Effect of eggshell nanoparticles and fluoride varnish, with and without diode laser, on dentinal tubule occlusion (An in vitro study).

BACKGROUND: Dentine hypersensitivity is a common clinical condition, affecting between 4% and 69% of the adult population. Management strategies are based on occluding patent dentinal tubules to reduce dentinal fluid movement. This in vitro study aimed to evaluate the effect of nano eggshell and fluoride varnish, alone and combined with diode laser, on dentinal tubule occlusion using scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDX). METHODS: Forty sound human maxillary premolars were sectioned to expose mid-coronal dentine and etched with 37% phosphoric acid to

Responses of intradental nerve fibres to stimulation of dentine and pulp.

In the present work responses of intradental nerve fibres to stimuli that induce fluid flow in dentinal tubules as well as to direct mechanical irritation of the exposed pulp were studied on 9 young adult beagle dogs. Under pentobarbitone anesthesia 31 single functional intradental fibre units were dissected from the mandibular nerve. Stimuli were applied to the lower left canine tooth. Exposed dentine surface was irritated by scraping, air blasts and dry absorbent paper and the pulp mechanically with a von Frey hair. Ten fibre units responded to stimulation of dentine. Six of them were also t

Low versus high pressure for in vitro determination of hydraulic conductance of human dentine.

The purpose of this study was to evaluate the effect of applied pressure and measurement time on the in vitro measurement of hydraulic conductance of human dentine. Dentine slices were prepared from 50 third molars. Water was forced through the slices under a constant hydrostatic pressure. Five pressures were tested: 1.3 kPa (n = 10), 13.3 kPa (n = 10), 26.6 kPa (n = 10), 40 kPa (n = 10) and 53.3 kPa (n = 10). The volume that went through the slices was recorded every 10 min for 3 h. The volume, the fluid flow rate and the hydraulic conductance under the five pressures were compared. The volum

Dentin-predentin complex and its permeability: physiologic overview.

The major channels for solute diffusion across dentin are the dentinal tubules. Since dentin permeation is proportional to the product of tubule number and diameter, both of which increase as the tubules converge on the pulp, we find that dentin permeability increases rapidly as the pulp chamber is approached. The presence of a smear layer of cutting debris on top of cut dentin decreases dentin permeability, especially when permeability is measured by fluid filtration. Further, intratubular material--such as mineral deposits, collagen fibrils, proteoglycan linings, bacteria, etc.--can greatly

Intratubular crystal formation in the exposed dentin from nano-sized calcium silicate for dentin hypersensitivity treatment.

The aim of this study is to evaluate intratubular crystal formation from the experimental material consisting of dicalcium silicate (C2S) and tricalcium silicate (C3S) with nano-scaled particle size. A total of twenty-four specimens were made by isolating 8 mm of the cervical part centered at the cementoenamel junction of extracted premolars. Twelve specimens were not treated and considered as control. The experimental material was applied to the other twelve specimens by brushing for 10,000 strokes. Each group was randomly divided into four subgroups according to the period of immersion in ph

An engineered dual-functional peptide with high affinity to demineralized dentin enhanced remineralization efficacy in vitro and in vivo.

Dental caries continues to be a major global public health problem. Remineralization of demineralized dentin is regarded as one of the hotspots in the current study in the treatment of dental caries. However, traditional remineralization agents, which usually lack the ability to bind to demineralized dentin collagen, are easily removed by the fluids in the oral cavity, thus decreasing the remineralization efficacy. Non-collagenous proteins (NCPs) have significant effects on the biomineralization of dentin due to their dual high binding capacity to the collagen fibers and minerals. But NCPs are

Clinical considerations of microleakage.

Most dental materials permit the microleakage of bacteria and bacterial products from oral fluids to reach dentin. Thus, an understanding of the clinical consequences of microleakage demands that we analyze the permeability characteristics of dentin. The more dentin surface that is exposed during tooth preparation, the greater the potential for microleakage. Thicker dentin is less permeable than thin dentin. Dentin over pulp horns is more permeable than central dentin. Similarly, the dentin making up axial walls is more permeable than dentin forming the pulpal floor of cavities. Coronal dentin

In vitro permeability of furcation dentin in permanent teeth.

