Multistate Survey of American Dog Ticks (Dermacentor variabilis) for Rickettsia Species.

Dermacentor variabilis, a common human-biting tick found throughout the eastern half and along the west coast of the United States, is a vector of multiple bacterial pathogens. Historically, D. variabilis has been considered a primary vector of Rickettsia rickettsii, the causative agent of Rocky Mountain spotted fever. A total of 883 adult D. variabilis, collected between 2012 and 2017 from various locations in 12 states across the United States, were screened for rickettsial DNA. Tick extracts were evaluated using three real-time PCR assays; an R. rickettsii-specific assay, a Rickettsia belli

Prevalence of Rickettsia species in Dermacentor variabilis ticks from Ontario, Canada.

Relatively little is known about the prevalence of rickettsial species in Dermacentor ticks in eastern Canada. In this study, Dermacentor ticks from the province of Ontario, Canada, were tested for the presence of spotted fever group rickettsial (SFGR) species, Coxiella burnetii and Francisella tularensis. Rickettsia rickettsii was not detected in any ticks tested, but R. montanensis was detected at a prevalence of 2.2% in D. variabilis (17/778). Two other SFGR species, R. parkeri and Candidatus R. andeanae, were detected individually in 2 Amblyomma maculatum ticks. Rickettsia peacockii, a non

Nonrandom distribution of vector ticks (Dermacentor variabilis) infected by Francisella tularensis.

The island of Martha's Vineyard, Massachusetts, is the site of a sustained outbreak of tularemia due to Francisella tularensis tularensis. Dog ticks, Dermacentor variabilis, appear to be critical in the perpetuation of the agent there. Tularemia has long been characterized as an agent of natural focality, stably persisting in characteristic sites of transmission, but this suggestion has never been rigorously tested. Accordingly, we sought to identify a natural focus of transmission of the agent of tularemia by mapping the distribution of PCR-positive ticks. From 2004 to 2007, questing D. varia

Lethal effect of Rickettsia rickettsii on its tick vector (Dermacentor andersoni).

Rickettsia rickettsii, the causative agent of Rocky Mountain spotted fever, was lethal for the majority of experimentally and transovarially infected Rocky Mountain wood ticks (Dermacentor andersoni). Overall, 94.1% of nymphs infected as larvae by feeding on rickettsemic guinea pigs died during the molt into adults and 88. 3% of adult female ticks infected as nymphs died prior to feeding. In contrast, only 2.8% of uninfected larvae failed to develop into adults over two generations. Infected female ticks incubated at 4 degreesC had a lower mortality (80.9%) than did those held at 21 degreesC (

Replication of tick-borne encephalitis (TBE) virus in ticks Dermacentor marginatus.

In the laboratory experiments, the virophoric period in D. marginatus ticks lasted 61 to 81 d, the premoulting period (nymphs - adults) amounted to 17 to 18 d. The titres of individually examined females and males for the presence of TBE virus ranged from 10(1) to 10(5.5) ic mouse LD50/0.03 ml between the 14th and 29th day after hatching. In the laboratory experiments 77% of ticks were positive. In the field experiments, the virophoric period in D. marginatus ticks lasted 79 to 114 d, the premoulting period amounted to 34 to 38 d. The titres of individually examined adults for the presence of

Spotted fever group rickettsiae in Dermacentor variabilis from Cape Cod, Massachusetts.

Spotted fever group (SFG) rickettsiae were identified by indirect immunofluorescent antibody tests in 1.1% of 6,956 specimens of Dermacentor variabilis collected on Cape Cod, Massachusetts. Forty-one of 45 rickettsial isolates were serotyped as Rickettsia montana, one as Rickettsia rickettsii, and three were unidentified. Studies of canines and humans with clinical evidence of spotted fever, and of healthy dogs with antibody to SFG rickettsiae, indicated that exposure of the canine population to R. montana was common but that R. montana was not involved in the etiology of spotted fever.

Temporal distribution of Ixodes ricinus, Dermacentor reticulatus and Haemaphysalis concinna in Hungary.

A survey was carried out over a 4-year period to describe the temporal distribution of three 'anthropophilic' tick species, Ixodes ricinus, Dermacentor reticulatus and Haemaphysalis concinna in Hungary. Altogether 4658 adult ticks belonging to the three species were collected from 1931 red foxes (Vulpes vulpes) killed in an area of about 70,000 km(2) representing all major climatic areas of the country. The seasonal activity of the three species was different. I. ricinus ticks were most active between April and June with an activity peak in May. A less marked increase of activity was also obse

Spatial distribution of Dermacentor reticulatus and Ixodes ricinus in Hungary: evidence for change?

