Dentinogenesis Imperfecta Type II in Children: A Scoping Review.

Dentinogenesis Imperfecta type II (DI2), also known as hereditary opalescent dentin, is one of the most common genetic disorders affecting the structure of dentin, not related with osteogenesis imperfecta, which involves both primary and permanent dentitions. The purpose of this article is to perform a scoping review of the published peer-reviewed literature (1986-2017) on DI2 management in children and to outline the most relevant clinical findings extracted from this review. Forty four articles were included in the present scoping review. According to the extracted data, the following are th

Dentinogenesis imperfecta type II in Swedish children and adolescents.

BACKGROUND: Dentinogenesis imperfecta (DGI) is a heritable disorder of dentin. Genetic analyses have found two subgroups in this disorder: DGI type I, a syndromic form associated with osteogenesis imperfecta (OI), and DGI type II, a non-syndromic form. The differential diagnosis between types I and II is often challenging. Thus, the present cross-sectional study had two aims: to (i) investigate the prevalence and incidence of DGI type II among Swedish children and adolescents and (ii) search out undiagnosed cases of DGI type I by documenting the prevalence of clinical symptoms of OI in these i

[Osteogenesis imperfecta and dentinogenesis imperfecta. Apropos of 4 cases].

Based on four personal cases of osteogenesis imperfecta and dentinogenesis imperfecta, the authors review the semiological elements constituting the syndrome, and assign each case to the Sillence (1979) classification. The author established his classification on clinical criteria and the genetic nature of the various forms of osteogenesis imperfecta, however, the most recent discoveries concerning collagen biochemistry confirm that the condition is without doubt more heterogenous than suggested clinically, thereby better explaining the wide range of mutations encountered.

Unequal Impact of COL1A1 and COL1A2 Variants on Dentinogenesis Imperfecta.

Dentinogenesis imperfecta (DI) is the main orodental manifestation of osteogenesis imperfecta (OI) caused by COL1A1 or COL1A2 heterozygous pathogenic variants. Its prevalence varies according to the studied population. Here, we report the molecular analysis of 81 patients with OI followed at reference centers in Brazil and France presenting COL1A1 or COL1A2 variants. Patients were submitted to clinical and radiographic dental examinations to diagnose the presence of DI. In addition, a systematic literature search and a descriptive statistical analysis were performed to investigate OI/DI phenot

A Review of Dentinogenesis Imperfecta and Primary Dentin Disorders in Dogs.

This review describes the clinical, radiographic and histologic characteristics of dentinogenesis imperfecta diagnosed in two unrelated young dogs without evidence of concurrent osteogenesis imperfecta. The dentition was noted to have generalized coronal discoloration ranging from grey-blue to golden brown. Clinical pulp exposure, coronal wear and fractures were observed as was radiographic evidence of endodontic disease, thin dentin walls or dystrophic obliteration of the pulp canal. The enamel was severely affected by attrition and abrasion despite histologically normal areas; loss was most

The Role of Dendritic Cells during Physiological and Pathological Dentinogenesis.

The dental pulp is a soft connective tissue of ectomesenchymal origin that harbors distinct cell populations, capable of interacting with each other to maintain the vitality of the tooth. After tooth injuries, a sequence of complex biological events takes place in the pulpal tissue to restore its homeostasis. The pulpal response begins with establishing an inflammatory reaction that leads to the formation of a matrix of reactionary or reparative dentin, according to the nature of the exogenous stimuli. Using several in vivo designs, antigen-presenting cells, including macrophages and dendritic

Assessment of dysplastic dentin in osteogenesis imperfecta and dentinogenesis imperfecta.

Two semiquantitative scoring systems, Clinical Radiographic Score (CRS) and Dysplastic Dentin Score (DDS), were introduced for analyzing degree of dysplastic manifestations in dentin. The utility of both systems was demonstrated in a large material of teeth from patients with dentinogenesis imperfecta (DI) and osteogenesis imperfecta (OI). Twenty teeth from healthy controls, 81 teeth from 40 patients with OI, and 18 teeth with DI without OI (DI type II) were examined. The degree of dysplasia was correlated with type and form of OI and type of DI. The median DDS did not differ between DI associ

Dentinogenesis Inperfecta. Report of three cases in an Indian family.

An Indian family with Type II Dentinogenesis Imperfecta is reported in which the pedigree was traced through four generations. Clinical and radiological examination was done in three individuals in the family studied, which showed variation in expression of colour and attrition. No clinical evidence of Osteogenesis Imperfecta was noted in any of the family members. Theoretical considerations regarding the development of this disorder and its clinical features are presented.

Aberrations in pulpal histology and dentinogenesis in transplanted erupting teeth.

Responses of pulpal tissues were assessed histologically in teeth that had been transplanted after root formation was moderately well advanced. Degeneration occurred in the coronal third, but the area was repopulated by cells proliferating from the center of the pulp. Apically, the tissues, including the odontoblasts, remained vital and functional. Widespread intrapulpal formation of osteodentin was prominent in later observation periods. Apical closure occurred prematurely by production of osteodentin and cellular cementum in the form of an apical diaphragm; these mineralized tissues were con

Effects of methylmercury chloride on rat incisor odontoblasts and dentinogenesis.

Methylmercury chloride, a well-known nerve cell toxicant, was administered to 40 rats in 2 groups, in doses of 10 mg/kg and 20 mg/kg. Histomorphologic investigation of maxillary incisors after 2 and 4 days revealed granular and hydropic changes in parts of the odontoblastema. Deposition of interglobular dentin was seen after 7 and 11 days. The most vulnerable cells seemed to be the mantle dentin-producing odontoblasts, and the alterations found are probably due to interference with the rough endoplasmic reticulum.