Extracted unerupted permanent third molars with the occlusal half of the crown and apical half of the roots removed were cemented to Plexiglas blocks. Using a positive pressure system in which the movement of fluid across the dental tubules could be measured, the permeability of furcation dentin was measured before and after alteration of the furcation thickness. Subsequently, measurements of reduction in thickness of cementum and dentin were performed. Furcation dentin permeability was found to increase as the cementum and dentin thickness was reduced and the smear layer removed. The permeabi

Cationic Poly(amino acid)/Black Phosphorus Hydrogel for Immediate and Durable Relief of Dentin Hypersensitivity Through Ion Flow Blockade, Photo-Thermal Occlusion, and Biomineralization.

Dentin hypersensitivity (DH) stems from exposed dentinal tubules, where external stimuli induce fluid movement, stimulate pulp nerves, and cause acute pain. A significant challenge in current DH management is that most commercially available desensitizing agents often lack both immediate and long-term efficacy. To address this, a black phosphorus (BP) nanosheet-composited methoxy poly(ethylene glycol)2k-block-poly(L-methionine)27-block-poly(L-lysine)5 (PMK5) cationic hydrogel (PMK5/BP) operates through three steps to relieve DH, including ion flow blocking, the photo-thermal effect of BP, and

Guided tissue remineralisation of partially demineralised human dentine.

Biomineralisation is a well-regulated process mediated by extracellular matrix proteins. Biomimetic remineralisation strategies should reproduce the dimension and structural hierarchy of apatite deposits within a demineralised collagen matrix. Interfibrillar and intrafibrillar remineralisation of phosphoric acid-etched human dentine was demonstrated in this study using a Portland cement/phosphate-containing fluid system in the presence of polyacrylic acid and polyvinylphosphonic acid as respective calcium phosphate- and collagen-binding matrix protein analogues. Metastable amorphous calcium ph

Mechanical and micromorphological evaluation of chlorhexidine-mediated dentin remineralization.

Chlorhexidine (CHX) has been reported to reduce self-degradation of collagen fibrils by inhibiting host-derived protease activity in demineralized dentin. Theoretically, if the collagen fibril scaffold of demineralized dentin maintains its original crosslinkage pattern on treatment with CHX and appropriate supplementation with necessary mineral sources, dentin remineralization may occur in demineralized lesions. In this study, we provide direct mechanical and micromorphological evidence for the ability of CHX to promote remineralization of demineralized dentin. Specifically, with respect to de

Dentin hypersensitivity and its management.

Dentin hypersensitivity is a common patient complaint that is more prevalent than the profession realizes. It is important for dentists to diagnose dentin hypersensitivity by exclusion and provide appropriate treatment recommendations for patients. Various treatment methods have been proposed but no universally accepted desensitizing agent or treatment has been identified. When a patient has symptoms that can be attributed to dentin hypersensitivity, a thorough clinical examination should be carried out to rule out other likely causes prior to diagnosis and treatment. Depending on the identifi

Influence of sodium hypochlorite on the permeability and structure of cervical human dentine.

The effect of 5% sodium hypochlorite and 35% hydrogen peroxide on dentine samples in vitro was investigated. It was demonstrated that both 5% NaOCl and 35% H2O2 depletes the dentine, which can be recorded as weight loss. Sodium hypochlorite removes approximately 14% of dry weight from dentine samples in 24 h; hydrogen peroxide is half as destructive. The hydraulic conductance (Lp) of dentine after treatment with NaOCl is increased over 100%. It is suggested that the use of high concentrations of sodium hypochlorite during root canal treatment may negatively affect the integrity of the root can

Dentin permeability of carious primary teeth.

BACKGROUND: Many in vitro studies have used dentine permeability to evaluate the efficacy of various restorative and preventative procedures. The easiest way to evaluate dentine permeability is to calculate its hydraulic conductance (Lp) using fluid filtration methods. Research has examined electronic hydraulic conductance measurement methods that give more precise and reliable results for the permeability of dentine than the classical method. To our knowledge, no study has examined the dentine permeability of carious primary teeth. AIM: This in vitro study determined the dentine permeability