A survey was conducted to investigate the spatial distribution of Ixodes ricinus and Dermacentor reticulatus in Hungary and to compare these data with the results of a previous large-scale survey. In the survey conducted in the 1950s, D. reticulatus adults were detected in two isolated areas of two counties, and the presence of these ticks in the collection was explained by accidental introduction. In the present survey, D. reticulatus became the second most common species occurring in all 16 counties involved in the monitoring and showed high prevalence. The change in the spatial distribution

[The occurrence of Coxiella burnetii in sheep and ticks of the genus Dermacentor in Baden-Wuerttemberg].

This study examined the occurrence of Coxiella burnetii (C. burnetii), the infectious agent of Q-fever, in sheep and sheep-ticks in Baden-Wuerttemberg, Germany, as a possible source of infection in Q-fever outbreaks. Using PCR, we examined a total of 1066 Dermacentor ticks from 23 herds and 49 samples of tick excrement from 18 herds for C. burnetii. We found the infectious agent in one non-engorged tick and in one sample of tick excrement from the same herd, in Efringen-Kirchen (district Loerrach). Sequencing the PCR-products confirmed the amplifications as specific for C. burnetii. Further se

Spotted fever group rickettsiae in Dermacentor marginatus from wild boars in Italy.

Following the increase in wild boar population recorded in urban and peri-urban areas throughout Europe, the present survey aimed to assess the occurrence of zoonotic tick-borne pathogens (TBPs) in wild boars living in southern Italy and in their ticks for evaluating the potential risk of infection for animals and humans. From October to December 2019, a total of 176 ticks collected from 93 wild boars andtheir spleen samples were molecularly screened for selected TBPs. Overall, all the wild boars were infested by ticks (mean intensity, 1.9) with Dermacentor marginatus and Ixodes ricinus being

Dermacentor everestianus Hirst, 1926 (Acari: Ixodidae): phylogenetic status inferred from molecular characteristics.

Dermacentor everestianus Hirst, 1926, is only reported in Northwestern China and Nepal. Few researches about this species have been involved, especially for molecular characteristics. The taxonomy studies of D.everestianus are mainly based on morphological features, and its taxonomic status is an ongoing controversy. To clarify the molecular characteristics and phylogenetic status of D.everestianus and other related species, the sequences of mitochondrial 16S ribosomal DNA (rDNA) and cox1 fragments were analyzed in the present study. Analysis of 16S rDNA and cox1 sequences showed 99.3-100% ide

Proteomics informed by transcriptomics identifies novel secreted proteins in Dermacentor andersoni saliva.

Dermacentor andersoni, known as the Rocky Mountain wood tick, is found in the western United States and transmits pathogens that cause diseases of veterinary and public health importance including Rocky Mountain spotted fever, tularemia, Colorado tick fever and bovine anaplasmosis. Tick saliva is known to modulate both innate and acquired immune responses, enabling ticks to feed for several days without detection. During feeding ticks subvert host defences such as hemostasis and inflammation, which would otherwise result in coagulation, wound repair and rejection of the tick. Molecular charact

Molecular study of transovarial transmission of Babesia canis in the Dermacentor reticulatus tick.

The Dermacentor reticulatus tick is a main vector of Babesia canis in Europe. The risk of canine babesiosis is unpredictable, due to significant differences in the prevalence of B. canis between ticks originating from closely situated regions. This phenomenon may be explained by vertical transmission of the pathogen in a vector population. Thus, molecular techniques were applied to investigate the occurrence of transovarial transmission in D. reticulatus ticks. DNA of B. canis was detected in 20.7% (6/29) of engorged female ticks collected from dogs, in every pool of eggs laid by positive fema

[The prevalence of Babesia canis canis in marsh ticks (Dermacentor reticulatus) in the Saarland].

An accumulation of autochthonous cases of canine babesiosis caused by Babesia canis has been registered in a small animal clinic in the Saarland since the beginning of 2006, some cases with fatal outcome. This study aims to contribute to the explanation of strong focal occurrence of infections with B. canis in this region. Therefore, patient owners who had presented their dogs in the years 2006 and 2007 because of babesiosis and who had claimed not having left the Saarland with their dogs at least six months before the outbreak of Babesiosis, were asked for their dog walking habits.
