure to HCV. All patients were anti-HCV and HCV RNA

Analytical transmission electron microscopy of the distribution of elements in human supragingival dental calculus.

Thin sections of supragingival calculus from lower anterior teeth of 26 subjects were examined. From each specimen, an average of 10 spectra was collected from a densely-mineralized area less than 100 micron diameter. Only spectra showing peaks for minor elements or unusual Ca:P peak ratios were collected, stored and analysed semi-quantitatively. Ca:P weight percentage ratios varied within each specimen. Although most lay within the range 1.6 to 1.8, ratios of less than 1 and greater than 22 were obtained. The latter indicated the presence of some P-free Ca salts. The minor elements Mg and Zn

Fluoride and magnesium concentrations in human dental calculus obtained from Japanese and Chinese patients.

Calculus deposited on a total of 68 permanent teeth from patients 30-60 years old from Nagoya in Japan and Beijing in China was investigated. An abrasive microsampling method was used to examine the fluoride (F) and magnesium (Mg) distribution, using a fluoride ion-specific electrode and atomic absorption spectrophotometry, respectively. F concentrations decreased from the surface towards the interior of the calculus. Mg concentrations, however, gradually rose towards the innermost surface adjacent to the tooth. In all parts of the depth profiles, the average concentrations of both magnesium a

Insights into demographic and cultural influences on the oral microbiome from historical Japanese dental calculus.

Recent advances in genomic technologies have enabled detailed analyses of ancient microbiomes using dental calculus. While most studies have focused on European and North American populations, ancient Japanese oral microbiomes remain largely unexplored. Here, we analyzed dental calculus primarily from Edo period individuals (17th-19th centuries) to investigate the compositional, functional, and phylogenetic diversity of ancient oral microbiomes. Our results revealed clear differences between ancient and modern Japanese microbiomes, as well as phylogenetic divergence between the Final Jomon (ca

Dental Calculus as a Potential Biosource for Human Papillomavirus Detection in Oral Squamous Cell Carcinoma.

OBJECTIVE: The infection of human papillomaviruses (HPVs) plays a role in the development of oral squamous cell carcinoma (OSCC). A poor oral hygiene and dental calculus may cause the infection to persist. Therefore, this study aimed to assess whether this dental calculus could serve as a potential biosource in early detection of HPVs in patients with OSCC. METHODS: DNA was isolated from the dental calculus of people diagnosed with OSCC, and MY09/11 primer set was used to detect the presence of HPV. The positive samples were further sequenced and aligned using megablast NCBI BLAST tool to ide

Ancient dental calculus reveals oral microbiome shifts associated with lifestyle and disease in Great Britain.

The prevalence of chronic, non-communicable diseases has risen sharply in recent decades, especially in industrialized countries. While several studies implicate the microbiome in this trend, few have examined the evolutionary history of industrialized microbiomes. Here we sampled 235 ancient dental calculus samples from individuals living in Great Britain (∼2200 BCE to 1853 CE), including 127 well-contextualized London adults. We reconstructed their microbial history spanning the transition to industrialization. After controlling for oral geography and technical biases, we identified multiple

Effectiveness of a Daily Honeycomb-Shaped Dental Chew in Reducing Calculus, Plaque, Gingivitis and Malodor in Dogs.

Periodontal disease in dogs is common. Client compliance with oral hygiene and oral care for pets is low. The gold standard is annual dental prophylaxis under general anesthesia with imaging followed by home care including daily brushing. Clients should be offered methods to reduce calculus, plaque, gingivitis, and resulting halitosis that are time efficient, cost-effective, and easy to administer between annual preventative dental prophylaxis with the goal to move into maintenance phase of managing periodontal disease. This study aimed to evaluate the efficacy of a honeycomb-shaped dental che

Use of the feed additive sodium hexametaphosphate to prevent dental calculus in squirrel monkeys (Saimiri spp.).

Dental calculus and associated periodontal disease are serious clinical problems in captive squirrel monkeys. Calculus begins to appear as early as 2 years of age, with subsequent development of periodontal disease, dental abscessation, tooth loss and other sequelae. When used as a feed additive, sodium hexametaphosphate (HMP) retards the growth of calculus on previously cleaned teeth in rhesus monkeys, lemurs, and other species. We wanted to determine whether HMP would reduce dental calculus in squirrel monkeys (Saimiri spp.) whose teeth had not been pre-cleaned. The study animals were divide

Dental Calculus as a Potential Early Sign of Oral Functional Decline: A Hypothesis-Generating Case Series.

Dental calculus is generally considered a consequence of inadequate plaque control. However, in clinical practice, calculus is sometimes observed repeatedly at specific intraoral sites despite reasonable oral hygiene. Such site-specific, recurrent calculus may reflect localized salivary stagnation associated with oral functional decline rather than hygiene alone. This hypothesis-generating case series describes three clinical presentations involving four patients and explores this possibility. The three presentations included two pediatric cases with localized lingual calculus on the mandibula
