Journal of bone and mineral research : the official journal of the American Society for Bone and Mineral Research2011eng
We describe results from a mutational analysis of the region of the dentin sialophosphoprotein (DSPP) gene encoding dentin phosphoprotein (DPP) in 12 families with dominantly inherited dentin diseases. In eight families (five mutations in the N-terminal third of DPP), the clinical and radiologic features were uniform and compatible with dentin dysplasia type II (DD-II) with major clinical signs in the deciduous dentition. In the other families (four mutations in the more C-terminal part), the permanent teeth also were affected, and the diseases could be classified as variants of dentinogenesis

The overall aim of this thesis was to study some aspects of the repair of the dentine barrier, especially in conjunction with dental pulp capping. Understanding the events leading to the healing of the dentine and pulp, and hence successfully preserving the vitality and functions of the tooth, would lead to a scientific basis for a less invasive treatment of pulp exposures than performing root canal treatments. The surfaces of the body have physiological barrier functions aimed at protecting the body from external noxious agents. In the tooth, the odontoblasts, which line the outermost part of

The remineralization of completely demineralized bovine dentin was studied in vitro by monitoring the velocities of fall of small particles through calcifying solutions. The density of a particle of dentin may be found from its velocity of fall through a fluid using Stokes' law. The minimum concentration product of calcium and acid phosphate ions of the solution in which remineralization would take place was 3.6 (mM)2 in the presence of 22 mM bicarbonate, pH 7.35, and ionic strength 0.1. This just above the solubility product of brushite (CaHPO4-2H2O). Incubation of decalcified dentin in a pho

Understanding light propagation in human dentin is critical for dental diagnostics, including optical coherence tomography (OCT) and early caries detection. Dentin's heterogeneous microstructure-a dense network of cylindrical tubules ( ≈ 2 μ m diameter) embedded in an organic-inorganic matrix-induces both isotropic and anisotropic light propagation. Previous isotropic-only models cannot capture the pronounced angular dependence observed experimentally, leading to substantial errors in the estimation of optical parameters. We present a combined isotropic-anisotropic Monte Carlo model for light

Nowadays, the occurrence of dentin hypersensitivity affects almost every person during their lifetime. Various stimuli – such as cold, heat, osmotic effects – can cause intense pain if the external structure of the tooth, enamel or cement layer are damaged or the anatomy of the enamel–cement border provides free dentin surface. Pain can trigger symptoms similar to tooth decay and its consequent diseases, so it is unclear to the patient what is causing the pain. The determination of dentin hypersensitivity is the task of the practising dentist. The prevention of the main root causes and the sol

Dentin dysplasia is a genetic defect of dentin formation inherited as an autosomal dominant trait. It is characterized by normal enamel but atypical dentin formation with abnormal pulpal morphology. Once thought to be a single entity, dentin dysplasia has now been divided into type I (radicular) and II (coronal). Type I is by far the more common. Both types include multiple/generalized involvement of primary and permanent dentition. Combinations of both types have also been described in the literature. Four distinct forms of dentin dysplasia type I and 1 form of dentin dysplasia type II are id

Dental hygienists frequently encounter clients who present with one or several sensitive teeth. These clients often rely on the dental hygienist to help determine the cause and to provide supportive treatment to relieve their discomfort. Therefore, it is essential for dental hygienists to fully understand the complexity of dentin sensitivity, be well informed about treatment approaches, and have an appreciation of the difficulties inherent in designing and conducting clinical hypersensitivity trials. Difficulties in designing hypersensitivity research have precluded a definitive answer concern

The situation with regard to current knowledge concerning dentinal sclerosis is examined. SEM and TEM examination of specimens of sclerotic dentin showed changes in diameter and tubular content leading to various degrees of obstruction of the lumen. There is agreement that this is due to the neo-apposition of peri-tubular dentin associated with intra-tubular deposition of crystals of Whitelockite, although the debate is still open on how such deposition may come about. Sclerotic casts are created in the tubules that may completely occlude the lumen or present a central cavity. In general these

The dentin-pulp complex is sensitive to infection. Restorative treatment should function as a Band-Aid protecting the dentin-pulp complex from bacteria and their byproducts. This article describes different ways of treating the exposed dentin-pulp complex in such a way that bacterial infection can be avoided.

