Berberine pretreatment enhances the homing and anti-inflammatory efficacy of dental pulp mesenchymal stem cells in TNBS-induced inflammatory bowel disease via activating the CXCR4/SDF-1 signaling pathway.

OBJECTIVE: Inflammatory bowel disease (IBD) is a chronic intestinal disorder characterized by immune dysregulation and persistent inflammation. Mesenchymal stem cell (MSC) therapy shows promise but is limited by low homing efficiency. This study aimed to investigate whether berberine (BBR) pretreatment enhances the homing capacity and therapeutic efficacy of dental pulp MSCs (DP-MSCs) in IBD, and to clarify the role of the CXCR4/SDF-1 signaling pathway in immunomodulation. METHODS: Human dental pulp mesenchymal stem cells were isolated and identified. Through in vitro screening experiments (b

Effects of human vascular endothelial growth factor on reparative dentin formation.

It is a challenge for dentists to save dental pulp in patients with pulp disease without resorting to root canal therapy. Formation of tertiary dentin to maintain pulp vitality is a key odontoblast response to dental pulp injury. Vascular endothelial growth factor (VEGF) is the most potent angiogenic and vasculogenic factor involved in tertiary dentin formation. It was hypothesized that VEGF may be used to treat pulp diseases such as pulpitis. To explore this hypothesis, the first step was to assess whether VEGF affects dental pulp cells to promote reparative dentin formation. In the current s

A Causal Association Study Between Chronic Kidney Disease and Oral Health: A Mendelian Randomization Study.

BACKGROUND AND AIMS: Previous studies have shown that chronic kidney disease (CKD) can lead to changes in oral health, but the conclusions remain controversial. Thus, we conducted a Mendelian randomization (MR) study from the perspective of genetic analysis to assess the causal association between CKD and oral health conditions, including dental caries, pulp diseases, periapical tissues, and diseases of the gum and periodontal tissues. METHODS: We performed inverse variance weighted (IVW) random effects MR analyses and several sensitivity MR analyses using summary statistics from genome-wide

Identification of the PP1 interactome in human dental pulp: Insights for regenerative strategies.

BACKGROUND: Dental caries are one of the most prevalent chronic diseases worldwide affecting all age groups. Current therapeutic strategies are technically demanding and invasive procedures that only address the restoration of dental pulp. Upon damage, mineralized tissues and dental pulp are affected, activating signalling cascades that promote their repair and regeneration through stem cells differentiation into odontoblasts-like cells. Protein phosphatase 1 (PP1) participates in a panoply of cellular events, including those related with regenerative outcomes, such as odontogenic differentiat

Creating a Microenvironment to Give Wings to Dental Pulp Regeneration-Bioactive Scaffolds.

Dental pulp and periapical diseases make patients suffer from acute pain and economic loss. Although root canal therapies, as demonstrated through evidence-based medicine, can relieve symptoms and are commonly employed by dentists, it is still difficult to fully restore a dental pulp's nutrition, sensory, and immune-regulation functions. In recent years, researchers have made significant progress in tissue engineering to regenerate dental pulp in a desired microenvironment. With breakthroughs in regenerative medicine and material science, bioactive scaffolds play a pivotal role in creating a s

Epigenetic regulation in dental pulp inflammation.

Dental caries, trauma, and other possible factors could lead to injury of the dental pulp. Dental infection could result in immune and inflammatory responses mediated by molecular and cellular events and tissue breakdown. The inflammatory response of dental pulp could be regulated by genetic and epigenetic events. Epigenetic modifications play a fundamental role in gene expression. The epigenetic events might play critical roles in the inflammatory process of dental pulp injury. Major epigenetic events include methylation and acetylation of histones and regulatory factors, DNA methylation, and

Human Atlas of Tooth Decay Progression: Identification of Cellular Mechanisms Driving the Switch from Dental Pulp Repair Toward Irreversible Pulpitis.

Dental pulp responses to dental decay, the most prevalent chronic disease worldwide, involve remodeling processes comparable to those observed in other human diseases. By combining volumetric imaging and single-cell analysis at various stages of the disease in human samples, the natural history of how dental pulp responds to decay is uncovered. During the early phases, an arterialization of capillary networks and a progressive outward remodeling of larger vessels are observed. Additionally, neurogenesis of nerve endings and the reprogramming of perivascular progenitor cells into fibroblasts, i

Methylenetetrahydrofolate reductase polymorphisms in dental caries-induced pulp inflammation and regeneration of dentine-pulp complex: Future perspectives.