Diagnosis complexity of dentinogenesis imperfecta involving DSPP genetic variants.

BACKGROUND: Variants in the dentin sialophosphoprotein (DSPP) gene are associated with dentin dysplasia type II (DD-II; OMIM # 125420) and dentinogenesis imperfecta (DI) types II (OMIM # 125490) and III (OMIM # 125500). DSPP encodes a precursor protein cleaved into three dentin matrix proteins: dentin sialoprotein (DSP), dentin phosphoprotein/phosphophoryn (DPP) and dentin glycoprotein (DGP). Exon 5 contains over 200 tandem 9-base pair repeats (DSS domain), complicating sequencing with standard methods. Regenerative Endodontic Treatment in Dentinogenesis Imperfecta-Induced Apical Periodontitis.

Pulp involvement of immature permanent teeth with dentinogenesis imperfecta is challenging and could lead to extraction. A case of dentinogenesis imperfecta-induced periapical periodontitis of an immature permanent tooth was treated with regenerative endodontic treatment (RET), and root maturation was observed in 12-month follow-up. An 8-year-old girl presented acute pain and swelling in central mandibular region. Clinical and radiographic examination revealed "shell teeth" appearance of teeth 31, 41, and 42. Periapical lesion of tooth 31 was observed. Tooth 41 was previously treated with apex

Dlx3 Ubiquitination by Nuclear Mdm2 Is Essential for Dentinogenesis in Mice.

Dentin is a major mineralized component of teeth. Odontoblasts are responsible for synthesis and secretion of dentin matrix. Previously, it has been demonstrated in a cell culture system that the E3 ubiquitin ligase, murine double minute 2 (Mdm2), promotes odontoblast-like differentiation of mouse dental papilla cells (mDPCs) by ubiquitinating p53 and the odontoblast-specific substrate Dlx3. However, whether Mdm2 plays an essential role in vivo in odontoblast differentiation and dentin formation remains unknown. In this study, we investigated the in vivo functions of Mdm2 using Dmp1-Cre;Mdm2fl

Aging of in vitro pulp illustrates change of inflammation and dentinogenesis.

INTRODUCTION: Dental pulp functions include pulp cell activity involvement in dentin formation. In this study we investigated the age-related changes in dental pulp cells that may influence pulp cell activity for restoring pulp function. METHODS: Human dental pulp cells (HDPCs) were serially subcultured until spontaneously arrested. Altered expression of chronic inflammatory molecules and age-related molecules were determined by Western blotting. Odontogenic functions impaired by senescence were assayed by Western blotting, reverse transcriptase polymerase chain reaction, alkaline phosphatase

Unveiling Novel DSPP Variants and Dental Phenotypes in Dentinogenesis Imperfecta.

BACKGROUND: Variants in the dentin sialophosphoprotein (DSPP) gene are known to cause hereditary dentin disorders, including dentinogenesis imperfecta (DGI), which is characterized by abnormal dentin development and structure. However, the full spectrum of clinical manifestations and the impact of specific variants remain to be fully elucidated. METHODS: Oral and radiological examinations were conducted on eight patients from two families. Physical characterization of deciduous DGI teeth and matched controls was performed. Exome and Sanger sequencing were employed for variant detection. RESU

Dentinogenesis Imperfecta and Caries in Osteogenesis Imperfecta among Vietnamese Children.

Osteogenesis imperfecta (OI) is a genetic disorder characterized by increased bone fragility and low bone mass, caused mainly by mutations in collagen type I encoding genes. The current study aimed to evaluate dentinogenesis imperfecta (DI), oral manifestations and caries status of OI children. Sixty-eight children (41 males, 27 females) aged from 3 to 17 years old (mean 9 ± 4.13) participated in the study. Participants were classified into three OI type groups (I-2 cases, III-31 cases and IV-35 cases). Clinical examination and an orthopantomogram were used to obtain prevalences and associatio

Non-Syndromic Dentinogenesis Imperfecta Caused by Mild Mutations in COL1A2.

Hereditary dentin defects can be categorized as a syndromic form predominantly related to osteogenesis imperfecta (OI) or isolated forms without other non-oral phenotypes. Mutations in the gene encoding dentin sialophosphoprotein (DSPP) have been identified to cause dentinogenesis imperfecta (DGI) Types II and III and dentin dysplasia (DD) Type II. While DGI Type I is an OI-related syndromic phenotype caused mostly by monoallelic mutations in the genes encoding collagen type I alpha 1 chain (COL1A1) and collagen type I alpha 2 chain (COL1A2). Dentinogenesis imperfecta type II: ultrastructure of teeth in sagittal sections.

The morphological abnormalities of the teeth of patients affected by dentinogenesis imperfecta type 2 (DI-II) may underlie the difficulties with the clinical restoration of such teeth. We therefore performed a scanning electron microscopy (SEM) study of four permanent first mandibular molars of four DI-II patients with periapical pathosis. The teeth were prepared for SEM evaluation by standard methods. In the crown, the enamel presented a highly irregular surface with a number of cracks and crevices. In some places, only granular remains of the enamel were found, while in other parts of the cr

Splicing mutation in DSPP causes dentinogenesis imperfecta and amelogenesis imperfecta.

BACKGROUND: Dentin sialophosphoprotein (DSPP) is an extracellular matrix protein, which is highly expressed in odontoblasts and transiently expressed in presecretory ameloblasts. DSPP mutations were related to dentinogenesis imperfecta (DGI), some of which can be accompanied by amelogenesis imperfecta (AI). However, the mechanism underlying DGI and AI caused by DSPP mutations is still unclarified. This study aimed to reveal the molecular pathogenesis in a Chinese family with both DGI and AI caused by the DSPP splicing mutation. METHODS: One Chinese family with DGI and AI was recruited.