Clinical trials remain the only conclusive research of dentin adhesive performance, but variables unique to the oral environment may influence the results. Dentin variability is one factor that may account for extreme variations in clinical evaluations. Despite the encouraging results of laboratory studies, clinical trials with adhesive systems have been disappointing. Current adhesives are reported to react with dentin surfaces by chemical and/or micro-mechanical mechanisms. Dentin surfaces, such as cervical abrasions, are subject to intraoral changes and dentin tubules commonly become obtura

Dentine becomes infected as a result of caries lesion formation on root surfaces and when lesions progress following cavitation of enamel lesions. However, this infection is unimportant because the driving force for lesion formation and progression is the overlying biofilm. This explains why root surface caries can be controlled by mechanical plaque control and fluoride, and restorations are not needed to arrest these lesions. Similarly, the infected dentine in cavitated coronal lesions does not have to be removed to arrest the lesion. If the lesion is either accessible or opened for cleaning

Dentinal hypersensitivity is generally reported by the patient after experiencing a sharp pain caused by one of several different stimuli. The pain response varies substantially from one person to another. The condition generally involves the facial surfaces of teeth near the cervical aspect and is very common in premolars and canines. The most widely accepted theory of how the pain occurs is Brannstrom's hydrodynamic theory, fluid movement within the dentinal tubules. The dental professional, using a variety of diagnostic techniques, will discern the condition from other conditions that may c

BACKGROUND: Inherited dentin defects comprise a group of rare genetic conditions that disrupt the normal formation, structure, and function of tooth dentin. Disorders such as dentinogenesis imperfecta and dentin dysplasia often have distinctive dental manifestations, including altered tooth color and translucency, abnormal crown and pulp morphology, foreshortened roots, and increased susceptibility to wear and fracture. The genetic sequence variations underlying selected dentin defects can also contribute to multisystem comorbidities, affecting skeletal, ocular, auditory, or kidney development

Dentin hypersensitivity is a common patient complaint that is more prevalent than the profession realizes. It is important for dentists to diagnose dentin hypersensitivity by exclusion and provide appropriate treatment recommendations for patients. Various treatment methods have been proposed but no universally accepted desensitizing agent or treatment has been identified. When a patient has symptoms that can be attributed to dentin hypersensitivity, a thorough clinical examination should be carried out to rule out other likely causes prior to diagnosis and treatment. Depending on the identifi

A major cause of tooth discoloration is sealer remnants in the pulp chamber after root canal treatment. The purpose of this study was to assess coronal distribution and color changes of four commonly used sealers placed in the pulp chamber after 2 yr. Fifty extracted premolars were cross-sectioned in the coronal third of the root. The chamber contents were removed, and instrumentation was via the canal; then freshly mixed sealer was placed in each chamber. Sealers evaluated were: AH 26, Kerr Pulp Canal Sealer, Roth 801, and Sealapex. The apical access was sealed with white sticky wax, and the

In hypersensitive teeth pain is typically induced by cold and hot fluids or foods brought to contact with the sensitive dentin or by osmotic and mechanical stimuli. The results of animal experiments indicate that intradental A-type nerve fibres are responsible for the sensitivity of dentin. They are most probably activated by the hydrodynamic mechanism. Thus, their activation is to a great extent dependent on the condition of dentin with either open or blocked dentinal tubules. Blocking of the tubules effectively prevents the nerve activation. Correspondingly, in human experiments, the conditi

This study determined the influence of dentine structure on the micro-tensile bond strengths between resin and dentine of two different dentine adhesive systems (Single Bond, 3M Dental Products, St Paul, MN; MF-102 (experimental self-etching primer), GC Corporation, Tokyo, Japan). The study was separated into two main parts: bond strength measurement and investigation of the bonding interface. Twenty-two human premolars were used for the bond strength measurement. Each tooth was cut vertically, separating the tooth into mesio-distal halves. One half of the tooth was used to bond to a surface p

The purpose of this study was to compare the biomineralization of circumpulpal dentine with that of mantle dentine by ultrastructural and element-analytical techniques. Forty upper second molar germs of 10-day-old albino rats were cryofixed in liquid nitrogen-cooled propane and embedded in resin after freeze drying. Semithin dry sections were cut for analyzing the calcium and phosphorous concentration in initial mantle dentine, at the mineralization front of circumpulpal dentine, in the middle region of circumpulpal dentine and in mantle dentine peripheral to circumpulpal dentine. For the morp