Dental caries (DC)-induced pulp infections usually undergo the common endodontic treatment, root canal therapy (RCT). Endodontically treated teeth are devitalized, become brittle and susceptible for re-infection which eventually results in dental loss. These complications arise because the devitalized pulp losses its ability for innate homeostasis, repair and regeneration. Therefore, restoring the vitality, structure and function of the inflamed pulp and compromised dentin have become the focal points in regenerative endodontics. There are very few evidences, so far, that connect methylenetetr

The Immunophenotype and the Odontogenic Commitment of Dental Pulp Stem Cells Co-Cultured with Macrophages Under Inflammatory Conditions Is Modulated by Complex Magnetic Fields.

Dental inflammatory diseases remain a challenging clinical issue, whose causes and development are still not fully understood. During dental caries, bacteria penetrate the tooth pulp, causing pulpitis. To prevent pulp necrosis, it is crucial to promote tissue repair by recruiting immune cells, such as macrophages, able to secrete signal molecules for the pulp microenvironment and thus to recruit dental pulp stem cells (DPSCs) in the damaged site. To date, root canal therapy is the standard for dental caries, but alternative regenerative treatments are gaining attention. Complex Multifrequency

Maintaining Tooth Vitality With Super Minimally Invasive Pulp Therapy.

In aging humans, tooth loss is a predictor of decreased longevity. Tooth loss is mainly caused by dental caries and periodontal disease. Pulpitis refers to inflammation of the dental pulp caused by bacterial infection secondary to dental caries. It is accompanied by severe toothache and has infectious disease-associated pathophysiology. Pulpitis is mainly treated by pulpectomy, which is aimed at removing the infected dental pulp and controlling pain by removing nociceptive nerve fibers. However, teeth without dental pulp have a poor prognosis. In this report, we proposed a novel "super minimal

Exosomes Derived from Stem Cells from the Apical Papilla Promote Dentine-Pulp Complex Regeneration by Inducing Specific Dentinogenesis.

Regenerative endodontic procedures (REPs) are a new option for the treatment of dental pulp or periapical diseases in permanent teeth with open apices. Histologically, the new tissues formed in the root canal after REPs are mainly cementum- or bone-like mineralised tissues, but not the real dentine-pulp complex. Therefore, how to promote dentine-pulp complex regeneration and improve the clinical effects of REPs has become a prominent research topic. Stem cells from apical papilla (SCAP) are derived from the dental papilla that can differentiate into primary odontoblasts and dental pulp cells t

Schwann cell-derived EVs facilitate dental pulp regeneration through endogenous stem cell recruitment via SDF-1/CXCR4 axis.

The dental pulp is critical for physiological vitality of the tooth, and dental pulp regeneration has great potential for rebuilding live pulp tissue after pulp disease. Schwann cells (SCs) play a critical role in the support, maintenance, and regeneration of nerve fibers in dental pulp. Extracellular vesicles (EVs), which possess cell homing and tissue repair potential, derived from SCs (SC-EVs), can regulate dental mesenchymal stem cells (MSCs) proliferation, multipotency, and self-renewal. However, the role of SC-EVs in dental pulp tissue regeneration remains unclear. To address this questi

Dental histology in familial expansile osteolysis.

The dental histopathology is described of patients suffering from Familial Expansile Osteolysis (FEO), a rare and unique autosomal dominant disease which has severe bony as well as dental consequences. The predominant dental features are resorption of the cervical and apical regions of the roots, premature depositions both of irregular secondary dentin, which causes narrowing of the pulp cavities, and of cellular cementum, which causes patchy narrowing of the periodontal ligament. The dental pulp shows age-like changes, including multiple pulp stones and the ligament vasculature appears to be

Interleukin-1beta activity and collagen synthesis in human dental pulp fibroblasts.

Immunopathologic reactions play a significant role in inflammatory diseases of dental pulp. Interleukin-1beta (IL-1beta) is recognized as a key player in mediating cellular immune response. In this study, we measured the content of IL-1beta and its effect on collagen synthesis in cultures of fibroblasts derived from healthy and diseased dental pulps. We found that diseased pulp fibroblasts contain 2.5-fold greater amounts of IL-1beta and synthesized 80% greater amounts of collagen compared with healthy pulp fibroblasts. However, exogenous IL-1beta failed to stimulate collagen synthesis by dise
